  • A thiol-specific spin label was attached to cysteine-102 of yeast cytochrome c and electron paramagnetic resonance (EPR) spectra were measured as a function of added cytochrome c oxidase concentration. The intensity decreased due to line broadening as cytochrome c formed a complex with cytochrome c oxidase and reached a minimum when the ratio of cytochrome c to cytochrome c oxidase became one. Replacement of either Lys-72 or Lys-87 of cytochrome c by Glu did not result in a significant change in binding affinity. Interestingly the K72E mutant, unlike K87E, had a much lower rate of electron transfer than the wild type. These results indicate that many positively charged residues as a group participate in complex formation but Lys-72 might be important for cytochrome c to be locked in an orientation for an efficient electron transfer. A stoichiometry of 1 was also confirmed by optical absorption of the cytochrome c-cytochrome c oxidase complex which had been run through a gel chromatography cloumn to remove unbound cytochrome c. The EPR spectrum of this 1:1 complex, however, was a mixture of two components. This explains a biphasic kinetics for a single binding site on cytochrome c oxidase without invoking conformational transition.

  • In this paper, EPR (ethylene propylene rubber) insulation material was accelerated degradation test at $121^{\circ}C$, $136^{\circ}C$, $151^{\circ}C$, and experiment the typical EAB (elongation at break) at mechanical characteristics analysis. It is shown that the failure-time at the point of 50% of the initial value of Elongation rate to obtain the activation energy. The failure-time was shown each 5,219 hr, 3,165 hr, and 668 hr at three temperatures. In order to derive the activation energy, Arrhenius methodology was applied. Also, we got the Arrhenius plot from three accelerated temperatures. The activation energy values got 0.98 eV from EAB test. The experimental data were evaluated for estimating the probability density, and the suitable distribution by using statistical program MINITAB. It is shown that EAB data by the acceleration thermal degradation is most suitable for the Weibull distribution.

  • Cr-Mo-V steel is widely used as a material for the turbine structural component in fossil power plants. It is well known that this material shows the various material degradation phenomenons such as temper embrittlement, carbide coarsening. and softening etc. or ins to the severe operation conditions as high temperature and high pressure. These deteriorative factors cause tile change of mechanical properties as reduction of fracture toughness. Therefor it is necessary to evaluate tile extent of degradation damage for Cr-Mo-V steel in life assessment of turbine structural components. In this paper. the electrochemical potentiokinetic reactivation(EPR) test in $50wt%-Ca(NO_3)_2$ solution is performed to develop the newly technique for degradation damage evaluation of Cr-Mo-V steel. The results obtained from the EPR test are compared with those in small punch(SP) tests recommended by semi-nondestructive testing method using miniaturized specimen. The evaluation parameters used in EPR test are tile reactivation current density$(I_R)$ and charge$(Q_{RC})$ reactivation rate$(I_R/I_{Crit},\;Q_R/Q_{Crit})$. The results suggest that $I_R/I_{Crit}$ in these parameters shows a good correlation with SP test results.

  • Production from copepodite IV to adult of two euchaetid species Euchaeta plana and Paraeuchaeta russelli was measured at the southeastern sea of Korea from April to November, 2014. The mean density was $2.0ind\;m^{-3}$ for E. plana and $4.1ind\;m^{-3}$ for P. russelli, with the high contribution of copepodite V to total density. The densities of total individuals, adult females and eggs were highest in November for both species. The mean egg production rate (EPR) was $1.7eggs\;female^{-1}\;d^{-1}$ for E. plana and $3.1eggs\;female^{-1}\;d^{-1}$ for P. russelli. Both of them showed the highest EPR in September but zero EPR in summer. The mean weight-specific EPR was $0.038d^{-1}$ for E. plana and $0.079d^{-1}$ for P. russelli. The mean total production rates of E. plana and P. russelli were $5.3{\mu}g\;C\;m^{-3}\;d^{-1}$ and $17.8{\mu}g\;C\;m^{-3}\;d^{-1}$, respectively, with the largest production in November. The mean Production/Biomass ratio was $0.06d^{-1}$ for E. plana and $0.07d^{-1}$ for P. russelli, with its peak in September for both. The total production of E. plana and P. russelli was positively correlated with the density of a copepod Oncaea venusta, rather than chl-a concentration, indicating that the two copepods might be carnivores. This study evaluates the contribution of euchaetids to the copepod community in the southeastern sea of Korea.

