The purpose of this study is to examine the effect of herbal medicine complex (HMC) containing Camellia sinensis L., Duchoesna chrysantha, Houttuynia cordata Thunberg, Poncirus trifoliata Rafinesque on skin inflammation and atopic dermatitis. First, we examined the anti-inflammatory effect of HMC in TNF-α induced human keratinocytes (HaCaT cell). Real-time PCR and western blotting were performed to evaluate the expression of inflammatory cytokines (e.g., iNOS, COX-2, IL-6, IL-8) mRNA and protein. Four-weeks old male NC/Nga mice were treated with 1% 2,4-dinitrochlorobenzene (DNCB) solution and used as an atopic dermatitis mice model. And, HMC (200 mg/kg or 400 mg/kg) was administered directly into the stomach of mice for 4 weeks, and blood or serum analysis, tissue staining were performed after oral gavage. As a result HMC inhibited the mRNA expression of iNOS, COX-2, IL-6, and IL-8, which had been increased by TNF-α in HaCaT cells. In addition, the protein expression was also significantly suppressed in the same way as the mRNA expression results. The in vivo experiment results showed that, HMC administration reduced thickening of the epidermis and infiltration of eosinophil into the skin stratum basale compared to DNCB treatment. In addition, HMC administration significantly reduced the inflammatory cytokines (IL-4, IL-5, IL-6, and IL-13) production and immunocyte (white blood cell, lymphocyte, neutrophil, and eosinophil) count compared to DNCB treatment. Moreover, the serum IgE and histamine level was decreased by HMC administration. These results suggest that HMC can be used as effective herbal medicine extract for skin inflammation and atopic dermatitis. And this study may contribute to the development of the herbal medicine-based drug for the treatment of skin inflammation and atopic dermatitis.
다양(多樣)한 재배양식(栽培樣式)하에서 파종(播種)및 이앙(移秧)한 5, 10, 15, 20일간 생장한 벼와 피를 굴취하여 해부학적(解剖學的) 특성차이(特生差異)를 검토(檢討)하고자 광학현미경(光學顯微鏡)으로 줄기, 잎, 뿌리의 구조(構造)를 각각(各各) 관찰(觀察)하였으며 그 결과(結果)를 요약(要約)하면 다음과 같다. 1. 줄기의 해부학적(解剖學的) 특성(特性) 차이(差異) 가. 5DAS/T째 벼와 피의 줄기종단(縱斷)을 통한 해부학적(解剖學的) 차이(差異)는 벼는 피보다 엽(葉)의 분화(分化)가 느리고 측아형성(側芽形成)도 적었으며, 분열조직(分裂組織) 신장(伸長)도 피보다 적었다. 