Soybean transgenic plants with ectopically expressed AtABF3 were produced by Agrobacterium-mediated transformation and investigated the effects of AtABF3 expression on drought and salt tolerance. Stable Agrobacterium-mediated soybean transformation was carried based on the half-seed method (Paz et al. 2006). The integration of the transgene was confirmed from the genomic DNA of transformed soybean plants using PCR and the copy number of transgene was determined by Southern blotting using leaf samples from $T_2$ seedlings. In addition to genomic integration, the expression of the transgenes was analyzed by RT-PCR and most of the transgenic lines expressed the transgenes introduced. The chosen two transgenic lines (line #2 and #9) for further experiment showed the substantial drought stress tolerance by surviving even at the end of the 20-day of drought treatment. And the positive relationship between the levels of AtABF3 gene expression and drought-tolerance was confirmed by qRT-PCR and drought tolerance test. The stronger drought tolerance of transgenic lines seemed to be resulted from physiological changes. Transgenic lines #2 and #9 showed ion leakage at a significantly lower level (P < 0.01) than ${\underline{n}}on-{\underline{t}}ransgenic$ (NT) control. In addition, the chlorophyll contents of the leaves of transgenic lines were significantly higher (P < 0.01). The results indicated that their enhanced drought tolerance was due to the prevention of cell membrane damage and maintenance of chlorophyll content. Water loss by transpiration also slowly proceeded in transgenic plants. In microscopic observation, higher stomata closure was confirmed in transgenic lines. Especially, line #9 had 56% of completely closed stomata whereas only 16% were completely open. In subsequent salt tolerance test, the apparently enhanced salt tolerance of transgenic lines was measured in ion leakage rate and chlorophyll contents. Finally, the agronomic characteristics of ectopically expressed AtABF3 transgenic plants ($T_2$) compared to NT plants under regular watering (every 4 days) or low rate of watering condition (every 10 days) was investigated. When watered regularly, the plant height of drought-tolerant line (#9) was shorter than NT plants. However, under the drought condition, total seed weight of line #9 was significantly higher than in NT plants (P < 0.01). Moreover, the pods of NT plants showed severe withering, and most of the pods failed to set normal seeds. All the evidences in the study clearly suggested that overexpression of the AtABF3 gene conferred drought and salt tolerance in major crop soybean, especially under the growth condition of low watering.

Changes of zinc speciation and dehydrogenase activity in soil were studied before and after a field scale phytoextraction by Fagopyrum esculentum at a paddy soil near a closed zinc mine. The concentrations of zinc in paddy soil, in which Fagopyrum esculentum was planted, ranged from approximately 600 mg/Kg(high Zn soil) to 300 mg/Kg(moderate Zn soil). Despite of severe growth inhibition by Zn at the high Zn region, Fagopyrum esculentum accumulated phytoavailable fraction of Zn absorbed from the soil, and enhanced soil dehydrogenase activity (DHA) that had been inhibited by Zn toxicity. After the plant cultivation of 2 months, the concentrations of phytoavailable Zn in the rhizosphere soil at high and moderate Zn region has decreased 25% and 75%, respectively. The amount of Zn reduction in the rhizosphere soil corresponds to that accumulated in plants (recovery 92$\sim$107%), which implies Fagopyrum esculentum removed Zn from the soil. DHA was inversely correlated to the total Zn concentrations in soil. Before plant cultivation, the DHA in the high Zn soil was twice lower than that in the moderate Zn soil. More than 35% of DHA increase was observed in both soils after the application of phytoextraction with Fagopyrum esculentum.
The seeds of naturally growing Heteropappus hispidus (Thunb.) were treated by nine different doses (0, 10, 20, 40, 80, 120, 160, 200, 300, 400 Gy) of gamma rays to investigate their germination rate and to quantity the characteristics of their germinated plants as like as leaf appearance and length, the formation rate of anthocyanin color in stem 30 days after germination, the formation rate of rosette leaf and multi-shoot, the flowering and seed-bearing, and shoot length. The germination rate at least up to 120Gy was not greatly affected but was rapidly decreased at over 160Gy. It seemed that lethal dose ($LD_{50}$) of germination was 160Gy. The leaf appearance and growth was also inhibited, but the formation rate of anthocyanin color in the flower stem was enhanced up to 30% with dose. The rosette plants were observed in plants irradiated with higher than 40Gy. Multi-shoots were developed over 80Gy. For a short shoot length and bundle of thin stem, it was considered that they can be selected as the potential pot flower plants, through genetic fixation. In particular, it was suggested that the formation of anthocyanin color in flower stem, rosette and multi-shoot plants induced by the high dose of gamma rays could be utilized as the morphological markers for the mutant selection of Heteropappus hispidus (Thunb.).

