PIF4 Integrates Multiple Environmental and Hormonal Signals for Plant Growth Regulation in Arabidopsis

  • As sessile organisms, plants must be able to adapt to the environment. Plants respond to the environment by adjusting their growth and development, which is mediated by sophisticated signaling networks that integrate multiple environmental and endogenous signals. Recently, increasing evidence has shown that a bHLH transcription factor PIF4 plays a major role in the multiple signal integration for plant growth regulation. PIF4 is a positive regulator in cell elongation and its activity is regulated by various environmental signals, including light and temperature, and hormonal signals, including auxin, gibberellic acid and brassinosteroid, both transcriptionally and post-translationally. Moreover, recent studies have shown that the circadian clock and metabolic status regulate endogenous PIF4 level. The PIF4 transcription factor cooperatively regulates the target genes involved in cell elongation with hormone-regulated transcription factors. Therefore, PIF4 is a key integrator of multiple signaling pathways, which optimizes growth in the environment. This review will discuss our current understanding of the PIF4-mediated signaling networks that control plant growth.

Biological Activities of Phytoestrogens in Plant and Foodstuff (작물에 함유된 Phytoestrogen의 특성과 생리활성)

  • Phytoestrogens are oestrogenic compounds found in plants and consist of isoflavones, lignans, and coumestans. The structural similarity of phytoestrogens to endogenous oestrogens has promoted the hypothesis that phytoestrogens exert hormonal or anti-hormonal effects relevant to the risk of hormone-dependent disease and/or their suitability as a dietary alternative to hormone replacement therapy. Epidemiological studies suggest that food stuffs containing phytoestrogens may have a beneficial role in protecting against a number of chronic disease and conditions. It is thought that these estrogen-like compounds may protect against chronic diseases, such as hormone-dependent cancers, cardiovascular disease and osteoporosis. Furthermore, phytoestrogens are used as a natural alternative to hormone replacement therapy and to reduce menopausal symptoms. Phytoestrogens are considered good candidates for use in natural therapies and as chemopreventive agents in adults. However safe and efficacious levels have yet to be established.

Change in Levels of Endogenous Hormone and Detection of Adventitious Bud-Related Protein during Culture of Hybrid Poplar Explants

  • Addition of plant growth hormones [0.01 mg/L NAA and 0.2mg/L benzyladenine (BA)] to a woody plant medium stimulated the adventitious bud formation of poplar explants during culture. Endogenous IAA content increased rapidly at the initial culture stage and then decreased, being followed by rapid increment again at the late culture. But the content of trans-zeatin riboside (t-ZR) increased continuously during the culture. Cytoplasmic soluble proteins were analyzed by one- and two-dimensional SDS-PAGE. Increased amount of 40 kD band was detected by one-dimensional electrophoresis using Coomassie Blue staining during the culture and two distinctive proteins whose mol wt is 40,000 were detected by two-dimensional electrophoresis using autoradiography and these proteins were synthesized continuously prior to the adventitious bud formation. When the midvein segments were transferred to the actinomycin D-containing medium, the spots of adventitious bud-related proteins(ABRPs) did not disappeared but weakened in intensity. So, it is concluded that genes coding for the ABRPs are regulated to some degree at the transcriptional level. Also, they were not observed in BA-free medium, suggesting that these proteins be regulated by cytokinin, which made then possible to form the adventitious bud.

Effect of Cucurbitaceous Crops and Rootstocks on the Xylem Sap, Amount Mineral Element and Plant Hormone (박과채소의 대목에 따른 복부분비액, 무기성분 및 식물호르몬의 함량)

  • Experiments were conducted to investigate the bleeding xylem sap characteristics, mineral contents and endogenous plant hormon contents in cucumbeer and origental melen plant grafted onto different rootstocks and various cucubitaceous crops developed for rootstocks. The volume of xylem sap, expressed as milliliter per 100g plant flesh weight, was high in the Heukzong rootstock plant as compared to the other rootstock plant,. Acidity(pH) and electric conductivity(EC) values in breeding xylem sap were not much influenced by the kind of rootstocks. Incucumber plant grafted onto different rootstocks, the contents of xylem sap in total nitrogen, phosphorus and K were increased by grafting, whereas those of Ca and Mg were decreased. Rootstocks had no influence on the total nitrogen contents in xylen sap of ‘Sineuncheon’, but decreased K Contents. Conecntration of cytokinin-like substances in origental melon xylem sap was a little higher in Sintozwa and ungrafted.

  • An immunoassay techniques using monoclonal antibodies have been developed of the quantitative analysis of endogenous hormones in plants. In order to be useful for immunoassay, both a high degree of specificity and a high affinity are required. A system is described for production of hybridomas which secrete antibodies against the plant hormone. Using the system we were able to produce hybridmas with the desired antibody specificity by cell fusion and culture method. For a number of obvious reasons, monoclonal antibodies(mAb) were superior to polyclonal antibodies.

