Differentiation of Lentinus edodes Isolates in Korea by Isozyme Polymorphisms and Random Amplified Polymorphic DNA (RAPD) Analysis (Isozyme Polymorphism 및 Random Amplified Polymorphic DNA(RAPD) Pattern에 의한 표고 버섯 품종간 비교)

  • Sixty-three isolates of Lentinus edodes obtained from Korea were used to assess the genetic similarity by isozyme polymorphisms and random amplified polymorphic DNA patterns. The activities of esterase, peroxidase and acid phosphatase displayed 10, 7 and 3 distinct isozyme patterns, respectively. By combining the isozyme patterns obtained with the 3 enzymes, every isolate showed its own distinct electrophoretic phenotypes. A distance matrix calculated between all pairs of 63 electrophoretic phenotypes based on the presence or abscence of isozyme bands were analyzed by the group-average method. Results of the cluster analysis assinged the 63 phenotypes into six major groups. In the analysis of random amplified polymorphic DNA patterns, all isolates of Lentinus edodes were devided into five RAPD groups.

Comparisons Among the Fishes of Genus Liobagrus in Korea by Their Morphology and Electrophoretic Patterns of Proteins (形態 및 蛋白質 電氣泳動像에 依한 韓國産 퉁가리屬 魚類의 比較)

  • Two species of fishes of Genus Liobagrus that had been collected from three separate river systems in Korea were compared by their morphology and gel electrophoretic patterns of water soluble proteins. In morphology L. andersoni from Han River was distinctly different from L. mediadiposalis collected from both Gum River and Nagdong River. But L. andersoni collected from Gum River not only showed partial similarity for both groups of fishes above in their classifying characteristics but also had a unique ratio of body width/standard length. Comparable similarity and difference among the three groups were also noted in their protein patterns of SDS polyacrylamide gel electrophoresis of various tissues. The difference was clearer and more distinct in the gel run with muscle proteins. A couple of more distinctly different low molecular polypeptides were detected by two dimensional gel electrophoresis. Since the protein patterns shown in this study are not only agreeable with the morphological results but also provide detailed comparisons, fishes of Liobagrus from various water sources can be classified reliably by gel electrophoresis. On the bases of tghe findings above, L. andersoni like fishes collected from Gum River should no longer be classified as L. andersoni. Before naming them as an independent species, the possibility of natural hybrid between the two defined species, or of a varient of L. andersoni by geograpic isolation should be tested.

  • The patterns of isoperoxidase in needle-leaves of 23 species of the genus Pinus were analyzed by means of starch gel electrophoresis. Each species had a unique band pattern. In all, 56 isoperoxidase bands were identified, of which 9 to 35 isoperoxidase bands were possessed by single species. No single band was common to all Pinus species but when band patterns were grouped into 7 types, type II was considered to be the specific to genus Pinus. The results of this experiment indicated that various Pinus species had their more or less specific band patterns of peroxidase.

  • The present researches were carried out to differentiate the species of Colletotrichum by elecrophoretic method. C. gloeosporioides, C. dematium and Gloeosporium fructigenum could be differentiated by the isozyme patterns of esterase, leucine aminopeptidase, acid phosphatase and glutamic oxaloacetic transaminase. Especially. G-strain and R-strain of C. gloeosporioides were differentiated by the enzyme patterns. The G­strain damaged all stage fruits (green and red fruits) of Capsicum annum. The R-strain could not infect the unripe (green) fruits, but it could damage only ripe (red) fruit of Capsicum annum.

  • Isozyme patterns of leucine aminopeptidase, esterase and protein from 16 isolates of Ganoderma lucidum were observed by electrophoresis for characterization of the isolates. Even in the same isolate, the patterns of the isozymes from mycelium and cap were different. The esterase patterns from the mycelium could differentiate the isolates. Of 16 isolates, four isolates showed identical patterns. It was assumed that these had the same genetic background.

Effects of Whole Body Ultraviolet -Light Irradiation on Serum Protein in Snake Head, Ophicephalus argus(CANTOR) (가물치의 혈청단백질에 미치는 자외선전신조사의 영향)

