Objectives : Electroacupuncture (EA)-induced analgesia has been known to be mediated through the activation of opioid, noradrenergic and serotonergic receptors. However, little study on serotonergic mechanism has been performed in an animal model of chronic pain. The present study was designed to elucidate the type of serotonergic receptors responsible for EA analgesia in the chronic pain model. Methods : In rats with complete Freund's: adjuvant-induced inflammation and spinal nerve injury, spinal wide dynamic range (WDR) cell responses to graded electrical stimulation of afferent C fiber were recorded before and after spinal application of selective 5-hydroxytryptamine (5-HT) receptor antagonists. EA stimulation (2Hz, 0.5msec, 3mA) was applied to the contralateral Zusanli point for 30 min. Results : In both models of chronic pain, WDR cell responses were greatly inhibited after EA stimulation. EA-induced inhibition of WDR celt responses was significantly attenuated by spinal application of non-selective 5-HT receptor antagonist, dihydroergocristine Of 5-HT receptor antagonists tested, 5-HT1A (WAY 100635) and 5-HT2 (LY53857) receptor antagonists strongly reduced an ability of EA stimulation to inhibit WDR cell responses. However, 5-HT1B (GR55562) and 5-HT3 (LY278584) receptor antagonists also had weak but significant blocking action on EA-induced inhibitory effect on chronic pain. Conclusions : Dorsal hem cell responses, afferent C fiber stimulation, chronic pain, electroacupuncture, serotonergic receptors.
Background or Objectives : The purpose of this study is to review oriental medicine journal clinical study in human that have been utilize Electroacupuncture in Korea. Methods : We investigated clinical studies in relation to electroacupuncture through 70 articles from OASIS excluded studies used rats and reviews. Results : Of 162 articles, totally 70 articles selected. There was differences ways of stimulation. In 70 articles, 19 type of instrument is used. About Stimulation time, 15 minutes form 40 percent, 20 minute form 37.1 percent, 30 minute form 11.4 percent. About stimulation intensity, there was 20 articles is unknown intensity. Only 21 articles present intensity by value. In frequency, 2 Hz frequency is used the most, 21 articles. Below 10 Hz frequency is used 54.2 % in 70 articles. There was 7 articles is unknown frequency. Conclusions : The result of this study could provide useful information of field of electroacupuncture is used in oriental medicine. Studies of electroacupuncture should be carried out in many different ways.
Purpose: The purpose of this study were to examine the anti-hyperalgesic effects of combination of electroacupuncture and microcurrent on anti-hyperalgesia in local hyperalgesia zone. Methods: It used 24 rats for experiment, divided them into control group, electroacupuncture group (EA group), microcurrent group (MC group), combination of electroacupuncture with microcurrent (EA+MC group), caused hyperalgesia by injecting ${\lambda}-carrageenan$ into hindpaw. Thickness of hindpaw, mechanical pain threshold (MPT), thermal pain threshold (TPT), noxious flexion withdrawal reflex (NFR) and somatosensory evoked potential (SEP) were measured immediately after induction, at 24 hours, 48 hours and 72 hours after induction. The electrical stimulation was given once a day for three days, 20min per session. Results: Change of thickness, MPT, and TPT showed significant difference in all groups compared to control group. In particular, there were remarkable difference in EA+ME group. In particular there were remarkable differences in EA group and EA+MC group. Change of NFR(% threshold, % reaction time, % RMS) and SEP showed mainly significant differences in EA group and EA+ME group compared to control group. In particular, there were remarkable difference in EA+ME group. Conclusion: The above results suggest that appropriate combination of microcurrent with electroacupuncture for pain control will be very desirable.
Purpose : To evaluate the effects of electroacupuncture on regional cerebral blood flow (rCBF) at acupoints suggested by oriental medicine to be related to the treatment of cerebrovascuiar diseases. Materials and Methods : Rest/electroaeupuncture-stimulation Tc-99m ECD brain SPECT using a same-dose subtraction method was performed on 5 normal male volunteers (age range from 27 to 30 years) using electroacupuncture at acupoint, ST 36. In the control study, needle location was chosen on a non-meridian focus 1cm posterior to the right fibular head. All images were spatially normalized and the differences between rest and acupuncture stimulation were statistically analyzed using SPM$^{(R)}$ for Windows$^{(R)}$. Results : Electroacupuncture applied at ST36 increased rCBF in the left hemisphere, that is, the left parietal lobe(angular gyrus), the left temporal lobe, the left inferior frontal lobe around rectus gyrus and the left cerebellar hemisphere, a part of the left inferior frontal lobe. In the control stimulation, no significant rCBF increase was observed. Conclusion : The results demonstrate that electroacupuncture increases rCBF in the contralateral cerebral hemisphere.
