Novel Dioxygenases, HIF-α Specific Prolyl-hydroxylase and Asparanginyl-hydroxylase: O2 Switch for Cell Survival

  • Studies on hypoxia-signaling pathways have revealed novel Fe(II) and $\alpha$-ketoglutarate-dependent dioxygenases that hydroxylate prolyl or asparaginyl residues of a transactivator, Hypoxia-Inducible $Factor-\alpha(HIF-\alpha)$ protein. The recognition of these unprecedented dioxygenases has led to open a new paradigm that the hydroxylation mediates an instant post-translational modification of a protein in response to the changes in cellular concentrations of oxygen, reducing agents, or $\alpha$-ketoglutarate. Activity of $HIF-\alpha$ is repressed by two hydroxylases. One is $HIF-\alpha$ specific prolyl-hydroxylases, referred as prolyl-hydroxylase domain(PHD). The other is $HIF-\alpha$ specific asparaginyl-hydroxylase, referred as factor-inhibiting HIF-1(FIH-1). The facts (i) that many dioxygenases commonly use molecular oxygen and reducing agents during detoxification of xenobiotics, (ii) that detoxification reaction produces radicals and reactive oxygen species, and (iii) that activities of both PHD and FIH-1 are regulated by the changes in the balance between oxygen species and reducing agents, imply the possibility that the activity of $HIF-\alpha$ can be increased during detoxification process. The importance of $HIF-\alpha$ in cancer and ischemic diseases has been emphasized since its target genes mediate various hypoxic responses including angiogenesis, erythropoiesis, glycolysis, pH balance, metastasis, invasion and cell survival. Therefore, activators of PHDs and FIH-1 can be potential anticancer drugs which could reduce the activity of HIF, whereas inhibitors, for preventing ischemic diseases. This review highlights these novel dioxygenases, PHDs and FIH-1 as specific target against not only cancers but also ischemic diseases.

A Study on the Improvement of Spatiotemporal Resolution about Fugitive Dust Activity Data in the Agriculture Field (농업분야 비산먼지 활동도 자료의 시공간 해상도 개선 연구)

  • The emission factor and activity data of fugitive dust in the domestic agricultural field have been applied to the US inventory system without reflecting the domestic environmental conditions (wind speed, humidity, etc.) and agricultural characteristics. In this study, the temporal resolution was improved for each region by deriving a monthly distribution factor through the application of wind speed and dry season and the spatial resolution was improved for each region by subdivided into dong and ri from ci·gun·gu. Through this study, it is judged that it can be used as an important data for improving the emission and activity data of fugitive dust in the agricultural field that currently exist.

Temperature-Dependency Urease Activity in Vibrio parahaemolyticus is Related to Transcriptional Activator UreR

  • Vibrio parahaemolyticus possessing urease-positive property is relatively rare, but such strains consistently exhibit the TDH-related hemolysin (TRH) gene. In this study, we examined the effects of incubation temperature on urease activity expression, using the TH3996 and AQ4673 strains where the enzyme activity is known to be temperature-dependent and -independent, respectively. In the TH3996 strain, $\beta$-galactosidase activity was 4.4-fold lower after $30^{\circ}C$ cultivation than after $37^{\circ}C$ in a ureR-lacZ fusion strain, but temperature dependency was not found in ureD- or nikA-lacZ fusion strains. However, ureR-, ureD-, and nikA-lacZ fusions of the AQ4673 strain was not influenced by incubation temperature. We compared the promoter sequences of ureR between the above two strains. Intriguingly, we detected mismatches of two nucleotides between the two strains located at positions -66 and -108 upstream of the methionine initiation codon for UreR. Additionally, urease activity was not affected by culture temperature at either $30^{\circ}C$ or $37^{\circ}C$ by allelic introduction of the AQ4673 ureR gene into the TH3996 ureR deletion mutant. Taken together, our study demonstrates that the transcriptional factor UreR is involved in the temperature dependency of urease activity, and two nucleotides within the ureR promoter region are of particular importance for the urease activity dependency of V. parahaemolyticus.

