This study was designed to prepare an animal model for partial thickness bum wound which can be employed for testing topical therapy. We first evaluated whether rabbit ear and mouse back skin wound model could differentiate the wound healing process in terms of degree of re epithelialization, required days for complete wound closure, presence of scarring. $2^{nd}$ degree wet bum were prepared on mouse back skin and rabbit ear by applying 5 mL hot water($85{\pm}0.1^{\circ}C$) for 7 see followed by 5 mL ice-cold 0.5% acrynol solution for cooling and disinfecting the inflicted area. After removing the dead epidermis layer at 24 hr, tested dressings were applied for specified time and wound progression was investigated. In mouse model, wound contraction was the primary wound closing mechanism, which is quite different from human wound healing process. In rabbit ear model, epidermal regeneration was the major wound healing process rather than wound contraction and the difference in wound healing property among tested dressings could be clearly demonstrated. A rabbit ear model could differentiate the wound progression among open, occluded and epidermal growth factor(EGF) treated wound. Four sites of circular wound(diameter: 1 cm) on the anterior part of rabbit ear could be employed for the comparative wound healing study. For obtaining reproducible bum wound, degree of bum depth and bum sites should be carefully controlled in addition, employing rabbits of same strain and weight. The result suggests that rabbit ear could be employed as a reliable and human-resembled wound model.
Scutellaria baicalensis Georgi (SB) has been widely studied to treat inflammatory diseases in east Asia. In the recent years, many studies have focused on modifying herbs to increase the pharmacological effects. Roasting alcohol absorbed SB is one of the traditional methods to increase the therapeutic effects. Currently there are no reports on the pharmacological effects of roasted SB. This study investigated the anti-inflammatory effects of roasted 30% ethyl alcohol absorbed SB extract (SR) on mice ear edema. After intra-gastric injection of dexamethasone (for positive control, 2 mg/kg) and SR (50, 100, 400 mg/kg), ear edema was provoked by croton oil (5% v/v in acetone, 10 ul/ear). Ear thickness was measured with a digital caliper to quantify the change in swelling. For histological study, we made paraffin sections and performed Phloxine-Tartrazine staining and Masson's trichrome staining to observe epidermis, dermis and subcutaneous region and collagen fiber of mice ear tissues. Ear thickness decreased dose-dependent manner in SR treated groups. Histological analysis compared with dexamethasone treated group, SR treated groups demonstrated a similar reduction in hypoplasia of epidermis and influx of inflammatory cells. Increase of subcutaneous layer and decrease of collagen fibers were significantly recovered in SR treated group (400 mg/kg) and dexamethasone treated group. In conclusion, treatment with SR ameliorates auricular inflammation induced with croton oil in mice. Experiments are now underway to understand the molecular mechanisms underlying anti-inflammatory activities of SR.
Finite element simulation with experimental analysis of Taguchi's orthogonal array was carried out to know the effects of material and forming parameters on the cup earing and skewness. It was revealed that the planar anisotropy was the most influencing factor in the cup ear formation whereas blank holding force and material properties such as strength and thickness deviation at the coil edge had a relatively high effect on the cup skewness.
The fluoroquinolones have been reported to cause, although at low frequency, severe phototoxicity which is due to singlet oxygen produced by ultraviolet-A (UVA; 320-400 nm) exposure. The objective of this study was to evaluate the phototoxicity based on plasma and tissue concentrations of commonly prescribed fluoroquinolones; lomefloxacin (LFLX), enoxacin (ENX), ofloxacin (OFLX), and ciprofloxacin (CPFX). The phototoxic potentials were investigated by measuring increments in ear thickness, 24 hrs after these fluoroquinolones were orally administered to Balb/c mice, which they were exposed to UVA 17.5 J/$\textrm{cm}^2$ for 2 hrs following drug administration. The fifty percent ear thickness increment-inducing doses ($ETID_{50}$), determined by single ascending dosing of each fluoroquinolone to mice, were calculated to be 50(LMFX), 250(ENX), 770(OFLX), 1100(CPFX) mg/kg. Post the administration of ETID$_{50}$, drug concentrations in plasma and ear tissue were measured at specified times and phototoxicities were quantified. Both peak plasma ($\mu\textrm{g}$/ml) and ear tissue ($\mu\textrm{g}$/g) concentrations were summarized as follows; 7.3/1.4 for LMFX, 15.0/1.6 for ENX, 90.1/18.4 for OFLX and 87.2/3.7 for CPFX. The degree of photo toxicity was more relevant to plasma concentrations than tissue concentrations. In order to assess the effect of irradiation time after drug administration on phototoxicity, the 2 hr UVA irradiation was given at 0, 1, 2, 3, and 5 hr after administering $ETID_{50}$, respectively and photo toxicities were evaluated. The shorter inteval between dosing and UVA exposure was, the higher risk of phototoxicity was produced.d.
In the present study, the anti-inflammatory effect of 'Jungcheonhwadamgangki -tang' water extract was tested in Xylene-Application mouse ear acute inflammation model. The test articles were once dosed before Xylene-Application, and the changes on body weight and ear weights and histopathological observation of induced ear were conducted with ear histomorphometry. The increases of absolute and relative ear weight detected in vehicle control compared to that of sham , were significantly and dose-dependently inhibited by Jungcheonhwadarn -gangki-tang in the present study. A classic acute inflammatory histological changes such as subcutaneous edema, hypertrophy and infiltration of inflammatory cells, was detected in vehicle control. However, these histological changes were significantly and dose-dependently inhibited by Jungcheonhwadam-gangki-tang. In addition, the increases of ear thickness half and thickness full detected in the vehicle control, were also dose-dependently decreased in the all Jungcheonhwadamgangki-tang-dosing groups. Base on these results, it is concluded that Jungcheonhwadamgangki-tang water extracts have clear anti-inflammatory effect on the acute inflammation, and about 500 mg/kg of Jungcheonhwadamgangki-tang water extracts have similar effect compared to that of Dicrofenac 15 mg/kg.

