NDP Kinases Suppressed Bax-Dependent Apoptosis in Yeast System

  • Many nucleoside diphosphate (NDP) kinases are ubiquitous enzymes responsible for the exchange of ${\gamma}$-phosphates between tri- and diphosphonucleosides. The catalytic Many nucleoside diphosphate (NDP) kinases are ubiquitous enzymes responsible for the exchange of ${\gamma}$-phosphates between tri- and diphosphonucleosides. The catalytic reaction follows a ping-pong mechanism in which the enzyme is transiently phosphorylated on a histidine residue conserved in all nucleoside diphosphate kinases. Beside their role in nucleotide synthesis, these enzymes present additional functions, possibly independent of catalysis, in processes such as differentiation, cell growth, tumor progression, metastasis and development. To clone murine nm23-M5, several expressed sequence tags (ESTs) of the GenBank data base, selected according to their homology to nm23-H5 cDNA, reconstituted a complete open reading frame (GenBank AF222750). To test whether murine NDPKs (1, 2, 3, 4, 5, and 6) can inhibit Bax-mediated toxicity in yeast, co-transformation was performed respectively. The yeast S.cerevisiae was transformed with a copy expression plasmid containing the histidine selection marker and expressing murine Bax under the control of a galactose-inducible promoter. Several clones were selected and found to be growth inhibited when Bax expression was induced with galactose. A representative clone was transformed again with a copy expression plasmid containing the tryptophane selection marker and expressing either murine Bcl-xL or NDPK under the control of a galactose-inducible promoter. Several subclones of the double-transformants were selected and characterized. The ability of Bcl-xL and NDPKs to suppress Bax-mediated toxicity was determined by growing yeast cells overnight in galactose media and spot-testing on galactose plates starting with an equal number of yeast cells as determined by taking the OD$_{600}$. Ten-fold serial dilutions were used in the spot-test. Plates were grown at 3$0^{\circ}C$ for 2-3 days. All murine NDPKs suppressed Bax dependent apoptosis. Futher study will be peformed whether Bax-toxicity inhibition was caused by NDP kinase activity or additional function.n.

Characteristic of Raindrop Size Distribution Using Two-dimensional Video Disdrometer Data in Daegu, Korea (2차원 광학 우적계 자료를 이용한 대구지역 우적크기분포 특성 분석)

  • This study analyzes Two-dimensional video disdrometer (2DVD) data while summer 2011-2012 in Daegu region and compares with Marshall and Palmer (MP) distribution to find out statistical characteristics and characteristics variability about drop size distribution (DSD) of Daegu region. As the characterize DSD of Daegu region, this study uses single moment parameters such as rainfall intensity (R), reflectivity factor (Z) and double moment parameters such as generalized characteristics number concentration ($N{_0}^{\prime}$) and generalized characteristics diameter ($D{_m}^{\prime}$). Also, this study makes an assumption that DSD function can be expressed as general gamma distribution. The results of analysis show that DSD of Daegu region has ${\log}_{10}N{_0}^{\prime}=2.37$, $D{_m}^{\prime}=1.04mm$, and c =2.37, ${\mu}=0.39$ on average. When the assumption of MP distribution is used, these figures then end up with the different characteristics; ${\log}_{10}N{_0}^{\prime}=2.27$, $D{_m}^{\prime}=0.9mm$, c =1, ${\mu}=1$ on average. The differences indicate liquid water content (LWC) of Daegu distribution is generally larger than MP distribution at equal Z. Second, DSD shape of Daegu distribution is concave upward. Other important facts are the characteristics of Daegu distribution change when Z changes. DSD shape of Daegu region changes concave downward (c =2.05~2.55, ${\mu}=0.33{\sim}0.77$) to cubic function-like shape (c =3.0, ${\mu}=-0.13{\sim}-0.33$) at Z > 45 dBZ. 35 dBZ ${\leq}$ Z > 45 dBZ group of Daegu distribution has characteristics similar to maritime cluster of diverse climate DSD study. However, Z > 45 dBZ group of Daegu distribution has a difference from the cluster.

