A modified method of urea adduct formation, in which water and organic solvent were used as the wetting agent and the reaction medium. respectively, is suggested to obviate methanolysis and to ease recovery in the separation of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) from fish oil. With the new method the fraction in which the total content of EPA. DHA and their precursors is more than 80% could be obtained. Although the total content of precursors in the concentrate was hardly affected by the kind of the wetting agent or the organic solvent, the content of EPA and DHA varied significantly depending on the organic solvent. This finding made it possible to selectively enrich the desired components. After DHA-enriched fraction (I)HA is 50%) was obtained by using pentane, EPA-enriched fraction ( EPA is 53%)) could be obtained from the residue of DHA-enriched fraction by using heptane.
The quality and quantity of food organisms in fish seed production are important. The marine microalgae Nannochloropsis oculata are used as initial food organisms in the field. We investigated the effects of salinity (0, 10, 20, 30, 40 and 50 psu) on the lipid and fatty acid composition of N. oculata. Cultivation of N. oculata at varying salinities showed the highest growth rate at 20 psu. Total lipid content ranged from 17.26 to 18.63% at salinities from 0 to 50 psu). The nonpolar lipid content increased markedly at 30 psu and was highest at 15.55%. The polar lipid content was lowest at 30 psu, by 84.45%. It was also found that the omega-3 and EPA contents were inversely proportional to salt concentration. For the polar and nonpolar lipid compositions, there was no significant effect of salinity. Omega-3 polyunsaturated fatty acid content especially the content of EPA in the seawater larvae is the essential fatty acid in this food organism. It is thus advantageous to culture N. oculata at 20 psu.
Proximate and fatty acid compositions in muscle tissue were studied in 14 species of coastal and offshore fishes in Korea. Lipid content ranged from 0.57 to 10.5% and was higher in migratory and reef dwelling fishes than in demersal fishes. Protein content ranged from 14.4 to 20.3% and moisture content ranged from 72.4 to 81.8% in all fish samples. There was a negative correlation between lipid and moisture content in all fish samples (r= -0.90, P<0.001). The prominent fatty acids were 16:0, 22:6n-3 (docosahexaenoic acid, DHA), 18:1n-9, 20:5n-3 (eicosapentaenoic acid, EPA), 16:1n-7, 18:0 and 18:1n-7. Most demersal fishes contained considerably higher amounts of 20:4n-6 and/or 22:5n-3 compared with migratory and reef dwelling fishes. The proportion of total polyunsaturated fatty acid (PUFA), including DHA and EPA, was higher in demersal fishes (38.6-54.0%) than in migratory and reef dwelling fishes (23.5-35.2%).
The bacterium producing EPA was isolated from the intestines of the marine fishes. The strains were studied for their identification and their culture conditions. The selected strain was gram negative, rod(0.7 $\times$ 2.4 $\mu m$ in size) and motile with a single polar flagellum. This strain was identified as a Pseudomonas sp. on the basis of its morphological, cultural and physiological characteristics. The strain showed a maximum productivity of phospholipid at $20^{\circ}C$ after 48 hours of culture time with an initia1 pH of 7.0 in the PYM-glucose medium. Under these culture conditions, the production of phospholipid was about 0.3 mg/ml and 0.06 mg/mg dry cell weight.
This study investigated the seasonal changes of body composition and elasticity between wild and cultured brown croaker, Miichthys miiuy. The wild fish were analyzed and compared with cultured fish in moisture, crude protein, lipid, ash and many kinds of amino acids. Cultured fish was higher in moisture content and lower in crude lipid and protein content than those of wild one. The wild fish were more abundant in the total amino acid compositions than those of cultured one. As result E/A ratio there was a little significant differences between wild and cultured. In highly unsaturated fatty acid, EPA (Eicosapentaenoic Acid) and DHA (Docosahexaenoic Acid) content of cultured fish were higher than wild one. On the other hand, the gel strength, max weight and hardness of wild fish were higher than cultured one.

N-3 지방산에 관한 연구는 최초 어유에 대한 역학조사(Dyerberg and Bang, 1978; Bang et al., 1980; Hirai et al., 1980)가 계기가 되어, 오늘날 오메가-3지방산의 다양한 생리작용이 밝혀지게 되었다. (중략)
The oil in mackerel muscle was extracted using an environment friendly solvent, supercritical carbon dioxide (SC-$CO_2$) at a semibatch flow extraction process and an n-hexane. The SC-$CO_2$ was maintained at a temperature of $45^{\circ}C$ under pressures ranging from 15 to 25 MPa. The flow rate of $CO_2$ (27 g/min) was constant during the entire 2 h extraction period. The fatty acid composition of the oil was analyzed using gas chromatography (GC). Significant concentrations of eicosapentaenoic acid (EPA) acid and docosahexaenoic acid (DHA) acid were present in the SC-$CO_2$ extracted oil. The oil extracted using SC-$CO_2$ exhibited increased stability compared with n-haxane extracted oil. Particles of mackerel oil together with the biodegradable polymer, polyethylene glycol (PEG) were formed using a gas saturated solution process (PGSS) with SC-$CO_2$ in a thermostatted stirred vessel. Different temperatures ($45-55^{\circ}C$), pressures (15-25 MPa) and a nozzle size $400{\mu}m$ were used for PGSS with a 1 h reaction time. The stability of mackerel oil in the particles did not changed significantly.