However, somewhat lower anti-inflammatory effects were detected in Haepyoeiin-tang plus Antler water extracts compare to that of Dethamethason and Dicrofenac. About 500 mg/kg of Haepyoeiin-tang plus Antler water extracts have similar effect compared to that of Dicrofenac 15 mg/kg.
두충(잎, 껍질, 줄기)의 추출물이 DNCB로 감작된 4주령 BALB/c mouse 암컷에 유도된 접촉성 피부염의 억제효과를 조사하였다. 림프절, 비장 및 흉선의 무게는 두충 투여군이 DNCB 대조군보다는 낮게 나타났다. 두충 부위별 추출물을 투여한 군에서 귀의 무게는 두충 추출물의 농도가 높을수록 감소하는 경향을 보였으며, 대조군과 비교하여 무게에 함량 변화가 있었다. 두충의 부위별 추출물을 1,000 mg/kg 농도로 투여하면 귀의 무게는 대조군과 유사한 수준까지 낮아졌으며, 귀의 두께는 두충 추출물을 투여한 군에서 시간이 경과할수록 두께가 감소하는 경향을 나타내었다. MDA 함량은 DNCB 대조군과 두충 투여군을 비교하였을 경우에, 간 조직에서 차이가 나타나지 않았으나, 염증이 발생한 귀 조직에서는 차이가 나타났으며(p<0.05), NO 함량은 SW, ST 및 BT 그룹에서 염증이 억제되어 대조군과 마찬가지로 측정되지 않았다.
The effects of DHU001, a mixed herbal formula consisted of 7 types aqueous extracts for treating respiratory disorders were observed on xylene-induced acute inflammation. The xylene was topically applied 60 min after administration of 500, 250 and 125 mg/kg of DHU001, and all animals were sacrificed 2 hrs after xylene application. The changes on ear weights, histolopathological analyses of ear were evaluated and compared to those of indomethacin and dexamethasone(15 mg/kg treated)-Both of drugs are well-known by anti-inflammatory agents. Xylene application resulted in marked increases in induced ear weights as compared with intact control ear. Severe vasodilation, edematous changes of ear skin and increase in the thickness of the ear tissues, neutrophil infiltration as acute inflammation were detected in xylene-treated control ears at histopathological observation. However, these xylene-induced acute inflammatory changes were dose-dependently decreased by oral treatment of DHU001. Therefore, it is concluded that DHU001 has favorable anti-inflammatory effects on xylene-applicated acute ear inflamed mice.
This study was carried out to identify the characteristics of 37 corn hybrids, which composed with 18 yellow colored kernel, 9 purple colored kernel, 8 white colored kernel. Days to silking of three colored waxy corn hybrids ranged from 61 to 73 days after sowing as midium-late ecotype. Ear length of hybrids ranged from 10.3 to 22.7 cm, and among them a total 22 waxy corn hybrids including CNU038 showed over 20 cm in ear length. The fresh weight per ear ranged from 58 to 289 g as the 32.9% of large coefficient of variance. In there results, we found yellow colored waxy corn hybrids showed relatively longer in ear length and heavier in fresh weight per ear compare to other colored waxy corn hybrids. In correlation relationships, plant height, ear height, stem diameter, ear length and ear weight showed positive correlation between themselves, while tip filling ratio did negative correlation with silking stage, plant height, ear length, ear weight and kernel thickness. Also, 100 kernel weight did positive correlation between traits related to ear and kernel.
Objectives The purpose of this study was to investigate the effects of AI on AD induced by DNCB in mice. AI has antiallergic property that is useful in treating allergy-related-diseases, such as asthma, anaphylactic shock, acute bronchitis and skin diseases, skin pruritus from gastrointestinal diseases. However, AI has not been studied intensively yet regarding anti-inflammatory effect on AD. Therefore, this study was conducted on 2,4-dinitrochlorobezene (DNCB)-induced mice to investigate effects of AI in AD. Methods In the experiment, we divided mice into four groups: a normal group (NOR), a control group (CON), an AI spread group (AI spread), and an AI spread and feeding group (AI spread & feeding). Then examined the changes in the body weight, weights of spleen and ear, thickness of dorsum skin and ear skin, clinical aspects on dorsum skin, historical assessments, proliferation of splenocytes in vitro and in vivo, and cytokine (TNF-${\alpha}$, IL-10). Results From the experiment, the ear weight of AI spread & feeding group was significantly dropped and the ear thickness of both AI spread and AI spread & feeding were decreased significantly. Dorsum skin thickness was also decreased significantly in both AI spread and AI spread & feeding group. Also, AI treatment improved the symptoms of AD, such as coloration, erythema and desquamation and had a better effect on AI spread & feeding group. In histopathological observation, thickened epidermis, hyperkeratosis, pigmentation, hypergranulosis, parakeratosis were diminished as well in both AI spread and AI spread & feeding group. In vitro, we could observe when AI was increased as proliferation rate of splenocytes were increased, too. Conclusions In conclusion, these data suggest that AI can decrease symptoms of AD and show AI can be useful herbal therapy for AD.
