Some Functional Properties of Extracts from Leaf and Fruit Stalk of Hovenia dulcis (헛개나무 잎과 과병추출물의 몇가지 가능성)

  • The present study was conducted to investigate antioxidant, nitrite scavenging and alcohol degradation effects of extracts from leaf and fruit stalk of Hovenia dulcis. Yields of various solvent extracts for Hovenia dulcis leaf and fruit stalk of were higher in water and methanol extract layer, respectively. Ethanol extracts of Hovenia dulcis leaf and fruit stalk of were fractionated with different solvents such as hexane, chloroform, ethyl acetate, butanol and water, yields of water fraction were highest. In the solvent extracts using methanol, ethanol, hexane, chloroform and water, ethanol extracts showed the most effective antioxidant and nitrite-scavenging effects. Ethanol extracts from Hovenia dulcis leaf and fruit stalk of were fractionated, the most natioxidant and nitrite-scavenging effects were ethyl acetate fraction. Alcohol degradation effects from different parts of Hovenia dulcis showed higher leaf and fruit stalk than xylem, branch and seed. Alcohol degradation effects from leaf and fruit stalk increased as time passed.

  • The antimicrobial substances from Rhodiola sachalinensis were extncted, isolated and identified. The highest level of antimicrobial activity and its yield were obtained in methanol extract. The minimum inhibition concentrations of the methanol extract were 500 $\mu\textrm{g}$mL on agar plate and 100 $\mu\textrm{g}$mL in broth media for four gram positive and four gram negative microbials. The methanol extract was fractionated by n-hexane, chloroform, ethyl ether, ethyl acetate, and butanol, orderly. The separate was developed on the TLC plate with different solvent system ratio of chloroform and methanol. Nine substances were isolated from chloroform and methanol mixture(9:1, v/v). Among them, three isolates showed antimicrobial activity. Three substances separated by HPLC were identified by GC/MS(EI) spectrum and $^1$H, /sup13/C-NMR spectrum. They were gallic acid, (-)-epicatechin and kaempferol. The antimicrobial activities of each substances were shown gallic acid, (-)-epicatechin, kaempferol orderly.

TOXICOLOGICAL EVALUATION OF ETHANOLIC EXTRACT FROM THE ROOT OF SCUTELLARIAE RADIX (황금(Scutellariae Radix)의 에타놀추출물과 프랄보노이드 성분들의 독성평가)

  • Flavonoids from Scutellariae Radix possessed a dual function both as an anti-inflammatory agent and an enhancer of cellular activity in gingival fibroblast. The purpose of this study was to evaluate on the toxicity of ethanolic extract from the root of Scutellariae Radix Georgi and its flavonoids, Wogonin, Baicalein, and Baicalin were isolated and purified by the following method. The crude drug was extracted with ethyl acetate and the residue was dissolved in ethyl alcohol. The ethyl alcohol soluble fraction was separated, concentrated, and then chromatographed on a silica gel column. The acute oral LD 50 in rats was determined for EtOH ex. of Scutellariae Radix and three compounds were evaluated with a single oral gavage at three graded dosage levels. The acute intravenous LD 50 was determined with a single intravenous injection via the jugular vein at three graded dosage levels. Groups of 5 male and 5 female rats, 6 week of age at the start of the study, were fed diets containing 3 graded dosage levels for 14 days. Groups of 5 male and 5 female hamster received O.5ml of the test article at once in a day for 5 days to the buccal cheek pouch for two minutes each. The acute oral LD50 for EtOH ex. of Scutellariae Radix is 1430mg/kg, and for Wogonin 1320mg/kg, for Baicalein 1250mg/kg, for Baicalin 1330mg/kg. The acute intravenous toxicity of EtOH ex. of Scutellariae Radix and its extracts was found to be 27mg/kg body weight No toxic effects were observed in rats fed up to 200mg/kg of EtOH ex. of Scutellariae Radix, Wogonin, Baicalein and Baicalin in the diet for 14 days. The acute Mucouse Membrane LD 50 in hamsters was found to be greater than 100mg/kg. These results suggested that EtOH ex. of Scutellariae Radix and its flavonoids are safe for oral care products using limited amount of extract.