  • The active sites of the nickel and iron-containing enzyme, carbon monoxide dehydrogenase (CODH) from clostridium thermoaceticum were investigated using Electron Paramagnetic Resonance (EPR) technique. CODH exhibits several spectral features called NiFeC, $g_{ave}=1.82$, $g_{ave}=1.86$. FCII signals which are originated from different clusters in this enzyme. CODH is know to catalyze two different kinds of reactions - acetyl-CoA synthesis and CO oxidation. The acetyl-CoA synthesis activity can be followed by monitoring CO/acetyl-CoA exchange. The addition of 1,10-phenanthroline (phen) to CODH selectively destroyed the CO/acetyl-CoA exchange activity and eliminated the NiFeC signal completely. CO oxidation activity and other EPR signals were unaffected. Such behavior demonstrates that CODH has two distinct active sites and that the NiFe complex is only responsible for the CO/acctyl-CoA exchange activity. Phen caused the removal of only 30% of Ni in the NiFe complex ($0.3Ni/{\alpha}{\beta}$) as shown by the quantitative metal analysis. The phen-treated CODH could be reactivated fully by incubation In $Ni^{2+}$ solution. Radioactive $^{63}Ni^{2+}$ was used to quantitate the amount of the $Ni^{2+}$ incorporated into phen-treated enzyme and showed that the amount was the same as the removed by the phen treatment. i.e. $0.3Ni/{\alpha}{\beta}$. This indicates that only 30% of NiFe complexes are labile and responsible for the CO/acctyl-CoA exchange activity, the other 70% are non-labile and have no exchange activity. This is the first clear evidence that the NiFe complex is heterogencous and labile and non-labile Ni sites arc interacting differently with substrates and chelating agents like phen.

  • The enhanced permeability and retention (EPR) effect is used extensively for the passive targeting of many macromolecular drugs for tumors. Indeed, the EPR concept has been a gold standard in polymeric anticancer drug delivery systems. This study investigated the tumoral distribution of self-assembled nanoparticles based on the EPR effect using fluorescein and radio-labeled nanoparticles. Self-assembled nanoparticles were prepared from amphiphilic chitosan derivatives, and their tissue distribution was examined in tumor-bearing mice. The size of the nanoparticles was controlled to be 330 run, which is a size suited for opening between the defective endothelial cells in tumors. The long-circulating polymer nanoparticles were allowed to gradually accumulate in the tumors for 11 days. The amount of nanoparticles accumulated in the tumors was remarkably augmented from 3.4%ID/g tissue at 1 day to 25.9%ID/g tissue at 11 days after i.v. administration. The self-assembled nanoparticles were sustained at a high level throughout the 14 day experimental period, indicating their long systemic retention in the blood circulation. The ${\gamma}$-images provided clear evidence of selective tumor localization of the $^{131}I$-labeled nanoparticles. Confocal microscopy revealed the fluorescein-labeled nanoparticles to be preferentially localized in the perivascular regions, suggesting their extravasation to the tumors through the hyperpermeable angiogenic tumor vasculature. This highly selective tumoral accumulation of nanoparticles was attributed to the leakiness of the blood vessels in the tumors and their long residence time in the blood circulation.

  • The Korea retrospective dosimetry (KREDOS)-electron paramagnetic resonance (EPR) group undertook an intercomparison investigation utilizing hydroxyapatite. This analysis involved four institutions: the Korea Institute of Radiological and Medical Sciences, Dongnam Institute of Radiological and Medical Sciences, Korean Association for Radiation Application, and Radiation Health Institute of Korea Hydro & Nuclear Power. Following the irradiation of the hydroxyapatite sample, the recorded build-up was analyzed. To validate the reliability of the EPR dosimetry findings and enhance its operational performance, a hydroxyapatite dose-response curve was plotted and dosimetry was performed for a blind sample. The proficiency of each laboratory was assessed by employing an interlaboratory comparison methodology. This involved a comparative analysis of the measurement results by calculating the relative bias, z-score, and En value. The results submitted by the participating laboratories demonstrated satisfactory ratings for doses of 1.006, 3.999, and 6.993 Gy. Following the second intercomparison, efforts to optimize their hydroxyapatite-EPR dosimetry systems are underway in the participating laboratories. The current assessment of hydroxyapatite dose yielded the foundational data required to establish the parameters of dental dosimetry. In future, the third intercomparison experiment will be conducted for exploring other materials.
