난황항체를 이용한 돼지 대장균설사증 방제기법 개발 II. 난황항체의 돼지 대장균증에 대한 치료효과 (Development of preventive method for enterotoxigenic colibacillosis using egg yolk antibodies II. Therapeutic effect of egg yolk antibodies against colibacillosis of piglets)

  • The present study describes the effectiveness of egg yolk antibodies (IgY) against enteric colibacillosis and edema disease in piglets. The antibodies were gained from the egg yolk of hens immunized with k88, k99, 987p fimbrial adhesin and heat-labile toxin antigens of enterotoxigenic Escherichia coli (ETEC). Orally-administered egg yolk antibodies solution protected against experimental challenge with ETEC $K88^+$ and $k99^+$ strains in neonatal piglets and mice. In field trial, a total of 598 diarrheal piglets were orally treated with 3ml of antibody once a day to determine for the therapeutic effect. Of them, 582 (97.3%) piglets were recovered from diarrhea in 3 days. We also experimentally treated with the egg yolk antibodies twice a day for 5 consecutive days for 94 weaning piglets with edema disease for the determination of therapeutic effects. Seventy four piglets (78.7%) were recovered from clinical edema signs. Theses findings indicate that egg yolk antibodies against k88, k99, 987p and LT of ETEC are useful source of passive immunity for enteric colibacillosis and edema disease of piglets.

  • Immunogenicity of Escherichia coli pilus and LT were evaluated in 20-week-old hens. The antigens were consisted of K88, K99, 987p pilus and heat labile toxin purified from enterotoxigenic Escherichia coli. The durations of antibody titers in sera and egg yolk were investigated by an enzyme-linked immunosorbent assay(ELISA). After first inoculation, antibody titers in sera reached at peak 2 weeks postinoculation. However, peak antibody titers in egg yolk were detected 4 weeks postinoculation, indicating that transfer of immunoglobulin from serum to egg yolk took about two weeks period. Although there were slight reduction in titers, the specific antibodies in egg yolk lasted up to 3 months. Immune responses against monovalent and combined antigens were showed as almost same patterns. The transfer rate of antibodies from serum to egg yolk didn't show any significant differences among three pilus antigens in this study. Considering the concentrations of antigens in each inoculated group, multivalent antigens containing heat labile toxin of E coli were found to be more immunogenic than monovalent antigen in producing specific antibodies. From this experiment, it was demonstrated that multivalent antigens containing three pilus and heat labile toxin could be a promising candidate for the production of egg yolk antibodies for prophylactic use in preventing swine colibacillosis in future.

Establishment of a linear regression equation for quantification of beta-hemolytic Escherichia coli in different media and survival of hemolytic Escherichia coli after blending with three different media

  • Pathogenic E. coli associated post-weaning diarrhea (PWD) and edema disease are common diseases in commercially-housed weanling pigs. An enterotoxigenic E. coli (ETEC) oral challenge model has been used to mimic the physiological responses observed in commercial conditions. However, an oral challenge procedure has two major limitations: (1) the ETEC cell density is unknown at the point of oral inoculation, and (2) blending ETEC with traditional TSB (trypticase soy broth) is not palatable and hence decreases acceptability by piglets. Therefore, the purposes of this study were to (1) establish a regression equation that can be used for estimation of ETEC concentration in dilution media using the spectrophotometric measurement of cell density; and (2) examine survival of ETEC after blending either with TSB, sweetener or dextrose. A strain of ETEC (serogroup beta-hemolytic E. coli O149; K91; F4; toxins LT, STa, STb) was grown in TSB for 3.5 hours, centrifuged, the supernatant was discarded, and the ETEC pellet was then blended either with TSB (100 mL), sweetener (60 mL TSB + 40 mL fruit flavored concentrate), or dextrose (50 mL TSB + 50 mL dextrose; 0.5g/mL dextrose). Cell density was measured using the colorimetric method and also plated on a 5% sheep blood agar for counting of ETEC colony forming units at 0, 5, 35, 65 and 125 min after blending. The optical density at 600 nm explained 83% of ETEC colony forming units, indicating that the established linear equation (y= 6E+08x - 4E+07, P<0.004) can be used for robust quantification of ETEC cell density in TSB, sweetener and dextrose media. When ETEC was blended with sweetener and dextrose, survival of ETEC was decreased by 45% and 72% within 5 min post-blending. Therefore, further research is required to find out the suitable medium that has potential to improve palatability without compromising survival of ETEC.