The Effect of Different Types of Leisure Activities' on the Satisfaction and Psychological Well-being of Primary School Students (초등학생의 여가참여유형이 여가만족도와 심리적 행복감에 미치는 영향)

  • The purpose of this study was to investigate the effect of different types of leisure activities' on the satisfaction and psychological well-being of primary school students. The subjects of this study consisted of 467(boys=191, girls=276) primary school students. The data was analyzed by one way ANOVA, factor analysis, Chi-square analysis, correlation analysis and multi-regression analysis. The main findings of this were as follows: first, there were statistically significant differences in the kinds of activities participated in according to gender. Second, among of the subscale of leisure satisfaction, boys showed higher scores in more education and physical satisfaction than girls, and in the subscale of psychological well-being, boys indicated a higher score in flow and happiness than girls. Third, there were significant differences in leisure satisfaction and psychological well-being in the types of leisure activity. Fourth, the participation motivation of leisure activity statistically influenced psychological well-being. Fifth, leisure satisfaction partially influenced on psychological well-being. Social and environmental satisfaction significantly influenced flow. Education and social satisfaction significantly influenced happiness and psychological well-being.

Proteomic Reference Map and Comparative Analysis between Streptomyces griseus S4-7 and wbiE2 Transcription Factor-Mutant Strain

  • Streptomyces griseus S4-7, a well-characterized keystone taxon among strawberry microbial communities, shows exceptional disease-preventing ability. The whole-genome sequence, functional genes, and bioactive secondary metabolites of the strain have been described in previous studies. However, proteomics studies of not only the S4-7 strain, but also the Streptomyces genus as a whole, remain limited to date. Therefore, in the present study, we created a proteomics reference map for S. griseus S4-7. Additionally, analysis of differentially expressed proteins was performed against a wblE2 mutant, which was deficient in spore chain development and did not express an antifungal activity-regulatory transcription factor. We believe that our data provide a foundation for further in-depth studies of functional keystone taxa of the phytobiome and elucidation of the mechanisms underlying plant-microbe interactions, especially those involving the Streptomyces genus.

Activation of Nrf2 by sulfuretin stimulates chondrocyte differentiation and increases bone lengths in zebrafish

  • Elongation of most bones occur at the growth plate through endochondral ossification in postnatal mammals. The maturation of chondrocyte is a crucial factor in longitudinal bone growth, which is regulated by a complex network of paracrine and endocrine signaling pathways. Here, we show that a phytochemical sulfuretin can stimulate hypertrophic chondrocyte differentiation in vitro and in vivo. We found that sulfuretin stabilized nuclear factor (erythroid-derived 2)-like 2 (Nrf2), stimulated its transcriptional activity, and induced expression of its target genes. Sulfuretin treatment resulted in an increase in body length of zebrafish larvae and induced the expression of chondrocyte markers. Consistently, a clinically available Nrf2 activator, dimethyl fumarate (DMF), induced the expression of hypertrophic chondrocyte markers and increased the body length of zebrafish. Importantly, we found that chondrocyte gene expression in cell culture and skeletal growth in zebrafish stimulated by sulfuretin were significantly abrogated by Nrf2 depletion, suggesting that such stimulatory effects of sulfuretin were dependent on Nrf2, at least in part. Taken together, these data show that sulfuretin has a potential use as supporting ingredients for enhancing bone growth.

  • In spite of the development of scientific and medical technology, the rate of prevalence and death of the chronic disease gets higher. Therefore people come to know prevention is more important than treatment in disease. In Western, it is nearly after 1970's when it is revealed that the major factor of chronic disease comes from habitual behaviors in everyday life. But that is common idea in Oriental Medicine since thousand years. In Oriental Medicine, the main cause of disease is composed of natural factor including climate condition, living-life factor, psycho-social factor, constitutional factor, inbody-pathologic-substance factor, and other factors including habits of using drug and medical service, injury, environmental pollutions, and occupational etiologic factors. Among these, the major factors of disease in modern society are lifestyle factors like the tendency of choosing and intaking food, the level of physical activity and labor, smoking and drinking alcohol, psychological tendency, the habits of using drug and medical service and so on. This is a practical aspect of oriental medicine where the most important etiologic factor comes from human behaviors, not from external factors like germ or virus. So that etiology in Oriental Medicine where improvement in the way of life is needed to prevent disease would play an important role in modern society.

