Shiranuhi is a fruit of Citrus species widely cultivated in Jeju Island, Korea. From an extract of Shiranuhi tree branches were identified five polymethoxyflavones possessing anti-inflammatory effects; nobiletin (1), sinensetin (2), tetramethylscutellarein (3), 6-hydroxy-5,7,3',4'-tetramethoxyflavone (4) and 5-desmethylsinensetin (5). Evaluation of the activities was conducted by monitoring the production of nitric oxide (NO), prostaglandin E2 and pro-inflammatory cytokines (TNF-α, IL-1β, IL-6) as well as the levels of iNOS and COX-2 protein expression in LPS-induced RAW264.7 cells. Among the isolates, the compound 4 exhibited the most significant NO inhibition, and suppressed the levels of iNOS and related cytokines. Therefore, it was suggested that the extract and constituents from Shiranuhi tree branches could be useful as anti-inflammatory ingredient.
To evaluate the antioxidant and anti-neuroinflammatory effects of defatted Perilla frutescens extract (DPE) in lipopolysaccharide (LPS)-stimulated BV-2 microglial cells. Cell viabilities were estimated by MTT assay. LPS-stimulated BV-2 microglia were used to study the expression and production of inflammatory mediators such as nitric oxide (NO), inducible NO synthase (iNOS), Cyclooxygenase-2 (COX-2), and prostaglandin $E_2$ ($PGE_2$). Pretreatment with DPE prior to LPS treatment significantly inhibited excessive production of NO (10, 25, 50, 75, and $100{\mu}g/mL$) in a dose-dependent manner, and was associated with down regulation of expression of iNOS and COX-2. DPE also suppressed the LPS-induced increase in $PGE_2$ level (10, 25, 50, 75, and $100{\mu}g/mL$) in BV-2 cells. Therefore, DPE can be considered as a useful therapeutic and preventive approach for the treatment of several neurodegenerative diseases.

Several lines of recent evidences have shown that several pro-inflammatory genes or mediators, such as inducible nitric oxide synthase (iNOS), cyclooxygenase-2 and cytokines (e.g., tumor necrosis factor $\alpha$ and interleukin-1$\beta$), are strongly expressed in the ischemic brain. Inflammation is now recognized as a significant contributing mechanism in cerebral ischemia because anti-inflammatory compounds or inhibitors of iNOS and cyclooxygenase-2 have been proven to reduce ischemic brain damage. This research was designed to evaluate the possible anti-inflammatory effects of Carex scabrifolia Steud. extract using RAW264.7 cells. The assessments of these effects were based on cell viability assay, mRNA expression levels of interleukin-1 alpha (IL-1α), interleukin-1 beta (IL-1β), IL-6, tumor necrosis factor alpha (TNFα), and levels of nitric oxide (NO)/prostaglandin E2 (PGE2) production. Quantitative real-time polymerase chain reaction showed that treatment with C. scabrifolia Steud. extract decreased the mRNA levels of iNOS, COX2, IL-1α, IL-1β, IL-6, and TNFα. Furthermore, from the production levels of PGE2/NO, it can be inferred that C. scabrifolia Steud. extract exhibited anti-inflammatory properties. These results suggest that C. scabrifolia Steud. extract contains anti-inflammatory compound(s), and consequently, that it may have applications as a potent cosmeceutical material.
Equisetum hyemale L. has been prescribed widely as a traditional medicine for the treatment of inflammatory diseases such as rheumatoid arthritis, conjunctivitis, pyelonephritis. In order to identification the mechanism, we examined an antioxidant and anti-inflammatory activity of 85% methanol extract of E. hyemale. In this study E. hyemale exhibited strong scavenging effect on 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical, superoxide radical, and nitric oxide. To elucidate the anti-inflammatory properties of E. hyemale, we investigated the inhibition effects of nitric oxide and IL-6 by E. hyemale in IFN-gamma and LPS-stimulated mouse peritoneal macrophages. E. hyemale suppressed nitric oxide, IL-6 production and iNOS expression dose-dependently without notable cytotoxic activity. These data suggest that E. hyemale might be useful in inflammatory diseases by inhibiting the free radicals and inflammatory mediators.