나. 담수조건(湛水條件)보다 건답(乾沓)에서 생장속도(生長速度)가 빨랐고 줄기종단(縱斷)에 의한 벼는 엽초가 두껍고, 최외층(最外層) 세포(細胞)가 정교(精巧)하며, 규칙적(規則的)으로 배열(配列)되어 있으나 피는 엽초가 짧고 거칠며 성기고 큰 세포(細胞)들이 불규칙적(不規則的)으로 배열(配列)되어 있는 것이 특징이었다. 다. 건답(乾沓)벼에서의 엽시원체(葉始原體)는 세포내용물(細胞內容物)이 있었던 반면 담수조건(湛水條件)에서는 세포내용물(細胞內容物)이 없는 통기세포(通氣細胞)가 발달하였다. 라. 이앙(移秧)벼에서는 직파(直播)벼에 비해 많은 엽시원체(葉始原體)가 분화(分化)되었고 큰 통기강(通氣腔)들이 발달하였다. 2. 잎의 해부학적(解剖學的) 특성(特性) 차이(差異) 가. 벼의 표피세포(表皮細胞)는 작고 치밀(緻密)한 배열(配列)을 하며 우두상세포(牛頭狀細胞)가 존재(存在)하고 엽물세포(葉肉細胞)에 엽록소(葉綠素)가 있는 반면, 피는 표피세포(表皮細胞)가 크고 성기게 배열(配列)되어 있으며 유관속초세포에 엽록소(葉綠素)가 있고 우두상세포(牛頭狀細胞) 없었다. 엽신(葉身)의 단위(單位)길이당(當) 유관속수(維管束數)는 벼끼리는 차이(差異)가 인정되지 않으나 피는 벼보다 더 많았고, 피는 건답(乾沓)보다 담수(湛水)가 더 많았다. 다. 5DAS/T째 엽신(葉身)의 두께는 이앙(移秧)벼보다는 직파(直播)벼, 건답조건(乾畓條件)보다는 담수조건(湛水條件)에서 피보다 벼가 더 컸으며 직파(直播)벼에 비하여 이앙(移秧)벼는 표피세포(表皮細胞)와 세포벽이 비후화(肥厚化)되었고 엽신(葉身)이 우두상세포(牛頭狀細胞)(기동세포(機動細胞)) 부위에서 굴곡(屈曲)되어 되어 전개(展開)되었다. 3. 뿌리의 해부학적(解剖學的) 특성(特性) 차이(差異) 가. 벼와 피의 근구조(根構造)의 기본적인 차이는 근단(根端)으로부터의 거리에 따라 보면 1mm 부위는 직경(直徑)이 피보다 벼가 크고, 벼의 표피세포(表皮細胞)는 좁고 긴 타원형(楕圓形) 세포(細胞)로 배열(配列)되어 있고, 표피(表皮) 아래층(層)에 외피층(外皮層)이 존재하며 후벽세포(厚壁細胞)가 형성(形成)된 것이 특징(特徵)이고 각(各) 세포(細胞)가 조밀(稠密)하게 배열(配列)된데 비해 피는 크고 둥근 타원형세포로 된 표피세포(表皮細胞)가 있고 외피층(外皮層)과 후벽세포(厚壁細胞)가 없으며 성기게 배열(配列)되어 있었다. 나. 재배양식(我培樣式)에 따른 차이에 있어서 근단(根端) 5mm 부위(部位)의 건답(乾沓짧)벼의 뿌리는 세포내용물(細胞內容物)이 있고 세포배열(細胞配列)이 규칙적(規則的)이며 세포간극(細胞問隙)이나 통기세포(通氣細胞)가 관찰되지 않았으나, 건담(乾沓) 및 담수(湛水)피는 핵(核)이 없고 세포배열(細胞配列)이 불규칙적(不規則的)인 경향(京鄕)이었다. 다. 담수(湛水)벼는 세포간극(細胞間隙)과 통기세포(通氣細胞)가 잘 발달(發達)되고 표피세포(表皮細胞)가 아주 작으며 조밀(稠密)하게 배열(配列)되고 그 아래엔 외피층(外皮層)이 형성되어 있는 것이 특징이었다. 라. 이앙(移秧)벼의 뿌리는 같은 담수상태(湛水狀態)의 직파(直播)벼와도 달리 치밀(緻密)하고 규칙적(規則的)인 표피세포(表皮細胞) 배열(配列)과 세포간극(細胞間隙)의 발달(發達), 핵(核)이 있는 왕성(旺盛)한 세포(細胞)들로 구성(構成)되어 있었다.
제주용암해수는 미네랄과 영양염류가 풍부한 물로 제주만이 보유한 지하수자원이다. 본 연구의 목적은 제주용암해수의 피부 보습효과를 확인하기 위한 것이다. 피부의 건조함을 막고 수분을 유지하기 위해서는 표피층의 장벽기능이 정상적으로 기능하고, 표피층 내 수분의 유지와 이동이 원활히 이루어져야 한다. 제주용암해수를 각질형성세포에 처리한 결과 표피층의 분화과정과 natural moisturizing factor (NMF) 생성과정에 관여하는 유전자인 필라그린과 caspase-14 유전자의 발현양이 증가되는 것을 확인할 수 있었다. 또한 막관통 단백질로 수분의 이동을 조절하는 aquaporin 3 (AQP3) 유전자 발현양과 단백질 발현양도 제주용암해수 처리에 의해 증가하였다. 인공피부를 이용한 실험에서 제주용암해수를 배지에 처리하고 배양한 결과 hyaluronic acid (HA)의 수용체인 CD44의 발현양이 증가하였다. 본 연구를 통해 제주용암해수는 피부 보습과 관련된 인자들의 발현양을 증가시켜 피부의 보습기능에 도움을 주는 것으로 사료되었다.