  • During the boom of museum building in the 1970s-80s two museum wee erected in Germany. They are James Stirling's Neue Stattsgalerie Stuttgart and hans Hollein's Stadtisches Museum Abteiberg Monchengladbach, These two museums share the folowing point of similarity : Stirling's Neue Stattsgalerie Stuttgart and Hollein's Staditisches Museum Abetiberg Monchenbladbach both manifest the conditions of the times in light of their respective cities' local characters and historical contexts without being bound to traditional formality. Stirling and Hollein attempted to grasp the meaning of the 기nuseum as a city in miniature." Taking into account the t two museums’s territorial characteristics of being situated on a slope, both Stirling and Hollein made the walker to pass t through their building complex and introduced the concept of a public square within them. As a result, the museums are not j just two large buildings but are architecture composed of a collage of various structures. S Stirling’s architecture employs the method of attaching additional elements on top of basic constituents, which is suggestive '||'&'||'#61551; of the historical fragment from Shinkel's Altes Museum. On the other hand, Hollein applies a collage-style method as if he w were doing urban planning, maintaining the distinctiveness of each of the various forms and materials of buildings. T The object style buildings of the two museums actively demonstrate the contrast of double meaning to represent the a ambiguous and multifarious characteristics of the modern times. Stirling explores the theme of opposition and coexistence in h his Neue Stattsgalerie Stuttgart by placing a series of opposing concepts, such as the past and present and histor${\gamma}$ and t technology, in one space. Thereby, the contradiction and its appeal are manifested. Hollein made use of the visual illusion and c contradiction in alluding to the irony of the modern reality induced by nature and culture and history and technology. F For the above reasons and methods, James Stirling ’ s Neue Stattsgalerie Stuttgart allowed the penetration of art into daily l life and became the general public’s favored museum for its free and unrestrained environment. Likewise, Hans Hollein’s S Stadtisches Museum Abteiberg Moncbengladbach contribute to the development of museum function as it an expression of art i in the form of a complex piece of scupture on its own.ts own.

Effects of Fructans on Blood Glucose, Activities of Disaccharidases and Immune Function in Streptozotocin-Induced Diabetic Mice (당뇨 유발 생쥐에서 Fructan이 혈당과 이당류분해효소 활성 및 면역능에 미치는 영향)

  • This study was conducted to investigate effects of fructans (chicory inulin, fructooligosaccharide and chicory inulin oligosaccharide) on blood glucose, activities of disaccharidases in small bowel and kidneys, and splenocyte proliferation in streptozotocin-induced diabetic mice. Sixty ICR male mice were divided into one normal group and four diabetic groups. Diabetes was induced by injecting streptozotocin after 2 weeks of experimental diets feeding. Experimental diets based on AIN93G diet were control diet, 6$ \%$ fructooligosaccharide (FOS) diet, 6$\%$ chicory inulin oligosaccharide (CIOS) diet, 6$\%$ chicory inulin (Cl) diet, and given for 25 days after streptozotocin injection. Plasma glucose was lower in Diabetic-Cl group as compared to Diabetic-control group. Plasma insulin level was not different among diabetic groups. Specific activities of jejunal maltase and sucrase in diabetic groups were about double as that of Normal group. Jejunal maltase activity and plasma glucose were positively correlated (r=0.643). However, specific activity of renal maltase in diabetic groups was not significantly different as compared to Normal group. Stimulation index of splenocyte proliferation by lipopolysaccharide (LPS) was significantly increased in Diabetic-CIOS as compared to Diabetic-control. Stimulation index of splenocyte proliferation by Concanavalin A (ConA) tended to be higher in Diabetic-CIOS group. Concentrations of interleukin-2 and interferon- $\gamma$ secreted from splenocytes induced by ConA were not significantly different among all groups. In conclusion, fructans may be effective for lowering plasma glucose, possibly by lowering disaccharidase activity and for increasing immune responses in diabetic con-ditions, where their effects can be different depending on degree of polymerization.

Difference in Isoproterenol-Stimulated Cyclic AMP Levels of Lymphocytes between the Subgroups of Depressive Disorders (우울장애 아형간에 Isoproterenol 자극후 임파구 Cyclic AMP Levels의 차이)

