Simultaneous Determination of Prostaglandin E1 and Prostaglandin E1 Ethyl Ester in Hairless Mouse Skin Homogenate by High-Performance Liquid Chromatography

  • A rapid and specific high-performance liquid chromatographic method was developed and validated for the simultaneous determination of prostaglandin $E_{1}\;(PGE_{1})$ and prostaglandin $E_{1}$ ethyl ester $(PGE_{1}-EE)$ in hairless mouse skin homogenate. The sample treatment procedure involved deproteination and precipitation by acetonitrile. $PGE_{1}$ and $PGE_{1}-EE$ in supernatant were separated in a reversed-phase C18 column without being interfered by other components present in hairless mouse skin homogenate. 9-Anthracenecarboxylic acid was used as an internal standard. The retention times of $PGE_{1}$, 9-anthracenecarboxylic acid and $PGE_{1}-EE$ were, 4.5, 9.5 and 18.0 min, respectively. The assay showed linearity from 1 to $40\;{\mu}g/ml$ for both $PGE_{1}$ and $PGE_{1}-EE$. Precision expressed as RSD ranged from 2.3 to 14.1 % for $PGE_{1}$ and 1.6 to 11.0% for $PGE_{1}-EE$. Accuracy ranged from 100.5 to 119.6 % for $PGE_{1}$ and from 98.0 to 103.7% for $PGE_{1}-EE$. This method was employed successfully to follow the time course of concentrations of $PGE_{1}$ and $PGE_{1}-EE$ in hairless mouse skin homogenate for stability study.

Canola Oil로부터 코코아버터와 유사한 Triacylglycerol 조성을 가진 재구성지질의 효소적 합성 최적화 연구 (Optimization of Lipase-Catalyzed Production of Structured Lipids from Canola Oil Containing Similar Composition of Triacylglycerols to Cocoa Butter)

  • 본 연구에서는 카놀라유(canola oil)를 이용하여 코코아버터 유사 재구성지질 합성의 최적조건을 연구하였다. 반응기질로는 카놀라유와 palmitic ethyl ester와 stearic ethyl ester를 이용하였으며, 효소는 Lipozyme TLIM을 사용하여 반응기질의 몰비율과 반응효소의 양, 반응시간을 독립변수로 하고, D-optimal 계획에 의한 반응표면분석에 따라 합성을 실시하였다. 반응을 통해 acyl-migration은 최소로 일어나며, POS 및 SOS, POP의 함량은 최대로 합성되는 조건을 최적화 조건으로 결정하였다. D-optimal 계획에 의한 반응표면분석을 실행한 결과, sn-2 위치 지방산 조성의 변화를 일으키는 acyl-migration($Y_1$)은 최소로 일어나고 POS($Y_2$)와 SOS($Y_3$), POP($Y_4$)는 최대로 일어나는 조건은 기질의 몰비율은 canola oil : palmitic ethyl ester : stearic ethyl ester=1:3:9이며, 반응효소의 양은 6%이고, 반응시간은 40 min이었다. 이와 같은 반응조건으로 합성된 재구성지질의 acylmigration(% of palmitic acid+stearic acid, $Y_1$)은 10.43%, POS($Y_2$)의 함량은 25.31%, SOS($Y_3$)는 19.79%, 그리고 POP($Y_4$)는 11.22%로 측정되었다.

  • External lyogels containing prostaglandin $E_1$ ethyl ester $(PGE_1-EE)$, a prodrug of prostaglandin $E_1\;(PGE_1)$ as a therapeutic agent for erectile dysfunction, were formulated to overcome the aqueous instability and enhance the percutaneous absorption. Lyogels of $PGE_1-EE$ were prepared with ethanol (EtOH)/proplyene glycol (PG) cosolvent system as a vehicle, cineol as an enhancer, and hydroxypropylcellusose as a gelling agent. In vitro percutaneous absorption studies were performed to determine the rate of $PGE_1$ absorption through rat or hairless mouse skin. The permeability of $PGE_1-EE$ lyogel with enhancer was 16-fold greater than that of lyogel without enhancer. Cosolvent produced 9-fold increase in percutaneous absorption. Pharmacodynamic effects of lyogels were evaluated in mature male cats in terms of intracavernosal pressure (ICP). Lyogels containing 0.1 % of $PGE_1-EE$ showed higher ICP compared to intraurethral preparation of $PGE_1$ (1 %) and enhancer-free control lyogel. The shelf-life $(t_{10%})$ of lyogel at refrigerated condition $(4^{\circ}C)$ was calculated as 928 days, which is 4.2 times longer than that of control hydrogel. As a result, $PGE_1-EE$ was formulated successfully to a lyogel system with a selective enhancer and cosolvent system for the topical delivery of $PGE_1$.

