Gonad Structure and Reproductive Cycle of the Smallmouth Scorpionfish, Scorpaena miostoma (Teleostei: Scorpaenidae) (쭈굴감펭 (Scorpaena miosfoma)의 생식소 구조 및 생식주기)

  • Gonad structure, germ cell development and reproductive cycle of the smallmouth scorpionfish, Scorpaena miostoma were investigated based on histological method. Samples were collected monthly in the vicinity of Suyoung Bay, Pusan, Korea from November 1995 to October 1996. The testis is seminiferous tubule type in internal structure. Seminiferous tubule consists of numerous testicular cysts which contain numerous germ cells in same developmental stage. The ovary consists of several ovarian lamellae originated from ovarian outer membrane. Oogonia originated from the inner surface of the ovarian lamella protrude to the ovarian cavity in oocyte stage, and they are suspended by the egg stalk. Biological minimum size of female and male were 12.5cm in total length. Gonadosomatic index (GSI) of female (3.81) and male (0.23) were the highest in October. Reproductive cycle was classified into the following successive stages: in female, growing stage $(May\~August)$, maturation stage $(September\~October)$, ripe and spawning stage $(November\~December)$, recovery and resting stage $(January\~April)$, and in male, growing stage $(June\~August)$, maturation stage $(September\~October)$, ripe and spent stage $(November\~January)$ and recovery and resting stage $(February\~May)$.

Sexual maturation of the bluespotted mud hopper, Boleophthalmus Pectinirostris(Linnaeus) (짱뚱어, Boleophthalmus pectinirostris(Linnaeus)의 성 성숙)

  • Sexual maturation of the bluespotted mud hopper, Boleophthalmus Pectinirostris(Linnaeus) was investigated histologically on the gonadal development, and studied by gonadosomatic index(GSI), egg diameter composition. Samples were collected in the intertidal zone of Wolyon-ri, Hoihyon-myon, Okku-gun, Chollabuk-do, Korea, from April to October in 1988 and from June to August in 1989. The ovary is a pair of sac-shaped organ. The testis is a pair of tubule-shaped organ and it is connected to the seminal vesicle which is located at the posterior end of the testis. In male and female, GSI began to increase from late May when the water temperature began to increase and reached the maximum value in June and July, respectively. It began to decrease from August, the highest water temperature season. Thereafter, maintained relatively low values until October. The annual reproductive cycle of this species could be classified into four sucessive developmental stages: growing stage$(April{\~}May)$, mature stage$(June{\~}early\;July)$, ripe and spent stage(late lune-early August), degenerative and resting stage$(late\;August{\~}March:\;the wintering\;period)$. According to the frequency distributions of egg diameters in the spawning season, Boleophthalmus Pectinirostris was species to spawn twice or more in the spawning season.

Reproduction and Embryonic Development within the Maternal Body of Ovoviviparous Teleost, Sebastes inermis (란태생 경골어류 볼락, Sebastes inermis의 생식과 체내자어발달)

  • Gonadal development, fertilization and egg development in the maternal body and reproductive cycle of ovoviviparous rockfish, Sebastes inermis, were investigated histologically. Gonadosomatic index(GSI) of male and female were increased from September and reached maximum values in December. In the male, GSI decreased from January, but in the female maintained high values till February and decreased from March. Hepatosomatic index(HSI) was related to GSI conversely. In both sex, HSI increased from February and reached maximum in August as the gonad were degenerating and resting, and began to decrease from September as gonad were glowing. This ovoviviparous rockfish copulates in December. Fertilization with sperms maintained between ovulated oocytes in the ovary occurs in January mainly. Egg development in the ovarian cavity and discharging of hatched preiarva occurs from January to February. The reproductive cycle includes the successive stages: Growing(September), Mature (October-November), Ripe and Fertilization(Decembr-Janua), Egg development and Discharging of hatched larva(January-February), Degeneration and Resting(February-August). According to the frequency distribution of egg diameter and histological observation, the ovoviviparous rockfish discharged the prelarva at a time in a spawning season. The sexual maturation is first attained at 2 ages. All females and males reaches first maturity at body length of 17.1cm and 15.1cm respectively. The mean number of the embryos increased with the increase of the total length of female.

The Spawning Behavior and Egg Development of Odontobutis interrupta IWATA and JEON, 1985 (얼룩동사리 (Odontobutis interrupta)의 산란행동 및 난발생에 관한 연구)