Purpose: This study investigated the effect of electroacupuncture stimulation on changes in blood biochemistry in spinal cord injury (SCI) rats whose cords were damaged by 6-hydroxydopamine (6-OHDA). Methods: Twenty-one Sprague-Dawley adult male rats were assigned to one of three groups: normal (n=7), control (n=7) and experimental group (n=7). The experimental group received electroacupuncture (Es-160, ITO, Japan) for 15 minutes in the form of low frequency (2 Hz) stimulation to the zusanli point. After treatment, we observed motor behavior recovery using the inclined plane test. We also measured serum and CNS concentrations and activities of enzymes including creatine kinase (CK), alkaline phosphatase (ALP), and Immunoglobulin G (IgG). Statistical analysis was done using one-way analysis of variance (ANOVA). Results: Concentrations of CK, ALP and IgG were lower in the experimental group than in the control group. Functional recovery was evaluated by the maximal angle of the inclined board on which rats could maintain their initial position. This allowed us to monitor progressive locomotor recovery. The maximal angles of the inclined plane test were higher in the experimental group than in the control group (p<0.05). Conclusion: The results of this study showed that electroacupuncture to the zusanli point has a therapeutic effect on functional recovery after SCI.
The effects of electroacupuncture(EA) on gastrointestinal motility were investigated in 6 horses. Three acupuncture points ; Guan Yuan Shu(BL-26), Wei Shu(BL-21) and Da Chang Shu(BL-25) were stimulated for 20 minutes by EA at separate occasions under varying condition ; 2V-1Hz, 2V-5Hz, 2V-30Hz, 4V-1Hz, 4V-5Hz and 4V-30Hz. Myoelectric activity of stomach and cecum was monitored to investigate the gastrointestinal motility. Electromyogram(EMG) recordings were carried out before, 0, 20 minutes after and 40 minutes after the EA stimulation. EMG bipolar electrode was surgically implanted in seromuscular layer of greater curvature in the stomach and between medial band and ventral band in the cecum. The EA stimulation and monitoring were not commenced until 15 days after electrode implantation. The EA stimulation of Wei Shu influenced on stomach motility and that of Da Chang Shu on, cecum motility. However, the EA stimulation of Guan Yuan Shu influenced on both the stomach and the cecum motility. The myoelectrical spike burst amplitude of the stomach and the cecum was significantly(p<0.05) increased by 2V-1Hz stimulation, but the myoelectrical spike burst frequence of the stomach and the cecum was significantly decreased by 2V-30Hz or 4V-30Hz stimulation. The myoelectrical spike burst duration of the stomach and the cecum was significantly lengthened by 4V-30Hz and 2V or 4V-30Hz stimulation, respectively.
Skeletal muscle injury occurs frequently in sports medicine and is the most general form of injury followed by physical impact. There are growth factors which conduct proliferation, differentiation, and synthesis of myogenic prodromal cells and regulate vascular generation for the continued survival of myocytes. The purpose of the present study was to confirm the effects of electroacupuncture (EA) and electrical stimulation (ES) on muscle recovery processes according to vascular endothelial growth factor (VEGF) expression. Eighteen Sprague-Dawley rats were separated into 2 experimental groups and a controlled group. All animals had suffered from crush damage in the extensor digitorum longus for 30 seconds and were killed 1, 3, and 7 days after injury. 30 Hz and 1 mA impulsion for 15 minutes was applied to the EA experimental groups Zusanli (ST36) and Taichong (LR3) using electroacupuncture and the same stimulation was applied to the ES group using an electrical node. Hematoxyline-Eosin staining and VEGF immunohistochemistry were used to ascertain the resulting muscle recovery. There were few morphological differences between the EA and ES groups, and both groups were observed to have tendencies to decrease atrophy as time passed. In the controlled group, gradually diminishing atrophy could be observed, but their forms were mostly disheveled. There were few differences in VEGF expression between the EA and ES groups, and tendencies to have an increased quantity of VEGF with the lapse of time were observed in both groups. In the controlled group, a little VEGF expression could be observed merely 7 days after injury. In conclusion, EA and ES contributed to muscle recovery processes and could be used for the treatment of muscle injury.
