통초추출물의 항산화 활성 및 항노화효과 (Antioxidative Activity and Antiaging Effects of Tetrapanax papyriferum extract)

  • In this study, we evaluated the anti-oxidative, whitening and anti-wrinkle effect of Tetrapanax papyriferum extract. Tetrapanax papyriferum was extracted by two different solvents which were n-Hexane, ethyl acetate. The anti-oxidant activity was measured by free radical scavenging activity using DPPH (1,1-diphenyl-2-picrylhydrazyl radical). And the inhibitory activities of tyrosinase for whitening effect and collagenase, elastase for anti-wrinkle were investigated. For anti-oxidant activity, ethyl acetate fraction of Tetrapanax papyriferum extract showed more significant activity than n-Hexane fraction of Tetrapanax papyriferum extract. For whitening activity, n-Hexane fraction of Tetrapanax papyriferum extract exhibited strong inhibition effects compared with reference. Therefore, Tetrapanax papyriferum extract may be useful as a new antioxidant and whitening agent.

  • The possible effects of garlic (Aliium sativurn L.) extract on growth and aflatoxin production by Aspergillus parasiticus R- 716 were investigated. Various solvent extracts of garlic strongly inhibited growth and sporulation by Aspergillus parasiticus R-716, and effective solvents used for extraction of garlic were chloroform, benzene, and water-chloroform. The growth and aflatoxin production decreased with the increase in extract concentration, and extract equivalent 1.5g of raw garlic weight in 25ml SLS medium completely inhibited, and at a level of 1.25g garlic, total aflatoxin was reduced 64% (472 ${\mu}g$/25ml) of that produced in the control (1, 352 ${\mu}g$/25 ml). During cultivation inhibitory rate of growth was reduced from 89.1% to 40% and aflatoxin $B_2$, $G_1$ production increased with the laps of time. Especially garlic extract appeared to have a stimulatory effect on lipid accumulation on the contrary aflatoxin production.

유황오리 추출물의 각종 암세포에 대한 생육억제 효과 (Growth Inhibition of Extract from Sulfur fed Duck Carcass against Various Cancer Cell Lines)

  • 유황오리 추출물의 각종 암세포에 대한 생육억제효과를 MTT assay를 이용하여 검토한 결과 10 mg/$m\ell$의 농도에서 KB(구강상피암) 89.5$\pm$0.7%, SNU-1(위암) 69.8$\pm$1.7%, K-562(백혈병) 79.8$\pm$2.8%, Farrow(흑색종) 82.7$\pm$2.6%, WiDr 결장암) 76.3$\pm$2.5%, SK-MES-1(폐암) 59.2 $\pm$4.4%, HL6O(백혈병) 60.5 +3.5%, Calu-3(폐암) 53.2$\pm$1.6%, HEP-2(후두암) 80.7$\pm$0.5%, P388(마우스 백혈병) 79.9$\pm$3.7%, 3LL(마우스 폐암) 87.2$\pm$3.3%의 효과가 있다는 사실이 판명되었다. 그리고 유황오리 추출물의 HP-20 column chromatography에서는 100% methyl alcohol 용출물이 HEP-2에 대한 생육억제효과가 있었으며, 10 mg/$m\ell$에서 99.1$\pm$0.4%의 생육억제효과가 나타났다. 또한 일반오리 추출물과 유황오리 추출물의 각종 암세포에 대한 생육억제효과를 비교하였을 때 거의 모든 암세포에서 유황오리 추출물의 효과가 더 높았다.

유산균 발효에 의한 톳 (Hizikia fusiforme) 추출액의 항산화 및 항염증 활성 증가 (Enhancement of the Antioxidant and Anti-inflammatory Activity of Hizikia fusiforme Water Extract by Lactic Acid Bacteria Fermentation)