This study was conducted to examine the effects of the seeding rate on the disease incidence of sharp eyespot(Rhizoctonia cerealis) on three different varieties and to select effective chemicals to control the disease. When the seeds were sown twice as many as the recommendation, the disease incidence increased by approximately 13%. However, the susceptible variety 'Jopummil' alone showed the significantly enhanced disease incidence at a two-fold seeding rate. Two chemicals such as Hexaconazole EC and Tebuconazole EC highly inhibited the fungal growth on agar medium. However, two strobilurin fungicides such as Pyraclostrobin EC and Trifloxystrobin SC were relatively weak. The fungicides tested displayed the similar in vivo antifungal activities as in vitro activities. Hexaconazole EC and Tebuconazole EC showed the strongest both protective and curative activities and the protective activities of the chemicals were generally higher than the curative activities. Hexaconazole EC and Tebuconazole EC controlled the disease by 64% and 73%, respectively, and the two chemicals reduced the disease by 45% and 39%, respectively, when they were applied one day after pathogen inoculation. These results indicate that both Hexaconazole EC and Tebuconazole EC could be used to control sharp eyespot on wheat.

Prostaglandin E2(PGE2), a major product of cyclooxygenase-2 (COX-2), plays an important role in the carcinogenesis of many solid tumors, including colorectal cancer. Because PGE2 functions by signaling through PGE2 receptors (Eps), which regulate tumor cell growth, invasion, and migration, there has been a growing amount of interest in the therapeutic potential of targeting Eps. In the present study, we investigated the role of EP4 on the effectiveness of cordycepin in inhibititing the migration and invasion of HCT116 human colorectal carcinoma cells. Our data indicate that cordycepin suppressed lipopolysaccharide (LPS)-enhanced cell migration and invasion through the inactivation of matrix metalloproteinases (MMP)-9 as well as the down-regulation of COX-2 expression and PGE2 production. These events were shown to be associated with the inactivation of EP4 and activation of AMP-activated protein kinase (AMPK). Moreover, the AMPK inhibitor, compound C, as well as AMPK knockdown via siRNA, attenuated the cordycepin-induced inhibition of EP4 expression. Cordycepin treatment also reduced the activation of CREB. These findings indicate that cordycepin suppresses the migration and invasion of HCT116 cells. Through modulating EP4 expression and the AMPK-CREB signaling pathway. Therefore, cordycepin has the potential to serve as a potent anti-cancer agent in therapeutic strategies against colorectal cancer metastasis.
The aim of this work was to establish the optimal conditions for the production of cellobiase by a marine bacterium, Cellulophaga lytica LBH-14, using response-surface methodology (RSM). The optimal conditions of rice bran, ammonium chloride, and the initial pH of the medium for cell growth were 100.0 g/l, 5.00 g/l, and 7.0, respectively, whereas those for the production of cellobiase were 91.1 g/l, 9.02 g/l, and 6.6, respectively. The optimal concentrations of $K_2HPO_4$, NaCl, $MgSO_4{\cdot}_{7H2}O$, and $(NH_4)_2SO_4$ for cell growth were 6.25, 0.62, 0.28, and 0.42 g/l, respectively, whereas those for the production of cellobiase were 4.46, 0.36, 0.27, and 0.73 g/l, respectively. The optimal temperatures for cell growth and for the production of cellobiase by C. lytica LBH-14 were 35 and $25^{\circ}C$, respectively. The maximal production of cellobiase in a 100 L bioreactor under optimized conditions in this study was 92.3 U/ml, which was 5.4 times higher than that before optimization. In this study, rice bran and ammonium chloride were developed as carbon and nitrogen sources for the production of cellobiase by C. lytica LBH-14. The time for the production of cellobiase by the marine bacterium with submerged fermentations was reduced from 7 to 3 days, which resulted in enhanced productivity of cellobiase and a decrease in its production cost. This study found that the optimal conditions for the production of cellobiase were different from those of CMCase by C. lytica LBH-14.
A pot experiment was conducted to find the nitrogen fixing activities associated with ice rhizoplane in nine varieties at various growth stage by acetylene reduction technique. Results obtained ware Summarized as follows. 1. Nitrog en fixing activites associated with rice rhizoplane differ by the rice variety and the growth stage. The activities increased with progressing of growing time, and maximum activities were obtained from the ear forming stage to the heading stage. 2. The application of rice straw did not greatly enhanced the nitrogen fixing activities associated with rice rhizoplane. However, somewhat increasing tendency was obtained in the Kataktara, TKM-6, Centrypartna. Daegujo, and Jodongji varieties. 3. The nitrogen fixed during the one rice cropping period was estimated as 0.55kg of N per 10a by Milyang-23 variety to 2.6kg of N per 10a by Jodongji variety. 4. The amount of rice root showed no significant relationship with the amount of nitrogen fixed during the growing period of rice plant.
The whole plant of crop maize was chopped and ensiled in airtight 1-L capacity glass jars to determine the influence of residual sugar on anaerobic yeast growth and on the fermentation of lactic acid by L. buchneri in whole crop maize silage. There were a total of six treatments used in this experiment as follow: added 25 g de-mineralised water per kg chopped maize serving as control (con), 37.5 g glucose solution containing 12.5 g glucose ($g_1$), 75 g glucose solution containing 25 g glucose ($g_2$), 25 g L. buchneri suspension intended for $10^6$ cfu $g^{-1}$ (L.b.), $g_1+L.b.$ and $g_2+L.b.$ All silos were stored in the dark at $20^{\circ}C$ until end of experiment. Jars were opened on duplicates at day 2, 7, 14, 28, 56 or triplicates at day 91 after ensiling for measuring the pH, microbiological enumeration and fermentative products. Results indicated that acidification rates for all silages were very fast, no difference occurred among treatments before day 28. After day 28 the pH values for silages inoculated by L. buchneri. with or without sugar tended to increase especially for treated only with L. buchneri, resulting in higher (p<0.01) finial pH than uninoculated silages. Compared with control silage, the added sugar significantly (p<0.01) increased dry matter (DM) loss, L. buchneri enhanced (p<0.01) DM loss further at different sugar existence. Silages inoculated by L. buchneri only or in combination with sugar addition contained less (p<0.01) lactic acid than the correspondent silages without inoculation with L. buchneri. In comparison with control, ethanol production is about 3 or 6 fold higher due to addition 12.5 or 25 g glucose per kg chopped maize at ensiling. The added sugar resulted in less acetic acid concentration (p<0.01) than control, but inoculation with L. buchneri increased (p<0.01) acetic acid than correspondent uninoculated silages at different sugar levels. No butyric acid and propionic acid were found in uninoculted silages, silages inoculated with L. buchneri. produced more propionic acid, 1-propanol and butyric acid. Lactobacilli counts were not influenced by added sugar, but increased (p<0.01) with inoculation of L. buchneri. The added sugar increased significantly (p<0.01) the yeast count, whereas L. buchneri showed the contrary effect. No differences were found in the aerobic stability among all treatments. In conclusions, 1) the added sugars encourage the growth of yeast and yeasts convert extra sugar into ethanol in maize silages. 2) The added sugars and L. buchneri do not influence the aerobic stability of silages stored in anaerobic silos.

