A Study on the Anti-Inflammatory Effect of Gyejigeojakyakgabuja-tang (계지거작약가부자탕(桂枝去芍藥加附子湯)의 항염증 효과에 대한 연구)

  • Objectives This study was conducted in in vitro environments to investigate the anti-inflammatory effect and expression mechanism of Gyejigeojakyakgabuja-tang (GJBT). Methods In the experiment, an inflammatory response was induced with lipopolysaccharide in macrophage RAW 264.7 cells treated with GJBT, and the production of nitric oxide (NO), prostaglandin E2 (PGE2), and cytokines were measured, and inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) were measured. The intracellular gene and protein expression levels of cytokines were measured. Results In vitro, GJBT significantly reduced intracellular NO, PGE2, and cytokine production in a concentration-dependent manner. The intracellular iNOS, COX-2, and Cytokine gene expression levels were significantly decreased, and the intracellular iNOS, COX-2, and cytokine protein expression levels were significantly decreased in a concentration-dependent manner. Conclusions These results show that GJBT has an anti-inflammatory effect.

  • Este estudo tem por objetivo analisar, do ponto de vista da alquimia, O Alquimia, considerado uma das obras mais representativas do escritor brasileiro Paulo Coelho. Para tanto, começamos a observar as características da alquimia à qual o próprio autor disse que dedicava 11 anos para estudá-la por volta da década de 1970 quando o mundo estava sofrendo grandes mudanças sociais. Desta observação, entendemos que, sendo uma projeção do 'inconsciente' conforme diz C. G. Jung, a essência da alquimia consiste resumidamente nos seguintes conceitos: união de elementos opostos, transformação e salvação. A partir daí, tentamos estudar como esses coneitos estão diluídos na estrutura de O Alquimia e como a experiência do Paulo Coelho com a alquimia interioriza-se no mundo literário dele. E entendemos que os conceitos simbólicos da alquimia e as mensagens que a experiência do escritor com a alquimia configura em suas obras literárias compartilham a quase mesma dimensão semântica. Percebemos também que em alguns aspectos as idéias do escritor consolidadas em torno da sua pesquisa da alquimia vão às vezes além dos conceitos genéricos da alquimia, atingindo assim uma dimensão bastante peculiar e fundamental para compreender melhor o seu universo literário: para ele que se declara católico, Deus não é católico nem islâmico, nem budista, nem hinduista. Por exemplo, ele diz que Deus é um só e é que o Seu nome varia dependendo da religião, porque cada religião precisa escolher um nome para chamá-lo.

The Anti-inflammatory Mechanism of the Peel of Zanthoxylum piperitum D.C. is by Suppressing NF-κB/Caspase-1 Activation in LPS-Induced RAW264.7 Cells

  • Zanthoxylum piperitum D.C. (ZP) peels has been used as a natural spice and herb medicine for hypertension reduction, for strokes, and for its anti-bacterial and anti-oxidant activity. However, the anti-inflammatory mechanisms employed by ZP have yet to be completely understood. In this study, we elucidate the anti-inflammatory mechanism of ZP in lipopolysaccharide (LPS)-induced RAW264.7 cells. We evaluated the effects of ZP in LPS-induced levels of inflammatory cytokines, prostaglandin E2 (PGE2), and caspase-1 using ELISA. The expression levels of inflammatory-related genes, including cyclooxygenase (COX)-2 and inducible nitric oxide synthase (iNOS), were assayed by Western blot analysis. We elucidated the effect of ZP on nuclear factor (NF)-κB activation by means of a luciferase activity assay. The findings of this study demonstrated that ZP inhibited the production of inflammatory cytokine and PGE2 and inhibited the increased levels of COX-2 and iNOS caused by LPS. Additionally, we showed that the anti-inflammatory effect of ZP arises by suppressing the activation of NF-κB and caspase-1 in LPS- induced RAW264.7 cells. These results provide novel insights into the pharmacological actions of ZP as a potential candidate for development of new drugs to treat inflammatory diseases.

  • Conchiolin is a complex protein which is secreted by the mollusc's outer epithelium to be the organic basis of mollusc shell. This study is to investigate a potential anti-inflammatory activity of conchiolin of oyster shell (COS). We tested the effects of COS on the lipopolysaccharide (LPS)-induced production of nitric oxide (NO) and prostaglandin E2 (PGE 2) in a murine macrophage cell line, RAW 264.7. COS inhibited production of NO and PGE2 in a dose dependent manner and also decreased the expression of inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$) and interleukin-6 (IL-6). These results suggest that COS can inhibit production of pro-inflammatory mediators and might be a useful source to treat inflammation.