Study on components of Geranium sibiricum L. (Geranium sibiricum L.의 성분에 대하여)

  • The herb of Geranium sibirium L, a drug knwon as "Chui Sonni Poul" distributed widely, has been used as a folk-medicine for the treatment of diarrhea. The dried entire herb is boiled with methanol and methanol is distilled of from the filtered methanol extract under reduced pressure. Then the extract is boiled with water and filtered off. From the filtrate, the following substances are isolated and identified by treating with organic solvents as ether, ethyl acetate, and etc.: 1. Gallic acid: a colorless needle crystal which is soluble in alcohol and water. mp.235.deg. C, positive (dark blue) against the ferric chloride reagent. 0.76 percent of gallic acid is yielded from the herb. 2. Quercetin: a light yellow crystal. mp.194.deg. C. negative against the ferric chloride reagent. 3. Ellagic acid: a light yellow crystal which is insoluble in ehter and acetone and slightly soluble in alcohol. Positive (blue) against the ferric chloride reagent. The crystal obtained by recrystalisation from pyridine, does not melt by 360.deg. C. Represent a yield of 0.03 percent from the herb. 4. Crude tannin: a approximately 7.6 percent of crude tannin is yielded by treating with ethyl acetate. gallic acid and querceetin are yielded by hydrosis with dliute sulfuric acid. Based on the above results, the following suggestion could be recommendable; Geraed in the Japanese Pharmacoeia VI.acoeia VI.

Effects of the Combined-administration of Acanthopanacis Cortex and Lycii Cortex Radicis on Immune Response (오가피(五加皮)${\breve{z}}$ -지골피(地骨皮) 병용투여가 면역반응에 미치는 영향)

  • Immunological activities of the combined-administration of Acanthopanacis Cortex and Lycii Cortex Radicis were examined in BALB/C mice. The 40% ethyl alcohol extract of Acanthopanacis Corter (AE) or the 40% ethyl alcohol extract of Acanthopanacis Coriex and Lycii Cortex Radicis (ALE) were administered p.o. once a day for 7 days, respectively. AE did not affect the viability of thymocytes, but ALE decreased the viability of thymocytes. ALE enhanced the viability of splenocytes increased by AE. Also, AE enhanced the population of cytotoxic T cell in thymocytes, and ALE enhanced the population of helper T cell compared with AE. Furthermore, AE increased the population of $Thy1^+$ cells in splenocytes, and increased the population of splenic $CD4^+$ cells. In addition, ALE enhanced the phagocytic activity which was decreased by AE ALE decreased the production of nitric oxide increased by AE in peritoneal macrophages. These results suggest that ALE enhance an immune-regulative action of AE.

Stem bark of Maackia amurensis Extract according to extraction Solvent (추출용매에 따른 다릅나무 수피 추출물의 항산화 활성)

  • Objectives : The objective of this research was to investigate the antioxidant activities of stem bark of Maackia amurensis extract.Methods : Stem bark of Maackia amurensis extract were prepared using 70% methanol. Methanol extracts were fractionated to hexane, chloroform, ethyl acetate, butyl alcohol, water fractions and investigated. The antioxidant activities of fractions was evaluated by four different assays as total polyphenol contents, total flavonoid contents, DPPH(1,1-diphenyl-2-picrylhydrazyl) free-radical scavenging activity and ABTS(2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) scavenging ability.Results : The yield of methanolic extracts from stem bark of Maackia amurensis was 10.16%, whereas those of its solvent fractions (hexane, chloroform, ethyl acetate, butyl alcohol, and water) were 5.45, 11.39, 13.88, 26.07, and 40.80%, respectively. The total polyphenol contents and electron donating ability of 70% methanol extracts from stem bark of Maackia amurensis were 15.44 mg/g and 194.15 μg/mL of its IC50, respectively. The 70% methanol extracts showed the highest antioxidant activity. The total polyphenol content and total flavonoid content of chloroform fractions were higher in each of 201.98 mg/g and 13.55 mg/g. The chloroform fraction showed the lowest levels of DPPH(IC50, 183.95 μg/mL) and ABTS scavenging activity(IC50, 10.0 μg/mL). The antioxidant activity was detected in methanol extract, chloroform fractions.Conclusions : These results indicate that 70% methanol extract and its fractions of stem bark of Maackia amurensis, especially chloroform fraction, have the properties of anti-oxidant suggesting stem bark of Maackia amurensis may be a candidate for natural and functional materials.

Effects of Heat and pH Treatments on Antioxidant Properties of Ishige okamurai Extract (패 추출물의 항산화능에 미치는 열과 pH의 영향)

  • This study was carried out to determine the optimum extraction conditions for Ishige okamurai by comparing the yields, total phenolic compound content (TPC), and antioxidant properties of its 95%, 70%, 50% fermented ethyl alcohol and water extracts. Additionally, the effects of heat and pH treatments on the antioxidant properties of the extracts were evaluated by their TPC and 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical scavenging capabilities. The yields of the extracts were greatest in the order of water > 50% > 70% > 95% fermented ethyl alcohol, and the TPC of the 70% (26.18%) and 50% fermented ethyl alcohol (27.56%) extracts were higher than those of the others. However, in terms of DPPH radical scavenging and ferrous-reducing power, the 70% fermented ethyl alcohol extract of Ishige okamurai showed the highest antioxidant effects. Additionally, in the results for the heat and pH treatments, the antioxidant properties of the 70% fermented ethyl alcohol extract were not influenced by the treatment conditions except at pH 10.

