The potential ability of Button mushroom compost (BMC) to solubilize heavy metals was estimated with metal contaminated soils collected from abandoned mines of Boryeong area in South Korea. The bacterial strains in BMC were isolated for investigating the mobilization of metals in soil or plant by the strains and identified according to 16S rRNA gene sequence analysis. When metal solubilization potential of BMC was assessed in a batch experiment, the BMC was found to be capable of solubilizing metals in the presence of metals (Co, Pb and Zn) and the results showed that inoculation of BMC could increase the concentrations of water soluble Co, Pb and Cd by 35, 25 and 45% respectively, than those of non-inoculated soils. BMC-assisted growth promotion and metal uptake in sunflower (Helianthus annuus) was also evaluated in a pot experiment. In comparison with non-inoculated seedlings, the inoculation led to increase the growth of H. annuus by 27, 25 and 28% respectively in Co, Pb and Zn contaminated soils. Moreover, enhanced accumulation of Co, Pb and Zn in the shoot and root systems was observed in inoculated plants, where metal translocation from root to the above-ground tissues was also found to be enhanced by the BMC. The apparent results suggested that the BMC could effectively be employed in enhancing phytoextraction of Co, Pb and Zn from contaminated soils.
Tall fescue (Festuca arundinacea Schreb.) is an important cool season forage plant that is not well suited to extreme heat, salts, or heavy metals. To develop transgenic tall fescue plants with enhanced tolerance to abiotic stress, we introduced an alfalfa Hsp23 gene expression vector construct through Agrobacterium-mediated transformation. Integration and expression of the transgene were confirmed by polymerase chain reaction, northern blot, and western blot analyses. Under normal growth conditions, there was no significant difference in the growth of the transgenic plants and the non-transgenic controls. However, when exposed to various stresses such as salt or arsenic, transgenic plants showed a significantly lower accumulation of hydrogen peroxide and thiobarbituric acid reactive substances than control plants. The reduced accumulation of thiobarbituric acid reactive substances indicates that the transgenic plants possessed a more efficient reactive oxygen species-scavenging system. We speculate that the high levels of MsHsp23 proteins in the transgenic plants protect leaves from oxidative damage through chaperon and antioxidant activities. These results suggest that MsHsp23 confers abiotic stress tolerance in transgenic tall fescue and may be useful in developing stress tolerance in other crops.

In attempt to avoid crop damage through wild bird's picking, this study was designed with aim to evaluate several pre-sowing soybean seed coatings for optimum yield in corn-soybean mixed forage. It was investigated under four cropping treatments, viz. 1) corn sole, 2) corn mixed with soybean without any coating, 3) corn with iron coated soybean and 4) corn with thiram coated soybean. Each treatment had three replicates and corn sole was control treatment. Pioneer (P1184) and crossbred ($PI483463{\times}Hutcheson$) seeds were used for corn and soybean, respectively. The trial was conducted under randomized block design from $5^{th}$ June to $23^{rd}$ September, 2015. Data were an alyzed through ANOVA technique using SAS9.1.3 software. Results depicted that survivability of soybean against wild birds damage was found better (p<0.05) in thiram coating which was higher than iron coating and control treatment but later on thiram coating had adverse effects on subsequent growth of soybean plants. Corn stalk height was decreased (p<0.05) in thiram coating, whereas corn ear height was reduced in iron coating treatment. Iron coating enhanced (p<0.05) height of soybean plant (p<0.05) better than that of thiram coating. Soybean seed coatings didn't influence dry matter yield and nutritive value in terms of total digestible nutrients yield in corn soybean mixed forage. Conclusively, although presowing thiram coating enhanced survivability of soybean plants against wild bird damage but had adverse effects on its subsequent growth. However, soybean seed coatings didn't influence yield and nutritive value of corn soybean intercropping forage.
