Effect of Glucose on Listeria monocytogenes Survival under Sequential Sublethal Stresses of Gamma Irradiation and NaCl

  • This study evaluated glucose effect on Listeria monocytogenes survival under gamma irradiation and NaCl stress. L. monocytogenes in phosphate buffered saline (PBS) plus glucose (0-4%) was treated with gamma irradiation (0-0.5 kGy), and the samples were then exposed to NaCl (0-9%) in tryptic soy agar plus 0.6% yeast extract. $D_{10}$ and $t_{3D}$ values were determined, and a model for prediction of $D_{10}$ values was developed. Cell counts of L. monocytogenes reduced as irradiation dose increased, and L. monocytogenes in PBS (no glucose) was more sensitive to irradiation and NaCl compared to those in PBS (2 or 4% glucose). $D_{10}$ values were 0.07-0.1, 0.12-0.16, and 0.13-0.15 kGy for 0, 2, and 4% glucose, respectively. The $t_{3D}$ values were 0.22-0.3 (0% glucose), 0.35-0.48 (2% glucose), and 0.40-0.44 (4% glucose). A model performance was acceptable. These results indicate that glucose in foods would increase the resistance of L. monocytogenes to gamma irradiation and NaCl stress.

Stability of Black Soybean Pigment Extract (검정콩 종피 색소추출액의 안정성)

  • To examine utilization of Korean black soybean as a natural pigment, the effects of temperature, carbohydrate, organic acid, vitamin C and metal ion were investigated. Korean black soybean pigment was more stable than other anthocyanin pigments when temperature was abused. There was no darkness effect in carbohydrate. In organic acid, the color intensity was increased. The vitamin C addition negatively affected on color of anthocyanin. Among the metal ions tested, $Cu^{2+}$ and $Zn^{2+}$ was not affected but $Mn^{2+}$ makes it unstable. Results indicate that Korean black soybean pigment was more stable than other anthocyanin pigment in various environment at pH 3.

Isolation and Characterization of Phytochemical Constituents from Soybean (Glycine max L. Merr.)

  • Four flavonoids 1-4 and one phytosterol 5 were isolated from methanol extract of Taekwangkong, one of the soybean cultivars, and the structures of these compounds were fully characterized by physical and spectral analysis. The content of compounds 1-4 as determined by $C_{18}$ reversed phase HPLC (high-performance liquid chromatography) coupled with diode-array detector were 12.1, 624.6, 18.0, and $219.6\;{\mu}g/g$, respectively, and the total phenolic content of this cultivar was measured as 3.7 mg gallic acid equivalent per g dry material (GAB/g). Also, compound 1 showed strong radical scavenging activity in the 2,2'-azino-bis-(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS) assay ($IC_{50}\;=\;47.6\;{\mu}M$), five-fold higher than seen in the 1,1-diphenyl-2-picrylliydrazyl (DPPH) assay. These results lead to the conclusion that soybean not only has many phytoestrogens but also has potent antioxidant activity.

The inhibitory effects of 3,4,5-Trimethoxy cinnamate thymol ester(TCTE, Melasolv$\circledR$) on Melanogenesis

  • To date, research on the regulation of melanogenesis has focused on factors which affect tyrosinase, the rate-limiting enzyme in the melanogenic pathway, by searching for chemicals which competitively inhibit tyrosinase function. Many types of tyrosinase inhibitors have been developed, but no satisfactory results have been made clinically until now, To find a new whitening agent, which effectively inhibits melanogenesis, we synthesized several compounds and selected compounds by cell-based assay system. Finally, 3, 4, 5-trimethoxy cinnamaie thymol ester(TCTE, Melasolv) was selected and the effects of TCTE on melanogenesis were investigated. Treatment of mouse-derived melanocyte melan-a cells with TCTE results in a marked down-regulation of tyrosinase activity. 80% decrease of tyrosinase activity occurs with 30uM TCTE treatment for 72 hours without affecting cell growth. The inhibition of tyrosinase activity is dose-dependent and melanin content was also decreased to 40%. From the in vitro tyrosinase assay using cell extract, TCTE does not act as a direct inhibitor of the enzyme. Treatment of melan-a cultures with TCTE blocks the increase in tyrosinase activity by either forskolin, 3-isobutyl-1-methtyl-xanthine. TCTE decreased the expression of tyrosinase, TRP-1 without effects on TRP-2 protein expression through the down regulation of tyrosinase and TRP-1 mRNA. From the results of cAMP immunoassays, intracellular levels of the cyclin nucleotide are unaffected in cells treated with TCTE. The inhibitory effects of melanin synthesis were also shown in reconstitute human epidermis model by topical application. These findings suggest that TCTE can be used for studying the regulation of melanogenesis and depigmenting agent.