  • Escherichia coli is recovered from a wide variety of infections in many animals species. It may be a primary or secondary agent. Nursing and young animals are particularly susceptible, and urinary tract infections are frequent. The various serotypes of E coli are intestinal inhabitants of animals including humans and probably infect most mammals and birds : therefore, they have a cosmopolitan distribution. Colibacillosis refers to any totalized or systemic infection caused entirely or partly by E coli. Collibacillosis in mammals is most often a primary enteric disease, whereas collibacillosis in poultry is typically a secondary located or systemic disease occurring when host defenses have been impaired or overwhelmed. Other opportunistic bacteria, which can be identified by culture, may play a similar role to that of I coli in secondary infections. Collectively, infections caused by E coli are responsible for significant economic losses to the animal performance. From the standpoint of pathogenic mechanisms and diseases, four major categories of E coli are recognized : enterotoxigenic(ETEC), enteropathogenic (EPEC), enteroinvasive(EIEC), and enterohemorrhagic(EHEC). In addition, two less-well-defined E coli categories are recognized in animals and humans : enteroaggregative and cytotoxin necrotizing factor-positive. The aforementioned categories are represented by different serotypes. Certain serotypes show a host preference and are encountered more frequently in some disease syndromes. Of the four major categories, ETEC is the most common cause of diarrhea in calves, lambs, and pigs. Strains in the other categories cause the less-common diarrhea and other disease syndromes. Enterotoxins and pilus antigens are the two most prominent virulence factors thus far identified for ETEC. Two enterotoxins, one heat-stable(ST) and one heat-labile(LT), are produced by enterotoxigenic strains of E coli : not all culture produce both of these plasmid-based enterotoxins.

전남 지방의 설사 자돈에서 분리된 병원성 대장균에 관한 연구 (Studies on the pathogenic Escherichia coli isolated from piglets with diarrhea in Chonnam)

  • Porcine E coli infection is a disease caused by Enterotoxin produced by Enterotoxigenic Escherichia coli(ETEC). Enteric colibacillosis has become an economically important disease in pigs as a result of increasing intensification of farrowing management. The present study undertaken to obtain the antibiotic sensitivity and distribution of serogroups and pili producibility test of ETEC from E. coli isolates in Chonnam. The results obtained were as follows. 1. A total of 71 isolates identified as E, coli employing IMViC system from rectal specimens of 54 piglets with diarrhea. 2. In antibiotic sensitivity test, isolates showed high sensitivity to AN, CM, Fox, GM, but resistance to EM, NA TC. 3. The distribution of 25 Isolates of serogroups were 0141:K85(11.3%), 08:K87(8.5%), 064:K (5.6%), 0138:K8l (4.2%), 0139 :K82(2.8%), 0157:K88ac(1.4%) and 0149:K9l (1.4%). 4. MRHA of guinea pig erythrocytes was detected in 8 out of 25OK serotypes and 9 out of 46 unidentified serotypes. MRHA titers of serotypes showed from 64 to 128 in 0141: K85, 2 in 0138:K8l and no titers in 0139:K82. 5. The production of heat labile enterotoxin of ETEC was detect 39 out of 52 isolates showed $\beta$-hemolysin, 7 out of 52 isolates showed ${\gamma}$-hemolysin and 6 out of 52 isolates showed ${\gamma}$-hemolysin by $GM_1$ganglioside ELISA. The distribution of LT toxin were in 12 isolate showed $\beta$-hemolysin, 2 isolates showed ${\alpha}$-hemolysin and 3 isolates showed ${\gamma}$-hemolysin in 25 OK serotypes.

장내 유해세균을 억제하는 양돈용 프로바이오틱스 개발을 위한 비피도박테리아 탐색 (Screening of Bifidobacteria for the Development of Probiotics Inhibiting Intestinal Pathogenic Bacteria)

  • 본 연구에서는 동물유래 시료들로부터 양돈 사료첨가용 유산균을 개발하기 위하여 enterotoxigenic E. coli, Salmonella Typhimurium, Clostridium perfringens 등 양돈산업에서 심각한 문제를 일으키는 인수공통 병원성 세균들과 Clostridium jejuni, Listeria monocytogenes, Staphylococcus aureus와 같은 식중독균을 강하게 저해하는 Bifidobacterium 유산균을 분리하고자 하였다. 이를 위하여 소의 반추위 내용물, 닭 창자 내용물, 돼지 분변 등의 시료들로부터 총 65주의 혐기성미생물을 분리하였다. 이 중에서 항병원세균활성이 가장 높은 4주를 선별하였는데, 이들은 16S rDNA 염기서열 분석방법에 의하여 3주의 B. boum과 1주의 B. thermophilum으로 동정되었다. 특히 B. thermophilum는 분리주들 중에서 가장 높은 돼지 장 상피세포에 대한 부착성을 보였고, 여러 인수공통병원세균들과 식중독균들에 대한 높은 항균력, 산과 담즙내성을 보여 양돈용 생균제 후보균주로서 우수한 특성을 나타내었다.

자돈 설사병에 대한 형질전환 당근백신의 방어 효능 평가 (Protective effects of a transgenic carrot vaccine on piglet diarrhea)

  • The study evaluated whether a transgenic carrot vaccine could induce a K88-specific immune response in sows and whether the resultant maternal antibody could protect piglets against enterotoxigenic Escherichia coli (ETEC) K88ac infection. Sows (n = 15) selected randomly from a farm in Korea were assigned to three groups (n = 5 per group: control [untreated]), group A (orally inoculated with a nontransgenic and transgenic carrot vaccines at 2 and 4 weeks ante partum, respectively), and group B (conventionally vaccinated according to the manufacturer's instructions). After 7 days of lactation, 5 piglets selected randomly from each group were challenged with $1{\times}10^{10}$ colony forming units/mL ETEC K88ac. Group C had the lowest mean fecal consistency score on post-challenge days 1 and 7. Histiologically, On post-challenge day 7, group C showed an increased duodenum and ileum villus:crypt ratio, compared to group A in the duodenum, with group B displaying the highest ratio. Groups B and C had more increased villus width than group A in the jejunum. Group C displayed the greatest increase in villus width in the ileum. The colostrums and serum from groups B and C displayed higher concentrations of IgA and IgG against ETEC K88, compared to group A. Based on the results, it was concluded that the transgenic carrot vaccine in sow per oral may have an effect on preventing piglet diarrhea as good as commercial recombinant vaccine.