Skin penetration enhancement of prostaglandin El and its ethyl ester for topical formulations

  • Purpose. To investigate the effect of different terpene enhancers on skin penetrations of prostaglandin El (PGE1) and its ethyl ester (PGE1-EE), a therapeutic agent for erectile dysfunction, external gel systems were formulated with the specific enhancers having different values in their lipophilicity (log P was ranged in 2.23-4.58). Topical gels containing PGEl (0.5 %) and PGEl-EE (0.1 %) were formulated with ethanol and propylene glycol as a vehicle, selective terpenes as a penetration enhancer, and HPC-H as a thickening agent. FT-IR과 탄소동위원소 분석을 통한 연어유의 구분에 관한 연구 (A Study of Salmon Oil Type Analysis by FT-IR and Carbon Isotopes Ratio)

  • This study analyzes the structure types of salmon oil to evaluate the purity of salmon oil products based on the 38 different types of imported salmon oil products distributed in the Republic of Korea. The major types of omega-3 foods in the salmon oil are ethyl ester (EE) and triglyceride (TG). If the salmon oil contained potential contaminants and was processed in order to remove it, EE type omega-3 fatty acids are found in concentration. This provides a good guide in assessing if products were made with EE type ingredients or re-esterified contaminated materials. The results of the FT-IR analysis showed significant difference in the C=O, C-O band positions in TG and EE. There were 19 TG type products and 19 EE type products. The analysis of carbon isotope ratio was performed on the types of TG and EE. There were different properties in the 19 TG type products. In one product, the carbon isotope ratio was -25.15 and the other 18 products showed -22.15~-23.96. The carbon isotope ratio of all 19 EE type products showed -21.91~-23.74. The results of the TLC analysis showed similar results with FR-IR. The re-esterified TG form was not detected in the TG type products, confirming that the TG type products contained natural salmon oil. This study aimed to provide the basic material in classifying the types of natural salmon oil and re-esterified salmon oil, by analyzing the pattern and proportion of FT-IR spectrum, carbon isotope ratio, and TLC.

  • 활성 빵효모를 이용한 생물학적 환원은 n-hexane, pentane과 석유에테르와 같은 유기용매에서 순조롭게 진행되어, ethyl(1)과 octyl 3-oxohexanoate(2)는 높은 %ee로 환원되었다. 활성 빵효모에 의한 생물학적 환원은 물보다 유기용매에서 생성물의 정제가 간편하고 경제적 효과가 크다는 이점이 있었다. 이러한 생물학적 환원방법은 광학활성 천연물의 합성에 이용할 수 있으리라 생각되어 진다.

Effect of Dietary Docosahexaenoic Acid on Maze-learning Ability in Aged Mice Fed N-3 Fatty Acid Deficient Diet

  • The effect of docosahexaenoic acid (DHA) on maze-learning ability in aged mice was investigated. Aged mice fed a diet deficient in n-3 fatty acid received a semi-purified diet of 0.5, 1 and 2% DHA-ethyl ester (DHA-EE) for 4 months. Maze-learning ability was assessed at 3 months after the start of the experiment. The time required to reach the maze exit and the number of times that a mouse strayed into blind alleys in the maze were measured in 3 trials conducted at 4-day intervals. The time was significantly less in the DHA-EE 0.5% and DHA-EE 2% groups than in the control group (p<0.05). The number of times mice strayed into blind alleys in the maze was significantly lower in the DHA-EE 2% group than in the control group (p<0.05). Mice fed DHA showed increased DHA and decreased level of arachidonic acid (AA) in the brain. These results suggest that the intake of a 2% DHA diet improves learning ability in aged mice, which is associated with a higher DHA content in the brain.