Comparison of Substance Change and Antibacterial Activity Before and After Fermentation Using Resource Plants for The Development of Natural Preservatives (천연방부제 개발을 위한 자원식물을 활용한 발효 전·후 물질 변화와 항균활성 비교)

  • Chemical preservatives have a good effect on antibacterial activity, but many side effects on the human body have been reported. Recently, the development of natural preservatives that are harmless to the human body and have preservative functions and self-efficacy is active. In addition, in order to increase the absorption rate of natural products by the human body, the method of fermentation using strains is also increasing. Therefore, this study selected varieties that are harmless to the human body and have good antibacterial activity. 1. The yield of origin, thickness and solvent was investigated. Scutellaria baicalensis Georgi was made in China and received a yield of 21.88% from 50% ethyl alcohol extract. Salvia miltiorrhiza Bunge was made in Korea and received a yield of 25.62% from 50% ethyl alcohol extract. Dryopteris crassirhizoma Nakai was made in China and received a yield of 6.50% from 70% ethyl alcohol extract. 2. The solid fermentation with the S. baicalensis and S. miltiorrhiza with B. Subtilis yield gained 24.40%, 39.30%, and D. crassirhizoma obtained 11.10% yield when fermented with L. casei. 3. After the liquid fermentation, a clear zone of 9mm was identified for the S. aureus strain in the S. baicalensis, and the antibacterial activity was not confirmed in S. miltiorrhiza and D. crassirhizoma. 4. When the S. baicalensis was fermented with L. Casei, it showed high antibacterial activity in C. albicans and S. aureus. S. miltiorrhiza showed antibacterial activity in S. aureus when it was solid with S. cerevisiae. When the spectators were solid with L. casei and S. cerevisiae, antibacterial activity was high in E. coli and S. aureus. Overall, the antibacterial activity after fermentation was much higher than when fermented. 5. The change in active ingredients was baicalin 101.57, baicalein 28.26, and wogonin 5.33mg/g in the S. baicalensis that did not ferment solid. When solid fermentation with S. cerevisiae, the content of baicalinin with baicalin 94.31, baicalein 30.41, and wogonin 3.57mg/g was found to have increased. S. miltiorrhiza that was not fermented, salvianolic acid A was 1.82mg/g, and when fermented with S. cerevisiae, it increased to 5.70mg/g. The active ingredients of the spectators were flavaspidic acid AP, flavaspidic acid PB, flavaspidic acid AB, and flavaspidic acid BB.

Preparation of Red Ginseng Extract Rich in Acidic Polysaccharide from Red Tail Ginseng Marc Produced After Extraction with 70% Ethyl Alcohol (홍미삼 알콜 추출박을 이용한 산성다당체 다량 함유 홍삼 엑기스 제조)

  • In this study, we investigated the appropriate conditions to extract acidic polysaccharide and to prepare red ginseng extract being rich in acidic polysaccharide from red tail ginseng marc produced after manufacturing alcoholic extract from red tail ginseng. Amount of acidic polysaccharide in red tail ginseng marc was about 11%. The best condition for the extraction of acidic polysaccharide from the marc was using of 3~5 mg of $\alpha$-amylase/g residue/15 ml of distilled water, and the amount of acidic polysaccharide in water extract of the residue treated with $\alpha$-amylase was about 27%. So, it is possible to manufacture red ginseng extract being rich in acidic polysaccharide using water extract of red tail ginseng alcoholic residue as extraction solvent. From the above results, we suggest that red tail ginseng residue produced by manufacturing alcoholic extract of red tail ginseng has high potencies in the utilization of waste material.

The Effects of Areca catechu Extracts on Tyrosinase Gene Expression in B16 Mouse Melanoma Cells (B16 마우스 흑색 종양 세포에서 빈랑(Areca catechu) 추출물이 티로시나아제 유전자에 미치는 효과)

  • In this study, we found that the methanolic extract of Areca catechu repressed expression of the tyrosinase gene in B16 mouse melanoma cells containing a tyrosinase promoter. Extract concentrations of 100 ${\mu}$g/ml and 500 ${\mu}$g/ml exhibited tyrosinase gene expression rates of aproximately 62% and 48%, respectively, compared to the control. The fraction layers consisting of ethyl acetate, butyl alcohol, and water showed repressive effects on the tyrosinase gene. In particular, the butyl alcohol fraction highly repressed at 100 ${\mu}$g/ml and 500 ${\mu}$g/ml. In the MTT assay, the methanolic extracts exhibited very low cytotoxicities at 1 ${\mu}$g/ml, 10 ${\mu}$g/ml, and 100 ${\mu}$g/ml.
