미숙사과 용매 추출물의 항산화성 (Antioxidant Properties of Unripened Apple Extracts)

  • The purpose of this study was to investigate the antioxidant properties of unripened apple extracts. The amount of total flavonoids in the peel and 5/30 samples were 2.7times and 5.0 times higher than the flesh and 6/30 samples, respectively. The degree of angintensin I converting enzyme (ACE) inhibition activity in hot water extracts was higher than the other extracts. In the 5/30 sample, a $0.1\%$ concentration of unripened apple powder showed almost the same electron-donating ability with a $0.5\%$ concentration of the 6/30 sample. More than $90\%$ of the electron-donating ability was observed from the peel extracts regardless of solvents used for extraction. The nitrite-scavenging effects of acetone and methanol extracts of the 5/30 sample were the strongest at pH 1.2, and especially the $80\%$ methanol extracts exhibited a powerful scavenging effect of more than $90\%$ at pH 3.0.

Isolation and Purification of Polyhydroxylated Alkaloids from Silkworm (Bombyx mori L.)

  • Several polyhydroxylated alkaloids were isolated from the extracts of freeze-dried silkworm powder, and purified by ion exchange chromatographic analysis. Through the HPLC analysis, we could identify 1-deoxynojirimycin (DNJ) and a kind of calystegin $B_2$ (HS-58) as well as a noble compound (HS-74) from the purified polyhydroxylated alkaloids. In order to know the characteristics of these isolated alkaloids as enzyme inhibitors, glycosidase inhibition activities of these identified alkaloids including other two non-purified alkaloids (SWP 3-1 and SWP 3-2) were investigated.

  • Maximum cellulase production was sought by comparing the activities of the cellulases produced by different Trichoderma reesei strains and Aspergillus niger. Trichoderma reesei Rut-C30 showed higher cellulase activity than other Trichoderma reesei stains and Aspergillus niger that was isolated from soil. By optimizing the cultivation conditions during shake flask culture, higher cellulase production could be achieved. The FP(filter paper) activity of 3.7U/ml and CMCase (Carboxymethylcellulase) activity of 60U/ml were obtained from shake flask culture. When it was grown in 2.5L fermentor, where pH and DO levels are controlled, the enzyme activities were 133.35U/ml (CMCase) and 11.67U/ml(FP), respectively. Ammonium sulfate precipitation method was used to recover enzymes from fermentation broth. The dried cellulase powder showed 3074.9U/g of CMCase activity and 166.7U/g of FP activity with 83.5% CMCase recovery.

Biological Activities and Stability of a Standardized Pentacyclic Triterpene Enriched Centella asiatica Extract

  • Pentacyclic triterpenes, mainly, asiatic acid, madecassic acid, asiaticoside, and madecassoside are the active constituents of Centella asiatica. A pentacyclic triterpene enriched C. asiatica extract (PRE) was prepared and standardized to contain a total pentacyclic triterpenes not less than 65% w/w. This work was focused on determination of antiinflammatory, antioxidant, and tyrosinase inhibitory activities of PRE and its stability. The PRE exhibited a satisfactory nitric oxide inhibitory effect, with an $IC_{50}$ value of $64.6{\mu}g/mL$. In addition, the PRE inhibited tyrosinase enzyme activity with an $IC_{50}$ value of $104.8{\mu}g/mL$. In contrast, the PRE possessed only weak antioxidant activity. The PRE was stable over a period of four months when stored as a dried powder but only in a well-closed container protected from light at $4^{\circ}C$. An aqueous alcoholic solution of the PRE was stable at pH values of 5.8 and 7.0, but was not stable at a pH of 8.2. Preparations of the PRE in an aqueous solution should be performed in acidic or neutral conditions.