Objectives : This study was aimed at producing emulsion by using butanol fractions of Sanguisorbae radix(SRA-B) which have high antioxidative and anti-inflammatory actions, and then evaluating stabilities of the emulsion. Methods : We measured antioxidant efficacy of SRA-B by using DPPH assay. Also, we checked the expressions of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) by using the Western blot to evaluate the anti-inflammatory effects of SRA-B3. We prepared emulsion containing SRA-B3(E-SRA-B3) and analysed its particle size distribution under a microscope. Also, we performed the test for stability of the emulsion. Results : SRA-B3 showed the highest efficacy in electronic donating abilities' activity. The Western blot's results indicated that the protein expression's amount of iNOS and COX-2 in macrophage stimulated by LPS were reduced by SRA-B3 treatment. The average particle size of E-SRA-B3 was $5{\sim}6{\mu}m$ in diameter and was $6.7{\mu}m$ in a view of the particle distribution. For a period of a observation, E-SRA-B3 has not made particular changes with storage temperature. It was observed that E-SRA-B3 could preserve its stable condition without a particular difference of viscosity during 28 days. Conclusions : From the above results, it was confirmed that SRA-B3 has potentiality enough to be applied to industrialization and could be utilized as antioxidative natural materials and anti-inflammatory cosmetics.
Under constant environmental pollution, the incidence of Atopic Dermatitis (AD) caused by air pollutants and allergens has increased. AD is an allergy inflammatory skin disease characterized by pruritus, eczema, and skin dryness. In herbal medicine, Anemarrhena asphodeloides (Anemarrhenae Rhizoma; AR) has been utilized to treat Alzheimer's disease, osteoporosis, hypertension, and inflammation. The purpose of study evaluated the effect of AR in a mouse model of 2,4-dinitrochlorobenzene (DNCB)-induced AD-like skin lesions. After acclimatization for 5 days, the mice (6-week-old, male Balb/c) were divided into five groups (n=6/group): NC (normal control), DNCB (control), Dex (5 mg·kg-1, p.o.), AR100 (100 mg·kg-1, p.o.), and AR300 (300 mg·kg-1, p.o.). On days 1 and 3, 1% DNCB was applied to the skin and ears. After 4 days, 0.5% DNCB was applied once every 2 days for 2 weeks. Then, skin and ears eczema area and severity index (EASI); skin nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), and prostaglandin E2 (PGE2) levels; and plasma immunoglobulin E (IgE) levels were examined. The AR groups showed lower EASI, skin and ear thickness, mast cell count, and IgE levels than the control groups. Moreover, AR reduced iNOS, COX-2, and PGE2 levels. Therefore, AR possesses anti-inflammatory properties and can improve skin damage, indicating its therapeutic potential against AD.
O presente estudo tem como objetivo principal analisar a influência de vocabulários de origem africana em uso no português brasileiro(PB). A fim disso, observamos os tipos e as características das línguas africanas. E ainda, para compreender a situação atual de uso desses termos no Brasil, apresentamos um panorama do registro dos vocábulos de origem africana no PB, e organizamos os vocabulários em três categorias, especialmente, nos registros dos séculos XIX e XX. Em primeiro, a categoria 1 pode ser usada em qualquer interação social, contrastando com as outras duas categorias. Em segundo, a categoria 2 é constuída de termos informais, de uso coloquial que, eventualmente, dependendo da situação, são substituídos por outras. Por último, a categoria 3 é de termos marcadamente informais, de uso restrito. Através disso, comparamos os registros anteriores com os dados obtidos da nossa pesquisa. Com os resultados, podemos reconhecer a estabilidade e o dinamismo do uso desses termos hoje no PB.
