Antioxidant Properties of 7 Domestic Essential Oils and Identification of Physiologically Active Components of Essential Oils against Candida albicans (식물정유 7종의 항산화능 분석 및 Candida albicans 생장 억제 정유의 생리활성 성분 구명)

  • In this study, we selected two essential oils, Citrus unshiu and Cinnamomum cassia with superior antioxidant effects from the essential oils of 7 wild plants in South Korea and examined their antimicrobial activity against Candida albicans, which causes dermatitis to identify the antimicrobial components in the essential oils. As a result of measuring DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity, SC50 value of the Citrus unshiu essential oil was 0.010 mg/mL, while for the Cinnamomum cassia essential oil, SC50 value was 0.09 mg/mL. In addition, when ABTS (2,2'-azinobis (3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging activity was measured, SC50 value of the Citrus unshiu essential oil was 0.09 mg/mL, while for the Cinnamomum cassia essential oil, it was 0.06 mg/mL, exhibiting high antioxidant activity. For the minimum inhibitory concentration (MIC), the essential oil of Cinnamomum cassia was 1.25 mg/mL and that of Citrus unshiu was 5 mg/mL, demonstrating a high antimicrobial activity of the Cinnamomum cassia essential oil. Through the thin layer chromatography (TLC) bioassay, we assessed the antimicrobial activity against C. albicans according to the fraction components of the two essential oils. Also, by using preparative TLC (prep. TLC), we obtained the active fractions, and by performing GC/MS analysis of the components with the same Rf value, we identified the antimicrobial-active components. As a result, the main components having antioxidant and antimicrobial activities were cinnamyl acetate, eucalyptol, linalool, and citral of the Cinnamomum cassia essential oil and linalool from the Citrus unshiu essential oil. Also, based on the analysis of the fractional components that showed antioxidant and antimicrobial activities in both of the two essential oils, it was found that linalool has antioxidant activity, while cinnamyl acetate, eucalyptol, citral, and geranyl acetate have antioxidant and antimicrobial activities.

Change of Essential Oil Constituents during Flue-curing Process in Flue-cured Tobacco, NC82 & KF114 (황색종 NC82와 KFl14의 건조단계별 정유성분의 변화)

  • Essential oil in tobacco leaves influences the taste and aroma of cigarette smoke and is important to tobacco quality. This study was conducted to investigate the change in the level of essential oil components during flue-curing process of two flue-cured tobaccos, NC82 and KEl14. Flue-curing process was divided by six steps; harvest stage, the end of yellowing stage, the middle of color fixing stage, the end of color fixing stage, the middle of midrib drying stage, full-cured stage. NC82 in each stage contained 0.28%, 0.30%, 0.35%, 0.36%, 0.40% and 0.42% essential oil, respectively, and KF114 were 0.29%, 0.31%, 0.34%, 0.36%, 0.39% and 0.41%, respectively. Almost all hydrocarbons on the basis of relative peak area were gradually increased in two varieties with curing, neophytadiene content in them was highest at the full-cured stage. Most of alcohols and esters with curing showed a declining trend, but benzyl alcohol was increased in two tobaccos. Ketones were largely increased at the midrib drying stage during the curing process, especially, the most largely increasing constituent was $\beta$-damascenone among them. The content of 2-butylterahydrofuran, heterocyclic compounds, was largely increased at tile color fixing stage. There was no considerable difference between NC82 and KFl14 at the GC profile of essential oil and the pattern of each components during flue-curing process.

Antimicrobial Activity of Essential Oil of Pinus koraiensis Seed Against Pathogens Related to Acne

  • The purpose of the present research was to evaluate the antimicrobial activity of the essential oil extracted from Pinus koraiensis seed against pathogens related to acne. The essential oil was extracted by steam distillation method. The chemical compositions of essential oil were analyzed by GC-MS. Alpha-pinene (29.87%), D-limonene (19.26%), betapinene (11.19%), beta-myrcene (3.84%), n-hexadecanoi acid (3.2%), beta-caryphyllene (2.72%), and cyclohexene (2.17%) were main components. This essential oil had antimicrobial activities against Malasseizia furfur, Propionibacterium acnes, and Staphylococcus epidermidis.

