양파 외피에서 추출한 수용성 색소의 분석 (Analysis and Dyeing of Natural Pigments Extracted from Onion Shell(Allium cepa))

  • 도시 Traditionally, dyes extracted from onion shells have been used as natural pigments but dyeing methods with reproducibility have not been developed. In this study, the dyes were analyzed by high performance thin layer chromatography(HPTLC) to obtain the ratio of pigments in water and ethyl acetate extract. Furthermore, the present study was also carried out to obtain the information characteristic of wavelength which proportionally decrease for continuous dyeing reaction. Scanning of water extract showed 4 peaks by the mobile phase of benzene/ethyl acetate/acetic acid(40/10/5) and the peak 1 exhibited yellow color with the maximal absorption spectra of 306nm and 309nm while ethyl acetate extract showed 9 peaks in the same mobile phase. The water extract after ethyl acetate extraction was adjusted to 0.5 of O.D.(optical density) at 550nm by adding of water and compared wavelength of the pigments from 200 to 600nm to find the proportional decrease of wavelength. As the result, it showed that wavelength of 306nm could be the standard of dyeing monitoring points.

Investigation of n-Butanol and Ethyl Acetate Extracts from Thermal Treatment Yam (Dioscorea batatas DECNE.) for their Antioxidant Activities

  • n-Butanol and ethyl acetate extracts of thermal treatment yam (Dioscorea batatas $D_{ECNE.}$) belonging to the family Dioscoreaceae were measured for their radical scavenging activity and lipid peroxidation inhibition ability. In this study, ethyl acetate extract showed the most potent antioxidant activity evaluated by ferrous ion chelating activity and NO radical scavenging activity. Nevertheless, n-butanol extract was more effective in inhibiting linoleic acid peroxidation. A significant difference between n-butanol extract and ethyl acetate extract in nitrite scavenging activity ${\beta}$-carotene bleaching assays could not be found. Also, the results of this study showed that thermal treatment yam could be used as easily accessible source of natural antioxidants and as a possible food supplement.

  • 갈대(Phragmites communis) 뿌리를 70% 에탄올 수용액으로 추출한 후 n-hexane, chloroform 및 ethyl acetate를 사용하여 순차적으로 분획하였다. 갈대 뿌리 에탄올 추출물의 분획물 중 ethyl acetate 분획의 폴리페놀 함량이 가장 높았다. 갈대 뿌리 추출물의 chloroform과 ethyl acetate 분획물은 우수한 아질산염 소거 능력을 나타내었다. 또한 각 용매 분획물은 10 mg/mL 농도에서 효과적인 지질과산화 억제능을 보여주었다.

엽차용 녹차 추출물 및 분획물의 항균효과 (Antimicrobial Activity of Extracts and Fractions of Green Tea Used for Coarse Tea)

  • 9월에 수확하여 엽차용 녹차로 시판되는 보성산 녹차를 온수, $80^{\circ}C$ 열수, 70% ethanol 용액으로 추출하고, 70% ethanol 추출물을 다시 hexane, chloroform, ethyl acetate, butanol로 분획한 다음 녹차 추출물과 분획물의 그램 양성균과 그램 음성균 8종에 대한 항균활성에 대하여 조사를 하였다. 녹차의 추출 수율은 70% ethanol 추출물은 33.0%, ethyl acetate 분획물은 11.2%를 보였다. 8종의 세균에 대한 녹차 추출물 간의 항균활성은 70% 에탄올 추출물의 경우 수 추출물에 비하여 Vibrio parahaemolyticus(V. parahaemolyticu) 및 Staphylococcus aureus(S. aureus)에 대하여 가장 높았다. 70% 에탄올 추출물로부터 여러 가지 용매를 달리하여 분획한 분획물 간의 항균활성은 70% ethanol 추출물에 비하여 ethyl acetate 분획물의 경우 Klebsiella pneumoniae, V. parahaemolyticus, S. aureus, Bacillus subtilis(B. subtilis) 및 Streptococcus mutans(S. mutans)와 같은 5종의 세균에 대하여 높았다. 70% ethanol 추출물 유래 ethyl acetate 분획물의 MIC는 B. subtilis의 경우 $5\;{\mu}L$/disc이었고, S. aureus 와 S. mutans의 경우 $3\;{\mu}L$/disc이었으며, V. parahaemolyticus의 경우 $3\;{\mu}L$/disc 이하로 아주 낮았다. Ethyl acetate 분획물의 첨가 농도에 따른 항균효과는 B. subtilis와 S. mutans 두 균종에서 확연히 나타났고 B. subtilis는 0.05% 이상의 첨가로 24시간 증식시켰을 때 control에 비하여 균증식이 90% 이상 억제됨을 보였으며, S. mutans는 동일한 균증식 억제 효과를 얻기 위하여 0.1% 이상의 ethyl acetate 분획물 농도가 요구되었다. Ethyl acetate 분획물의 열처리($121^{\circ}C$, 20분)는 균종에 따라 10%정도의 항균활성을 감소시켰다. 이와 같은 결과를 종합할 때 9월에 채엽된 저렴한 녹차로 천연 항균제 혹은 식품보존료의 개발은 충분한 잠재력이 있는 것으로 판단되었다.