Effect of Chinese Yam on Benzo[a]pyrene Hydroxylase Activities in Rats Fed Dietary Benzo[a]pyrene

  • To investigate the effect of yam on the activity of benzo[a]pyrene hydroxylase(BPH), the key enzyme associated with polycyclic aromatic hydrocarbons(PAHs) metabolism, rats were fed a fiber free diet for 7 days, whereupon they were switched to experimental diets for another 7 days. Diets contained benzo[a] pyrene(BP, 400 mg/kg diet) and 25% or 50% yam powder (freezer dried and hot air dried ). Diets containing pectin and cellulose were compared with diets containing yam. BPH activities were assessed in the liver, lung, kidney, stomach, small intestine and large intestine of rats. BP induced BPH activities in various tissues ; 8 fold in liver, 28 in lung and stomach , and 32 in large intestine. The addition of yam significantly lowered BPH activity in liver, lung and stomach and hot air dried yam was nmor eeffectivie than freeze dried yams. These data suggested that yam containing diet may influence carcinogen metabolism in liver and extrahepatic target tissues by altering activities of BPH and may reduce exposure of these tissues to dietary carcinogens.

Enzymatic preparation and antioxidant activities of protein hydrolysates from hemp (Cannabis sativa L.) seeds

  • Hemp (Cannabis sativa L.) seeds have recently been attracting attention as a new high-value-added food material owing to their excellent nutritional properties, and research on the development of functional food materials using hemp seeds is actively progressing. This study aimed to evaluate the antioxidant properties of hemp seed protein hydrolysates. Protein hydrolysates were prepared from defatted hemp seed powder (HS) by enzymatic hydrolysis using five different proteases (alcalase, bromelain, flavourzyme, neutrase, and papain). 2,4,6-trinitrobenzene sulfonic acid (TNBS) assay and SDS-PAGE analysis revealed that HS showed a high degree of hydrolysis after treatment with each enzyme except papain. The total polyphenol content of the protein hydrolysates (<3 kDa) and the RC50 values obtained from two different antioxidant tests showed that alcalase hydrolysate (HSA) had a relatively high level of antioxidant capacity. In addition, treatment with HSA (25-100 ㎍/mL) significantly inhibited linoleic acid peroxidation. These results suggest that hemp seed protein hydrolysates are potential sources of natural antioxidants. Future studies will focus on the identification of active peptides from HSA.

Protaetia brevitarsis larvae extract protects against lipopolysaccharides-induced ferroptosis and inflammation by inhibiting acid sphingomyelinase

  • BACKGROUND/OBJECTIVES: Inflammation and ferroptosis are implicated in various diseases and lipopolysaccharides (LPS) have been linked with these disorders. Recently, many edible insects, such as Gryllus bimaculatus, Protaetia brevitarsis larvae (PB) and Tenebrio molitor larvae, have been recommended as alternative foods because they contain lots of nutritional sources. In this study, we explored the potential of PB extract in preventing LPS-induced inflammation and ferroptosis in Hep3B cells. MATERIALS/METHODS: PB powder was extracted using 70% ethanol and applied to Hep3B cells. Co-treatment with LPS was conducted to induce ferroptosis and inflammation. The anti-inflammatory and anti-ferroptosis mechanisms of the PB extract were confirmed using Western blot, enzyme-linked immunosorbent assay, and real-time polymerase chain reaction analysis. RESULTS: PB extract effectively prevented LPS-induced cell death and restored LPS-induced inflammatory cytokine production, NF-κB signaling, endoplasmic reticulum (ER) stress and ferroptosis. Interestingly, PB extract reduced LPS-induced ceramide increase and acid sphingomyelinase (ASMase) expression. The use of the ASMase inhibitor, desipramine, also demonstrated a reduction in these pathways, highlighting the pivotal role of ASMase in inflammation and ferroptosis. Treatment with each inhibitor revealed that ferroptosis causes ER stress and that NF-κB and MAP kinase pathways are involved in inflammation. CONCLUSION: PB emerges as a potential functional food with inhibitory effects on LPS-induced inflammation and ferroptosis, making it a promising candidate for nutritional interventions.