  • Early life history and spawning for Odontobutis interrupta were observed in the laboratory during May-August 1999 with a condition of natural habitats in the field. Optimal water temperature for spawning was between 17.5 and 22.$0^{\circ}C$ and appropriate water depth and current velocity in the natural habitat ranged 0.3-0.6 m and 0.1-0.3 m/sec, respectively. Ovary maturation index peaked at about 100mm in the total length and their values gradually decreased after the size. Male fishes showed a territory and courtship behavior to adult females and the males frequently pushed upper-ventral part of females for egg releases. After females spawned, the males guarded the egg masses and supplied dissolved oxygen using pectoral fins. According to observation of egg development in the laboratory, blastodisc formed in 1hr 17 min after the fertilization, cleavaging at 36-minute interval regularly. Blastulation occurred in 7 hr 12 min after the fertilization, with gastrulation after 11 hr 11 mins and formation of york plug after 32 hr 48 min. Embryo was formed in 33 hr 45 min after fertilization and optic vesicles appeared in 47 hr 27 mins when 30-31 somites were formed. Cardiac primordium was formed in 65 hr 15mins and its beat averaged 44- 48 time/min. Pectoral fins were formed in 138 hr 40 min, air-bladder and black vesicles were observed in lower portion of young fish. Embryo hatched from she-11 membrane after about 10 days and juvenile was 5.8$\pm$0.2mm in total length 3.0$\pm$0.5mg in weight.

  • The seasonal reproductive cycle of Gomphina melanaegis collected in the coastal area of Jumunjin, between April 2006 and March 2007, was analyzed. Maturation cycle parameters such as the gonad index (GI), ovarian egg diameter, frequency of developmental stages, protein content, and RNA/DNA variation in the gonads were analyzed monthly for the 40 samples. According to the indices from histological sections, the frequency of gonad developmental stages, and the oocyte diameter, this clam has a long-term partial spawning pattern from March to October. However, GI and nucleic acid values showed that the mature stage is from March to July and that the main spawning season is August. The peak RNA and DNA contents were good indicators of sexual maturation in females and males, respectively. The variation in protein content corresponded with the RNA/DNA ratios.

Ecdysteroid Titer during Metamorphosis and the Effect of Ecdysteroid on Oocyte Develoment on Phormia regina (검정금파리의 변태기에 따른 엑디스테로이드와 난세포성숙에 미치는 엑디스테로이드의 효과)

  • The ecdysteroid titers of representive developmental stages of the blackblow fly, Phormia regina, were determined by radioimmunoassay and the effect of ecdysteroid on the oocyte maturation was investigated. Prior to every molts ecdysteroid levels began to increase sharply, suggesting ecdysteroid was the major component for egg-larval, larval-larval, and larval-pupal transformation. A difference in the levels of ecdysteroid between male and female was ob¬served during adult life span. Following the protein meal, ecdysteroid in the females increased rapidly to a maximum at 96 hr of age when terminal oocyte fully matured. Effect of ecdysteroid on oocyte development was determined for control and ecdysone-treated female flies after the liver-feeding. The growth of oocyte in the flies treated by $\mu$g of ecdysone, along with the control flies, was not facilitated. When the flies treated by 5 $\mu$g of ecdysone, however, duration of oocyte maturation was shorter than those of other two groups. This can be suggested that oocyte development in P. regina is due to the critical level of ecdysone.

Studies on Serum Estradiol (E2), Androstenedione (ADD) and Testosterone (T) Levels in the Hyperstimulated Ovulatory Cycles (과배란유도 월경주기에서 혈중 Estradiol Andrstenedione 및 Testerone농도의 동태에 관한 연구)

  • It is now common practice to attempt ovarian hyperstimulation in vitro fertilization and embryo transfer (IVF-ET) to promote the development of multiple preovulatory follicles and to maximize the number of mature egg available. There are several drugs for hyperstimulation such as clomiphene citrate only, clomiphene citrate and human menopausal gonadotropin (HMG) and HMG only. Accumlated experience has shown that the hyperstimulation of the ovary in IVF-ET results in high pregnancy rate. But the hyperstimulation of the ovary in IVF-ET may cause the hyperandrogenism, so we must consider the adverse effect on pregnancy rate of the hyperandrogenism. Little is known about the functional significance of androgen for the follicular growth, however, the hyperandrogenism might interfere with oocyte maturation. The aim of the present investigation was to determine the serum profiles of estradiol, androstenedione and testosterone during the hyperstimulated menstrual cycles in IVF. The results were summarized as follows: 1. There was a gradual increase in the mean levels of serum estradiol, androstenedione, and testosterone approaching follicular maturation. 2. The mean serum estradiol levels in the hyperstimulated groups were significantly higher than that in the control group in late follicular phase and ovum retrieval (ovulation) day (p<0.01). 3. The mean serum androstenedione levels in the clomiphene citrate groups were significantly higher than that in the control group in late follicular phase (p<0.01). There was no statistically significant different in the mean serum androstenedione levels between the control group and the HMG group (p>0.05). 4. There was no statistically significant difference in the mean levels of testosterone among each group (p>0.05). 5. There was no statistically significant different in the mean levels of estradiol, androstenedione and testosterone between the fertilized patients and non-fertilized patients in clomiphene citrate and HMG group (p>0.05).

