Removal Effects of Microorganism and Pesticide Residues on Chinese Cabbages by Electrolyzed Water Washing (전기분해수 세척에 따른 배추의 미생물 및 잔류농약 제거효과)

  • This study investigated the washing efficiency of electrolyzed water for the removal of microorganisms and pesticide residues from Chinese cabbage. Initial total bacteria and coliform counts were 6.64 and 3.56 log cfu/g respectively. After washing, total bacteria count of tap water (TW) were 5.97 log cfu/g and low alkaline electrolyzed water (LAlEW) and strong acidic electrolyzed water (SAcEW) were 1.63-4.67 log cfu/g. Especially SAcEW-100 was found to the most effective method of washing the cabbages. After washing, the coliform count was dramatically reduced. The removal rate of pesticide residues by NaClO treatment (36.93-50.13%) was greater than that of TW treatment (32.28-38.46%). The removal rate of LAlEW-100 and SAcEW-100 was 63.79 and 78.30% respectively, and was higher than those of TW and NaClO treatments. The vitamin C content of the Chinese cabbages after all treatments did not differ significantly. Consequentially, the electrolyzed water was found to be effective to remove bacteria and pesticide residues from Chinese cabbage without affecting quality.

Antimicrobial Activities of Berry Extract of Domestic Plants on 4 Kind of Pathogenic Microorganism (4종의 식품 부패 미생물에 대한 국내산 자생 식물 열매 추출물의 항균성 탐색)

  • To select new useful domestic plants with antimicrobial activities, thirty five samples of berry plant were distributed from Plant Diversity Research Center in Korea Research Institute of Bioscience and Biotechnology. Their extracts with methanol were tested against Escherichia coli, Salmonella typhimurium, Staphylococcus aureus, Listeria monocytogenes by paper disc method. The methanol extracts from Viburnum dilatatum, Viburnum erosum, Aralia continentalis, Camellia japonica, Acer trumcatam, Arasaema takesimense and Rhysocarpus intermedius were effective against S. aureus and the results were as follows: Viburnum erosum 6.5 mm, Viburnum dilatatum and Aralia continentalis 7.0 mm, Rhysocarpus intermedius 8.0 mm, Acer trumcatum 9.0 mm, Camellia japonica and Arisaema takesimense 9.5 mm. And only the methanol extract from Camellia japonica was effective against L. monocytogenes, 7.0 mm. Antimicrobial activities of E. coli and S. typhymurium were not detected. The minimum inhibitory concentration(MIC) of berry of Camellia japonica was examined as 1,250 $\mu\textrm{g}$/mL against S. aureus and as 1,250∼2,500 $\mu\textrm{g}$/mL against L. monocytogenes.

Improvement of the Phosphate Solubilization Microorganism by the Introduction of Glucose Dehydrogenase Gene into Aeromonas hydrophila DA33. (Glucose dehydrogenase 유전자의 Aeromonas hydrophila DA33으로의 도입에 따른 인산가용화 균주의 개량)

  • Aeromonas hydrophila DA33 was isolated from cultivated soils as a bacteria having high abilities to solubilize inorganic phosphate. Glucose dehydrogenase gene (gdh) was cloned from Escherichia coli. The recombinant plasmid, pGHS containing glucose dehydrogenase gene was introduced into A. hydrophila DA33 in order to improve the activity of phosphate-solubilizing. The transformant harboring the gdh gene, A. hydrophila pGHS/DA33 increased enzyme activity. The strain also increased the gluconic acid generation that was effective for phosphate solubilization. It was possible that the strain containing pGHS produced higher solubilized phosphate with tri-calcium phosphate as the unique (P) source, in comparison with that of wild type without plasmid. These results suggest that the strain, A. hydrophila pGHS/DA33 is expected as effective biofertilizer for phosphate solubilization.

Control Effects of Micromonospora sp. AW050027 by Media Optimization and Microbial Treatment Against Pine Wood Nematode (Micromonospora sp. AW050027 균주의 배지최적화 및 미생물제제 처리에 의한 소나무재선충 방제효과)

  • Nematicidal activity against pine wood nematode, Bursaphelenchus xylophilus, was tested in the pot and field by the treatment of microbial formulation after media optimization. The optimized media composition was glycerol 10 g, soybean meal 10 g, NaCl 1 g, $CaCO_3$ 2 g, $K_2HPO_4$ 0.125 g per liter and microbial complex formulation was made with liquid and powder type. Most effective antibiotics against symbiotic microorganism with nematode, kanamycin, was added to the formulation. The control effects against pine wood nematodes were checked by pot test and field test. In the result of treatment by trunk injection, five times treatment was more effective than one time and the treatment with the formulation of concentrated culture supernatant was the most effective in the nematicidal activity showing below 10% mortality in pine tree.

