Objectives : The present study was designed to investigate effects of Ssanghwa-tang (Shu$\bar{a}$nghu$\bar{a}$-g$\bar{e}$ng) on oxidation/reduction reaction-related and aging-related enzymes $in$$vitro$. Methods : We performed MTT assay, collagenase inhibition assay, elastase inhibition assay, tyrosinase inhibition assay, DPPH free radical scavenging assay, SOD-like activity and xanthine oxidase (XO) inhibition assay. Results : The 50% ethanol (EtOH) extract of Ssanghwa-tang (SHT) showed 55% inhibition of collagenase activity, and 42% inhibition of elastase activity at 1 mg/ml concentration. Also it's treatment showed 18% inhibition of tyrosinase activity, to relate whitening effect, at the same dose of 50% ethanol extract of SHT. Antioxidant activities were determined by DPPH radical scavenging, XO inhibiting activity and SOD-like activity. These scavenging, XO-inhibiting and SOD-like activities were measured in 80%, 75%, and 28% inhibitions, respectively, at a 1 mg/ml treated dose, compared to those of control. The inhibitory effects of 50% EtOH extract on aging and oxidation-related enzyme activities were higher than those of water extract and 95% EtOH extract. Conclusions : Taken together, our findings suggest that the SHT has potential and applicable benefits for development of cosmetics to have anti-aging (anti-wrinkle and whitening) and anti-oxidation functions.

Inhibitory effects of 200 medicinal plants on elastase activity were screened. Among the 200 plants, six plant extracts exhibited more than 65% of inhibition on elastase activity by their total methanol extracts at 1, 000$\mu\textrm{g}$/$m\ell$ as a final concentration. For six active plants including Areca catechu, Cinnamonum cassia, Myristica frograns, Curcumma longa, Alponia katsumadai, and Dryopteris crassirrhizoma, the inhibitory effects of their comsituents were examined on the activity of human leukocyte elastase, hyaluronidase and lipid peroxidation. In lipid peroxidation assay using TBA method, three of the 6 plants including Curcumma longa, Areca Catechu and Alponia katsumadai exhibited more than 70% of inhibition at the concentration of 1, 000$\mu\textrm{g}$/$m\ell$, but only one plant, Areca Ctechu showed high inhibitory effect on hyaluronidase activity. The results suggest that medicinal plants showing several biological activities may be potent inhibitors of anti-aging process on skin and that might be useful for application in cosmetics.

Objectives : Recently, the demands for the effective and safe depigmentating and anti-aging agents of the skin have increased due to the medical, pharmaceutical and cosmetic reasons. The aim of this study is to search new materials from the traditional herbal medicines which inhibit the aging process of skin in vitro.Methods : Human dermis cell (HS68) was used to test the effect of Rhus verniciflua Stokes (RVS). 80% ethanol or water extracts were screened for their inhibitory activities against elastase. Elastase inhibition effect was tested by microplate reader instrument. And MMP-1 suppression effect of RVS was tested by western blot. These cells were investigated the viability by MTS assay. And also the inhibition effect of tyrosinase by RVS was tested.Results : RVS (final concentrstion 1 ㎎/㎖ appeared over 30% of inhibition of elastase activity. So we are investigated anti wrinkle effects of Rhus verniciflua Stokes look through MMP-1 inhibition activity, also Extracts of RVS showed higher anti-tyrosinase activity than arbutin as final concentration 1 mg/ml. These results suggest that herbal medicines could be strong potential sources of inhibition of anti-aging and whitening effects for the skin.Colclusions : RVS was the best suppressor candidate of elastase activity among other control oriental drugs. It was found that RVS did not have toxicity to cells. We found proper concentration of RVS to treat in HS68 culture and investigated not only wrinkle care effect but also whitening effect of RVS.