GC-MS Analyses of the Essential Oils from Ixeris dentate(Thunb.) Nakai and I. stolonifera A. Gray (GC-MS를 이용한 씀바귀 및 좀씀바귀의 정유 성분 분석)

  • The volatile flavor compounds of the essential oils from Ixeris dentate (Thunb.) Nakai and I. stolonifera A. Gray were investigated. The essential oils were extracted by hydro distillation extraction method. Ninety-three volatile flavor components were identified from I. dentate (Thunb.) Nakai essential oil. Hexadecanoic acid(33.73%) was the most abundant compound, followed by (Z,Z,Z,)-9,12,15-octadecatrienoic acid(18.59%), 6,10,14-trimethyl-2-pentadecanonel(10.39%) and phytol(5.21%). Ninety-seven volatile flavor components were identified from the essential oil of I. stolonifera A. Gray. Hexadecanoic acid was the most abundant component(39.7%), followed by (Z,Z,Z)-9,12,15-octadecatrienoic acid(12.63%), 9,12-octadecadienoic acid, ethyl ester(12.36%), pentacosane(5.2%) and 6,10,14-trimethyl-2-pentadecanone(3.18%). The volatile composition of I. dentate (Thunb.) Nakai was characterized by higher contents of phytol and phthalides than those of I. stolonifera A. Gray. The volatile flavor composition of I. stolonifera A. Gray can easily be distinguished by the percentage of sesquiterpene compounds against I. dentate (Thunb.) Nakai essential oil.

Biological Activities of the Essential Oil from Angelica acutiloba

  • Angelica acutiloba is one of the most intensively cultivated medicinal plants in Korea. The roots of this plant have been used as an important herbal drug, especially for the treatment of various female disorders, as the traditional therapy in Korea and other Asian countries. Consumption of its fresh leaves as a healthy vegetable has recently increased. In this study, essential oil fractions were extracted from the roots and leaves of this plant by steam distillation. Compositions of the two oils were compared by gas chromatography-mass spectrometry (GC-MS). The antibacterial activities of the essential oil were determined against three strains of Escherichia coli. DPPH radical scavenging and reducing power tests were performed to evaluateits antioxidant activities. The cytotoxic activities of the essential oil against a human breast and a uterine cancer cell line were estimated by MTT tests. Additionally, the morphological changes after treatment of the oil fraction were observed under a microscope. The essential oil fraction and its main components, Z-ligustilide and butylidene phthalide, inhibited the growth of three E. coli strains examined, with minimum inhibiting concentrations (MICs) ranging from 1.0 mg/ml to 8.0 mg/ml. Additionally, the essential oil fraction of A. acutiloba exhibited significant DPPH free radical scavenging activity and reducing power. Significant cytotoxic activities of the A. acutiloba essential oil were observed for human uterine (Hela) and breast (MCF-7) cancer cell lines.

DPPH Radical Scavenging Activity and Composition of Essential Oil from the Herbs of Jeju Agastache rugosa (제주산 배초향 지상부에서 얻은 정유의 조성과 항산화 효과)

  • The essential oil obtained by steam distillation of the herbs of Jeju Agastache rugosa has shown significant DPPH radical scavenging activity. For the elucidation of its volatile components, the essential oil was analyzed by GC-MS. Each component was identified by GC or mass spectral analysis. The compounds identified were as follows: pulegone, L-menthone, DL-limonene, isopulegone, beta-caryophyllene, beta-myrcene, and estragole. Several abundant components with phenylpropanoid-type structures may have contributed to the activity. The results suggest that the essential oil may be suitable for development as a food preservative and alternative antioxidant.