Achyranthes japonica extract (AJE) is derived from a medicinal plant Achyranthes japonica, known for its anti-inflammatory, antioxidant, and antimicrobial properties. AJE contains multiple bioactive compounds, including saponins, triterpenoids, phytoecdysteroids, 20-hydroxyecdysone, and inokosterone. The aim of this investigation was to examine the impact of AJE as a phytogenic feed additive on growth performance, nutrient digestibility, excreta microbial count, noxious gas emissions, breast meat quality in broilers. About three hundred and sixty, day-old broilers (Ross 308) were assigned into four treatments (five replication cages/treatment, and 18 birds/cage). Dietary treatments: CON, basal diet; 0.02% AJE, basal diet with 0.02%; 0.04% AJE, basal diet with 0.04% AJE, and 0.06% AJE, basal diet with 0.06% of AJE. Body weight gain increased linearly (p < 0.05) through the inclusion of AJE during days 7 to 21, 21 to 35, as well as the entire experimental period. Besides, feed intake increased (p < 0.05) linearly during days 21 to 35 and the entire experiment with the increased AJE doses in broiler diet. Dry matter digestibility was increased (p < 0.05) linearly along with increasing amounts of AJE. With increasing AJE supplementation, nitrogen and energy utilization tended to improve (p < 0.10). In summary, the addition of AJE in the corn-soybean meal diet led to higher body weight gain and increased feed intake as well as enhanced nutrient digestibility, among them the highest improvement was found in 0.06%-AJE indicating the acceptance of AJE as a phytogenic feed additive.
To investigate the characteristics of plant growth and flower quality of gerbera 'Sunny Lemon' by amount of nutrient solution, young seedling plants, 'Sunny Lemon' were transplanted to rock-wool and medium of peat moss and perlite mixed with 1 to 2 and they were acclimatized in greenhouse during about 1 month. Nutrient solution supplied to the plants is sonneveld solution of 1/2 concentration and treatments launched June 24, 2010 when average plant height was $20{\pm}1cm$. Nutrient contents as a standard for starting point of irrigation by time domain reflectometry (TDR) were determined with 60-65%, 70-75%, and 80-85%. Results of growth during vegetative growth, plant height, leaf width and leaf number increased by 10% in rockwool, but they were not significantly different. As for plant growth depending on nutrient content, 80-85% treatment showed the highest values. Leaf number increased by 60%, and leaf width and plant height had a about 40% increase than initial growth. Effectiveness for flower quality, yield and days to flowering were superior when nutrient content of media was higher than in the others. Especially, average days to flowering in 80-85% content was advanced by 7-10 days compared to the day in 60-65% treatment. The total amount of nutrient supply per plant was higher in mixed medium than in rockwool, but change patterns of EC and pH were enhanced in rockwool. Based on our results, we recommended that growth, cut flower, and yield of gerbera 'Sunny Lemon' were more effective when nutrient content of mixed medium was maintained at 80-85%.

All tested Korean populations of the diamondback moth, Plutella xylostella, are known to be resistant especially against pyrethroid insecticides by mutation in its molecular target, para-sodium channel. Moreover, P. xylostella is able to develop resistance against most commercial insecticides. This study was performed to develop an efficient control technique against P. xylostella by a combined treatment of an endoparasitoid wasp, Cotesia plutellae, and a microbial insecticide, Bacillus thuringiensis. To investigate any parasitism preference of C. plutellae against susceptible and resistant P. xylostella, five different populations of P. xylostella were compared in insecticide susceptibilities and parasitism by C. plutellae. These five P. xylostella populations showed a significant variation against three commercial insecticides including pyrethroid, organophosphate, neonicotinoid, and insect growth regulator. However, there were no significant differences among five P. xylostella populations in their parasitic rates by C. plutellae. Moreover, parasitized larvae of P. xylostella showed significantly higher susceptibility to B. thuringiensis. As an immunosuppressive agent, viral ankyrin genes (vankyrins) encoded in C. plutellae were transiently expressed in nonparasitized larvae. Expression of vankyrins significantly enhanced the efficacy of B. thuringiensis against the third instar larvae of P. xylostella. Thus an immunosuppression induced by C. plutellae enhanced the insecticidal efficacy of B. thuringiensis. These results suggest that a combined treatment of C. plutellae and B. thuringiensis may effectively control the insecticide-resistant populations of P. xylostella.

