Bacillus licheniformis 단백질 분해 효소에 의한 정연 견사의 특성 I. 정연 견사의 이화학적 특성 (Physicochemical Characteristics of Silk Fibroin Degummed by Protease in Bacillus licheniformis I. Physicochemical Characteristics of Degummed Silk Fiber)

  • 견사의 정연 방법으로는 알카리 용액에서 열을 이용하는 방법과 단백질 분해 효소를 이용하는 방법으로 크게 나눌 수 있는데 본 실험은 Bacillus licheniformis 단백질 분해 효소로 정연한 견사의 이화학적 특성과 열 처리 영향 및 효소 정연 견 피브로인 수용액의 특성을 비누 정연 견사와 비교하여 실험한 결과 다음과 같은 결과를 얻었다. 1. 정연 견사의 표면을 주사 전자현미경으로 관찰한 결과 정연 방법에 따른 뚜렷한 차이가 없었으며 정연 견사의 아미노산 조성을 분석한 결과에서도 정연 방법에 따른 큰 차이가 없었다. 2. 효소 정연 견사는 비누 정연 견사에 비하여 강력과 신도가 약간 저하되었고 점도도 비누 정연 견사의 0.59에 비하여 효소 정연 견사는 0.56으로서 저하된 반면 ($\alpha$$\varepsilon$) amino group content는 비누 정연견사의 49.5$\mu$mo1/g보다 효소 정연 견사는 53.6 $\mu$mol/g로서 증가되었으므로 효소 정연 견사는 비누 정연 견사보다 degradation이 더 일어난 것으로 추측된다. 3. 효소 정연 견사는 비누 정연 견사에 비하여 복굴절도가 높았으며 DSC thermogram에서 열 분해 온도도 약 1$0^{\circ}C$ 높았다. 또 효소 정연 견사의 IR 및 X-ray crystallinity도 각각 0.70과 0.53으로서 비누정연 견사의 0.67과 0.50에 비하여 약간 높았으므로 효소 정연 견사의 경우 결정성 및 배향성이 비누 정연 견사보다 높은 것으로 판단된다.

  • The pure-separation of calcium chloride-treated fibroin hydrolysates could be carried out using gel filtration chromatography. Also, the effect of its enzymatic hydrolysis was investigated in order to find out the enhancement of their functionality. The average molecular weight(Mw), solubility and free amino acid compositions of three hydrolysates samples (calcium chloride, calcium chloride-flavourzyme and calcium chloride-thermoase)were measured to compare their characteristics. The molecular weight of calcium chloride hydrolysate was about Mw 46,800 and it can be reduced to Mw 12,500 and 1,070 upon the enzymatic hydrolysis by flavourzyme and thermoase, repectively. A solubility of calcium chloride-treated samples shows about 60% while calcium chloride/enzyme-treated samples are perfectly soluble (100% solubility). The total amino acid composition of calcium chloride enzymatic hydrolysates are much higher than that of calcium chloride hydrolysate.

  • Cellulolytic enzymes were prepared from alkaline resistant microorganisms which were newly screened from calcic soil. Characteristics of enzymes and enzymatic deinking efficiency of wastepaper were investigated. The results were summarized as fellows: 1. The recovery rate of crude enzyme was 93.7% in Bio-B and 57.4% in Bio-F. 2. The protein content in crude enzymes was lowest and the thermal stability of crude enzymes was highest in Bio-F. 3. The brightness gain of Bio-F deinked pulp was best in ONP and Bio-B deinked pulp was best in MOW. 4. The reject yield was increased with enzymatic deinking flotation process. 5. The residual ink area of paper was increased with enzymatic deinking and large size of ink particles were remained in paper.

  • 글루코스 생산을 위한 효과적인 방법을 개발하기 위해서 cellulase에 의한 현사시 나무의 효소가수분해를 실시하였다. 목재의 효소가수분해는 미생물이 생산한 효소를 사용하여 글루코스를 생산하는 반응이다. 이렇게 얻어진 글루코스는 발효에 의해 쉽게 에탄올로 변환시킬 수 있다. 에탄올은 아세톤이나 부탄올, 시트릭산 그리고 락틴산을 제조하는 원료물질이나 석유자원을 대체할 수 있는 대체에너지 자원이다. 셀룰로오스의 효소가수분해 기작은 endo-cellulase, exo-cellulase 그리고 β-D-glucosidase라는 세 개의 서로 다른 형태의 효소가 연속적인 반웅에 의해 일어난다고 설명되어지고 있다. 본 실험의 목적은 다양한 농도의 수산화나트륨으로 현사시나무를 전처리 하여 이러한 전처리가 셀룰로오스의 결정화도와 리그닌함량에 미치는 영향과 가수분해율과의 관계를 조사하였다.

섬유소계 바이오매스의 분별을 위한 다양한 알칼리 전처리 특성 (The Characteristics of Alkaline Pretreatment Methods of Cellulosic Biomass)

  • 이 연구는 목질계 바이오매스에 대한 알칼리 용액의 침지와 침출 전처리의 효능을 비교한다. 볏짚과 보리짚과 같은 다양한 바이오매스는 수산화나트륨 용액, 수산화칼륨 용액, 암모니아수 그리고 탄산나트륨 용액에 의해 침지 공정으로 수행되었다. 암모니아수에 의해 전처리된 볏짚과 보리짚의 효소 소화율은 80% 이상으로 나타났다. 전처리된 유칼립투스 부산물, 낙엽송 그리고 리기다 소나무의 효소 소화율은 상대적으로 낮은 범위로 나왔다. 하지만 전처리된 유칼립투스 부산물은 초기 바이오매스에 비해 효소 소화율이 약 5배 증가되었다. 또한 침출 공정으로 전처리된 유칼립투스 부산물의 효소 소화율은 약 12배가 증가되는 것을 확인하였다.