HaCaT 세포에서 UVB와 $IFN-{\gamma}/TNF-{\alpha}$에 의한 염증 관련 인자의 활동에 복분자 씨앗 추출물의 항염증 소재로서의 가능성을 알아보고자 하였다. 복분자 씨앗 추출물은 HaCaT 세포에서 UVB와 $IFN-{\gamma}/TNF-{\alpha}$에 의한 ROS 유도 활성과 interleukin-$1{\beta}$, interleukin-6, interleukin-8의 발현을 억제 하였다. 또한 염증 매개인자인 cyclooxygenase-2 (COX-2)의 발현 또한 억제 시켰으며, COX-2에 의해 증가되어 지는 $PGE_2$의 발현 또한 억제 시키는 것으로 확인 되었다. 마지막으로 복분자 씨앗 추출물의 피부장벽의 주요 인자인 filaggrin의 발현을 측정해 본 결과 농도 의존적으로 손상된 filaggrin의 발현을 증가 시키는 것을 확인할 수 있었다. 이를 통하여 복분자 씨앗 추출물이 표피 층의 손상을 회복함으로써 염증을 보호하는 효능이 있음을 확인 할 수 있었다. 이상의 결과로 부터 복분자 씨앗 추출물은 UVB로부터 발생되어지는 염증을 개선시킴으로써 항염증에 효능이 있는 추출물임을 확인 수 있었다.
Objectives : Atopic dermatitis is a chronic inflammatory skin disease with frequent relapses. This study was to investigate the effects of Gammakdaejo-tang(GMD) in DNCB induced atopic dermatitis mice. Methods : The study was divided into five comparion groups. 2,4-dinitrochlorobenzene(DNCB) solution was applied to Nc/Nga mice to induce atopic dermatitis, followed by normal group, negative control group with distilled water, positive control group with Dexamethasione and GMD 200mg/kg or 400mg/kg. The control group was orally administered 200㎕ once daily for 4 weeks. Visual skin condition, Immunoglobulin E, Histamine, Cytokine, Immune cells, Tissue biomarkers were observed. Results : As a result of the dermatitis score evaluation, it was confirmed that the GMD-administered group improved symptoms compared to the negative control group. As a result of measuring IgE, the GMD-administered group significantly decreased compared to the negative control group. As a result of measuring Histamine, GMD group except 200mg/kg of GMD significantly decreased compared to negative control group. As a result of measuring cytokine, GMD 200mg/kg significantly reduced IL-1β, IL-6 and TNF-α compared to the negative control. 400mg/kg significantly reduced IL-1β, IL-4, IL-5, IL-6, IL-10, TNF-α and significantly increased IL-2, IFNγ. As a result of confirming the immune cells, all experimental groups showed no difference in basophil, GMD group significantly reduced monocyte and eosinophil compared to negative control group, and GMD 400mg/kg group significantly reduced white blood cell and neutrophil. And significantly increased lymphocytes. As a result of measuring the gene expression level, all GMD group significantly increased TGF-β1 compared with the negative control group, and filaggrin, VEGF and EGF were significantly increased in GMD 400mg/kg group. Epidermis, dermis thickness, and eosinophil infiltration were found to be decreased in all GMD groups compared with the negative control group. Conclusions : GMD is effective in atopic dermatitis by reducing imbalance of immune response of T cells (Th1 / Th2) and reducing skin tissue damage and inflammatory response.
깽깽이풀의 발생학적 연구가 수행되었으며, 이를 매자나무과내의 근연속 들과 비교연구하였다. 깽깽이풀의 발생학적 특징은 4포약, basic type의 약벽형성, 2핵성의 glandular tapetum, 약벽 표피세포의 숙존, 내피층이 섬유상조직으로 발달, 판개형의 약 열개, 화분모세포의 연속형 세포질분열, 정사면체형 화분사분체, 약 열개시 화분은 2세포성, 양주피성, 도생배주, 후층성주심, 한개의 포원세포, 마디풀형 배낭형성, 성숙배낭은 타원형, 핵형 배유형성, 배 발생은 Onagrad 형태, 종자는 종의가 있고, 종피는 외종피외층형 등의 특징을 나타내었다. 매자나무과내의 다른 속들과 비교한 결과 깽깽이풀속은 융단세포의 핵수, 감수분열시 세포질의 분열패턴, 그리고 외종피외층의 두께 등에서 매자나무속이나 Mahonia속 보다는 삼지구엽초속과 Vancouveria 속에 좀 더 닮았다. 또한, 약벽형성 패턴, 융단세포의 핵수, 주심cap의 형성, 반족세포의 행동패턴등에서 한계령풀속과도 유사한 형질이 있었다. 그럼에도 불구하고, 한계령풀속은 다른 많은 발생형질과 분자 data에서 깽깽이풀속과는 다르다. 따라서 발생학적 증거들은 깽깽이풀속이 삼지구엽초속(Epimedium)과 Vancouveria 속에 가장 가까운 유연관계라는 것을 제시하고 있다.