탁주양조중 Riboflavin의 소장에 관한 연구 (Studies on the Quantitative Changes of Riboflavin during Takju Brewing)

  • 탁주 양조에 사용되는 각종 원료중의 Riboflavin을 정량하고 아울러 쌀, 소맥분및 옥수수분을 각각 주원료로 일반 탁주양조법에 따라 담금하여 숙성시까지 그들 술덧중의 Riboflavin의 소장을 측정하여 다음과 같은 결과를 얻었다. 1. 각원료 100g중의 Riboflavin함량 (${\gamma}$)은 쌀 46.9, 소맥분 67.3, 옥수수분 73.9, 누룩 169.0, 소맥분입곡 87.1, 옥수수분입곡 $84.4{\gamma}$이었으며 쌀소맥분 및 옥수수분중의 함량은 증자에 의하여 약 $30{\sim}40%$파괴됨을 보였다. 2. 누룩단용법의 쌀및 소맥분구의 각술덧에서 Riboflavin의 소장은 같은 경향을 보여 3일까지 감소하였다가 이후 다시 증가하며 5일째에 가서 급격히 증가하여 6일째에 약간 감소하는 소장을 보여 결국 양 숙성술덧 100ml중의 Riboflavin함량은 누룩단용법의 쌀구에서 $82{\gamma}$, 소맥분구에서 $72{\gamma}$를 보였으며 누룩및 입곡혼용법의 소맥분및 옥수수분구의 각술및에서는 초기부터 감소하지 않고 기복이 있는 증가의 경향을 보이다가 역시 5일째에 현저히 증가되며 6일째에 다시 감소하여 결국 양 숙성술덧 100ml중에는 소맥분구에서 $88.6{\gamma}$ 옥수수분구에서 $83.6{\gamma}$이었다. 3. 각숙성술덧으로 제성한 탁주 100ml중에는 Riboflavin 이 누룩단용법구들에서 $28.8{\sim}41{\gamma}$, 누룩및 입곡혼용법구들에서 $33.6{\sim}44.5{\gamma}$ .함유됨을 알 수 있었다. 5. 누룩단용법의 쌀구및 소맥분구의 지게미 무수물 100g중에는 Riboflavin이 각각 $47.3{\gamma}$$66.2{\gamma}$함유되었으며 누룩및 입곡혼용법의 소맥분구및 옥수수분구의 것에는 각각 $64.5{\gamma}$$62.9{\gamma}$ 함유되었다.

탁주 양조중 Thiamin의 소장에 관한 연구 (Studies on the quantitative changes of thiamin during Takju brewing)

  • 탁주 양조에 사용되는 각종원료중의 Thiamin과 쌀, 소맥분 및 옥수수분을 주원료로 일반 탁주양조법에 따라 담금하여 숙성시까지 술덧중의 Thiamin의 소장을 측정하여 다음과 같은 결과를 얻었다. 1. 각원료 100g중의 Thiamin 함량 $({\gamma})$은 쌀 107.8 소맥 분 185.0 옥수수분 410.2 누룩 347.4 소맥분입곡 170.1 옥수수분입곡 257.,3 이었으며 쌀, 소맥분 및 옥수수분중의 함량은 증자에 의하여 $40{\sim}50%$ 파괴됨을 보였다. 2. 쌀, 소맥분 및 옥수수분을 주원료로 하여 각각 담금한 술덧에서 Thiamin의 소장은 각 술덧에서 같은 경향을 보여 누룩단용법의 양구에서 담금 후 1, 2일 사이에 급격히 감소하며 이후 숙성시까지 큰 변동이 없었고 입곡혼용법의 양구에서는 담금초기에 누룩단용법구들에 비하여 감소율이 훨씬 적었다. 또한 각 숙성 술덧 100ml중의 함량은 누룩단용법의 쌀구에서 약 6${\gamma}$, 소맥분구에서 7.5${\gamma}$였고 입곡혼용법의 소맥분구에서 약 $12,4{\gamma}$, 옥수수분구에 서 15.4 ${\gamma}$ 였다. 3. 각숙성 술덧으로 제성한 탁주 100ml중에는 Thiamin이 누룩단용구들에서 $2.4{\sim}3.5{\gamma}$, 입곡혼용법구들에서 약 $5{\sim}7{\gamma}$ 함유됨을 알 수 있었다. 4. 각숙성 술덧의 지게미 100g중에는 Thiamin이 누룩단용법의 쌀구에서 약 $43.7{\gamma}$, 소맥분구에서 $56.1{\gamma}$ 그리고 입곡혼용법의 소맥분구에서 약 $57.5{\gamma}$. 옥수수분구에서 $81.4{\gamma}$ 함유되었다.

