쑥의 건조 방법에 따른 품질 변화 (Influence of Dry Methods on Qualities of Artemisia sp.)

  • This study is on different dry methods for Artemisia sp. which were not affected by the dry matter percent. Essential oil components such as 1,8-cineol and terpineol were highly affected by the dry method. 1,8-cineol and terpineol components were decreased by adaptation of the heated oven-drying method and the freeze-drying method. Borneol components were increased through shady sun-drying, sun-drying and freeze-drying. Heated oven drying method decreased the vitamin C content. Vitamin C content was decreased in oven-drying about 50% during this period, but only 10% decreased in the freezing dry method. Qualities were changed by different drying methods. Therefore, different dry methods of Artemisia sp. were applied to the flavorgen.

Antileishmanial and Cytotoxic Effects of Essential Oil and Methanolic Extract of Myrtus communis L.

  • Plants used for traditional medicine contain a wide range of substances that can be used to treat various diseases such as infectious diseases. The present study was designed to evaluate the antileishmanial effects of the essential oil and methanolic extract of Myrtus communis against Leishmania tropica on an in vitro model. Antileishmanial effects of essential oil and methanolic extract of M. communis on promastigote forms and their cytotoxic activities against J774 cells were evaluated using MTT assay for 72 hr. In addition, their leishmanicidal activity against amastigote forms was determined in a macrophage model, for 72 hr. Findings showed that the main components of essential oil were ${\alpha}$-pinene (24.7%), 1,8-cineole (19.6%), and linalool (12.6%). Findings demonstrated that M. communis, particularly its essential oil, significantly (P<0.05) inhibited the growth rate of promastigote and amastigote forms of L. tropica based on a dose-dependent response. The $IC_{50}$ values for essential oil and methanolic extract was 8.4 and $28.9{\mu}g/ml$ against promastigotes, respectively. These values were 11.6 and $40.8{\mu}g/ml$ against amastigote forms, respectively. Glucantime as control drug also revealed $IC_{50}$ values of 88.3 and $44.6{\mu}g/ml$ for promastigotes and amastigotes of L. tropica, respectively. The in vitro assay demonstrated no significant cytotoxicity in J774 cells. However, essential oil indicated a more cytotoxic effect as compared with the methanolic extract of M. communis. The findings of the present study demonstrated that M. communis might be a natural source for production of a new leishmanicidal agent.

Antibacterial Mode of Action of Cinnamomum verum Bark Essential Oil, Alone and in Combination with Piperacillin, Against a Multi-Drug-Resistant Escherichia coli Strain

  • This study aims to investigate the mechanism of action of the cinnamon bark essential oil (CB), when used singly and also in combination with piperacillin, for its antimicrobial and synergistic activity against beta-lactamase TEM-1 plasmid-conferred Escherichia coli J53 R1. Viable count of bacteria for this combination of essential oil and antibiotic showed a complete killing profile at 20 h and further confirmed its synergistic effect by reducing the bacteria cell numbers. Analysis on the stability of treated cultures for cell membrane permeability by CB when tested against sodium dodecyl sulfate revealed that the bacterial cell membrane was disrupted by the essential oil. Scanning electron microscopy observation and bacterial surface charge measurement also revealed that CB causes irreversible membrane damage and reduces the bacterial surface charge. In addition, bioluminescence expression of Escherichia coli [pSB1075] and E. coli [pSB401] by CB showed reduction, indicating the possibility of the presence of quorum sensing (QS) inhibitors. Gas-chromatography and mass spectrometry of the essential oil of Cinnamomum verum showed that trans-cinnamaldehyde (72.81%), benzyl alcohol (12.5%), and eugenol (6.57%) were the major components in the essential oil. From this study, CB has the potential to reverse E. coli J53 R1 resistance to piperacillin through two pathways; modification in the permeability of the outer membrane or bacterial QS inhibition.

사인으로부터 추출한 정유와 용매 추출물의 휘발성 물질 및 금속성분 분석 (Analysis of volatile compounds and metals in essential oil and solvent extracts of Amomi Fructus)