Shikonin Isolated from Lithospermum erythrorhizon Downregulates Proinflammatory Mediators in Lipopolysaccharide-Stimulated BV2 Microglial Cells by Suppressing Crosstalk between Reactive Oxygen Species and NF-κB

  • According to the expansion of lifespan, neuronal disorder based on inflammation has been social problem. Therefore, we isolated shikonin from Lithospermum erythrorhizon and evaluated anti-inflammatory effects of shikonin in lipopolysaccharide (LSP)-stimulated BV2 microglial cells. Shikonin dose-dependently inhibits the expression of the proinflammatory mediators, nitric oxide (NO), prostaglandin $E_2$ ($PGE_2$), and tumor necrosis factor-${\kappa}B$ (TNF-${\alpha}$) as well as their main regulatory genes and products such as inducible NO synthase (iNOS), cyclooxygenase-2 (COX-2), and TNF-${\alpha}$ in LPS-stimulated BV2 microglial cells. Additionally, shikonin suppressed the LPS-induced DNA-binding activity of nuclear factor-${\kappa}B$ (NF-${\kappa}B$) to regulate the key regulatory genes of the proinflammatory mediators, such as iNOS, COX-2, and TNF-${\alpha}$, accompanied with downregulation of reactive oxygen species (ROS) generation. The results indicate that shikonin may downregulate the expression of proinflammatory genes involved in the synthesis of NO, $PGE_2$, and TNF-${\alpha}$ in LPS-treated BV2 microglial cells by suppressing ROS and NF-${\kappa}B$. Taken together, our results revealed that shikonin exerts downregulation of proinflammatory mediators by interference the ROS and NF-${\kappa}B$ signaling pathway.

  • Objectives This experiment is about Jojoongikgi-tang(JIT) exerts anti-inflammatory effects in BV2 microglial cells, and the effect of JIT on Nitric oxide(NO) production in lipopolysaccharide(LPS)-stimulated BV2 microglial cells were also demonstrated. Methods To investigate the anti-inflammatory effects of JIT, NO production, expression level of iNOS mRNA, PGE2 synthesis, expression COX-2 mRNA, cell viability, $TNF-{\alpha}$ mRNA expression were examined. Results The expression level of inducible nitric oxide synthase(iNOS) was decreased by JIT, and the production of Prostaglandin E2(PGE2) and the expression of Cox-2 mRNA also were inhibited by JIT. Proinflammatory mediators, such as $TNF-{\alpha}$, $IL-1{\beta}$, IL-12, were inhibited by JIT in a dose-dependent manner. Conclusions JIT have anti-inflammatory effects in BV2 microglial cells and could be used in inflammatory disease.

  • Prostaglandins biosynthesis and nitric oxide production have been implicated in the process of inflammation. In this study we investigated on the effects of ethyl acetate extract of Angelica Koreana Radix (EAKR) on the activities of prostaglandin $E_2\;(PGE_2)$, proteoglycan (PG) degradation and nitric oxide synthase (NO) in inflammation cytokines-activated rabbit articular chondrocytes. EAKR exhibited inhibitory activities on NO production and $PGE_2$ production as $73.08\%\;and\;89.49\%$, respectively at $20{\mu}g/ml$ and inhibited the degradation of PG in a concentration-dependent manner Zelatin zymography analysis demonstrated that EAKR significantly inhibited MMP-2, 9 expression in chondrocytes. In vivo, EAKR was shown to have inhibitory effects on acetic acid-induced pain. This study suggests that modulation of $PGE_2$, NO, PG degradation and MMP-2, 9 by EARK may be important in the prevention of inflammation and osteroarthritis.

Anti-Inflammatory Effects of Chrysanthemum indicum Water Extract in RAW 264.7 Cell as a Whole Plant

  • Purpose: Chrysanthemum indicum (CHI) has been used for edible and medical purposes for a long time in Korea. The purpose of this study was to evaluate the anti-inflammatory effects of CHI water extract in lipopolysaccharides (LPS)-induced RAW 264.7 macrophage cells. Methods: To investigate the anti-inflammatory effects on LPS-induced RAW 264.7 macrophage cells, CHI extract as a whole plant was used in this study. RAW 264.7 cells were treated with various concentrations of CHI extract (1, 10, and $100{\mu}g/mL$). After that Nitric Oxide (NO), inducible nitric oxide synthase (iNOS), interleukin (IL)-$1{\beta}$, cyclooxygenase (COX)-2 and prostaglandin $E_2$ ($PGE_2$) expression level were measured. Results: CHI extract significantly suppressed the LPS-induced NO production and decreased the level of iNOS, IL-$1{\beta}$, COX-2 messenger ribonucleic acid (mRNA) expression and also the down regulation of $PGE_2$ expression in a dose-dependent manner. Conclusion: The present study suggested that CHI extract can be substituted for anti-inflammatory drugs and provide a safe and effective non pharmacological therapeutic approach.

In vitro Antiinflammatory Activity of the Essential oil Extracted from Chrysanthemum sibiricum in Murine Macrophage RAW 264.7 Cells

  • This research was undertaken to find the in vitro inflammatory action of the essenetial oil (CS-oil) extracted from Chrysanthemum sibiricum (Compositae) herbs. We investigated the effects of the CS-oil not only on the formation NO, $PGE_2$, and $TNF-{\alpha}$ but also on inducible nitric oxide synthase and cyclooxygenase-2 (COX-2) in lipopolysaccharide (LPS)-induced murine macrophage RAW 264.7 cells. The data obtained were consistent with the modulation of iNOS enzyme expression. A similar fashion was also observed when LPS-induced $PGE_2$ release and COX-2 expression were tested. The significant inhibitory effects were shown in concentration-dependent manners. In addition, CS-oil also mildly but significantly reduced the formation of TNF-a. These findings support the application of CS-oil as an antiinflammatory essential oil.