Antioxidative and Hepatoprotective Effects of Ethanol Extracts from Different Barley Cultivars (보리 품종별 주정 추출물의 항산화 활성 및 간 보호 효과)

  • Barley's nutritional value as a health food is increasing due to its excellent nutritional functionality. In this study, the levels of β-glucan, total polyphenols, and total flavonoids were analyzed in the ethanol extracts of different barley cultivars (Hinchalssal, Heuksoojeongchal, Betaone, Ganghochung, and Saechalssal). Also, the free radical scavenging abilities of 2,2-diphenyl-1-picryl-hydrazil (DPPH) and 2,2'-azino-bis-3-ethylbenzo-thiaxoline-6-sulfonic acid (ABTS) were measured to determine their antioxidant activity. The results confirmed that Betaone extract contained highly activefunctional components and exhibitedantioxidant activity. Next, we evaluated the hepatoprotective and inhibitory effects of reactive oxygen species (ROS) generated by barley ethanol extracts after inducing oxidative stress with tert-butyl hydroperoxide (tBHP) in HepG2 cells. Hinchalssal and Saechalssal extracts showed the most significant cytoprotective effect and also reduced ROS production significantly. These results suggest that Hinchalssal, Saechalssal, and Betaone represent potential natural antioxidant and hepatoprotective agents.

Changes of Antioxidant Activity as affected by cultivation period in Buckwheat (Fagopyrum species) Sprouts (재배기간에 따른 쓴메밀(Fagopyrum tataricum Gaertner)싹의 항산화 활성 및 생리활성 평가)

  • Buckwheat (Fagopyrum species) has long been an excellent functional food. Besides, buckwheat sprouts contain various functional substances. In this study, we investigated the antioxidant activity of buckwheat sprouts in the context of cultivars harvested after different cultivation periods (0, 3, 5, 7, 9, 13, and 15 days after planting). Buckwheat sprouts were cultivated at 25℃ for up to 15 days and then extracted with ethanol. Antioxidant components were then extracted from sprouts and leaves using a freeze dryer. The total polyphenolic content, flavonoid content, and antioxidant activity were then analyzed. The total polyphenol content increased from 32.26 mg GA eq/100 g for raw buckwheat to 114.75 mg GA eq/100 g after 7 days of cultivation. Also, the flavonoid content increased from 20.61 mg catechin eq/100 g (0 days) to 56.54 mg/g after 9 days of cultivation. The DPPH radical scavenging activity (concentration of extract at 0.25 mg/mL) increased from 7.89% at day 0 to 53.48% after 9 days of cultivation. Additionally, the ABTS radical scavenging activity increased from 10.26% at day 0 to 32.89% after 7 days of cultivation; of note, the activity decreased afterward. These results suggest that the best buckwheat sprouts with higher biological activities are those cultivated for 7-9 days. For a complete understanding of the potential of buckwheat sprouts as functional foods, we plan to further analyze their antioxidant activity in the future.

  • Modern weapon systems are composed of complex systems(System of Systems) and require a complex and advanced operational concept that performs missions through interoperability with various weapon systems. In order to derive the operational concept of the weapon system that the military wants to acquire (i.e., single mission, component operation, Joint and Alliance operations), it is necessary to identify the system related to the weapon system, environmental factors and restrictions of the weapon system to be developed. Through the derivation of the operational concept, the weapon system acquisition agency can reasonably and accurately extract various and complex requirements. In this paper, we propose a complementary method of using MND-AF to OCD and ORD required in weapon system acquisition process. MND-AF can increase the understanding and consensus of business stakeholders (users, acquirers, developers, etc.) by showing the results of weapon system analysis from various perspectives. We compare the items in the standard form of OCD and ORD with the MND-AF outputs.