  • Ethylene, known as a stress hormone regulate wide developmental processes including germination, root hair initiation, root and shoot primordial formation and elongation, leaf and flower senescence and abscission, fruit ripening. The acceleration of ethylene biosynthesis in plant associated with environmental and biological stresses. 1-Aminocycloprophane-1-carboxlyate deaminase(ACCD) is an enzyme that cleaves ACC into and ammonia, a precursor of the plant hormone ethylene. Plant growth-promoting rhizobacteria (PGPR) having ACCD can decrease endogenous ACC level of tissue, resulting in reduced production of ethylene in plants. ACC deaminse was a key enzyme for protect stressed plants from injurious effects of ethylene. ACCD gene was encoded from Pseudomonas flourescens, PGPR and was cloned in Escherichia coli. We expressed the recombinant ACCD(rACCD) containing 357 amino acids with molecular weight 39 kDa that revealed by SDS-PAGE and western blot. The rACCD was purified by Ni-NTA purification system. The active form of rACCD having enzyme activity converted ACC to a-ketobutyrate. The optimal pH for ACC deaminase activity was pH 8.5, but no activity below pH 7.0 and a less severe tapering activity at base condition resulting in loss of activity at over pH 11. The optimal temperature of the enzyme was $30^{\circ}$ and a slightly less severe tapering activity at 15 - 30$^{\circ}$, but no activity over $35^{\circ}$. P. flourescens ACC deaminase has a highly conserved residue that plays in allowing substrate accessibility to the active sites. The enzymatic properties of this rACCD will provide an important reference for analysis of newly isolated ACCD and identification of newly isolated PGPR containing ACCD.

Isolation of Bacillus amyloliquefaciens MJ-3 and Its Effect on the Early Growth Promotion of Red Pepper Plug Seedlings in Compost (고추 플러그묘 초기 생육을 촉진시키는 Bacillus amyloliquefaciens MJ-3의 분리 및 상토내 처리 효과)

  • The effect of useful rhizobacterium added in bed soil on the early growth promotion of red pepper plug seedlings was investigated. Total 540 colonies of rhizobacteria from 385 samples of eggplant family roots were isolated. Among these, 5 isolates were selected for antifungal activity against pathogenic fungi such as Alternaria solani, Botrytis cinerea, Fusarium oxysporium, Phytophthora capsici, and Sclerotia sclerotiorum. Of all the isolates, MJ-3 having the most pronounced growth-promoting ability for red pepper was finally selected and identified as Bacillus amyloliquefaciens through characterization of biochemical and bacteriological aspects and 16S rDNA sequence. The plant height, stem diameter, root length and fresh weight of red pepper plants which were grown with inoculation of B. amyloliquefaciens MJ-3 were higher than those without inoculation. Especially the root weight of the inoculated red pepper plant increased by 44.3%, the content of endogenous plant hormone (CA$_1$) being 0.556 ng/g (dry weight).

Changes in Mineral Uptake and Hormone Concentrations in Rice Plants Treated with Silicon, Nitrogen and Calcium Independently or in Combination (규소, 질소, 칼슘 단독 및 혼합처리가 벼 식물체 내 무기성분 흡수 및 식물호르몬 함량 변화에 미치는 영향)

  • To elucidate the physiological responses of rice plants to the essential mineral silicon (Si), we assessed the effects of treatments with Si, nitrogen ($NH_4NO_3$; ammonium nitrate), and calcium ($CaCl_2$; calcium chloride), independently or in combination on mineral uptake rates and levels of the hormones abscisic acid (ABA), gibberellin ($GA_1$) and jasmonic acid (JA). We found that nitrogen and calcium uptake was inhibited by Si application. However, solo application of nitrogen or calcium did not affect Si uptake. Compared to the untreated plants, the application of Si, $NH_4NO_3$ or $CaCl_2$ increased the endogenous hormone levels in treated plants. In particular, the concentrations of $GA_1$ and JA increased significantly after the application of Si or $NH_4NO_3$. The level of $GA_1$ observed after a treatment (solo or combine) with Si, and $NH_4NO_3$ was higher than that of the control. By contrast, independent application of $CaCl_2$ or a combined treatment with Si and $CaCl_2$ did not alter $GA_1$ levels. The highest level of $GA_1$ was present in plants given a combination treatment of Si and $NH_4NO_3$. This effect was observed at all time points (6 h, 12 h and 24 h). Endogenous JA contents were higher in all treatments than the control. In particular, a combination treatment with Si and $NH_4NO_3$ significantly increased the JA levels in plants compared to other treatments at all time points. A small increase in JA levels was observed after 6 h in plants given the $CaCl_2$ treatment. However, JA levels did not differ between plants given a $CaCl_2$ treatment and controls after 12 h or 24 h of exposure. We conclude that treatment with $CaCl_2$ alone does not affect endogenous JA levels in the short term. Endogenous ABA contents did not show any differences among the various treatments.