  • We previously reported that fermentation by Lactobacillus brevis LB-20 isolated from Kimchi resulted in improvement of the sensory quality of Hizikia fusiforme water extract. This study was conducted to evaluate the possible application of lactic acid bacteria fermentation to improve the functional qualities of H. fusiforme extract. L. brevis LB-20 was inoculated and cultivated in H. fusiforme extract. The antioxidant and anti-inflammatory activities of extract were then assayed both before and following fermentation for two days. Antioxidant activity was determined by assaying levels of radical scavenging activity against 1,1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl, superoxide, and alkyl radical. Lactic acid bacterial fermentation of H. fusiforme extract resulted in enhancement of antioxidant activity. The greatest enhancement of antioxidant activity was seen in the hydroxyl radical scavenging assay that incorporated 0.5 mg/mL of raw and fermented H. fusiforme extract. Fermented extract exhibited greater (21.95%) inhibition of nitric oxide synthesis than did raw extract (14.66%) at a concentration of 1 mg/mL. The fermented extract exerted its potent anti-inflammatory activity via attenuation of expression of inflammation-related cytokine proteins (TNF-${\alpha}$ and iNOS).

  • 中國産 집오리, Anas platyrhynchos의 肝 alkaline phosphatase의 特性에 대해 觀察하여, 다음과 같은 結果를 얻었다. 1. 이 酵素는 分子의 크기가 다른 2가지 酵素種을 가짐을 보였다. 各 酵素分子는 各各 電氣泳動 速度가 다른 2가지 分子로 이루어 짐을 알았다. 2. 이 효소의 最適 pH는 9.0임이 판명되었고, 다른 포유류 酵素에 比해 熱에 대한 安定度가 낮았다. 3. $Mg^2+$은 이 酵素의 活性을 약 2배 증가 시켰으나, $Ca^2+$은 活性에 거의 影響을 주지 않았다. 4. Phosphate 이온은 이 효소에 대해 competitive, L-phenylalanine은 uncompetitive 阻害劑로 作用함을 알았다.

DHA와 EPA에 대한 초임계 유체의 분할추출 (Fractional Extraction of DHA and EPA by Supercritical Fluid)

  • 초임계유체를 사용하여 네 종류의 추출조건에서 EPA가 20%, DHA가 15%인 물고기 기름을 구성하는 지방산에스터의 혼합물에 대하여 네 단계 분할추출실험을 수행하여 분할단계별 extract와 raffinate내의 성분무게조성, 추출되는 양, 누적추출량의 변화특성을 규명하였다. 네 가지 추출조건중 온도 60$^{\circ}C$, 압력 101 bar, 유속 1.0 mL/min, 추출시간 50 min, 추출평형시간 15 min인 경우에 extract와 raffinate내로 DHA와 EPA가 분리 농축되는 경향이 가장 뚜렷하였다. 이 경우 네 번째 단계의 추출에서 extract내로 EPA가 50%까지 농축되었으며 raffinate내로 DHA가 40%까지 농축되었다. 이때 extract내로 추출되는 EPA의 양은 초기 주입양의 34%이였으며, raffinate내로 농축되는 DHA의 양은 초기 주입양의 73%이였다. 동일한 추출조건으로 DHA의 무게 조성이 34%인 혼합물은 네 단계 분할 추출한 경우 최종 단계의 raffinate내에 DHA가 70%이상으로 농축되었으며 그 양은 초기 주입양의 80%이상이었다.

인진수근오미자(茵蔯水芹五味子) extract가 알콜성 간질환과 바이러스성 간염환자의 LFT에 미치는 영향 (The Effects of Injinsugunomija Extract on Liver Function Test of Alcoholic Liver Disease and Viral Hepatitis Patients)

  • Objectives : This study was conducted to evaluate the effects that Injinsugunomija Extract affected Liver function test of Alcoholic liver disease and Viral hepatitis patients. Methods: We adminstered eighty packets of Injinsugunomija Extract to some kinds of liver disease patients for one month. And we investigated the changes of their liver function test. Results: The results from this study were summarized as followed. In the 10 patients of Alcoholic Liver Disease (ALD), 3 cases improve prominent and 4 cases improve effective and 3 cases are ineffective. Then 70% 01 ALD patients has improved by Injinsugunomija Extract In the 9 patients of Viral hepatitis, 1 case improves prominent and 4 cases improve effective and 1 case become worse and 1 case become worst and 2 cases are ineffective. Then 55.6% of Viral hepatitis patients has improved by Injinsugunomija Extract. Conclusions: From these results, Injinsugunomija Extract showed the meaningful effects on improving liver function test of ALD patients and Viral hepatitis patients.