  • Effects of aqueous ammonia soaking treatments to yellow poplar (Liriodendron tulipifera L.) were investigated to focus on chemical compositional changes and enzymatic hydrolysis characteristics changes by this treatment. Treatment temperature and time were main variables. At 3 different levels of aqueous ammonia soaking temperature and time ($145^{\circ}C$ -1 h, $90^{\circ}C$ -16 h and $45^{\circ}C$ - 6 days), lower temperature and longer soaking time led to more xylan removal based on carbohydrate compositional analysis. However, at higher temperature treatment led to more enzymatic saccharification of cellulose to glucose by commercial cellulose mixtures (Celluclast 1.5L and Novozym 342 from Novozyme, Denmark). Cellulose hydrolysis was gradually increased with increasing enzymatic hydrolysis time but xylan hydrolysis was leveled out at early stage (less than 10 h) of enzymatic hydrolysis.

Improved Functional Characteristics of Whey Protein Hydrolysates in Food Industry

  • This review focuses on the enhanced functional characteristics of enzymatic hydrolysates of whey proteins (WPHs) in food applications compared to intact whey proteins (WPs). WPs are applied in foods as whey protein concentrates (WPCs), whey protein isolates (WPIs), and WPHs. WPs are byproducts of cheese production, used in a wide range of food applications due to their nutritional validity, functional activities, and cost effectiveness. Enzymatic hydrolysis yields improved functional and nutritional benefits in contrast to heat denaturation or native applications. WPHs improve solubility over a wide range of pH, create viscosity through water binding, and promote cohesion, adhesion, and elasticity. WPHs form stronger but more flexible edible films than WPC or WPI. WPHs enhance emulsification, bind fat, and facilitate whipping, compared to intact WPs. Extensive hydrolyzed WPHs with proper heat applications are the best emulsifiers and addition of polysaccharides improves the emulsification ability of WPHs. Also, WPHs improve the sensorial properties like color, flavor, and texture but impart a bitter taste in case where extensive hydrolysis (degree of hydrolysis greater than 8%). It is important to consider the type of enzyme, hydrolysis conditions, and WPHs production method based on the nature of food application.

Preparation of Diacylglycerol from Lard by Enzymatic Glycerolysis and Its Compositional Characteristics

  • The aim of this study was to prepare diacylglycerol (DAG) by enzymatic glycerolysis of lard. The effects of reaction parameters such as lipase type, reaction temperature, enzyme amount, substrate molar ratio (lard/glycerol), reaction time, and magnetic stirring speed were investigated. Lipozyme RMIM was found to be a more active biocatalyst than Novozym 435, and the optimal reaction conditions were 14:100 (W/W) of enzyme to lard substrate ratio, 1:1 of lard to glycerol molar ratio, and 500 rpm magnetic stirring speed. The reaction mixture was first incubated at $65^{\circ}C$ for 2 h and then transferred to $45^{\circ}C$ for 8 h. At the optimum reaction conditions, the conversion rate of triacylglycerol (TAG) and the content of DAG in the reaction mixture reached 76.26% and 61.76%, respectively, and the DAG content in purified glycerolized lard was 82.03% by molecular distillation. The distribution of fatty acids and Fourier transform infrared spectra in glycerolized lard samples were similar to those in lard samples. The results revealed that enzymatic glycerolysis and molecular distillation can be used to prepare more highly purified DAG from lard.

  • An enzymatic process was developed to produce protein hydrolysater form defatted soya protein. Various unit operations were tried, and the effects of pre- and post-treatments on the product characteristics such as degree of hydroylsis (DH), free amino acid content (%FAA) and average molecular weight (MW) were investigated. The use of acid washes showed no difference in %DH. Increasing pH during pre-cooking gave lower %DH. Alkaline cooking made too much insoluble protein, thus the protein yield was too small. A better hydrolysis with more acceptable taste was obtained when the combination of Neutrase/Alcalase/Flavourzyme was used in place of Alcalase/Flavourzyme combination; Untoasted defatted soya was more effective on the proteolysis than toasted one. The MW of the evaporated and spray dried product was higher than that of undried product, due to precipitation of low-solubility components. When ultrafiltration and the product concentration carried out the product separation by reverse osmosis, the solubility and the taste of the product were improved. The difference between enzyme hydrolysate and acid hydrolysate was significant in free amino acid composition, especially in tyrosine, phenylalanine, glutamine and asparagine.

Galactose epimerase결손 Salmonella pullorum 변이주의 효소활성 (Characterization of enzymatic activity of galactose epimerase-less mutant of Salmonella pullorum)

  • Uridine diphosphate(UDP)-galactose-4-epimerase-less mutants of Salmonella pullorum were isolated after mutagenic treatment with ethidium bromide. When isolated gal E mutants of S. pullorum A2 and D1 were grown in the presence of galactose(0.1 W/V), they exhibited marked bacteriolysis in heart infusion broth. The mutant strains were further investigated the characteristics of enzymatic activities in the Leoloir galactose pathway. Isolated A2 and D1 strains were completely deficient in UDP-galactose-4-epimerase activity. And the activity of other enzymes involved in galactose metabolism were reduced significantly.