Ultrastructural Changes of Oocytes from Vitellogenesis into Maturation in Korean Catfish, Silurus asotus (난황형성부터 성숙까지 메기(Silurus asotus) 난모세포의 미세구조)

  • This study was carried out to investigate the histomorphological changes and the electrophoretic patterns of egg components, obtained from 100 of 2-year-old female catfishes (Silurus asotus). Female Korean catfishes collected in the vicinity of Chollabukdo have synchronous ovaries which discontinously ovulate eggs during the breeding season (from late May until early July). The fishes were killed by severing the spinal cord just posterior to the head after immobilization with tricaine methanesulfonate (MS 222). Especially, the light microscopic and ultrastructural changes of ooplasm and follicular membranes of oocytes, were examined by means of light and transmission electron microscopy. The size of the nucleoli and number of the yolk granules increased as the oocyte groved. Yolk granules were deposited in the oocyte as fluid Due to tile presence of large early and late maturing oocytes, their ovaries were enlarged, transparent, granular and greenish in color. As the percentages of fish in late maturing oocyte (LMO) and ripe oocyte (RO) stage increased from March to April, mean gonadosomatic index (GSI) values (19.95%) increased. Zona radiata changed from squamous into cuboid in shape in the early maturing oocyte (EMO) stage. Processes, microvilli, from the zona radiata and from the oocyte grow, and make contact with each other in the pore canals of the zona radiata during vitellogenesis, but are withdrawn as the zona radials becomes more compact and devoid of pore canals during oocyte maturation. Seasonal changes in the microscopic appearance of the ovaries were well correlated with those in both gonadosomatic index and macroscopic appearance. The main cytological changes such as increase in size of cell, nucleus, nucleolus, and increase in number of nucleoli and mitochondria demonstrated with electron microscopy in the previtellogenic oocytes of Korean catfish, provided evidences for important synthetic processes in an early preparatory phase of oocyte development. The electrophoretic pattern of major band in mature stage was much thicker (24 k, 66 k, 90-110k dalton) than that in previtellogenic phase.

Reproductive Ecology of the Bladder Moon, Glossaulax didyma (Gastropoda: Naticidae) in Western Korea (한국 서해산 큰구슬우렁이, Glossaulax didyma (복족강: 구슬우렁이과) 의 번식생태)

  • The reproductive cycle, egg capsules in the egg-mass, first sexual maturity, and sex ratio of the bladder moon, Glossaulax didyma ($R\ddot{o}ding$) were investigated. The gastropods collected from the intertidal zone of Biin Bay, Seocheon, Korea were studied by using histological analysis and morphometric data. The gonadosomatic index (GSI) of females and males began to increase in March and reached maximum in May. Then their values sharply decreased from late in May to August due to spawning. The condition index (CI) began to increase in February and reached maximum in May, then gradually declined in the spawning period. The CI calculated for determination of the spawning period was coincided with changes in the GSI and gonadal phases. Spawning occurred between late in May to August in females and early in May to August in males. Spawning peak was observed between July and August when the seawater temperature rose to 19 $^{\circ}C$. Reproductive cycle with the gonadal development phases of this species can be divided into five successive stages in females and four in males: in females, early active stage (December to February), late active stage (February to March), ripe stage (April recovery stage (August to November); in males, active stage (December to March), ripe stage (March to July), copulation stage (early May to August), and recovery stage (August to January). Fully matured oocytes were approximately 250-270 ${\mu}m$ in size. The egg-mass was a hat in shape, and a number of egg capsules were found in an egg-mass. An egg capsule was 0.53-0.57 mm in size. An embryo (veliger larva) hatched from an egg capsule. Percentage of first sexual maturity in females and males were over 50% for individuals that are 40.1-45.0 mm in shell radius, and 100% for those that are over 45.1 mm. The sex ratio of female to male was significantly different from 1:1 $(x^2\;=\;57.22,\;p\;<\;0.05)$.

Maturation Induction of Mouse Immature Oocytes by Fusion (생쥐 미성숙 난자의 융합에 의한 성숙 유도)

  • The research of fused oocytes was conducted to investigate the in vitro mejotic maturation of immature oocytes (GV oocytes) fused with oocytes in germinal vesicle breakdown (GVBD oocytes) in the presence of dbcAMP which is known as one of the strong inhibitors to GVBD. The immature oocytes fused together as well as those fused with GVBD oocytes proceeded to GVBD in 3 hr culture in plain medium. But in the medium containing dbcAMP (100$\mu$g/ml), the immature oocytes fused together did not show any GVBD and thus the fusion itself could not affect the inhibitory activity of dbcAMP. However, all of the immature oocytes fused with GVBD oocytes underwent GVBD in 3 hr culture despite of the presence of dbcAMP. When the culture was extended to 20 hr, nearly all of the immature oocytes fused together were still arrested at the GV stage in the presence of dbcAMP. But most of the fused oocytes which had shown GVBD during 3 hr culture developed to metaphase II stage extruding one or two polar bodies regardless of the presence of dbcAMP. In this experiment, it was found that two sets of the metaphase chromosomes were somewhat concomitant with a pair of the polar bodies in the fused egg. Upon the results of the present studies, it is assumed that there may be a maturation promoting factor(s) in the cytoplasm of the GVBD occytes, and this factor(s) possibly nullifies the function of dbcAMP.