Inhibition of Metarhizium anisopliae infection of Protaetia brevitarsis seluensis larvae using several effective microorganisms

  • The purpose of this study was to determine the best method for minimizing the occurrence of Metarhizium anisopliae infection of Protaetia brevitarsis seluensis during mass breeding on agricultural farms. There is a high demand for the use of P. b. seluensis larvae in animal feed and as food for humans. However, mass breeding results in the entomopathogenic fungal (usually M. anisopliae) infection of P. b. seluensis. A mixture of microorganisms (Bacillus subtilis, Lactobacillus plantarum, and Saccharomyces cerevisiae) delayed fungal infection by M. anisopliae, which infected fewer P. b. seluensis when the microorganism mixture was added to sawdust as feed for P. b. seluensis. When sawdust with the effective microorganisms (EM) was given to P. b. seluensis for 30 d, their mortality rate was approximately 35 % less than that of the control group, which was fed sawdust without the EM. In addition, the growth of M. anisopliae on agar media spread with each bacterium as inhibited by up to 80 % more than those spread with 4 % sodium hypochlorite, which is a harmless fungal inhibitor generally used in agricultural farms for disinfection.

Applications of Agro-Based Materials for Water Dropwort (Oenanthe stolonifera DC) Organic Farming (미나리 유기재배를 위한 활용자재 시용효과)

  • Organic farming is a type of agricultural practices based on naturally occurring processes excluding or strictly limiting the use of synthetic fertilizers, pesticides, and other chemicals. This study was conducted to investigate the influences of agro-based materials, effective microorganisms (EM), liquid silicate (LS), and organic liquid fertilizer (OLF) for water dropwort (Oenanthe stolonifera DC.) cultivation. Soil pH, soil organic matter, and plant available phosphorous decreased with LS application. Exchangeable Ca and Mg decreased with EM application, and electrical conductivity and exchangeable Ca and K decreased with OLF application. Most of essential nutrient contents in water dropwort were reduced with the treatments of LS, EM, and OLF as compared with those in control plot, except nitrogen and phosphorus. However, diseases and insect pests were almost not observed in the water dropwort in the agro-based material application plots, except cluster caterpillar (Spodoptera litura). Productivity of water dropwort tended to be reduced: its higher productivity in the OLF and EM+LS plots and lower in the LS and control plots.

Natural Purification Treatment using Soil Brick with Combined Effective Microorganisms and Emergent Plants (복합유용미생물 및 수생식물을 활착시킨 흙블록을 이용한 자연정화 처리방법 연구)

  • In this study, using soil brick with combined effective microorganisms and emergent plants was identified which it can increase the effect of conservation and improvement of water. Lab-test was consist of four kind of reactors and each of reactors were A(rawwater), B(soil brick), C(emergent plant) and D(soil brick+emergent plant). Iris pseudoacorus, Phargmites australis, Typha angustifolia and Zizania latifolia were used for emergent plant. Evaluation of application on various environment were performed on agricultural waterway and pond. The pH measurement test of soil brick was performed due to evaluate whether a strong alkaline water flows out of the soil brick. Result of lab-test, removal efficiency of D was better than removal efficiency of A presenting 20.9%, 27.9% 21.5%, 33.8% and 58.4% for $COD_{Cr}$, $BOD_5$, TN, TP and TSS respectively. Removal efficiency of soil brick on agricultural waterway was revealed to be 49.5%, 45.0%, 43.7%, 37.3% and 28.6% for $COD_{Cr}$, $BOD_5$, TN, TP and TSS respectively. And removal efficiency of soil brick on the pond was revealed to be 12.7%, 10.5%, 9.32%, 10.4% and 36.3% for $COD_{Cr}$, $BOD_5$, TN, TP and TSS respectively. Result of pH measurement test of soil brick was neutral which was about 6 to 8.