Root extracts of Angelica gigas Nakai were tested to see the possibility for functional cosmetic agents. From ethanol extraction method, 97% of decursin and decursinol angelate was obtained, and concentration ratio of decursin to decursinol angelate was about 3:2. To test possibility as a functional cosmetic agent, DPPH free radical scavenging assay, UVA/B absorption, tyrosinase inhibition assay, melanogenesis inhibition assay, elastase inhibition assay and MTT assay were done. Root extracts of Angelica gigas Nakai showed $45.2{\pm}3.9%$ tyrosinase inhibition of tyrosine, and $24.2{\pm}12.0%$ melanin inhibition at $15{\mu}g/ml$ extract concentration, so that it indicated good whitening effect. DPPH free radical scavenging activity was $40.9{\pm}9.1%$ at $240{\mu}g/ml$ concentration, which is relatively good. Anti-wrinkle effect was poor such that it was $12.7{\pm}6.8%$ at $100{\mu}g/ml$. UVA/B absorption was also negligible. From the research, root extracts of Angelica gigas Nakai showed good potential as a whitening agent.
The main objective of this work is to prepare Rehmannia glutinosa extract (RE) incorporated functional chitosan (CH) based biomaterials and evaluate their physical properties, RE release properties, inhibitory effect of melanogenesis, and antioxidant and elastase inhibitory activities. RE incorporated CH based biomaterials were synthesized by a casting method and UV curing process. The surface and cross sections of prepared biomaterials were characterized by a field emission scanning electron microscope (FE-SEM). The physical properties such as tensile strength and elongation at break were also investigated. To apply the transdermal drug delivery system, RE release properties were examined with pH 4.5, 5.5, and 6.5 buffer solutions and artificial skin test at 36.5 ℃. Results indicated that RE release of RE incorporated biomaterials with/without the addition of plasticizers [glycerol (GL) and citric acid (CA)] at pH 6.5 was about 1.10 times higher than that of at pH 4.5. In addition, results of the artificial skin test verified that RE was released constantly for 6 h. To verify the applicability of the prepared biomaterials, tyrosinase, 2,2-diphenyl-1-picrylhydrazyl (DPPH), and elastase assays were investigated. Results indicated that RE incorporated biomaterials added CA exhibited tyrosinase activation, DPPH radical scavenging activity rate, and elastase activation of 45.12, 89.40, and 59.94%, respectively.

Background: To date, the anti-wrinkle efficacy of phellodendri cortex has not been defined. In this study, we investigated the nitric oxide (NO) production and elastase inhibitory activities of 80% methanol extract of Phellodendri cortex and its ethyl acetate fraction. Methods and Results: We prepared 80% methanol extract, and its fractions from phellodendri cortex. The treatment of RAW 264.7 cell with $25{\mu}g/m{\ell}$ 80% methanol extract and ethyl acetate fraction resulted in no toxicity. We conducted assays of nitric oxide (NO) production and elastase inhibition. In the NO production assay, the ethyl acetate fraction showed an inhibitory effect approximately 17 times stronger than the 80% methanol extract. In elastase inhibitory assay, the ethyl acetate fraction also showed a stronger effect than the 80% methanol extract. In order to standardize the extract and fraction, we used TLC to separate the extract and observed the plate under UV light. We confirmed that the known pharmacological ingredients berberine, and palmatine in the 80% methanol extract and the ethyl acetate fraction. Conclusions: These results indicated that phellodendri cortex extract and its ethyl acetate fraction produced strong inhibitory effect on elastase and NO production.

In this study, plant extracts including 25% Saururus chinensis, 20% Morus bombycis stem and 20% Morus papyrifera stem were tested to use as possible cosmetic agent. As cosmetic effect test, cell toxicity test, elastase inhibition assay, tyrosinase inhibition assay, anti-oxidation assay and temperature stability test were done as cosmetic assays. Plant extract showed low cell toxicity, excellent elastase inhibition and anti-oxidation effect, however, tyrosinase inhibition effect was lower. Skin, lotion and essence formulation was made using plant extracts and temperature stability test was done. Skin and essence indicated good stability, however, lotion showed phase separation and severe viscosity variation, which means lotion formulation change. From the research, plant extracts including Saururus chinensis, Morus bombycis stem and Morus papyrifera stem were suitable for possible cosmetic agent.