  • This study was designed to examine the basal cyclic AMP levels and the $10^{-5}mol/L$ isoproterenol-stimulated cyclic AMP levels of lymphocytes, by which ${\beta}$-adrenoceptor function was shown, between to normal controls and 17 drug free patients(8 major depresive patients and 9 dysthymic patients), who were diagnosed by DSM-III-R. The severity of depression was assessed by Hamilton Rating Scale for Depression (HDRS). Cyclic AMP levels were measured by radioimmunoassay(double antibody). The results were as follows ; 1) HDRS score was significantly higher in major depressive patients($41.8{\pm}4.6$) than in dysthymic patients($24.0{\pm}4.2$)(p<005). 2) There was no Significant difference in basal cyclic AMP levels among normal controls($3.9{\pm}1.7pmol/10^6cells/10min$), major depressive patients($2.1{\pm}0.5pmol/10^6cells/10min$), and dysthymic patients($3.9{\pm}1.8pmol/10^6cells/10min$). 3) There was significant difference in net cyclic AMP levels($10^{-5}mol/L$ isoproterenol-stimulated cyclic AMP levels minus basal cyclic AMP levels) among normal controls($16.5{\pm}6.0pmol/10^6cells/10min$), major depressive patients($3.0{\pm}1.4pmol/10^6cells/10min$), dysthymic patients($10.9{\pm}4.4pmol/10^6cells/10min$)(p <005). 4) The net cyclic AMP levels were significantly correlated with HDRS scores in major depressive patients(${\gamma}=-0.8^6$, p<0.05), but not in dysthymic patients(${\gamma}=0.43$, p=0.25). In conclusion, we suggested that the dysthymic disorder might differ from the molar depressive disorder not only in the severity of depressive symptoms but also in ${\beta}$-adrenergic responsiveness of lymphocytes, which was regarded as a biological marker of depressive disorder.

Financial Fraud Detection using Text Mining Analysis against Municipal Cybercriminality (지자체 사이버 공간 안전을 위한 금융사기 탐지 텍스트 마이닝 방법)

  • Recently, SNS has become an important channel for marketing as well as personal communication. However, cybercrime has also evolved with the development of information and communication technology, and illegal advertising is distributed to SNS in large quantity. As a result, personal information is lost and even monetary damages occur more frequently. In this study, we propose a method to analyze which sentences and documents, which have been sent to the SNS, are related to financial fraud. First of all, as a conceptual framework, we developed a matrix of conceptual characteristics of cybercriminality on SNS and emergency management. We also suggested emergency management process which consists of Pre-Cybercriminality (e.g. risk identification) and Post-Cybercriminality steps. Among those we focused on risk identification in this paper. The main process consists of data collection, preprocessing and analysis. First, we selected two words 'daechul(loan)' and 'sachae(private loan)' as seed words and collected data with this word from SNS such as twitter. The collected data are given to the two researchers to decide whether they are related to the cybercriminality, particularly financial fraud, or not. Then we selected some of them as keywords if the vocabularies are related to the nominals and symbols. With the selected keywords, we searched and collected data from web materials such as twitter, news, blog, and more than 820,000 articles collected. The collected articles were refined through preprocessing and made into learning data. The preprocessing process is divided into performing morphological analysis step, removing stop words step, and selecting valid part-of-speech step. In the morphological analysis step, a complex sentence is transformed into some morpheme units to enable mechanical analysis. In the removing stop words step, non-lexical elements such as numbers, punctuation marks, and double spaces are removed from the text. In the step of selecting valid part-of-speech, only two kinds of nouns and symbols are considered. Since nouns could refer to things, the intent of message is expressed better than the other part-of-speech. Moreover, the more illegal the text is, the more frequently symbols are used. The selected data is given 'legal' or 'illegal'. To make the selected data as learning data through the preprocessing process, it is necessary to classify whether each data is legitimate or not. The processed data is then converted into Corpus type and Document-Term Matrix. Finally, the two types of 'legal' and 'illegal' files were mixed and randomly divided into learning data set and test data set. In this study, we set the learning data as 70% and the test data as 30%. SVM was used as the discrimination algorithm. Since SVM requires gamma and cost values as the main parameters, we set gamma as 0.5 and cost as 10, based on the optimal value function. The cost is set higher than general cases. To show the feasibility of the idea proposed in this paper, we compared the proposed method with MLE (Maximum Likelihood Estimation), Term Frequency, and Collective Intelligence method. Overall accuracy and was used as the metric. As a result, the overall accuracy of the proposed method was 92.41% of illegal loan advertisement and 77.75% of illegal visit sales, which is apparently superior to that of the Term Frequency, MLE, etc. Hence, the result suggests that the proposed method is valid and usable practically. In this paper, we propose a framework for crisis management caused by abnormalities of unstructured data sources such as SNS. We hope this study will contribute to the academia by identifying what to consider when applying the SVM-like discrimination algorithm to text analysis. Moreover, the study will also contribute to the practitioners in the field of brand management and opinion mining.