Evaluation of the Antioxidant Activity of Wheat Sprouts Produced by Different Cultivation Methods and Harvest Times (새싹밀의 재배방법과 수확시기에 따른 항산화 성분 및 활성 평가)

  • In this study, the antioxidant activity of methanol extracts of wheat sprouts grown in wild fields and cultivated fields and harvested at various times were analyzed to compare the quality characteristics of the wheat sprouts as a function of their cultivation and harvest time, as well as for the development of functional materials. The total polyphenolic and flavonoid content, as well as antioxidant activity of the wheat extracts, were subsequently analyzed. The ABTS radical scavenging activity of the wheat extract increased from 16.97 mg TEAC/g sample on cultivation day three in cultivated field wheat sprouts to 25.99 mg TEAC/g sample after seven days of wild field cultivation. The total polyphenol content increased from 17.08 mg GA eq/100 g in cultivated field wheat sprouts grown for three days to 28.70 mg GA eq/100 g after seven days of wild field cultivation. In addition, the flavonoid content increased from 7.02 mg catechin eq/100 g (7 days) to 8.47 mg/g after 12 days of wild field cultivation. Notably, the activity subsequently decreased. These results suggest that the wheat sprouts with higher biological activity were those produced from the wild field after 20 days.

Multi-class, Multi-residue Analysis of 59 Veterinary Drugs in Livestock Products for Screening and Quantification Using Liquid Chromatography-tandem Mass Spectrometry

  • BACKGROUND: The objective of this study was to develop a comprehensive and simple method for the simultaneous determination of 59 veterinary drug residues in livestock products for safety management. METHODS AND RESULTS: For sample preparation, we used a modified liquid extraction method, according to which the sample was extracted with 80% acetonitrile followed by incubation at -20℃ for 30 min. After centrifugation, an aliquot of the extract was evaporated to dryness at 40℃ and analyzed using liquid chromatography combined with tandem mass spectrometry. The method was validated at three concentration levels for beef, pork, chicken, egg, and milk in accordance with the Codex Alimentarius Commission/Guidelines 71-2009. Quantitative analysis was performed using a matrix-matched calibration. As a results, at least 52 (77.6%) out of 66 compounds showed the proper method validation results in terms of both recovery of the target compound and coefficient of variation required by Codex guidelines in livestock products. The limit of quantitation of the method ranged from 0.2 to 1119.6 ng g-1 for all matrices. CONCLUSION(S): This method was accurate, effective, and comprehensive for 59 veterinary drugs determination in livestock products, and can be used to investigate veterinary drugs from different chemical families for safety management in livestock products.

Immunomodulatory Effect of Eleutherococcus Senticosus Stem Extract by Cultivars in RAW 264.7 Macrophage Cells (RAW 264.7 대식세포에서 산지별 가시오가피 줄기 추출물의 면역 증강 효과)

  • Global interest in natural functional materials to strengthen human immunity is increasing due to the increase in immune-related diseases associated with COVID-19 and the aging population. In this study, we determined the potential therapeutic effect of Eleutherococcus senticosus stems on immune enhancement according to the cultivation region. The contents of eleutheroside B and E, which are chemical components of E. senticosus stems, were analyzed. We showed that the eleutheroside B content of E. senticosus stems in different cultivation regions ranged from 2.96±0.11 to 6.24±0.05 mg/g and from 1.11±0.05 to 2.11±0.03 mg/g in 70% ethanol and hot water extracts, respectively. The eleutheroside E content ranged from 4.93±0.20 to 10.79±0.03 mg/g and 1.75±0.14 to 3.64±0.05 mg/g in 70% ethanol and hot water extracts, respectively. In addition, the immunomodulatory effect of E. senticosus stems was evaluated using RAW 264.7 macrophages. The 70% ethanol extract of E. senticosus stems showed no cytotoxicity up to 200 ㎍/mL, and the hot water extract showed no cytotoxicity up to 500 ㎍/mL. Additionally, the E. senticosus stem extract significantly increased the production of nitric oxide and cytokines (TNF-α, IL-6, and IL-1β) compared to their production in the control group. These results suggest that E. senticosus stem extracts are a potential functional food material and ingredient to enhance the immune response.
