Odor Characteristics of Essential Oil of Valeriana fauriei var. dasycarpa HARA (광릉쥐오줌풀의 정유향 특성)

  • This study was conducted to know the odor characteristics and compounds in neutral, acidic and basic fraction from the essential oil of Valerliana fauriei root. Among the fraction from the essential oil of V. fauriei root, the content of neutral fraction was 92%, that of the acidic fraction, 6%, and that of basic and phenolic fractions, less than 1%, respectively. The neutral fraction was characterized by sweet-balsamic, woody, musky and medicinal odor. The acidic fraction had sweat-socks, valeric-like and cheese-like odor characters. The netural fraction of essential oil was fractionated by solvents with different polarities, resulting in 44% of ethyl ether fraction, 34% of pentane-ethyl ether fraction, 11% of pentane fraction and 11% of methanol fraction. The ethyl ether fraction was characterized by woody and medicinal odor and the pentane-ethyl ether fraction, by sweet-balsamic, woody and herb-like odor. The fractions were fractionated again by different solvents, and 12 chemical components including valeranone, 26 components including bornyl acetate, and 43 components including camphene were identified in the ethyl ether fraction, the pentane - ethyl ether fraction, and pentane fraction, respectively. In the acidic fraction, n-butanoic acid and other 44 components were identified, among which the most abundant ones were 3-methyl butanoic acid(42.1%), dimethoxy-2-propanoic acid(11.5%), and 5-­ethyldihydro-2H-furanone(5.7%).

  • The effects of ginseng extract, crude saponin and ether layer fraction on the alcohol fermentation and the growth of Zymomonas mobilis studied. The growth of Zymomonas mobilis was inhibited by the addition of ginseng extract, crude saponin and ether layer fraction, of which the last was shown to have the most inhibitory effect. The pH cultural broth recorded a significant decrease during 36 hrs alcohol fermentation, followed by a slower decrease in pH after 36 hrs. The alcohol fermentation was stimulated most remarkably by the addition of 0.305% crude saponin, while inhibited most by the addition of 0.228% ether layer fraction.

  • Experiments were performed to investigate the effect of Panax ginseng petroleum ether fraction on delayed type hypersensitivity, rosette formation, phagocytic activity and histophathological influence in lead acetate treated mice. Lead acetate was administered in the drinking water and ginseng pet. ether fraction was injected i.p.. Mice were sensitized and challenged with sheep red blood cells. Erythrocyte(I) rosette formation and DTH reaction were significantly depressed in lead acetate treated mice, and those were restored administration of ginseng fraction. Ginseng pet. ether fraction administration did not have any effect on decreased phagocytic activity. Follicular and parafollicular areal destruction of spleen, and destruction of thymus were finded in lead acetate exposed-mice. Small dose of ginseng pet. ether fraction (5 mg/kg, 10 mg/kg), administraction inhibited those histopathological changes, but large dose (20 mg/kg) didn't.

  • Experiments were performed on mice to investigate the effect of Panax ginseng petroleum ether fraction on the immunotoxicity of lead acetate. Lead acetate was administered in the drinking water and ginseng p-ether fraction was injected intraperitoneally, Mice were sensitized and challenged with sheep red blood cell. Humoral immune responses were evaluated by antibody production and Arthus reaction. Pathotoxicological influences were measured by serum protein, alkaline phosphatase and total cholesterol. The weight of liver, spleen and thymus were measured. Lead acetate exposure significantly decreased hemagglutination titer, hemolysin titer, Arthus reaction, spleen and thymus weight. Ginseng p-ether fraction administration significantly restored or potentiated reduced humoral immune response, spleen and thymus weight. Reduced serum A/G ratio, total cholesterol and alkaline phosphatase activity were restored or increased by ginseng p-ether fraction administration.

  • Experiment was performed to investigate the immunotoxicity of cadmium administered orally and the effect of ginseng petroleum ether fraction on it. Mice were given 3, 30, or 300 ppm cadmium as cadmium chloride orally in the drinking water and injection of ginseng petroleum ether fraction intraperitoneally for 4 weeks. Mice were sensitized and challenged u'ith sheep red blood cells (5-HBC). Immune response was evaluated by delayed type hypersensitivity (DTH), Rosette forming cell (RFC), phagocyte activity, and natural killer cell activity (NK cell activity). In the present study, cadmium suppressed the cellular immunity, It also depressed phagocyte activity very significantly in all cadmium-administered groups, NK cell activity in the cadmium-300 ppm administered group. Ginseng petroleum ether fraction showed restoring effect on the decrease in RFC by cadmium-administration. Remarkably, it showed very significant restoring effect on the depression of phagocyte activity induced by cadmium-administration. From this result, we suppose that the anti-tumor effect of ginseng ether or petroleum ether extract, which has been reported by some other researchers, is mainly due to the increase of phagocyte activity by it's administration.