국내 한 대학병원의 임상검체에서 분리된 Bacteroides fragilis 독소 유전자의 특성 (Characterization of bft Genes among Enterotoxigenic Bacteroides fragilis Isolates from Extraintestinal Specimens at a University Hospital in Korea)

  • Enterotoxigenic Bacteroides fragilis (ETBF)는 병독소 인자로 알려진 장독소를 생성하는 균종이다. B. fragilis enterotoxin (BFT)은 bft-1, bft-2 및 bft-3 세 개의 유전자 아형들이 밝혀졌다. 본 연구에서는 임상 검체에서 분리된 B. fragilis에서 bft 유전자의 유무와 BFT 음성 및 양성 균주의 항균제 내성을 조사하였다. 국내의 한 대학병원에서 8년간(2006~2013년) 장외 검체에서 분리된 B. fragilis는 총 537주이었다. 다중중합연쇄반응으로 시험하여 bft 유전자 아형을 확인하였다. BFT 음성 74주와 양성 33주를 포함한 B. fragilis 107주의 항균제 감수성은 CLSI 한천희석법으로 시험하였다. 임상 검체에서 분리된 B. fragilis의 bft 유전자 검출율은 30% 이었고, 이 중 혈액과 혈액 외 장외 검체 분리주에서는 각각 33%, 29% 이었다. ETBF 중에서 가장 흔한 아형은 bft-1 이었고, 그 다음은 bft-2, bft-3 순이었다(bft-1: 77%, bft-2: 14%, bft-3: 9%). BFT-음성과 양성 균주의 내성률은 일부 항균제에 대해서 차이가 있었다(BFT-음성 균주: piperacillin-tazobactam 3%, cefoxitin 5%, imipenem 1%, clindamycin 38%; BFT-양성 균주: piperacillin-tazobactam 3%, cefoxitin 6%, imipenem 3%, clindamycin 42%). BFT-음성 및 BFT-양성 균주 모두는 chloramphenicol과 metronidazole에 대한 내성은 없었다. 결론적으로, 혈액 분리주에서의 ETBF 검출율은 혈액 외 장외 검체 분리주에서와 비슷하였고, 가장 흔한 유전자 아형은 bft-1 이었다. 항균제 내성은 BFT 양성 균주가 음성 균주보다 대체로 높았으나 통계학적으로 유의한 차이는 없었다.

  • 식품으로 인한 질병은 개인의 생명을 위협할 뿐 아니라 건전한 노동력의 상실로 생산성을 감소시키고 의료비를 증가시켜 개인의 행복 및 국가 경제에 부담을 준다 (Buzby et al. 1996, Mead et at. 2000). HACCP와 여러 식품안전에 관한 규제의 시행 등의 많은 노력에도 불구하고 Salmonella serotypes, Staphylococcus aureus, Camplobacter jejuni, Campylobacter coli, enterotoxigenic and enteroinvasive Escherichia coli, Clostridium perfringens, Bacillus cereus와 같은 식품기원 병원성 세균으로 인한 질병은 줄어들고 있지 않다. 오히려 외식 위주로 변해가는 생활습관은 식품을 통한 이들 병원성 세균의 전파가능성을 증가시키고 있다. 게다가 새로운 병원성균 (Yersinia enterocolitica, Listeria monocytogenes, E. coli 0157:H7, Aeromonas spp., Plesiomonas spp.) 의 출현 또는 특정 식품과 연관되어 나타나는 특정 subtype (Salmonella serotype Enteritidis) 의 출현은 이제까지 통용되어왔던 식품기원 세균을 위한 제어프로그램과는 다른 각도로 재원을 활용해야 할 동기를 부여하고 있다.

  • Escherichia coli strains associated with diarrhea have been divided into the following six major categories on the basis of pathogenic mechanisms: enterotoxigenic E. coli (ETEC), enteroinvasive E. coli (EIEC), enteropathogenic E. coli (EPEC), enterohemorrhagic E. coli (EHEC), enteroaggregative E. coli (EAggEC) and diffusely adherent E. coli (DAEC).$\^$15,18/ EPEC, EAggEC, and DAEC strains were classified by their ability to produce distinct patterns of adherence to cultured epithelial cells in virto: localized (LA), aggregative (AA), and diffuse (DA) adherence. The objective of this study was to investigate the relationship of the adherence patterns with AIDA-positive E. coli isolated from diarrheic pigs. (omitted)