  • 한약재 중 항산화 작용이 있다고 알려진 사인(Amomi Fructus)을 SDE (simultaneous distillation extraction) 방법으로 정유(essential oil)를 추출하고 GC-MS (gas chromatography-mass spectrometry)를 이용하여 39개의 성분을 규명하였으며, 주요 성분은 camphor, borneol acetate, borneol, D-limonene 및 camphene이었다. 유기용매 3가지(헥세인, 다이에틸 에터 및 다이클로로메테인)를 이용하여 얻은 용매 추출물에서 정유성분 이외에 추가로 4개의 성분을 규명하였으며, 주요 5개 성분은 동일하였으나 상대적인 면적 비율은 차이가 있었다. 특정온도와 휘발 조건에서 정유와 용매 추출물에서 휘발하는 물질들의 종류와 상대적인 비율을 측정하기 위하여 dynamic headspace를 이용하여 특정 조건에서 휘발하는 물질들을 트랩에 포집한 후 GC-MS로 분석하였다. 사인으로부터 SDE 방법에 의해 정유를 추출하는 방법의 회수율을 장뇌(camphor)를 이용하여 측정하였으며, 장뇌의 회수율은 82.0% 이었다. 사인에서 Hg의 함량은 수은 분석기로 측정하였으며, Cd, Pb, Cr, Mn, Co, Ni, Cu 와 Zn은 ICP-MS (Inductively coupled plasma-mass spectrometer)로 측정하였다. 사인에서 Pb, Cd 및 Hg이 각각 0.72 mg/kg, <0.10 mg/kg 및 0.0023 mg/kg 검출되었으며, 이는 순도시험 허용기준치 이하의 값들이었다. 사인에서 Cr, Mn, Co, Ni, Cu와 Zn의 함량을 조사한 결과 Mn (213 mg/kg), Cu (8.29 mg/kg) 및 Zn (31.0 mg/kg)등이 비교적 높은 함량으로 검출되었으며, 정유와 용매 추출물에서도 Mn (0.65~9.08 mg/kg), Cu (1.16~4.40 mg/kg) 및 Zn (1.10~3.80 mg/kg)이 검출되었다. 이들 금속성분이 사인에서 기인하는 것인지 아니면 유통과정이나 취급과정에서 오염된 것인지의 여부 규명이 필요하며, 또한 이런 금속들이 생물학적 활성 연구에 어떤 영향 주는지도 규명할 필요가 있다.

털두꺼비하늘소(Moechotypa diphysis)로에 대한 18종 식물정유와 주요성분의 훈증효과 (Fumigant Toxicity of 18 Essential oils and Their Major Compounds against Adult Oak Longicorn Beetle, Moechotypa diphysis (Coleoptera: Cerambycidae))

  • 털두꺼비하늘소(Moechotypa diphysis) 성충에 대한 18종 식물정유의 훈증독성을 조사하였다. 그들 중 10 $\mu\ell/\ell$ (공기)의 농도에서 eucalyptus oil, rosemary oil, 그리고 pennyroyal oil은 100%, sage oil은 85%의 살충활성을 나타내었다. 4종 정유의 치사속도를 조사한 결과, eucalyptus oil과 rosemary oil이 처리 후 6시간 이내에 100%의 살충률을 나타내었다. 각 정유에 대한 주요 구성성분을 GC와 GC/MS로 분석한 결과, eucalyptus oil, rosemary oil, sage oil의 주성분인 1,8-cineole과 sage oil의 또 다른 주성분인 thujone과 pennyroyal의 주성분인 pulegone 모두 20 $\mu\ell/\ell$(공기)의 농도에서 100%의 살충효과를 나타내었다.

Comparison of Chemical Compositions and Antimicrobial Activities of Essential Oils from Three Conifer Trees; Pinus densiflora, Cryptomeria japonica, and Chamaecyparis obtusa

  • The chemical compositions, and antibacterial and antifungal effects of essential oils extracted from three coniferous species, Pinus densiflora, Cryptomeria japonica, and Chamaecyparis obtusa, were investigated. Gas chromatography mass analysis of the essential oils revealed that the major components and the percentage of each essential oil were 16.66% $\beta$-phellandrene and 14.85% $\alpha$-pinene in P. densiflora; 31.45% kaur-16-ene and 11.06% sabinene in C. japonica; and 18.75% bicyclo [2,2,1] heptan-2-ol and 17.41% 2-carene in Ch. obtusa. The antimicrobial assay by agar disc diffusion method showed that $2.2{\mu}g$ of Ch. obtusa oil inhibited most effectively the growth of Escherichia coli ATCC 33312 and Klebsiella oxytoca ATCC 10031, whereas the C. japonica oil gave weak antimicrobial activity. The minimal inhibitory concentration(MIC) values for bacterial strains were in the range of 5.45-21.8 mg/ml depending on essential oils, but most Gram-negative bacteria were resistant even at 21.8 mg oil/ml. P. densiflora oil showed the most effective antifungal activity and the MIC values for Cryptococcus neoformans B42419 and Candida glabrata YFCC 062CCM 11658 were as low as 0.545 and 2.18 mg/ml, respectively. Cryp. neoformans B42419 was the most sensitive to all essential oils in the range of 0.545-2.18 mg/ml. Our data clearly showed that the essential oils from the three conifers had effective antimicrobial activity, especially against fungi.

