주거환경 및 개인 생활습관에 따른 화학물질 노출수준 차이 - 국민환경보건기초조사 (Differences of Chemical Exposure Levels according to Residential and Personal Life-style Characteristics of Korean adult population - from Korean National Environmental Health Survey)

  • Objectives: The aim of this study was to determine environmental chemical exposure related to residential and personal lifestyle characteristics in the adult Korean population. The observations of this study can provide information useful for developing reduction approaches for exposure to chemicals among the general adult population. Methods: The second stage of the Korean National Environmental Health Survey (KoNHES) was conducted from 2012 to 2014, with 6,478 persons participating. Using the results of the survey, the relationship between exposure levels of heavy metals and organic chemicals and exposure factors, e.g. residential and personal lifestyle characteristics, were analyzed. Results: The exposure levels of VOCs and PAHs were significantly lower in participants living at a distance of more than 100 m from roads versus living closer to roads. Home ventilation lowered VOC and PAH exposure but did not lower chemical exposure from household products. Use of public transportation showed lower exposure to heavy metals, VOCs, and PAHs. Current smoker was significantly higher for levels of heavy metals, VOCs, and PAHs, and the exposure trend was similar for current drinkers. Physical activity was related with higher exposure to phthalates and environmental phenols. Conclusion: Our observations based on a nationally representative population for Korea show that exposure to chemicals varies by residential and personal lifestyle, and this should be considered for developing appropriate mitigation measures and policies. Given the health concerns surrounding environmental chemicals, it is necessary to develop comprehensive measures to reduce chemical exposure.

OECD 대량생산화학물질 위해성평가 및 대책 (Risk Assessment in OECD High Production Volume Chemicals Program and its Countermeasure)

  • The risk assessment is the qualitative or quantitative evaluation of the risk posed to human health and the environment by the actual or potential presence or release of hazardous substances, pollutants or contaminants. The environmental impact assessment (EIA) is assessed by the environmental criteria, and risk assessment is assessed by the risk rate. Risk rate based on dose-response values may not be easy to apply on regulatory basis like EIA for uncertainty. Internationally there is an example of OECD program. Risk assessment of High Production Volume (HPV) Chemicals has started since the OECD Program with the 1990 Council Act on the Co-operative Investigation and Risk Reduction of Existing Chemicals. These HPV chemicals include all chemicals produced or imported at levels greater than 1,000 tonnes per year in at least one Member country or in the European Union region. The SIDS called the Screening Information Data Set is regarded as the minimum information needed to assess an HPV chemical to determine whether any further work should be carried out or not. All the data elements of SIDS including assessment for environment and health are prepared as three formats of the full SIDS Dossier, the SIDS Initial Assessment Report (SIAR), and the SIDS Initial Assessment Profile (SIAP) of an HPV chemical. In 1998 the global chemical industry through the International Council of Chemical Associations (ICCA) has joined to work with OECD. The OECD has assessed approximately 1,000 chemicals from 1991 through 2004 with ICCA. Till the February of 2005, 592 chemicals of those chemicals completed SIDS reports. Member countries have been targeted the goal of 1,000 new chemicals from 2005 to 2010 and Korea shared 36 chemicals from the 1,000 new chemicals. Currently Korea has completed SIDS reports of 7 chemicals among sponsored 24 chemicals. In conclusion SIDS project will be linked to national program for outputs application with more reliable production. Both the OECD and industry will carry out their commitment to complete assessments for more and the remaining chemicals assessment. The major outputs will contribute to cope with international chemical management.

  • Objectives: Human biomonitoring (HBM) is a measurement of the chemicals and their metabolites in human biological samples and has been successfully employed to determine the exposure levels of environmental chemicals. In this study, we analyzed seasonal variations of the blood or urinary levels of chemicals, and assessed that these differences could affect the results of association study. Methods: The Korea National Environmental Health Survey (KoNEHS) is a nationwide survey that analyzes exposure levels of environmental pollutants, 19 kinds of chemicals including heavy metals and organic chemicals, and the exposure factors in the general population. Based on KoNEHS data, we analyzed the levels of chemicals concentrations over the total survey period (2012-2014) and each season, and assessed the association of thyroid measures with phthalate metabolite and BPA. Results: Exposure levels of blood mercury and lead were lower in summer compare to winter. Bisphenol A and PAHs metabolites were higher in spring and summer, but lower in autumn. VOCs metabolites were generally lower in summer and autumn. Phthalate metabolites were higher in all other seasons than in winter. Pyrethroid metabolite, 3-PBA, was higher in summer and autumn. Regarding seasonal variation of chemical exposures, the statistical significance and size of effects between thyroid measures and phthalate and BPA were changed with season. Conclusion: Seasonal variations of chemical exposure and health outcome should be considered for interpreting biomonitoring results from a public health context.