Objective: The aim of this study was to investigate the effects of simultaneous supplementation of laying hens with alpha-linolenic acid (ALA) resources (flax, perilla, and Eucommia ulmoides [E. ulmoides] seeds) and eicosapentaenoic acid/docosahexaenoic acid (EPA/DHA) resources (Schizochytrium sp.) on egg quality and n-3 polyunsaturated fatty acids (PUFA) profile. Methods: Dietary treatments were as follows: i) diet C (control diet); ii) diet F (diet C+10% flaxseeds); iii) diet P, (diet C+10% perilla seeds); iv) diet E (diet C+10% E. ulmoides seeds); v) diet A (diet C+1.5% microalage); vi) diet AF (diet C+10% flaxseeds+1.5% microalage); vii) diet AP (diet C+10% perilla seeds+1.5% microalgae); viii) diet AE (diet C+10% E. ulmoides seeds+ 1.5% microalage). Results: Egg weight, yolk weight and production ratio were not significantly affected by either algae or in combination with seeds (p>0.05). No significant difference was observed in ALA and DHA concentration in eggs between flaxseed, perila, and E. ulmodies seeds supplementation alone (p>0.05). N-3 PUFA in eggs was slightly improved by microalgae supplementation. The best supplementation, a combination of microalgae and perilla seeds, elevated (p<0.05) ALA from 19.7 to 202.5 mg/egg and EPA+DHA from 27.5 to 159.7 mg/egg. Highest n-3 PUFA enrichment (379.6 mg/yolk) was observed with supplementation of a combination of perilla seed and microalgae (362.2 mg/yolk), followed by a combination of flaxseed and microalgae (348.4 mg/yolk). The ALA, EPA, and DHA content obtained with a combination of microalgae and seeds surpassed the total sum of that obtained with microalgae or ALA-seeds alone. Conclusion: It is feasible to enrich eggs with n-3 PUFAs by perilla or E. ulmodies seeds instead of flaxseeds. Simultaneous supplementation of microalgae and seeds helped improve the transfer from EPA and docosapentaenoic acid into DHA.
This experiment was conducted to examine the effects of fish oil supplementation with low does on the lipid concentration and fatty acid composition of plasma and the fatty acid composition of plasma phospholipid and erythrocyte of infants. Among 18 breast-fed infants, 6 were in control group and 12 were in fish oil groups. The subjects in fish oil groups were nursed by their mothers who supplemented with fish oil 1.96g/d or 3.92g/d, respectively for 2 weeks from 10 to 12 weeks postpartum. The nursing mothers consumed their usual diets at home. Blood samples were collected at the final day of experiment. There were no significant changes in daily intakes of total lipid, triglyceride, free fatty acid, phospholipid and cholesterol of infants by fish oil supplementation. However, the content of EPA (eicosapentaenoic acid)increased and that of ARA (arachidonic acid) decreaed significantly in plasma PC(phophatidylchline). And also, there were tendencies to increase triglyceride concentration and to decrease cholesterol and phopholipid concentrations of plasma. As the above results, atherogenic index (AI) showed a tendency to decrease, but not significant. DHA (docosahexaenoic acid) and EPA contents in plasma PC and PE (phosphatidylethanolamin) as well as those of erythrocyte tended to increase. In these results, we concluded that fish oil supplementation with low dose to lactating women does not obviously affect of the plasma lipid concentrations and fatty acid composition of plasma PC and PE as well as erythrocyte. However the increase of EPA content of plasma PC and the tendency to increase DHA and EPA contents of plasma as well as erythrocyte membrane indicate that there may be some beneficial effect on infant lipid metabolism of fish oil intake of nutsing mother were increased.
This study investigated the hypolipidemic and antithrombotic effects of linolenic acid derived from Korean perilla oil. The experimental rats(male, Sprague-Dawley) were divided into 5 groups using a Randomized Complete Block Design and fed one of the five following diets : DO*/O#. D4/O, D4/4, D4/8, or D4/20(D*/# represents the ratio of linoleic to linoenic acid as the percentage of total dietary energy intake) for 4 or 8 months. Bleeding time and whole blood clotting time were determined and the composition of serum and platelet lipids analyzed. Comparisons from the DO/O to the D4/20 group showed that serum lipids (total lipid, triglyceride, total cholesterol, and HDL-cholesterol) gradually decreased with increasing linolenic acid intake - the hypolipidemic effect. The composition of platelet fatty acids[the ratio of eicosapentaenoic acid(EPA)/arachidonci aci(AA)] increased gradually with increasing linolenic acid intake. Higher linolenic acid intake increased bleeding time and whole blood clotting time, and decreased malondialdehyde(MDA) production in the platelets, though no significant differences. These results suggest that linolenic acid derived from perilla oil appears to suppress the conversion of linoleic acid to AA and the EPA transformed from linolenic acid appears to suppress the conversion of AA to TXA2. Since TXA2 is a platelet-aggregating and vasoconstricting agent, the redulction of TXA2 released by platelets with increasing intake of perilla oil containing a lot of linolenic acid confers an antithrombotic effect.