  • The content ratios of gingerol in 4 fractions (hexane, ether, ethylacetate and hexane-ether (1:1, v/v) fraction) extracted from ginger (Zingiber officinale Roscoe) were analyzed using high performance liquid chromatography (HPLC). The content ratios of 6-gingerol in 4 fractions were hexane fraction; 49.5%, ether fraction; 20.74%, ethylacetate fraction; 21.43% and hexane-ether (1 : 1, v/v) fraction; 93.70%. Antioxidant activities of soybean oil added 0.2% of each ginger fraction and 0.02% of BHT were determined by peroxide value during storage at 45$^{\circ}C$. And relative antioxidant effectiveness (RAE) was calculated as the ratio of the induction period of a given substrate oil to that of a control oil. RAE of each fraction was hexane fraction; 2.60, ether fraction; 2.33, ethylacetate fraction; 2.07 and hexane-ether (1 : 1, v/v) fraction; 2.75, BHT; 1.74. Inhibition effect of each fraction on the rat liver microsomal lipid peroxidation showed hexane fraction; 93%, ether fraction; 92%, ethylacetate fraction; 86% in 350 g/$m\ell$ and hexane-ether (1 : 1, v/v) fraction; 89% in 20 g/$m\ell$.

Pharmacological Studies on Ether Fraction of Corni Fructus (산수유 에텔분획물의 약리작용에 관한 연구)

  • The ether fraction obtained from dry fruit of Cornus officinalis was investigated for the anti-inflammatory action, acute toxicity and central nervous system activities. From several pharmacological examinations, it was found that the ether fraction suppressed considerably carrageenin edema at the dose of 600mg/kg p.o. in rats, inhibited the granuloma formation in rats as given 100mg/kg p.o. for 7 days and decreased the swelling of both of complete adjuvant injected and noninjected (contralateral) paws of the rats at the dose of 100mg/kg p.o. given for 14 days. The $LD_{50}$ of the fraction are estimated to be more than 2,000mg/kg p. o. and 642mg/kg i. p. in mice. And the fraction did not show any sedative, stimulative, analgesic and anticonvulsant action but exhibited hypothermic action. These results might be concluded that the ether fraction of Corni fructus showed anti-inflammatory action in both of acute and chronic type models without any considerable central nervous depressant activity and exhibited very weak acute toxicity in mice.

Mice were sensitized and challenged with sheep red blood cells (S-RBC). Humoral immune response was evaluated by antibody production and Arthus reaction. Body weight gain, spleen weight, thymus weight, and liver weight were also measured. In the present study, cadmium generally suppressed the humoral immunity. There was decrease in the rate of body weight gain and liver weight to body weight by cadmium-administration. Ginseng petroleum ether fraction showed restoring effect, to some extent, on the decrease in PFC and in the rate of liver weight to body weight by cadmium-administration. But it more suppressed Arthus reaction.

  • It has been known that ethyl acetate fraction and petroleum ether fraction prepared from ginseng are inhibitory to the L5178Y and sarcoma 180 cell at the concentrations o! 0.Imgfml or 0.2mg/ml. The shiny was carried out to examine effects of the two fractions on the activities of RNA polymerase, succinate dehydrogenase (SDH) and malate dehydrogenase (MDH) present in normal rabbit liver. The ethyl acetate fraction did not show any inhibitory effect on the RNA polymerase and SDH activity at the concentrations of 0.Imgfml and 0.2mglml, but inhibited malate dehydrogenase activity by 12.3% and 15.5%, at the same concentrations, respectively. The fraction also inhibited all the three enzymes at higher concentrations tested, but stimulated the succinate dehydrogenase activity at 0.024mg/ml to increase the enzyme activity by 14.6%. The petroleum ether fraction activated the SDH activity by 12.9% and 20.8%, at the concentration of 0.1mg/ml and 0.2mg/ml respectively. But the fraction did not affect the MDH activity at the same concentration. The fraction, however, inhibited the MDH activity and activated the SDH activity by 13.5% and 18.2%, at the concentration of 0.8mg/ml respectively.

  • The methanol extract of Thalictrum uchiyamai Nakai was fractionatedi nto four parts; (1) ether soluble nonphenolic fraction (2) phenolic tertiary bases fraction, (3) chloroform soluble tertiary bases fraction and (4) aqueous quaternary bases fraction. The fractions of this plant showed a total of twenty-four spots on the silica gel TLC. Ether soluble nonphenolic fraction and ether soluble phenolic fraction showed nine alkaloid spots respectively, chloroform soluble fraction showed three alkaloid spots and quaternary bases fraction showed three alkaloid spots. The antimicrobial activity of the four extracts was shown as follows; nonphenolic tertiary bases inhibited the growth of Micrococcus dutica at the concentration of $50\mu$g/ml, in phenolic tertiary bases $500\mu$g/ml concentration was inhibitory to Micrococcus dutica and to Staphyloccus aureus. In aqueous quaternary bases, $500\mu$g/ml concentration was inhibitory to Klebsiella pneumonae. Chloroform soluble tertiary fraction showed slight activity. All of bases showed reproducible activity against tested microorganisms.