The Expression Patterns of Estrogen-responsive Genes by Bisphenol A in the Wild Medaka (Oryzias sinensis)

  • Gene expression levels of choriogenin, vitellogenin and estrogen receptor were determined using Reverse transcription (RT)-PCR technique after exposure to estrogenic chemical bisphenol A in the Korean wild medaka (Oryzias sinensis). These genes have been known to be induced in male test fish when the fish are exposed to estrogenic chemicals. Therefore they can be suggested as a possible biomarker of endocrine disruption in fish, however, relatively little has been known about these genes expression by estrogenic chemicals in Korean wild fish. Mature male Oryzias sinensis were treated with bisphenol A at nominal concentrations of 0.02, 0.2 and 2 mg/L for 6 days and total RNA was extracted from the livers of treated fish for RT-PCR. When the five biomarker genes were amplified by RT-PCR in the same condition, mRNA induction level of each gene was elevated with different sensitivities. Conclusively, the results of this work indicated that measurement of vitellogenin and choriogenin using RT-PCR is effective as a simple tool for the screening of estrogenic chemicals and suggested that O. sinensis would be a suitable model fish for the environmental risk assessment of potential endocrine disruptors.

사고대비물질 개인보호구 선정에 관한 연구(1): 물질유해성 및 작업위해성 분석 (A Study on Selecting Personal Protective Equipment for Listed Hazardous Chemicals (1): Analysis of Hazard Ranks and Workplace Exposure Risks)

  • Objectives: According to the new Chemical Control Act from the Korean Ministry of Environment (2014-259), workers handling hazardous chemicals should wear personal protective equipment (PPE). However the act simply states in basic phrases that every worker handling one or more of the 69 listed chemicals should wear PPE and does not consider the unique hazard characteristics of chemicals and work types. The main purpose of this study is to provide basic data to revise the act to suit particular work processes and situations. Methods: The hazard rank of the substances was classified based on hazardous characteristics such as LC50 and vapor pressure using matrix analysis. The workplace exposure risk of the substances was also determined through a matrix analysis based on the previously determined hazard ranks and the demands of manual handling together with the likelihood of accident frequency of the operation combined with the exposure of workers during spill accidents. Results: To meet the demands for developing subsequent guidelines for the risk-based application of PPE in hazardous workplaces, this study sorted the 69 listed chemicals into five hazardous categories based on their LC50 and vapor pressures, and also assigned exposure categories according to exposure vulnerability for various types of work which are frequently performed throughout the life cycle of the chemicals. Conclusion: In the next study, an exposure risk matrix will be produced using the hazard rank of chemicals and workplace exposure risk, and then PPE will be selected to suit the categories of the exposure risk matrix.

반도체 산업의 웨이퍼 가공 공정 유해인자 고찰과 활용 - 화학물질과 방사선 노출을 중심으로 - (Review of Hazardous Agent Level in Wafer Fabrication Operation Focusing on Exposure to Chemicals and Radiation)

  • Objectives: The aim of this study is to review the results of exposure to chemicals and to extremely low frequency(ELF) magnetic fields generated in wafer fabrication operations in the semiconductor industry. Methods: Exposure assessment studies of silicon wafer fab operations in the semiconductor industry were collected through an extensive literature review of articles reported until the end of 2015. The key words used in the literature search were "semiconductor industry", "wafer fab", "silicon wafer", and "clean room," both singly and in combination. Literature reporting on airborne chemicals and extremely low frequency(ELF) magnetic fields were collected and reviewed. Results and Conclusions: Major airborne hazardous agents assessed were several organic solvents and ethylene glycol ethers from Photolithography, arsenic from ion implantation and extremely low frequency magnetic fields from the overall fabrication processes. Most exposures to chemicals reported were found to be far below permissible exposure limits(PEL) (10% < PEL). Most of these results were from operators who handled processes in a well-controlled environment. In conclusion, we found a lack of results on exposure to hazardous agents, including chemicals and radiation, which are insufficient for use in the estimation of past exposure. The results we reviewed should be applied with great caution to associate chronic health effects.

음용수 중 유해 화학 물질에 대한 위해성 평가에 관한 연구 - II. 비발암성 화학 물질을 중심으로 - (Study on Health Risk Assessment of Non-carcinogenic Chemicals in Drinking Water)

  • The purpose of this research is to estimate a safe environmental level of human exposure to thresholding-acting toxicants in drinking water and recommend the acceptable levels and management plans for maintaining good quality of drinking water' and protecting health hazard. This research has been funded as a national project for three years from 1992 to 1995. This study(the second year, 1993-1994) was conducted to monitor 39 species of noncarcinogenic chemicals such as volatile organic compounds(VOCs), polynuclear aromatic hydrocarbens(PAHs), pesticides and heavy metals of drinking water at some area in six cities of Korea, and evaluate health risk due to these chemicals through four main steps (hazard identification, exposure assessment, dose-response assessment and risk characterization) of risk assessment in drinking water. In hazard identification, 39 species of non-carcinogenic chemicals were identified by the US EPA classification system. In the step of exposure assessment, sampling of tap water from the public water supply system had been conducted from 1993 to 1994, and 39 chemicals were analyzed. Inclose-response assessment for non-carcinogens, reference doses(RfD) and lifetime health advisories(HAs) of lifetime acceptable levels were calculated. In risk characterization of detected chemicals, the hazard quotients of noncarcinogens were less than one except those of manganese and iron in D city.