Evaluation of extraction methods for essential oils in mugwort (Artemisia montana) using gas chromatography-mass spectrometry

  • Mugwort (Artemisia montana), which is a perennial plant mainly distributed throughout Northeast Asian regions, has been used as a preferred source of various foods and traditional medicines in Korea. In particular, as essential oils extracted from mugwort were reported to be biologically active, its steam distillate has been widely used to treat various conditions, such as itching, hemorrhoids, and gynecological inflammation. Therefore, efforts have been devoted to develop effective methods for the collection of bioactive essential oils from mugwort. In this study, five mugwort extracts were obtained using different extraction conditions, namely, 6 % ethanol at room temperature and at 80 ℃, pure ethanol, n-hexane, and an adsorbent resin. To evaluate the five extracts of mugwort, area-under-the-curve values (AUCs), chemical profiles, and major bioactive essential oil contents were investigated using gas chromatography-mass spectrometry (GC-MS). An overall assessment of the volatile components, including essential oils, in the five extracts was conducted using AUCs, and the individual essential oil in each extract was identified. Furthermore, the four major essential oils (1,8-cineole, camphor, borneol, and α-terpineol), which are known to possess anti-microbial and anti-inflammatory activities, were quantified using authentic chemical standards. Based on the evaluation results, pure ethanol was the best extractant out of the five used in this study. This study provides evaluation results for the five different mugwort extracts and would be helpful for developing extraction methods to efficiently collect the bioactive oil components for medical purposes using chemical profiles of the extracts.

광릉쥐오줌풀의 정유향 특성 (Odor Characteristics of Essential Oil of Valeriana fauriei var. dasycarpa HARA)

  • 광릉쥐오줌풀 뿌리의 정유성분 조성 및 정유의 특성을 더욱 정밀하게 조사하기 위하여 수증기 증류에 의해 얻어진 정유성분을 산성, 페놀성, 염기성 및 중성성분 분획으로 분리하여 분획별 향의 관능평가 및 분획물의 조성을 조사한 결과는 다음과 같다. 1. 광릉쥐오줌풀 뿌리의 정유성분분획물 각각의 조성은 중성성분 분획물 92%, 산성성분 분획물 6%, 염기성성분 및 페놀분획물은 각각 1% 미만이며 각 분획물의 향특성은 중성성분 분획은 sweet-balsamic, woody, musky, medicinal 향이며 산성성분 분획은 sweat-socks, valeric-like, cheese-like 향이었다. 2. 중성성분분획물을 용매의 극성을 달리하여 분획한 결과 용출 용매별 분획 비율은 ethyl ether분획물이 44%, pentane : ethyl ether분획물 34%, pentane과 methanol분획물은 각각 11%였다. 분획별 향 특성은 ethyl ether 분획물은 woody, medicinal 향이며 pentane :. ethyl ether 분획물은 sweet-balsamic, woody, herb-like향 특성을 나타내었다. 극성을 달리한 용출용매별 분획물에서 확인된 성분은 ethyl ether 분획물에서 valeranone 등 12종, pentane : ethyl ether(9 : 1) 분획물에서는 bornyl acetate등 26종, pentane 분획물에서 camphene등 43종을 확인하였다. 3. 산성성분분획에서는 n-butanoic acid 외 44종을 확인하였고 그 중 양적으로 많은 성분은 3-methyl butanoic acid 42.1%, dimethoxy-2-propanoic acid 11.5%, 5-ethyldihydro-2H-furanone 5.7% 등 이었다.

부지화 미숙과 에센셜 오일의 항산화 및 항균 활성 효과 (Influence of Essential Oil in 'Shiranuhi' Immature Fruit on Antioxidant and Antimicrobial Activities)

  • This study was designed to analyze the chemical composition of essential oil in 'Shiranuhi' immature fruit and to test their biological activities. 'Shiranuhi' immature essential oils (SIEO) were obtained by steam distillation from fruits collected from Jeju Island and were analyzed using gas chromatograph (GC)-flame ionization detectors (FID) and GC-MS. Fourteen components were identified in the essential oil. Limonene (75.21%) and terpineol (8.68%) were the major components in SIEO. Since acne vulgaris is the combined result of a bacterial infection and the inflammatory response to that infection, we examined whether SIEO possessed antibacterial against skin pathogens. As a result, SIEO showed excellent antibacterial activities against drug-susceptible and -resistant Propionibacterium acnes and Staphylococcus epidermidis, which are acne-causing bacteria. In this study, SIEO was examined on DPPH radical scavenging activities, which showed moderate antioxidant activity ($SC_{50}$, $15.36{\mu}L/mL$). In order to determined whether SIEO can be safely applied to human skin, the cytotoxicity effects of SIEO were determined by colorimetric MTT assays in normal human fibroblasts and keratinocyte HaCaT cells. They exhibited low cytotoxicity at $0.5{\mu}L/mL$ in both celllines. Based on these results, we suggest the possibility that essential oil of 'Shiranuhi' maybe considered as an antibacterial and antioxidant agent.