상황버섯 자실체 메탄올 추출물과 분말의 지방소화효소 억제 및 항산화 활성 (Lipase-Inhibitory and Anti-Oxidative Activity of the Methanol Extract and the Powder of Phellinus linteus)

  • 상황버섯의 지방소화효소 억제와 항산화 활성을 구명하기 위해 상황버섯 메탄올 추출물의 췌장 lipase 억제 활성과 항산화 활성을 in vitro에서 측정하고, 흰쥐에 고지방식이와 함께 상황버섯 분말을 투여하여 지방소화효소 활성 저해를 통한 지방 흡수 억제와 항산화 효과를 in vivo에서 관찰하였다. 메탄올 추출물의 췌장 지방소화효소 활성 저해능과 DPPH 라디칼 소거능을 측정한 결과, $IC_{50}$가 각각 $36.3\;{\mu}g/mL$$20.1\;{\mu}g/mL$로 나타났으며, DPPH 소거능은 비타민 C의 $IC_{50}$$18.3\;{\mu}g/mL$와 비슷하였다. 상황버섯 분말을 고지방식이에 각각 2%와 5% 첨가한 식이를 공급하고 8주간 사육한 결과, 상황버섯 첨가로 인해 식이섭취량은 증가하였으나 체중과 체지방량에는 차이가 없었으며 따라서 식이효율은 낮았지만 유의적이지는 않았다. 그러나 상황버섯 섭취로 인해 지질의 흡수가 감소되어 분변으로의 지질 배설량이 증가한 것으로 조사되었다. 혈장 지질의 경우, 상황버섯 첨가군에서 혈장 총 콜레스테롤과 LDL 콜레스테롤, 간의 총 지질과 총 콜레스테롤 함량이 대조군에 비해 낮아진 것으로 조사되었다. 혈장 GPT 활성은 상황버섯 2%와 5% 첨가군에서 각각 대조군에 비해 43%와 45% 감소하였다. 혈장, 적혈구 및 간의 항산화 효소 활성을 측정한 결과, 상황버섯의 첨가로 T-SOD 활성은 영향을 받지 않았으나, GSH-Px는 간과 혈장에서 활성이 증가하였다. 실험식이를 공급하고 2주, 4주 및 8주에 림프구를 분리하여 DNA 손상 정도를 측정한 결과, 상황버섯을 2주간 섭취한 후의 림프구 DNA 손상이 대조군에 비해 큰 것으로 조사되었으나, 4주간 섭취한 군에서는 차이가 없었고, 8주에서는 DNA 손상이 억제되었으며 2% 첨가군에서 DNA 손상 억제 정도가 가장 크게 나타났다. 림프구에 $H_2O_2$로 산화스트레스를 가하고 DNA 손상 정도를 측정한 결과, 상황버섯 첨가군의 DNA 손상이 대조군에 비해 식이섭취기간에 비례해서 낮아졌고, 5% 첨가보다는 2% 첨가군에서 억제 효과가 더 높은 것으로 조사되었다. 이상의 결과들로부터, 상황버섯은 생체 내 항산화 효소계의 활성유도와 산화스트레스로 인해 생성된 라디칼의 활발한 소거를 통해 세포의 산화적 손상을 억제하는 효과가 있는 것으로 생각된다.