Although the parallel prescription of Sopoongsangagambang (SG) administration along with external treatment such as spraying or ointment application on the skin is clinically used for the treatment of atopic dermatitis (AD), molecular mechanism underlying its effectiveness is unknown. Thus in the present study, diverse immune responses in terms of chemical mediators related to AD were investigated using an atopic mouse model NC/Nga after SG administration and external treatment (ET), and major findings are summarized as follows. 1. The clinical severities in 16 and 20 week old NC/Nga mice with SG and ET treatment were decreased to 72.2% and 62.3% respectively compared to the control NC/Nga mice with no drug treatment. 2. IgE, IL-4, IL-5, IL-6, IL-13, IgM, IgG1 and IgG2a levels in the serum of SG and ET treated NC/Nga mouse group were significantly decreased compared to the untreated control mice. In contrast, $IFN-{\gamma}$ showed a significant increase in the experimental group compared to the untreated control group. 3. The spleen weight of SG and ET treated NC/Nga mice was significantly decreased compared to the untreated control group. 4. The B/T ratio in the lymph node of SG and ET treated NC/Nga mice was increased compared to the untreated control group. $CD4^+\;and\;CD8^+$ cell numbers in the lymph node of SG and ET treated NC/Nga mice were significantly increased compared to the untreated control group, but $CD69^+\;and\;CD11a^+$ cells were significantly decreased. 5. mRNA expression levels of IL-4, IL-5, and CCR3 in the skin tissues of SG and ET treated NC/Nga mice were significantly decreased, and expression levels of IL-6, IL-13, $CD69^+/CD3{\varepsilon}^+\;and\;CD19^+/CD44^+$ in the skin tissues of SG and ET treated NC/Mga mice were significantly decreased compared to the untreated control group. $IFN-{\gamma}$ mRNA expression levels were increased compared to the untreated control group. 6. Histological observation of the ear and neck skin tissues showed that the extents of inflammation and infiltrated immune cells in the epidermis and dermis of SG and ET treated NC/Nga mice were highly reduced compared to the untreated control group. 7. Lymphokine assay showed a significant decrease in IL-4 levels in SG and ET treated NC/Nga mice compared to the untreated control group, but the levels of $IFN-{\gamma}$ secretion were significantly increased drug treated NC/Nga mice.
Objective : This Experimental study was done to investigate the Effect of Taklisodok-um and Hwangryunhaedok-tang(TH) on Atopic Dermatitis. Methods : We assessed effects of TH on the IgE, IL-4, IL-5, IL-6, IgM, IgG1, IFN-${\gamma}$ in vivo, on the IL-4, IL-5, CCR3 in the skin tissues of ear and dorsum with NC/Nga mice. And we assessed effects of TH on the COX-2, IL-$1{\beta}$, TNF-${\alpha}$, IL-6 with RAW 264.7 cell. Results and Conclusion : 1. IgE, IL-4, IL-5, IL-6, IgM, IgGl levels in the serum of TH treated NC/Nga mouse group were decreased compared to the untreated control mice. IFN-r showed a increase in the experimental group compared to the untreated control group. The spleen weight of TH treated NC/Nga mice was decreased compared to the untreated control group. 2. mRNA expression levels of IL-4, IL-5 and CCR3 in the skin tissues of TH treated NC/Nga mice were decreased, and expression levels of IL-6 in the skin tissues of TH treated NC/Nga mice were decreased compared to the untreated control group. IFN-${\gamma}$ mRNA expression levels were increased compared to the untreated control group. 3. Judged from that IL-$1{\beta}$, TNF-${\alpha}$, IL-6 express of gene, effect of inflammatory Cytokines revelation were decreased compared to the untreated control group. 4. Depend on the strength of TH, inflammatory RAW 264.7 in the serum of TH were inhibited compared to the untreated control serum that leaded a COX-2 activity model. 5. Histological observation of the ear and skin tissues showed that the extents of inflammation and infiltrated immune cells in the epidermis and dermis of TH treated NC/Nga mice were highly reduced compared to the untreated control group.