Objective : Zusanli(ST36) and Hoku(Li4) are analgesic acupuncture points frequently used for acupuncture in Oriental medicine. The present study was conducted to see the antinociceptive effects produced by electroacupuncture combined two frequencies(Low, High) and two different acupuncture points(LI4, ST36) in the rat tail flick test. Method : In this study the Rats (Sprague-Dawley, 250-300g) were partially anesthetized with thiopental sodium(40mg/kg, i.p.). The basal reaction time for the tail-flick was 3${\pm}$0.5 sec. Low frequency(3Hz, 5V, biphasic) and high frequency(100Hz, 5V, biphasic) were applied to the inserted needle for the period of insertion(twenty minutes). Experimental groups are divied as follow; a) electroacupuncture stimulation groups at Hoku with or high frequency(L-EA, H-EA), b) electroacupuncture stimulation groups at Zusanli with low or high frequency(1-EA, h-EA), c) low frequency at Hoku and Zusanli(LIEA), d) low frequency at Hoku and high frequency at Zusanli(LhEA), e) high frequency at Hoku and low frequency at Zusanli(HIEA), f) high frequency at Hoku and Zusanli(HhEA) Results : The individual stimulation at either Hoku or Zusanli with low frequency has stronger and longer analgesic effect than high frequency stimulation. In addition, the combined stimulation at Hoku and Zusanli with low frequency has superior effect to individual stimulation with low frequency. LhEA and LIEA have superior effect to other stimulation groups among the combined groups. In order to determine the involvement of opioid system on the different antinociceptive effects, Naloxone, an opioid antagonist, was used in the combined groups. LIEA is the most sensitive when naloxone was administrated among study groups. HhEA is the least sensitive in the administration of naloxone. Conclusion : From results, this study confirmed that the opioid system is involved in analgesic effect of low frequency stimulation of acupuncture point, and we also can suggest the stronger analgesic effect of combining stimulation points is due to the theory of spatial summation in the nervous system.
To elucidate the effect of dorsal acupoints for canine electroacupuncture anesthesia, mongrel dogs(6 heads: 2.6-10.5kg, 3 males and 3 females) were used. After needling perpendicularly to Tian-Ping(GV-5) and Bai-Hui(GV-20), the electrodes were connected with positive to Tian-Ping and negative to Bi-hui, respectively using with veterinary electroacupuncture apparatus(TEC AM-3000). The electric conditions were 2-4V and 30 Hz. The pain of body surface was checked by hemostatic forceps. The effect of electroacupuncture andesthesia was appreciated by application of laparotomy(2 heads), ear trimming and tail amputation(1 head), castration(2 heads) and enterotomy(1 head), respectively. Induction time, clinical findings and operation findings were investigated under the electroacupuncture anesthesia. The induction time of electroacupuncture anesthesia was very rapid, approximately 1 minute and the pain of body surface including the extremities was not detected. As for clinical findings during electroacupuncture anesthesia, consciousness was evident, pupil reflex was weak and blepharoreaction was existed. As for operation findings, pain was not detected during incision and suture, and bleeding was relatively small volume. The grade of electroacupuncture anesthesia was excellent in all cases applied with ear trimming and tail amputation, laparatomy, castration and enterotomy. Considering above all findings, it was thought that electroacupuncture stimulation at Tian-Ping and Bai-hui examined in the present study induced excellent electroacupuncture anesthesia for dogs.
Objective : The aim of the present study is to investigate the effect of stimulation on different sites of the body on MBP(mean blood pressure) and HR(heart rate). Methods : Six healthy men have participated in this study. Before and after 10 min, exercise on a running machine of l0km/hr, acupressure, plain acupuncture and electroacupuncture(50Hz) stimulation was practiced on GV20, LI4, ST36, BL40 and non-acupoints on T4 and T10 respectively for 20 min. and in a control group without any treatment. The changes of MBP and HR after exercise have been observed for 20 min. at 5 minute intervals. Results : Compared with control, no significant difference was observed in research of the blood pressure measurement regardless of methods nor sites of stimulation. But there were trends of reduction in the heart rates in all experimental groups. Especially, in the group of acupressure on T10 before exercise, GV20, T10 after exercise and electroacupuncture on GV20, LI4, BL40 after exercise there was statistically significant decreases in heart rates. Conclusion : From the present experiment, it is concluded that somatic stimulation has effect on the heart rates but not on the blood pressures, and the presence of effective sites on the decreases of heart rates suggest that this effect may depend on sites of stimulation.