Effect of Seafood Amino Acid Fertilizer and Korean Effective Microorganisms on the Fruit Quality of Fuji Apple (해양부산물 아미노산액비 및 유용미생물(KEM) 시용이 후지사과의 품질에 미치는 영향)

  • This study was carried out to compare and analyze the effect of Korean effective microorganism(KEM) and seafood amino acid fertilizer(SAF) application on apple on functional substances of fruit. Vitamin C in fruit of control and KEM/SAF treated were 29.3 and, 39.8 mg, respectively. Also active oxygen scavenging ability of those treatments measured with DPPH method were 40.6 and 54.1%, respectively. Vitamin E contents of those treatments were 2.51 and 2.50 mg, respectively. Sitosterol, a phytosterol, contents of those treatments were 4.79 and 5.41 mg, respectively. Proportion of sugars, fructose, glucose, and sucrose in the fruit of control were 36.1, 15.3 and 17.8%, respectively. Those of the fruit of KEM/SAF treated were 45.9, 13.2 and 18.8%, respectively. Malic acid contents of control and KEM/SAF treated fruits were 53.7 and 43.8%, respectively.

The Antimicrobial Effect of Water Soluble Chitosan (수용성 키토산의 항균효과)

  • Structure of water soluble chitosan (WSC) was confirmed by Fourier transform infrared spectrometer (FT-IR), X-ray diffractometer and thermal analyser. The viscosity average molecular weight of WSC ranged from $3.0{\times}10^{4}$ to $4.5{\times}10^{4}$. Using the WSC having viscosity average molecular weight of $3.0{\times}10^{4}$, the antimicrobacterial effects against microorganism and oral microorganism showed 81.7% and 80.6% for Staphyloccus aureus and Bacillus subtilis, respectively, while the anitmicrobacterial effect exhibited 100% and 73.8% against Streptococcus mutans and Streptococcus sanguis, respectively. Therefore it is concluded that WSC is more effective against oral microorganism that microorganism in terms of antimicrobacterial effects. WSC sample with the viscosity average molecular weight of $4.5{\times}10^{4}$ exhibited a half of the antimicrobacterial effect of the low MW sample, indicating that the WSC with low MW was better than that with high MW. Chitin and chitosan showed a drastic decrease of acidity from pH 7.0 to 4.9 after 8 minute incubation time and reached an equilibrium after that. WSC, however, restrained pH of the sample from lowering up to about 16 minutes of incubation and reached an equilibrium after that. WSC obviously showed a buffering effect against pH change.

Arthroscopic Treatment of Septic Knee Arthritis in Old Aged Group: Prognostic Factor (50세 이상에서 발생된 화농성 슬관절염의 관절경적 치료: 예후 인자)

  • Purpose: The purpose of this study was to assess the result of arthroscopic treatment in septic knee arthritis and evaluate the prognostic factor over 50 years old. Materials and Methods: Fifty-two patients were treated by arthroscope for septic knee arthritis from January, 2002 to August, 2005. The mean follow-up period was 27.5months. We assessed Lysholm score as functional result, CRP normalized period as laboratory result, and knee range of motion as clinical result. We evaluated patient's age, underlying disease, causative organism, previous knee status (Kellgren stage), clinical status of septic arthritis (G$\ddot{a}$chter stage) and history of intra-articular injection as prognostic factor. Results: Mean Lysholm score was improved from 40.7 to 67.1. And the mean CRP normalized period was 38.7days. At last follow-up, almost patient (92%) were recovered to prior knee full range of motion and 45 patient (74%) were completely cured by one stage operation. The microorganism isolated were MSSA (n=13), MSSE (n=3), MRSA/MRSE (n=4),no microorganism (n=27) and others (n=5). In Lysholm score, young age (42.8(preop.)$\rightarrow$83.5(postop.)), Kellgren stage 0 ($45.5{\rightarrow}84.2$), G$\ddot{a}$chter stage I ($39.3{\rightarrow}73.1$) and no microorganism (442.1{\rightarrow}72.6$) were more increased than old age (439.3{\rightarrow}61.7$), Kellgren IV ($28.3{\rightarrow}43.7$), G$\ddot{a}$chter stage IV ($40.2{\rightarrow}67.1$) and MRSA/MRSE ($40{\rightarrow}58.75$). In case of old age (42.3days), G$\ddot{a}$chter stage IV (55.5), Kellgren stage IV(43.7), DM patient (42.1) and intra-articular injection history (52.1), the CRP titer normalized period was longer than mean period. MRSA/MRSE(n=3,75%) were not normalized in CRP titer at last follow-up. Conclusion: Arthroscopic treatment of septic knee would be an effective and satisfactory procedure. Age, previous knee status (Kellgren stage), underlying disease (DM), intra-articular injection history, microorganism and Ga¨chter stage effect end result outcome.