Objective : The aim of this study is to determine the effects of Dendrobii herba extract and Punica granatum extract on skin disease and skin beauty. Methods : To investigate in vitro anti-oxidant activity assay, ethanol extracts of medicinal plants tested by DPPH radical, xanthine oxidase activity. In the next experiment, to investigate anti-inflammatory activity assay, examined by relations in NO synthesis, IL-$1{\beta}$, IL-6, TNF-${\alpha}$, NF-${\kappa}B$, COX-2, MAP kinase. To study Skin wrinkle formation effect, we were examined by tyrosinase activities, melanin synthesis in MNT-1 cell. Results : 1. In an anti-oxidant test, Dendrobii and Punica granatum extract showed high radical scavenging activity. 2. In an anti-inflammatory test, Dendrobii herba and Punica granatum extract weakly inhibited the lipopolysaccharide(LPS)-induced nitric oxide(NO) release from RAW 246.7 macrophage cells. Dendrobii herba and Punica granatum extract also inhibited LPS-induced IL-$1{\beta}$ and COX-2 expressions. The inhibitory effect of Dendrobii herba and Punica granatum extract on macrophage activation were via the inhibition of NF-${\kappa}B$, evidenced by transient transfection assay. however, Dendrobii herba and Punica granatum extract did not have any effects about activation of Jun-N-terminal kinase(JNK) and inhibition of p38 MAP kinase in RAW 264.7 cells. 3. In the skin wrinkle formation assay, Dendrobii herba and Punica granatum extract weakly inhibited collagenase and elastase, however it was not statistically significant. 4. In the skin whitening assay, Dendrobii herba and Punica granatum extract weakly inhibited tyrosinase activity, however, it was not statistically significant. They did not have any effect on melanin synthesis, indicating that they could not be applicable for skin whitening. Conclusion : Dendrobii herba extract and Punica granatum extract may play a significant role in skin disease and skin beauty.

In this paper, we present to evaluate physiological activity of Asparagopsis armata extraction. After extraction with Asparagopsis armata using hydrothermal and supercritical carbon dioxide, various physiological activities were examined. The total concentration of polyphenol compounds was determined to be 18.85 mg/g of hydrothermal Asparagopsis armata extraction and 14.74 mg/g of supercritical Asparagopsis armata extraction. In DPPH radical scavenging assay, ascorbic acid was used as positive antioxidant control. In ABTS radical scavenging assay, ascorbic acid was used as positive antioxidant control. The percentage of inhibition and IC50 were measured. The IC50 of Asparagopsis armata extraction is 261.44ppm and the IC50 of supercritical Asparagopsis armata extraction is 153.98 ppm. The elastase inhibitory assay showed concentration dependence and the IC50 of hydrothermal Asparagopsis armata extraction is 3387 ppm and the IC50 of supercritical Asparagopsis armata extraction is higher than 2500 ppm. In mushroom tyrosinase inhibition experiments, tyrosinase inhibition's IC50 of supercritical Asparagopsis armata extraction was 248.06. In the SOD-like experiments, the concentration-dependent results were showed and IC50 of hydrothermal Asparagopsis armata extraction is 845.29 ppm. In the antimicrobial experiments, maximum clear zones of supercritical Asparagopsis armata extraction represented 23.00 mm in Propionibacterium acnes. In the other hand, in experiments with the same conditions, hydrothermal Asparagopsis armata extraction had no effect in all strains.

In this study, we investigated the anti-oxidant, anti-aging, and skin whitening effects of Korean mistletoe (Viscum album var. coloratum). The mistletoe fraction was composed of four types: hexane (HX), ethyl acetate (EA), butanol (BU), and methylene chloride (MC). In total phenol content assay, HX showed the highest phenol content among four fractions. In addition, EA significantly increased 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging and catalase-like activities, and MC significantly increased superoxide dismutase (SOD)-like activity. When we compared IC50 value in the hyaluronidase and elastase inhibition assay, MC had the lowest IC50. In addition, we also performed tyrosinase inhibition assay to demonstrate the possibility of Korean mistletoe as a cosmetic component. HX showed the highest tyrosinase inhibition rate among the fractions.