Phaeophyta Extracts Exhibit Antiviral Activity against Feline Calicivirus

  • The objective of this study was to evaluate the antiviral activity of Phaeophyta extracts against feline calicivirus (FCV), used as a norovirus surrogate. A bioassay-guided cytotoxicity and virus infectivity assay revealed that methanolic extracts of Phaeophyta possessed significant antiviral activity against FCV. Among them, Eisenia bicyclis extract exhibited the highest antiviral activity against FCV. The 50% effective concentration of the extract ($EC_{50}$) inhibiting FCV viral replication by 50% was $80{\mu}g/mL$. The extract also showed the highest selectivity index, calculated from the ratio of the median cellular cytotoxicity concentration ($CC_{50}$) and $EC_{50}$, indicating antiviral efficacy against FCV. In addition, significant interruption of FCV infection was observed by pretreatment of host Crandall-Reese feline kidney cells with the E. bicyclis extract ($200{\mu}g/mL$) prior to virus infection, in a dosedependent manner.

Cross-reactivity between sera from dogs experimentally infected with Dirofilaria immitis and crude extract of Toxocara canis

  • This study was performed to investigate whethere there is cross-reactivity between Dirofilaria immitis and three intestinal nematodes of dogs. In ELISA, D. immitis- infected dog sera obtained at the 4th molting stage (9-11 weeks) and microfilaremic stage (25-30 weeks) were shown to be highly reactive with crude extract of T. canis. In immunoblotting, some antigenic fractions, 44, 57 88, 100 kDa of crude extract of T. canis, were found to be positive reaction with sera of dogs infected with D. immitis. However, little or no cross-reaction were observed between sera of D. immitis-infected dogs and crude extract antigen of T. vulpis or A. caninum. These result suggest that there are partial cross reaction between sera of D. immitis-infected dogs and the antigen of T. canis.

천연 염지제을 이용한 훈제오리의 품질특성 (Quality characteristics of smoked duck using natural curing agent)

  • 본 연구는 천연 소재를 이용하여 개발한 천연훈제오리의 품질 특성을 일반훈제오리와 비교하여 합성 염지제를 천연물로 대체 가능한지를 알아보았다. 또한 소비자가 믿고 찾을 수 있는 안전하고 위생적인 식품으로 더 적합한 훈제오리를 개발하고자 하였다. 식품첨가물 잔류량, 저장기간 동안 훈제오리의 pH, 미생물량 및 색도변화, 관능평가, 항산화활성 등을 조사하였다. 천연물을 이용하여 개발한 천연훈제오리에서는 아질산나트륨, 산화방지제, 글루타민산나트륨 등의 합성첨가물은 검출되지 않았다. 저장 기간 동안 두 군 모두 pH가 감소하는 경향을 보였으며 대장균군은 발생하지 않았다. 훈제오리의 명도와 황색도는 두 군 간에 차이가 없었으나 저장기간 동안 두군 모두 감소하였으며, 적색도는 저장 전 기간 동안 일반 훈제오리가 유의적으로 높았다. 훈제오리에 대한 기호도 평가는 저장 기간이 길어질수록 외관, 색, 맛, 후미, 질감, 전체적인 품질이 유의적으로 감소하였으나 맛이나, 질감, 전체적인 품질에 대한 평가는 저장 기간 내내 유의적으로 천연훈제오리의 평가가 더 높았다. 총 폴리페놀 함량과 항산화 활성은 천연훈제오리가 일반훈제오리보다 높았다. 이상과 같은 결과로 볼 때 배당침액, 셀러리파우더, 비타민 C를 포함한 천연염지제는 일반훈제오리 염지제를 대신할 수 있으며, 천연물을 첨가하여 개발한 천연훈제오리는 식품첨가물이나 미생물 등이 검출되지 않고 항산화 활성이 우수하여 일반 소비자가 더 믿고 찾을 수 있는 위생적이고 안전한 건강식품으로써 적합하다고 사료된다.