산업안전보건법 상 관리수준 검토를 위한 화학물질 유해성.위험성 평가대상 후보물질 선정에 관한 연구 (A Study on the Priority for the Hazard and Risk Evaluation of Chemicals (HREC) According to the Industrial Safety and Health Act (ISHA))

  • Object: The aim of this study is to suggest a list of priority chemicals for the Hazard & Risk Evaluation of Chemicals (HREC) controlled by the Industrial Safety and Health Act (ISHA). Method: Screening assessment was done for 642 chemicals whose exposure threshold limit values were set by the Ministry of Employment and Labor (MOEL). Hazard data were collected from Korea Occupational Safety & Health Agency (KOSHA) and/or other toxicity database. Exposure data were obtained from KOSHA internal database. The hazard and exposure scores of chemicals were listed by order of priority in accordance with GHS classification and exposure index data. Result: From the result of screening risk assessment for 642 chemicals, we extracted a list of 13 priority chemicals for HREC performed by the ISHA. A priority list of 27 chemicals which have carcinogen, mutagen and/or reproductive toxicity but not controlled by the ISHA was suggested for additional evaluation as "chemicals for special management".

화학사고 시 수용체 보호를 위한 독성끝점 농도와 급성독성 자료를 활용한 우려농도 예측값 조사 (Investigation of the Guidance Levels for Protecting Populations from Chemical Exposure and the Estimation of the Level of Concern Using Acute Toxicity Data)

  • Objectives: To protect individuals working at the site as well as the surrounding general population from a chemical accident, several emergency exposure guidance levels have been used to set a level of concern for certain chemicals. However, a level of concern has not been established for many substances that are frequently used or produced in large quantities in Korean workplaces. In the present study, we investigated the guidance levels for protecting populations from chemical exposure and the estimation of level of concern using acute inhalation and oral toxicity data. Methods: The number of chemicals to which emergency exposure guidance levels (e.g., ERPG-2, AEGL-2, PAC-2, and IDLH) can be applied were determined among 822 hazardous chemicals according to the 'Technical Guidelines for the Selection of Accident Scenarios (revised December 2016)'. The ERPG and AEGL values were compared across all three tiers for the 31 substances that appeared on both lists. We examined the degree of difference between the emergency exposure guidance levels and the estimates of level of concern calculated from acute inhalation or acute oral toxicity data. Results: Among the 822 hazardous chemicals, emergency exposure guidance levels can be applied to 359 substances, suggesting that the estimates of level of concern should be calculated using acute toxicity data for 56.3% of the hazardous chemicals. When comparing the concordance rates of ERPG and AEGL for 31 substances, the difference between the two criteria was generally small. However, about 40% of the substances have values diverging by more than three-fold in at least one tier. Such discrepancies may cause interpretation and communication problems in risk management. The emergency exposure guidance levels were similar to the estimates of level of concern calculated using acute inhalation toxicity data, but the differences were significant when using acute oral toxicity data. These results indicate that the level of concern derived from acute oral toxicity data may be insufficient to protect the population in some cases. Conclusion: Our study suggests that the development of standardized guidance values for emergency chemical exposure in the Korean population should be encouraged. It is also necessary to analyze acute toxicity data and fill the information gaps for substances that are important in Korean workplace situations.

  • Background: The occurrence of environmental disease is known to be associated with chronic exposure to toxic chemicals, including waterborne contaminants, air/indoor pollutants, asbestos, ingredients in humidifier disinfectants, etc. Objectives: In this study, we reviewed toxicological studies related to environmental disease as defined by the Environmental Health Act in Korea and toxic chemicals. We also suggested a direction for future toxicological research necessary for the prevention and management of environmental disease. Methods: Trends in previous studies related to environmental disease were investigated through PubMed and Web of Science. A detailed review was provided on toxicological studies related to the humidifier disinfectants. We identified adverse outcome pathways (AOPs) that can be linked to the induction of environmental diseases, and proposed a chemical screening system that uses AOP, chemical toxicity big data, and deep learning models to select chemicals that induce environmental disease. Results: Research on chemical toxicity is increasing every year, but there is a limitation to revealing a clear causal relationship between exposure to chemicals and the occurrence of environmental disease. It is necessary to develop various exposure- and effect-biomarkers related to disease occurrence and to conduct toxicokinetic studies. A novel chemical screening system that uses AOP and chemical toxicity big data could be useful for selecting chemicals that cause environmental diseases. Conclusions: From a toxicological point of view, developing AOP related to environmental diseases and a deep learning-based chemical screening system will contribute to the prevention of environmental diseases in advance.