  • The present paper deals with the changes in serum protein fraction, total serum protein, hematocrit, red blood cell, haemoglobin, and weight of liver or kidney : body weight ratio of irradiated and non-irradiated snake head, Ophicephalus argus(CANTOR) . Irrardiation doses are 2537$\AA$-7 minutes (7M) and 2537$\AA$-15 minutes (15M). Serum electrophoretic patterns showed a marked decrease in albumin fraction at 1, 3, 9, 12 and 15 days on 7M group and 15 M group. On both experimental groups percentage increases in $\alpha$1 and $\beta$ fractions occurred at different time periods in general but are interpreted as only apparent changes accompanying the greater albumin fall. ${\gamma}$-Globulin decreased at 1, 6 and 15 days on 7M group. and at 1, 3, 6 and 9 days on 15M group. Also, A/G ratio was significantly low in groups subjected to above conditions as compared to the controls. A/G ratio decreased at 3 and 15 days remarkably on both groups. On the average, the reductions in the A/G ratio were not proportional to themagnitude of ultraviolet-light. Total protein of serum changed according to suggestive changes in electrophoretic patterns of serum . Total protein of serum declined at 1, 3, 6, 9 and 12 day periods on 7M groups and at 1, 3, 9, 12 and 15 day-periods on 15 M group, and increased approaching control values at 15 and 18 day-periods on both groups. Hematocrit increased remarkably at 1, 3, 6 and 12 days and decreased at 9 and 15 days on 7M group, and increased throughout the sampling period on 15M . Red blood cell decreased throughout the sampling period and increased slightly at 6 days on 7 M group and decreased at 1, 6, 9, 15 and 18 days and increased remarkably at 3 and 12 days on 15M group. Hemoglobin decreased remarkably at 1 day-period and increased at other days on both groups. The liver weight was not remarkably changed after whole body irradiation on both groups. and kidney was increased from 1st day on 7M group and 3 rd day on 15M group respectively. It appears that changes in electropphoretic patterns of serum, A/G ratio, total protein of serum, hematocrit, red blood cell, hemoglobin, and liver or kidney weight act to the detriment of the animal following non-ionizing irradiation.

Proteinase activity in the isolates of Trichomonas vaginalis according to their pathogenicity (질트리코모나스의 병원성과 단백 분해 효소와의 상관성)

  • Ten axonic isolates of Trichomonns uaginolis were subcutaneously injected to the BALB/c mice in order to assess their pathogenicity by means of so-called "mouse assay" method. All the isolates revealed neutral and acid proteinase activities both in their Iysates and in culture media, but the specific activities of both proteinases in the severely pathogenic group were significantly higher than the mildly pathogenic group (p < 0.05). In the SDS-PAGE system in which the electrophoretic gels contained 0.4% gelatin as the substrate, five different handing patterns of trichomonal proteinases were detected, and the patterns were closely related with the pathogenicity of the isolates of T. vosinalis. All five bands might be regarded as cysteine proteinases group in the inhibitor assays. The cytotoxicity of the Iysates of T. vaginalis to the target Chinese hamster ovarian (CHO) cell line was also significantly different according to the pathogenicity of the isolates, and generally lower in the Iysates treated with cysteine proteinase inhibitors than in the control Iysates. In summarizing the results, it might be considered that the proteinases of T.vaginalis showing five electrophoretic banding patterns are closely related with the pathogenicity and cytotoxicity of the isolates of T. voginolis.

  • Physiological and morphological differences in larvae and female adults of Thecodiplosis Japonensis from Haenam, Chullanamdo, and Chunsung, Kangweondo, were studied by means of electrophoretic technique and scanning electron microscope (SEM). On zymograms of whole body, third instar larvae of T. japonensis showed geographic differences in the band patterns of esterase and MDH iszymes, but patterns and the staining density of general proteins were similar in tow populations. In female adults, the populations revealed geographic differences in general proteins and esterase isozymens. In external ultrastructures, especially in genital segments, each population had distinctive structures in the 2nd segment of ovipositor.

  • A comparative electrophoretic study on protein and several important enzymes of calli induced from stem, intercostal area and minor vein area were conducted in Pinella ternata (Thunb.) Breit. Soluble protein band patterns of calli induced from stem, intercostal area and minor vein area were distinctly different from those of the corresponding plant parts. Esterase isozyme patterns of calli induced from stem, intercostal area and minor vein area were different from those of the corresponding plant parts. Glutamate oxalo-acetate transaminase isozyme patterns of calli grown for 4 weeks induced from 3 plant parts were similar to those of the corresponding plant parts. But a high molecular weight isozyme band appeared in the calli grown for 8 weeks. Alow molecular weight isozyme band disappeared on the peroxidase isozyme patterns of calli grown for 4 weeks appeared on those of the corresponding plant parts.

  • The present researches were carried out to classify the species of ginseng by electrophoretic methods with isozyme patterns of LAP. esterase, GOT, phosphatase, peroxidase and proteins. All variants of Korean and Japanese ginseng had identical band patterns of the investigated enzymes in roots as well as in seeds. However, American ginseng had different patterns from those of Korean or Japanese.