Changes in Endogenous Abscisic Acid, Jasmonic Acid and Sucrose Content during Bulb Development in the Cold-type Cultivar of Garlic (Allium sativum L.) of Korea (한지형 마늘의 인경 발육과정에서 식물내생호르몬 Abscisic Acid, Jasmonic Acid 및 당 함량변화)

  • This study was performed to investigate the role of plant growth substances on the bulbing of cold type of garlic (Allium sativum L. cv. Uiseongmaneul) during long and short day conditions. The change in endogenous plant hormones such as abscisic acid (ABA), jasmonic acid (JA) and sugar contents in leaf blade and sheath was examined during the growth stage from bulb differentiation (starting at April 16) to bulbing (April 24 to May 18) in cold type of garlic. In the long day condition, ABA contents were higher than short day condition and ABA contents of leaf sheath were higher than leaf blade. ABA contents of growth stage in garlic were not changed during growth stage from April 16 to May 2, however it rapidly increased during bulbing (May 2 to May 18). On the other hand, endogenous JA contents in short day condition did not change in long day condition, it increased from April 16 to May 2. JA contents in the leaf sheath (33.85-62.04 $ng{\cdot}g^{-1}$ DW) were higher than leaf blade (15.39-30.04 $ng{\cdot}g^{-1}$ DW). These results showed that garlic bulb differentiation and bulbing was induced by JA in leaf sheath. In long day condition, total sugar contents in the leaf blade were increased from bulb differentiation (April 16) to bulbing (May 4) and it was decreased during bulbing (May 4 to May 18) while the total sugar content in leaf sheath were gradually increased from bulb differentiation (April 16) to bulbing (May 18). In conclusion, our results showed that there is a significant correlation between the bulb development of garlic and hormonal content in the leaf sheath.

Regulation of Chilling Tolerance in Rice Seedlings by Plant Hormones

  • Since the major important factors limiting plant growth and crop productivity are environmental stresses, of which low temperature is the most serious. It has been well known that many physiological processes are alterant in response to the environmental stress. With regard to the relationship between plant hormones and the regulation of chilling tolerance in rice seedlings, the major physiological roles of plant hormones: abscisic acid, ethylene and polyamines are evaluated and discussed in this paper. Rice seedlings were grown in culture solution to examine the effect of such plant hormones on physiological characters related to chilling tolerance and also to compare the different responses among tested cultivars. Intact seedlings about 14 day-old were chilled at conditions of 5$^{\circ}C$ and 80% relative humidity for various period. Cis-(+)-ABA content was measured by the indirect ELISA technique. Polyamine content and ethylene production in leaves were determined by means of HPLC and GC respectively. Chilling damage of seedlings was evaluated by electrolyte leakage, TTC viability assay or servival test. Our experiment results described here demonstrated the physiological functions of ABA, ethylene, and polyamines related to the regulation of chilling tolerance in rice seedlings. Levels of cis-(+)-ABA in leaves or xylem sap of rice seedlings increased rapidly in response to 5$^{\circ}C$ treatment. The tolerant cultivars had significant higher level of endogenous ABA than the sensitive ones. The ($\pm$)-ABA pretreatment for 48 h increased the chilling tolerance of the sensitive indica cultivar. One possible function of abscisic acid is the adjustment of plants to avoid chilling-induced water stress. Accumulation of proline and other compatible solutes is assumed to be another factor in the prevention of chilling injuies by abscisic acid. In addition, the expression of ABA-responsive gene is reported in some plants and may be involving in the acclimation to low temperature. Ethylene and its immediate precusor, 1-amincyclopropane-1-carboxylic acid(ACC) increased significantly after 5$^{\circ}C$ treatment. The activity of ACC synthase which converts S-adenosylmethionine (SAM) to ACC enhanced earlier than the increase of ethylene and ACC. Low temperature increased ACC synthase activity, whereas prolonged chilling treatment damaged the conversion of ACC to ethylene. It was shown that application of Ethphon was beneficial to recovering from chilling injury in rice seedlings. However, the physiological functions of chilling-induced ethylene are still unclear. Polyamines are thought to be a potential plant hormone and may be involving in the regulation of chilling response. Results indicated that chilling treatment induced a remarkable increase of polyamines, especially putrescine content in rice seedlings. The relative higher putrescine content was found in chilling-tolerant cultivar and the maximal level of enhanced putrescine in shoot of chilling cultivar(TNG. 67) was about 8 folds of controls at two days after chilling. The accumulation of polyamines may protect membrane structure or buffer ionic imbalance from chilling damage. Stress physiology is a rapidly expanding field. Plant growth regulators that improve tolerance to low temperature may affect stress protein production. The molecular or gene approaches will help us to elucidate the functions of plant hormones related to the regulation of chilling tolerance in plants in the near future.