Botulinum neurotoxin A (BoNT/A) has been used therapeutically to treat muscular hypercontractions and sudomotor hyperactivity and it has been reported that BoNT/A might have analgesic properties in headache. PEP-1 peptide is a known carrier peptide that delivers fulll-ength native proteins in vitro and in vivo. In this study, a BoNT/A gene were fused with PEP-1 peptide in a bacterial expression vector to produce a genetic in-frame PEP-1-BoNT/A fusion protein. The expressed and purified PEP-1-BoNT/A fusion proteins were efficiently transduced into cells in a time- and dose-dependent manner when added exogenously in a culture medium. In addition, immuno-histochemical analysis revealed that PEP-1-BoNT/A fusion protein efficiently penetrated into the epidermis as well as the dermis of the subcutaneous layer, when sprayed on mice skin. These results suggest that PEP-1-BoNT/A fusion protein provide an efficient strategy for therapeutic delivery in various human diseases related to this protein.
In order to improve efficacy of oriental medicines and to study the application of fermented oriental medicine in clinicals, the efficacy of CYT and CYTBH on atopic dermatitis were evaluated. The results and conclusions are as follows. CYT and CYTBH significantly improved the atopic dermatitis symptoms in NC/Nga mice by naked eye evaluation and significantly decreased clinical index in both groups. CYT and CYTBH both decreased the cell numbers of CD3+, CD11b+Gr-1+ cells in dorsal skin. Of the cells, CYT significantly decreased CD11b+Gr-1+ cells whereas CYTBH significantly decreased all immune cells. CYT and CYTBH both decreased the production rate of IL-4 and IFN-${\gamma}$ activated by CD3/CD28. In the case of CYTBH, significant decrease in all cases was observed. CYT and CYTBH decreased the production rate of IL-5, IL-13 and IL-17 in serum. Significant decrease of IL-5 in the case of CYT and IL-5 and IL-13 in the case of CYTBH were observed. CYT and CYTBH significantly decreased transcription of IL-5 mRNA and IL-13 mRNA in skin. Significant decrease in IgG1 and IgE immunoglobulins in serum were oberved in both groups. Significant decrease was only observed in the case of CYTBH. Both CYT and CYTBH significantly decreased the secretion of histamine. Both CYT and CYTBH suppressed erythema, hemorrhage, edema, excoriation, erosion of skin tissues of NC/Nga mice resulting in the decrease of thickness of epidermis. Significant decrease of infiltration of obese cells was also observed. The results above indicated that both CYT and CYTBH had significant efficacy in the treatment of atopic dermatitis through immune modulation. Animal studies showed that CYTBH had superior activity than that of CYT suggesting further and continuous studies on the changes in ingredients or absorption improvement by fermentation should follow.

ID의 비밀번호 관리소홀, 부정사용에 의하여 발생하는 모든 결과에 대한 책임은 회원에게 있습니다.

제 4 장 서비스의 이용
제 12 조 (서비스 이용 시간)
① 서비스 이용은 당 사이트의 업무상 또는 기술상 특별한 지장이 없는 한 연중무휴, 1일 24시간 운영을
원칙으로 합니다. 단, 당 사이트는 시스템 정기점검, 증설 및 교체를 위해 당 사이트가 정한 날이나 시간에
서비스를 일시 중단할 수 있으며, 예정되어 있는 작업으로 인한 서비스 일시중단은 당 사이트 홈페이지를
통해 사전에 공지합니다.
② 당 사이트는 서비스를 특정범위로 분할하여 각 범위별로 이용가능시간을 별도로 지정할 수 있습니다. 다만
이 경우 그 내용을 공지합니다.
제 13 조 (홈페이지 저작권)
① NDSL에서 제공하는 모든 저작물의 저작권은 원저작자에게 있으며, KISTI는 복제/배포/전송권을 확보하고
있습니다.
② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