  • The essential oil obtained by steam distillation from a medicinal plant of Artemisia princeps. The essential oil was analyzed by GC-MS. The heat treatments in roasted condition of temperatures were $80^{\circ}C$, $110^{\circ}C$, and $230^{\circ}C$, and the lengths of the time were 6 min, 10 min, and 14 min, respectively. The blanching conditions($100^{\circ}C$) of the treatments lasted 1 min, 2 min, and 5 min while the times of oven dry ($50^{\circ}C$) was 5 min. As the result, the essential oil content of the control plot was higher than the others, and that of the roasted ones was the second highest in low temperature with short treatment time. The major components were eucalyptol, cyclohexadience, phenol, terpineol, and caryopbyllene.

  • Since Houttuynia cordata is well known as a medicinal herb, due to its antibacterial activity on various microorganisms, present investigation was performed to identify the flavor compounds for volatile essential oil. Volatile essential oil was collected by simultaneous distillation-extraction(SDE), and then the oil components were separated on HP-5 capilliary column$(25m{\times}0.25mm\; i.d.)$ and identified those components by GC-MS. Fifty two compounds were isolated from the volatile essential oil of Houttuynia cordata and forty four were positively identified by GC-MS. The volatile compounds were composed mainly of terpenoids(25 classes), aldehydes(7 classes), alcohols(4 classes), ketones(3 classes), acids(1 class) and miscellaneous compounds(4 classes). Of these, the major compounds were ${\beta}-myrcene$, ${\beta}-ocimene$, decanal, 2-undecanone and geranyl propionate.

  • Contents of essential oil and its composition in aerial part of Agastache rugosa were investigated. Essential oil was obtained from the dried stems, leaves and flowers by steam distillation and fractionated into hydrocarbon and oxygenated hydrocarbon by silica gel column chromatography. Each isolate or fraction was identified by GC and GC-MS. The contents of essential oil were 0.29% in leaves of Agastache rugosa on dry basis, 0.38% in flowers. Major components were ${\beta}-caryophyllene(59.3%)$, limonene(13.1%), ${\delta}-cadinene(10.7%)$ among 13 kinds of components confirmed in hydrocarbon fraction of essential oil isolated from leaves. Also, major components in oxygenated hydrocarbon fraction were methyl chavicol (79.1%) and cis-3(1-propenyl) phenol(4.5%) among 44 kinds components confirmed. The highest content among the components identified was methyl chavicol in both leaves and flowers. On the other hand, limonene, trans-2-hexenal, 1-octen-3-one, 6, 10, 14- trimethyl pentadecane- 2-one and phytol were detected typically in leaves, but jasmone and ${\rho}-methoxyacetophenone$ were detected mainly in flowers with small quantity.

Variation of the Essential oil and Main Component from Different Origin of Levisticulum officinale Koch

  • The essential oils were isolated by hydrodistillation and their composition determined capillary GC method with standards. The essential oil content showed significant differences between the two populations on the vegetative organs. The essential oil level of the leaves and roots was considerably higher in the Korean population at full flowering and waxy ripening stage but essential oil content of the roots was significantly higher in the Hungarian taxon at leaf rosette stage. We observed the essential oil accumulation tendency was mianly dependent on plant organs and intra-specific taxon during the vegeation period. Butylidene-phthalide was proved to be the main component of the oil in both population roots (50.9-73.3%), while dimethyl-acetate was showed as a major compound on the over-ground parts (56.7-62.0%). The qualitative composition of the essential oil in the reproductive organs concerning the identified compounds was the same as the vegetative parts with the main component ${\alpha}-phellandrene$ (4.8-28.1%) and butylidene-phtalide (9.7-16.1%), The quantitative composition showed some changes during the ontogenesis phases. Most characteristic ones are the decreasing proportion of dimethyl-acetate (from 7.3% to 1.1%) and the appearance of ${\alpha}-pinene$ (from 0.5% to 1.5%) only after fruit setting in both population.