Regulation of Activity of the Response Regulator RssB (Response Regulator RssB의 활성 조절)

  • Against environmental stresses, many bacteria utilize the alternate sigma factor RpoS that induces transcription of the specific set of genes helpful in promoting bacterial survival. Intracellular levels of RpoS are determined mainly by its turnover through proteolysis of ClpXP protease. Delivery of RpoS to ClpXP strictly requires the adaptor protein RssB. The two-component-type response regulator RssB constantly interacts with RpoS, but diverse environmental changes inhibit this interaction through modification of RssB activity, which increases RpoS levels in bacteria. This review discusses and summarizes recent findings on regulatory factors in RssB-RpoS interactions, including IraD, IraM, IraP anti-adaptor proteins of RssB and phosphorylation of N-terminal receiver domain of RssB. New information shows that the coordinated regulation of RssB activity in controlling RpoS turnover confers efficient bacterial defense against stresses.

Keap1 represses nuclear activation of antioxidant responsive elements by Nrf2 through binding to the amino-terminal Neh2 domain

  • Transcription factor Nrf2 is essential for the antioxidant responsive element (ARE)-mediated induction of phase II detoxifying and oxidative stress enzyme genes. Detailed analysis of differential Nrf2 activity displayed in transfected cell lines ultimately led to the identification of a new protein, which we named Keap1, that suppresses Nrf2 transcriptional activity by specific binding to its evolutionarily conserved amino-terminal regulatory domain. The closest homolog of Keap1 is a Drosophila actin-binding protein called Kelch, implying that Keap1 might be a Nrf2 cytoplasmic effector. We then showed that electrophilic agents antagonize Keap1 inhibition of Nrf2 activity in vivo, allowing Nrf2 to traverse from the cytoplasm to the nucleus and potentiate the ARE response. We postulate that Keap1 and Nrf2 constitute a crucial cellular sensor for oxidative stress, and together mediate a key step in the signaling pathway that leads to transcriptional activation by this novel Nrf2 nuclear shuttling mechanism. The activation of Nrf2 leads in turn to the induction of phase II enzyme and antioxidative stress genes in response to electrophiles and reactive oxygen species.

Activity Factors and Differences in Activity by Period of Red-eared Slider Turtles (Trachemys scripta elegans) in Ansan Reed Wetland (안산갈대습지에 서식하는 붉은귀거북(Trachemys scripta elegans)의 활동 요인과 시기에 따른 활동 차이)

  • The purpose of this study was to understand the factors that affected activities and differences in seasonal activities of the invasive red-eared slider turtle (Trachemys scripta elegans) at Ansan reed wetland in Sangnok-gu, Ansan, Gyeonggi Province, the Republic of Korea. We installed nine nets in the study region to capture turtles and checked the nets twice a day for 154 days from April 28 to September 28 2013. In total, 60 individuals, including 12 juveniles, 26 adult males, and 22 adult females, were captured in 96 of 308 net checks. The multiple regression analysis of 12 environmental factors was conducted to analyze activity factors of red-eared slider turtle by assuming the number of turtles captured for a week to be the indicator of the activeness of the turtles. The result showed that the daylength was the only environmental factor that significantly affected the turtles' activity (p<0.001), and none of the other factors showed a significant correlation. The highest activity was recorded at the end of May, which was the beginning of the reproductive period. The daylength during the breeding season was significantly different from that during other seasons (Kruskal-Wallis test, p<0.001). The information about the relationship between environmental factors and species in habitat can be useful for the management of invasive alien species in the future.