등골나물(Eupatorium japonicum)은 식용으로 이용되어온 식물이지만 생리활성에 대한 연구가 없었다. 따라서 본 연구진은 등골나물 에탄올추출물이 쥐 대식세포인 Raw264.7 세포에 LPS로 유도된 염증 반응에 미치는 영향에 대하여 연구를 수행하였다. 등골나물 꽃 부분에 70% 에탄올을 가하여 얻은 등골나물 에탄올추출물(EJE)을 Raw264.7 세포에 LPS와 함께 0, 1, 2.5, 5, 10 mg/L로 처리하여 세포를 배양하였다. LPS에 의해 생성된 NO 및 $PGE_2$ 분비는 EJE를 처리함에 따라 감소하였고 이 결과는 EJE의 독성에 의한 것이 아님이 증명되었다. Raw264.7 세포에 LPS에 의해 생성된 iNOS, COX-2의 단백질과 mRNA의 발현이 EJE의 농도 의존적으로 억제되었으며, 염증 반응시 생성되는 IL-6, IL-${\beta}$, TNF-${\alpha}$의 mRNA 발현도 등골나물 추출물에 의해 현저히 억제되었다. 더욱이 EJE의 분획물 중 EA와 MC 분획물이 독성이 적으면서 효과적으로 NO의 생성을 억제하는 것으로 나타났다. NO 생성 억제효과가 뛰어난 이들 분획물 내의 생리활성 물질에 대한 연구가 추가적으로 수행되어야 한다고 사료된다. 등골나물 꽃을 추출물로 항염증효과를 연구한 논문이 없고 다른 식용 가능한 천연재료들의 항염증효과와 비교하여(data not shown) EJE 분획물의 항염증효과가 낮은 농도에서 탁월한 점을 미루어 볼 때, 등골나물 추출물은 상당히 낮은 농도에서 뛰어난 항염증 물질을 가진 단일물질을 포함할 가능성이 높다고 사료된다. 따라서 위의 결과는 등골나물 추출물이 독성과 부작용이 적은 염증 치료제로 활용될 수 있는 가능성이 높음을 제시한다.
목적: 6-cyano-7-nitroquinoxalin-2,3-dione (CNQX)는 저산소성허혈성뇌병증, 외상성뇌손상, 흥분독성과 같은 뇌 손상동물 모델에서 보호 효과가 있다고 발표되고 있지만 신생아기에 그 효과에 대해서는 아직 제한적이다. 저자들은 CNQX를 이용하여 저산소성허혈성뇌손상을 동반한 발달 과정에 있는 흰쥐 뇌에 산화질소합성효소 중재를 통한 뇌 보호 효과를 알아보고자 하였다. 방법: 생체내 실험으로 생후 7일된 신생 흰쥐의 좌측 총 경동맥을 결찰한 후 6개군(정상산소군, 수술없이 저산소군, sham수술 후 저산소군, 수술 후 저산소군, vehicle 투여 후 저산소군, CNQX 투여 후 저산소군)으로 나누었고, 저산소 손상은 특별히 제작한 통속에서 2시간 동안 8% $O_2$에 노출시켰다. CNQX은 뇌손상 전 30분에 체중 kg당 10 mg를 투여하였다. 생체외 실험으로 재태기간 18일된 태아 백서의 대뇌피질 세포를 배양하여 정상산소군은 5% $CO_2$ 배양기(95% air, 5% $CO_2$)에 두었고, 저산소군과 뇌손상 전 CNQX 투여군(10 ${\mu}g$/mL)은 1% $O_2$). 배양기(94% $N_2$, 5% $CO_2$)에서 16시간동안 뇌세포손상을 유도하였다. 생체내 외 실험 모두 산화질소합성효소와 관련된 iNOS, eNOS, nNOS 항체와 primer를 이용하여 western blotting과 실시간중합효소연쇄반응을 시행하였다. 결과: 산화질소합성효소와 관련된 생체외 내 실험에서 iNOS와 eNOS의 발현은 저산소군에서 정상산소군보다 감소하였으나 CNQX 투여군에서는 저산소군보다 증가하였다. nNOS의 발현은 반대로 표현되었다. 결론: 본 연구에서 CNQX는 산화질소합성효소 중재를 통하여 주산기 저산소성 허혈성 뇌손상에서 신경보호 역할을 하는 것을 알 수 있었다.
