Overexpression of Mutant Galactose Permease (ScGal2_N376F) Effective for Utilization of Glucose/Xylose or Glucose/Galactose Mixture by Engineered Kluyveromyces marxianus

  • Mutant sugar transporter ScGAL2-N376F was overexpressed in Kluyveromyces marxianus for efficient utilization of xylose, which is one of the main components of cellulosic biomass. K. marxianus ScGal2_N376F, the ScGAL2-N376F-overexpressing strain, exhibited 47.04 g/l of xylose consumption and 26.55 g/l of xylitol production, as compared to the parental strain (24.68 g/l and 7.03 g/l, respectively) when xylose was used as the sole carbon source. When a mixture of glucose and xylose was used as the carbon source, xylose consumption and xylitol production rates were improved by 195% and 360%, respectively, by K. marxianus ScGal2_N376F. Moreover, the glucose consumption rate was improved by 27% as compared to that in the parental strain. Overexpression of both wild-type ScGAL2 and mutant ScGAL2-N376F showed 48% and 52% enhanced sugar consumption and ethanol production rates, respectively, when a mixture of glucose and galactose was used as the carbon source, which is the main component of marine biomass. As shown in this study, ScGAL2-N376F overexpression can be applied for the efficient production of biofuels or biochemicals from cellulosic or marine biomass.

Biomass Estimation of Gwangneung Catchment Area with Landsat ETM+ Image

  • Spatial information on forest biomass is an important factor to evaluate the capability of forest as a carbon sequestrator and is a core independent variable required to drive models which describe ecological processes such as carbon budget, hydrological budget, and energy flow. The objective of this study is to understand the relationship between satellite image and field data, and to quantitatively estimate and map the spatial distribution of forest biomass. Landsat Enhanced Thematic Mapper (ETM+) derived vegetation indices and field survey data were applied to estimate the biomass distribution of mountainous forest located in Gwangneung Experimental Forest (230 ha). Field survey data collected from the ground plots were used as the dependent variable, forest biomass, while satellite image reflectance data (Band 1~5 and Band 7), Normalized Difference Vegetation Index (NDVI), Soil-Adjusted Vegetation Index (SAVI), and RVI (Ratio Vegetation Index) were used as the independent variables. The mean and total biomass of Gwangneung catchment area were estimated to be about 229.5 ton/ha and $52.8{\times}10^3$ tons respectively. Regression analysis revealed significant relationships between the measured biomass and Landsat derived variables in both of deciduous forest ($R^2=0.76$, P < 0.05) and coniferous forest ($R^2=0.75$, P < 0.05). However, there still exist many uncertainties in the estimation of forest ecosystem parameters based on vegetation remote sensing. Developing remote sensing techniques with adequate filed survey data over a long period are expected to increase the estimation accuracy of spatial information of the forest ecosystem.

Optimization of Medium Composition for Biomass Production of Lactobacillus plantarum 200655 Using Response Surface Methodology

  • This study aimed to optimize medium composition and culture conditions for enhancing the biomass of Lactobacillus plantarum 200655 using statistical methods. The one-factor-at-a-time (OFAT) method was used to screen the six carbon sources (glucose, sucrose, maltose, fructose, lactose, and galactose) and six nitrogen sources (peptone, tryptone, soytone, yeast extract, beef extract, and malt extract). Based on the OFAT results, six factors were selected for the Plackett-Burman design (PBD) to evaluate whether the variables had significant effects on the biomass. Maltose, yeast extract, and soytone were assessed as critical factors and therefore applied to response surface methodology (RSM). The optimal medium composition by RSM was composed of 31.29 g/l maltose, 30.27 g/l yeast extract, 39.43 g/l soytone, 5 g/l sodium acetate, 2 g/l K2HPO4, 1 g/l Tween 80, 0.1 g/l MgSO4·7H2O, and 0.05 g/l MnSO4·H2O, and the maximum biomass was predicted to be 3.951 g/l. Under the optimized medium, the biomass of L. plantarum 200655 was 3.845 g/l, which was similar to the predicted value and 1.58-fold higher than that of the unoptimized medium (2.429 g/l). Furthermore, the biomass increased to 4.505 g/l under optimized cultivation conditions. For lab-scale bioreactor validation, batch fermentation was conducted with a 5-L bioreactor containing 3.5 L of optimized medium. As a result, the highest yield of biomass (5.866 g/l) was obtained after 18 h of incubation at 30℃, pH 6.5, and 200 rpm. In conclusion, mass production by L. plantarum 200655 could be enhanced to obtain higher yields than that in MRS medium

Biotechnological improvement of lignocellulosic feedstock for enhanced biofuel productivity and processing