To underatand in vitro regulation of differentiation, the effects of growth regulators and nitrogen source on metabolism of cell wall polysaccharides in suspension culture of kidney bean (Phaseolus vulgaris L.) were investigated. The suspension cells (cell clusters) were directly induced from the epicotyl segments of the seedlings, which were cultivated in MS medium supplemented with 1.0mg/L of 2,4-D and 0.5 mg/L of kinetin. When compared with cell wall sugar contents of the epicotyl segments, the cellulose content of the suspension-cultured cells decreased; while the pectin and hemicellulose content increased; suggesting increases of rhamnogalacturonan I and arabinogalactan IIduring the dedifferentiation, respectively, The effects of growth regulators(2,4-D, 1.0mg/L and kinetin, 0.5mg/L) and nitrogen source (potasium nitrate, 19.0mg/L and ammonium nitrate, 16.5 g/L) in the medium on the proliferation and the turnover of the cell wall polysaccharides were investigated for 30 days. In the medium with growth regulators and without nitrogen source, the proliferation rate was extremely high (16 folds). Growth regulators and nitrogen source increased the pectin content. Analysis of neutral sugar composition of pectin fraction showed that nitrogen source enhanced rhamnose level remarkably, suggesting that rhamnogalacturonan I was the one most likely synthesized. In hemicellulose fraction, growth regulators reduced arabinose level, suggesting that arabinogalactan II was degraded. And nitrogen source reduced galactose level, suggesting that xyloglucan was also degraded.
Effects of difference between day and night temperatures (DIF) and cytokinins on in vitro growth of 'Campbell Early' grape plantlets were investigated, Shoot growth was suppressed by -7DIF (18/$25^{\circ}C$). Effects of BA and TDZ were not affected by DIF. 0DIF (25/$25^{\circ}C$) enhanced the growth with increasing cytokinin level. Kinetin at 10 $\mu$M was the most effective for shoot growth with 87 mm among cytokinins used except for control. Zeatin and kinetin each at 5 $\mu$M significantly increased shoot growth to respective 162 and 110 mm. In 0DIF, two-fold increase of 162 mm was obtained by 5 $\mu$M zeatin. In control, no further shoot proliferation was observed regardless of DIF. In 0 and +7DIF (25/18$^{\circ}C$), 4 shoots were observed with 10 $\mu$M BA. Zeatin, 2iP and kinetin resulted in poor shoot proliferation while BA at 10 $\mu$M resulted in profuse branching in all DIFs.
Tomato (lycopersicon esculentum Mill.‘Sunroad’) seedlings grafted onto the four different rootstocks, ‘Anchor-T’, ‘Kagemushia’, ‘Joint’ and ‘Vulcan’, and fed with different amount of nitrogen were investigated for their susceptibility to the root rot Fusarium wilt (Fusarium oxysporum Schl. f. sp. lycopersici Snyder et Hansen, race J$_3$) disease, plant growth, and fruit quality. The flowering of the first flower cluster was promoted in grafted plants (GP) as compared to non-grafted (NG) ones. The plant height was increased by the grafting, and the growth and stem diameter were enhanced with increased nitrogen levels. Grafted plants were little affected by the Fusarium wilt disease except ‘Anchor-T’rootstock (64.7% infection). The rate of malformed, underdeveloped, and gray-mold infected fruits were reduced in the grafted plants regardless of the rootstocks. Soluble solid content, sugars, ascorbic acids and organic acids did not show any difference between grafted and non grafted plants, but ascorbic acid tends to be reduced in the high nitrogen treatment. Content of N, Ca and Mg in leaves were greatly increased in the GP grown with high nitrogen level (30kg.㏊$^{-1}$ ).
New Guinea impatiens (Impatiens hawkeri) shows very sensitive responses to different light conditions. Due to these phenomena, testers of DUS (distinctness, uniformity and stability) for granting plant variety protection right often have problems distinguishing genetic or physiological differences. New Guinea impatiens 'Fishlimp 149' was grown under several light intensities in a rain-sheltered vinyl house to observe differences of plant growth and flowering. As compared with the control (avg. $1,010{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$), treatments of shade-1 (avg. $599{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$) and shade-2 (avg. $88{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$) showed increased plant height, plant width, leaf size and pedicel length. On the contrary, these growth parameters decreased in shade-3 (avg. $30{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$) with the lowest light intensity. Shade-1 treatment enhanced flower characteristics such as flower diameter, upper petal width, side petal width and lower petal length. However, these characteristics were suppressed by lower light intensities than that in shade-1. Anthocyanin contents of shoot, leaf and pedicel decreased with increasing shading, but that of flower petal was the greatest of the shade-1 treatment. Shade-1 treatment showed the greatest Hunter a value analyzed by a colorimeter, and L and b values increased with increasing shading. Shade-1 treatment seemed to be provided the most proper light condition for DUS test of New Guinea impatiens. Additionally, anthocyanin accumulation on New Guinea impatiens during DUS test was due to not genetic differences but physiological phenomenon.