This study was conducted to examine the effects of cow manure originated from the clay mineral (porphyrite, zeolite and bentonite) feeding on the growth of Zea mays GK729. CEC of the manure after bentonite, zeolite, and porphyrite feeding were 147.5, 137.0, and 114.0 cmol/Kg, respectively. These values were higher than that of non-mineral treated manure, 107.5 cmol/Kg. After 3 month growing, there was significant difference among treatment in the biomass prodcution and the content of minerals in plant tissue of Z. mays, but the physico-chemical properties of soils were not showed significant difference. For these results suggested that the clay mineral enhanced the grwoth of Z. mays according to the unknown mechanism, the further studies on the physiological changes will be needed.

This study was conducted at the forage experiment field of SNU, Snweon to investigate effects of sowing time and seeding rate on growth characteristics, winter survival and dry matter yield of forage rye (Secale cereale L.) as fresh-cut forage. Treatments involved 9 combinations of sowing times (September 8, 28 and October 18) and seeding rates(80, 130 and 180kg $ha^{-1}$). Plant height was not influenced by the time of sowing, but forage rye had a tendency to be taller as increasing seeding rate when sown on September 8 and October 28 and harvested in fall and spring, respectively, Winter survival was reduced by fall harvesting in the plot sown on September 8, however, total dry matter yield was increased by the fall harvesting. Average dry matter yields of 5884, 4993 and 3525kg $ha^{-1}$ were obtained when forage rye was sown on September 8, 28 and October 18, respectively. Dry matter yield was decreased as delaying the time of sowing, but a tendency toward higher yield of forage rye by increasing seeding rate was observed. Yield difference by seeding rates was markedly larger when forage rye was sown on October 18 than September 8 and 28. Based on the results of this study, it appears that the dry matter yield of forage rye could be enhanced by sowing early to late September under upland condition in the middle plain are of Korea. The seeding rates of 80 to 130 kg $ha^{-1}$ and 180kg $ha^{-1}$ would be suitable for early to mid fall and late fall sowings, respectively.