  • Secondary walls have recently drawn research interest as a primary source of sugars for liquid biofuel production. Secondary walls are composed of a complex mixture of the structural polymers cellulose, hemicellulose, and lignin. A matrix of hemicellulose and lignin surrounds the cellulose component of the plant's cell wall in order to protect the cell from enzymatic attacks. Such resistance, along with the variability seen in the proportions of the major components of the mixture, presents process design and operating challenges to the bioconversion of lignocellulosic biomass to fuel. Expanding bioenergy production to the commercial scale will require a significant improvement in the growth of feedstock as well as in its quality. Plant biotechnology offers an efficient means to create "targeted" changes in the chemical and physical properties of the resulting biomass through pathway-specific manipulation of metabolisms. The successful use of the genetic engineering approach largely depends on the development of two enabling tools: (1) the discovery of regulatory genes involved in key pathways that determine the quantity and quality of the biomass, and (2) utility promoters that can drive the expression of the introduced genes in a highly controlled manner spatially and/or temporally. In this review, we summarize the current understanding of the transcriptional regulatory network that controls secondary wall biosynthesis and discuss experimental approaches to developing-xylem-specific utility promoters.

FERPM을 적용한 저등급 고체연료의 기공발달 모델 특성 연구 (Study of Pore Development Model in Low Rank Solid Fuel Using FERPM)

  • Due to the increasing demand of high rank coal, the use of low rank coal, which has economically advantage, is rising in various industries using carbonaceous solid fuels. In addition, the severe disaster of global warming caused by greenhouse gas emissions is becoming more serious. The Republic of Korea set a goal to reduce greenhouse gas emissions by supporting the use of biomass from the Paris International Climate Change Conference and the 8th Basic Plan for Electricity Supply and Demand. In line with these worldwide trends, this paper focuses on investigating the combustibility of high rank coal Carboone, low rank coal Adaro from Indonesia, Baganuur from Mongolia and, In biomass, wood pellet and herbaceous type Kenaf were simulated as kinetic reactivity model. The accuracy of the pore development model were compared with experimental result and analyzed using carbon conversion and tau with grain model, random pore model, and flexibility-enhanced random pore model. In row lank coal and biomass, FERPM is well-matched kinetic model than GM and RPM to using numerical simulations.

아세톤을 사용한 목질계 바이오매스의 가용매 분해에 대한 연구

  • 목질계 바이오매스의 아세톤상 가용매분해는 목재의 구성성분인 셀룰로오스, 헤미셀룰로오스, 리그닌중에 가장 전환율이 낮은 리그닌의 전환율을 증가시킴으로 전체적인 목재의 전환율의 증가에 기여하였다 그러나 $300^{\circ}C$ 이상에서는 리그닌의 전환율이 감소하였다. 아세톤의 첨가에 관계없이 셀룰로오스와 헤미셀룰로오스의 전환율의 변화는 온도에 의존하기 때문에 주어진 열분해 온도에 따라 생성물을 선택적으로 얻을 수 있었다. 자일란의 경우 $250^{\circ}C$ 이하에서 전환되고 셀룰로오스의 경우 $250^{\circ}C$이하에서는 거의 전환되지 않지만 $250^{\circ}C$$300^{\circ}C$ 사이에서 급격한 전환율의 증가가 나타났다. 생성물은 주로 방향족류와 페놀류이며, 특히 리그닌의 아세톤 상 가용매 분해의 경우 $300^{\circ}C$에서의 메톡시 페놀류가 가장 많이 얻어졌다. $400^{\circ}C$이상의 반응에서는 메톡시기가 없는 methyl-, dimethyl-, trimethyl-, phenol 류가 생성되는 것으로 나타났다. 이것은 메톡시기를 제거시킬 만한 에너지가 $350^{\circ}C$ 이상에서 공급되는 것으로 사료된다.

북동 태평양수역에서 위도에 따른 부유 원생동물의 분포 (The Distribution of Planktonic Protists Along a Latitudinal Transect in the Northeast Pacific Ocean)

  • As a part of Korea Deep Ocean Study program, we investigated the distribution of planktonic protists in the upper 200 m of the northeast Pacific from $5^{\circ}N$ to $17^{\circ}N$, along $131^{\circ}30'W$. Area of divergence was formed at $9^{\circ}N$ which is boundaries of the north equatorial counter current (NECC) and the north equatorial current (NEC) during this cruise. Chlorophyll-a concentration was higher in NECC than in NEC area. Pico chl-a(<$2\;{\mu}m$) to total chl-a accounted for average 89% in the study area. The contribution of pico chl-a to total chl-a was relatively high in NEC area than in NECC area. Biomass of planktonic protists, ranging from 635.3 to $1077.3\;mgC\;m^{-2}$(average $810\;mgC\;m^{-2}$), was most enhanced in NECC area and showed distinct latitudinal variation. Biomass of HNF ranged from 88.7 to $208.3\;mgC\;m^{-2}$ and comprised 15% of planktonic protists. Biomass of ciliates ranged from 123.6 to $393.0\;mgC\;m^{-2}$ and comprised 25% of planktonic protists. Biomass of HDF ranged from 407.2 to $607.8\;mgC\;m^{-2}$ and comprised 60% of planktonic protists. HDF was the most dominant component in both NECC and NEC areas. Nano-protist biomass accounted for more than 50% of total protists in the both areas. The contribution of nanoprotist to total protists biomass was relatively higher in NEC area than in NECC. The biomass of planktonic protists was significantly correlated with phytoplankton biomass in this study area. The size structure of phytoplankton biomass coincided with that of planktonic protists. This suggested that the structure of the planktonic protists community and the microbial food web were dependent on the size structure of the phytoplankton biomass. However, biomass and size structure of planktonic protist communities might be significantly influenced by physical characteristics of the water column and food concentration in this study area.