양제근 추출물 및 분획의 항산화 활성과 Tyrosinase 저해 활성 (Antioxidant and Tyrosinase Inhibitory Activity of Extract Rumex japonicus HOUTT Root and Its Fractions)

  • Background: We investigated the antioxidative and tyrosinase inhibitory activities of 70% ethanol extract, and its fractions, of the root of Rumex japonicus HOUTT. Methods and Results: The total phenolic compound contents of the 70% ethanol extract and ethyl acetate fraction were 168.99 mg/g and 651.78 mg/g, respectively. The antioxidant activity was compared through the DPPH radical and nitric oxide (NO) scavenging assays. The ethyl acetate fraction showed the highest DPPH radical and NO scavenging abilities, which confirmed the antioxidant activity. Specifically, the ethyl acetate fraction showed a higher DPPH radical scavenging ability than ascorbic acid. These results were related to the total phenolic compound content of the ethyl acetate fraction. Moreover, in the tyrosinase inhibition assay, the ethyl acetate fraction exhibited stronger inhibitory activity than arbutin, which was used as the positive control. The cell viability of L929 cells was analyzed by MTT assay after treatment with 70% ethanol extract and all fractions; no changes in viability were observed, which demonstrated the nontoxic nature of the extract and fractions. Conclusions: These results suggested that the extract from the root of R. japonicus and its ethyl acetate fraction could be a novel resource for the development of a cosmetic with antioxidant and tyrosinase inhibitory activity.

석류 내피 용매별 추출물의 항산화 활성 (Antioxidant Activities of Solvent Extracts from Pomegranate Endocarp)

  • 본 연구는 가식부위가 20~30%로 과즙 착즙 시 주로 버려지고 있으나 석류의 부위별 추출물 중 가장 항산화성이 우수한 천연식품 소재인 석류 내피를 에탄올로 추출하여 천연항산화제로서의 활용 가능성을 조사하였다. 석류 내피 추출물을 극성이 낮은 용매에서 극성이 높은 용매로 순차적으로 계통 분획하여 항산화능을 분석하였다. 각 측정 방법 별로 항산화 활성에 있어 약간의 차이를 보였으나 전반적으로 ethyl acetate 분획의 항산화능이 높게 측정되었다. DPPH 라디칼 소거활성 측정 결과 ethyl acetate와 butanol 분획물에서 $IC_{50}$이 각각 2.40 ${\mu}g$/mL, 2.88 ${\mu}g$/mL로 기준 물질인 ascorbic acid와 유사한 항산화 활성을 나타내는 것으로 나타났다. Superoxide anion 소거활성에서 ethyl acetate과 butanol 분획물의 $IC_{50}$은 각각 58.94 ${\mu}g$/mL, 61.91 ${\mu}g$/mL로 기준물질인 tannin acid보다 항산화능이 우수하였다. $ABTS^+$ 소거활성에서 ethyl acetate와 butanol 분획물의 $IC_{50}$은 각각 39.26 ${\mu}g$/mL, 40.95 ${\mu}g$/mL로 나타났으며 환원력을 측정한 결과 ethyl acetate 분획물(0.1 mg/mL)의 흡광도가 1.404로 기준물질인 ascorbic acid보다 활성이 높게 측정되었다. SOD 유사활성과 아질산염 소거능에서는 유사한 결과를 나타냈는데 SOD 유사활성은 $42.88{\pm}0.66%$(dichloromethane), $15.43{\pm}0.85%$(butanol), $12.82{\pm}0.68%$(water)이었으며 아질산염 소거능에서 dichloromethane 분획물의 $IC_{50}$은 115.54 ${\mu}g$/mL로 기준물질인 ascorbic acid보다 높게 나타났다. 따라서 본 연구결과 석류 내피 추출물은 높은 항산화 활성을 가지고 있으며, 폐자원으로써 버려지는 석류 내피는 식품첨가물 등의 기능성 소재로의 이용가능성이 있을 것으로 여겨진다.

The ethyl acetate soluble fraction of the M. japonicus methanol extract showed the best DPPH radical scavenging activity. The ethyl acetate fraction was measured for the activity of superoxide dismutase (SOD), catalase, and oxidative stress tolerance by using C. elegans along with reactive oxygen species (ROS) level. In addition, to confirm that the regulation of the stress response gene is responsible for the increased stress tolerance of C. elegans treated by the ethyl acetate fraction, SOD-3 expression was measured using a transgenic strain. As a result, the ethyl acetate fraction increased SOD and catalase activity, and decreased ROS accumulation in a dose-dependent manner. In addition, the ethyl acetate fraction-treated CF1553 worm showed higher SOD-3::GFP intensity than the control worm.

  • Oat, the seeds of Avena sativa L. (Gramineae), is an important dietary staple for people in many countries. Previous studies reported that A. sativa had various pharmacological effects such as anti-inflammatory, antitumor, neurotonic, and antispasmodic activities. In this study, Caenorhabditis elegans model system was used to investigate the antioxidant activity of methanol extract of oat. The ethyl acetate soluble fraction of the oat methanol extract showed the best DPPH radical scavenging activity. The ethyl acetate fraction was measured for the activity of superoxide dismutase (SOD), catalase, and oxidative stress tolerance by using C. elegans along with reactive oxygen species (ROS) level. In addition, to confirm that the regulation of the stress response gene is responsible for the increased stress tolerance of C. elegans treated by the ethyl acetate fraction, SOD-3 expression was measured using GFP-expressing transgenic worm. As a result, the ethyl acetate fraction increased SOD and catalase activities, and decreased ROS accumulation in a dose-dependent manner. In addition, the ethyl acetate fraction-treated CF1553 worm showed higher SOD-3::GFP intensity compared to the control.

In addition, to confirm that the regulation of the stress response gene is responsible for the increased stress tolerance of C. elegans treated by the ethyl acetate fraction, SOD-3 expression was measured using a transgenic strain (CF1553). As a result, the ethyl acetate fraction increased SOD and catalase activity, and decreased ROS accumulation in a dose-dependent manner.