방사선 조사선량에 따른 lipase의 interesterification 반응특성 변화 연구 (Studies of interesterification properties of irradiated lipases)

  • This study was conducted to prove the effect of irradiation on lipases (lipase AK, lipase AH, lipase PS-D, Lipozyme TLIM, Lipozyme RMIM and Novozyme SP435) which were used for interesterification reaction using batch type reactor. Through such interesterification, structured lipid (1(3)-palmitoyl-2-oleoyl-3(1)-stearoyl, POS) was synthesized by lipase treated with irradiation at different doses (0, 3, 7, 14, 29 and 59 kGy) using canola oil, palmitic ethyl ester (PEE) and stearic ethyl ester (StEE). After the reaction, fatty acid composition of triacylglycerol (TAG) in structured lipid was analyzed to compare the lipase activity. The results showed that activity of the irradiated lipase AH, PS-D and Novozyme SP435 with certain dose (3 kGy) were slightly improved. Such change of lipase activity suggested that irradiation might affect on the interesterification properties. Especially, Lipase AK, Lipozyme TLIM and Lipozyme RMIM after at 3 kGy irradiation showed that content of stearic acid ($C_{18:0}$) was increased while palmitic acid ($C_{16:0}$) decreased in the interesterified products.

PEI(Polyethyleneimine)를 이용하여 음이온계 레진에 고정화된 Lipase AH 제조 및 효소적 Interesterification을 통한 반응 특성 연구 (Immobilization of Burkholderia cepacia Lipase on Weak Base Styrene Resin Using Polyethyleneimine with Cross-linking)

  • 본 연구는 polyethyleneimine(PEI)을 이용하여 음이온계 레진에 비 고정화 효소인 lipase AH(Burkholderia cepacia)를 고정화한 후 효소적 interesterification을 통해 고정화 효소의 반응 특성을 확인하고자 하였다. 효소적 interesterification에 canola oil, palmitic ethyl ester와 stearic ethyl ester를 기질로 사용하였으며 합성물의 TAG 조성 분석을 통해 특성 연구를 진행하였다. 또한 두 가지 고정화방법(multi layer, mono layer)으로 고정화를 진행하였으며 사용된 lipase solution의 농도와 pH를 달리하여 실시하였다. 먼저 효소와 support 간의 친화력을 확인하기 위하여 단백질 함량을 측정하였다. 그 결과 고정화에 사용된 lipase solution의 농도가 8 mg/mL일 때 친화력이 좋았으며 MUIM의 경우 pH 7.5에서, MOIM의 경우 pH 8.0에서 고정화 효율이 가장 좋았다. 또한 고정화 효소의 활성을 확인하기 위하여 interesterification을 진행하였고 TAG와 sn-2 position 분석을 통하여 활성을 비교하였다. 이러한 결과를 참고하여 가장 친화력이 좋았던 조건에서 고정화를 진행하였다. 그 결과 MUIM의 경우 total saturated fatty acid(${\Sigma}SFA$)의 함량이 16.79 area%였으며 MOIM의 경우는 ${\Sigma}SFA$의 함량이 26.17 area%로 나타났다. 반면에 sn-2 position 분석결과 MUIM과 MOIM 모두 palmitic acid와 stearic acid의 함량이 증가한 경향을 나타내었는데, 이는 기존의 sn-1, 3 특이성을 가지는 lipase AH가 고정화를 통해 무작위 특이성으로 변화한 것으로 생각되었다. 또한 RP-HPLC를 이용하여 TAG의 조성을 확인한 결과, 기질은 canola oil이 주로 OOO와 LOO로 조성되었으나 고정화 효소의 interesterification 반응 후 합성물은 주로 POO/SLO, LPO/LOP와 SOO, SOS 등으로 이루어진 것으로 확인되었다. 위와 같은 결과를 바탕으로 최종 생산된 MUIM과 MOIM을 이용하여 interesterification 반응을 온도, 시간을 달리하여 진행하였다. 전체적으로 MOIM의 활성이 가장 높았으며 lipase AH, MOIM과 MUIM 모두 반응시간이 증가할수록 ${\Sigma}SFA$의 함량은 증가하였으나 12시간 이후로는 시간대비 효율은 크게 증가하지 않았다. 또한 시간이 증가함에 따라 sn-2 position의 ${\Sigma}SFA$ 함량이 높아졌다. 한편 반응온도가 높을수록 ${\Sigma}SFA$의 함량도 증가하였는데, 이는 상온인 $25^{\circ}C$보다 높은 melting point를 가지는 stearic ethyl ester와 palmitic ethyl ester가 semi solid 형태로 존재하여 활성이 저해된 것으로 생각된다. 또한 lipase AH는 재사용이 불가능하였지만 고정화 효소인 MUIM과 MOIM은 5번을 재사용하여도 그 효소의 활성이 유지되는 것을 확인할 수 있었다. 이러한 결과를 토대로 음이온계 레진에 PEI를 이용하여 lipase AH를 고정화하였을 경우 비 고정화 효소인 lipase AH보다 MUIM과 MOIM의 활성이 더 높아 산업적으로 이용 가능할 것으로 생각된다.