Biomass and Molecular Characteristics of Multi-tillering Miscanthus Mutants

  • Compared to wide ranges of genetic variation of natural populations, very limited Miscanthus cultivar has been released. This study was the first report on the development of Miscanthus cultivar by means of radiation breeding. Seeds of M. sinensis were initially exposed to gamma rays of 250 Gy for 24 h, generated from a $^{60}Co$ gamma-irradiator. The irradiated seeds were sown and then the highly tiller-producing mutants were selected for this study. Biomass-related parameters including tiller number, plant height, stem diameter, and leaf number were measured. Ploidy level and internal transcribed spacer (ITS) were investigated to characterize the mutants compared to wild type (WT) Miscanthus. Plant height and tiller number were negatively related, where multi-tillering mutants were relatively short after 4 month growth. However stem diameter and leaf number were greater in mutants. All the materials used in this study were diploid, implying that the mutants with greater tiller numbers and stem diameter were not likely related to polyploidization. Based on the sequence of ITS regions, the mutants demonstrated base changes from the gamma irradiation where G+C content (%) was decreased in the ITS1, but increased in ITS2 when compared to WT sequence. ITS2 region was more variable than in ITS1 in the mutants, which collectively allows identification of the mutants from WT. Those mutants having enhanced tillers and allelic variations might be used as breeding materials for enhanced biomass-producing Miscanthus cultivars.

Effect of Carbon Source on the Hydrolytic Ability of the Enzyme from Fomitopsis pinicola for Lignocellulosic Biomass

  • In this study, effect of carbon source on the hydrolytic ability of the enzyme from Fomitopsis pinicola, a brown rot fungi, for lignocellulosic biomass were examined on two lignocellulosic biomasses (rice straw and wood) without any pretreatment. Cellulase activities of crude enzyme from F. pinicola, which was cultured on softwood mixture as a carbon source, were 19.10 U/$m{\ell}$ for endo-${\beta}$-1,4-gulcanase (EG), 36.1 U/$m{\ell}$ for ${\beta}$-glucosidase (BGL), 7.27 U/$m{\ell}$ for cellobiohydrolase (CBH), and 7.12 U/$m{\ell}$ for ${\beta}$-1,4 xylosidase (BXL). Softwood mixture as a carbon source in F. pinicola comparatively enhanced cellulase activities than rice straw. The optimal pH and temperature of the cellulase was identified to pH 5 and $50^{\circ}C$for the hydrolysis of rice straw. Under these condition rice straw was hydrolyzed to glucose by the cellulase up to $32.0{\pm}3.1%$ based on the glucan amount of the rice straw for 72 h, while the hydrolytic capability of commercial enzyme (Celluclast 1.5${\ell}$) from rice straw to glucose was estimated to $53.7{\pm}4.7%$ at the same experimental condition. In case of addition of Tween 20 (0.1% w/w, substrate) to the cellulase the hydrolysis of rice straw to glucose was enhanced to $38.1{\pm}2.0%$.

화학적으로 변형된 하수슬러지를 이용한 반응성염료의 생물흡착 (Biosorption of Reactive Dyes using Chemically Modified Sewage Sludge)

  • 생물흡착은 염색폐수로부터 염료를 제거하기 위한 기술로서 현재 사용되고 있는 기술을 대체할 수 있는 유망한 처리 방법이다. 본 연구에서는 생물흡착제로써 저가이면서 풍부한 소재중의 하나인 하수 슬러지를 이용하였다. 본 연구의 목적은 바이오매스의 변형을 통하여 흡착능력을 향상시키는데 있다. FT-IR 분석과 적정실험을 통하여 흡착에 관여하는 작용기는 카르복실 그룹, 인산 그룹, 아민 그룹으로 판명하였으며 그 중에서 반응성 염료(Reactive Red 4, RR 4)를 흡착할 수 있는 작용기는 아민 그룹임을 알 수 있었다. 또한 음이온성 염료인 RR 4의 흡착을 저해하는 것으로 생각되는 카르복실 그룹을 제거함으로써 흡착성능을 향상시킬 수 있었다. 그 결과, 카르복실 그룹이 제거된 바이오매스의 최대 흡착량이 변형 전에 비해 pH 2에서는 130%, pH 4에서는 210% 증가하였다. 그러므로 화학적으로 변형시킨 하수 슬러지는 산업폐수내 염료제거에 효과적이면서 값싼 생물흡착제로 이용될 수 있을 것으로 기대된다.
