  • The investigation was aimed to study effects of Shirhyung-tang extract and Kamishirhyung-tang extract on the diuretic, antipyretic, anti-inflammatory, analgesic and the injured kidney induced by $HgCl_2$. The extract was administered to the experimental animals. The following results have been obtained. Duiretic effect was noted to be significant in extracts of Shirhyung-tang and Kamishirhyung-tang. Antipyretic effect was noted to be significant according to the density of Shirhyung-tang and Kamishirhyung-tang. Anti-inflammatory effect was noted to be significant in extract of Shirhyung-tang and Kamishirhyung-tang. Analgesic effect was noted to be significant in extracts of Shirhyung-tang and Kamishirhyung-tang.

  • To investigate the recorded efficacy in Oriental medical references and effects in clinics, experimental studies were undertaken to estimate 'Palmiwhan' to the diuretic effect in normal rats, rabbits and rats with $HgCl_2-induced$ acute renal failure and hypotensive effects on the nephrogenous hypertension in rabbits with $HgCl_2-induced$ acute renal failure. The results of this study were summarized as follows; Increase in urinary volume, urinary $Na^+\;and\;Cl^-$ excretions were significantly recognized in normal rats and rabbits. Increase in urinary volume, urinary $Na^+\;and\;Cl^-$ excretions were significantly shown in rats with 2mg/kg $HgCl_2-induced$ acute renal failure. Although plasma electrolyte and creatinine levels were little affected, plasma urea nitrogen levels were significantly elevated in normal rats and 2mg/kg $HgCl_2-induced$ acute renal failure of rats. Although the blood pressure was not affected in normal rabbits and cats, hypotensive actions due to the diuretic effect were recognized in rabbits $HgCl_2-induced$ acute renal failure.

  • It has been known that Gentianae macrophyllae Radix is reputed to be effective in removal of 'Pungseub' and 'Heoyeol', and to be analgesic. Since ancient times, however, there have been many examples of substituting Lycoctoni Radix for Gentianae macrophyllae Radix. For that reason, this experiment was conducted to compare the diuretic, analgesic, anti-inflammatory, anti-pyretic, anti-bacterial effect, and toxicity of Lycoctoni Radix with those of Gentianae macrophyllae Radix. The results obtained in this work were summarized as follows; In the test of acute toxicity, Lycoctoni Radix was found to be toxic in comparison to Gentianae macrophyllae Radix which was found to be non-toxic. Both herbs were found to be diuretic and Gentianae macrophyllae Radix was more effective than the other and to be ineffective for the blood pressure and respiration. Lycoctoni Radix was found to be more analgesic than the other. Gentianae macrophyllae Radix was found to be more anti-inflammatory of the two and both demonstrated anti-pyretic effect. Anti-bacterial effect was detected only in Lycoctoni Radix. It is concluded that Gentianae macrophyllae Radix has mild diuretic, analgesic, anti-inflammatory and anti-pyretic effect, but Lycoctoni Radix, due to its toxicity, can not be substituted for Gentianae macrophyllae Radix.

  • The pharmacological effects of an Ayurvedic diuretic drug 'Treenoponchomul' (TPM) was investigated in animal model. The pharmacological actions of the test drug along with that of the components thereof, on the Central Nervous System (CNS) were studied. The drug under study TPM showed little effect on the CNS, the same can not be said about the components. The most prominent CNS depressant effect was observed with Saccharum officinarum Linn. (EE) in that it lowered the spontaneous motor activity as well the exploratory -behavior of the animals. An exploration retarding effect of moderate degree, was evident with Imperata cylindrica Beauv. (UU), and Phragmites maxima Blatter & McCann (NN). Although the test drug did not alter the normal locomotor and/ or exploratory behavior of the treated animals, it did significantly (p<0.01) lower the locomotion of the amphetamine induced hyperactive animals. TPM along with its components (especially Desmostachya bipinnata Stapf. Root, KU), significantly reduced the gastro-intestinal motility of the treated animals (p<0.01). The test drug and its components lowered the body weight of the treated animals, on being administered chronically (30 days), with EE being the only exception.

Effect of 5-Hydroxytryptamine(5-HT) on Renal Function in Dog (5-Hydroxytryptamine(5-HT)이 개의 신장기능에 미치는 영향)

  • 5-Hydroxytryptamine(5-HT, serotonin), when given into the vein, produced antidiuretic action accompanied with reduction of glomerular filtration(GFR), renal plasma flow(RPF), osmolar clearance(Cosm) and amounts of sodium or potassium excreted in urine( $E_{Na}$ , $E_{K}$), with the augmented reabsorption rates of sodium and potassium in renal tubules. 5-HT, when infused into a renal artery, exhibited diuretic action accompanied with the augmented RPF and increased $E_{Na}$ and $E_{K}$ in only infused kidney. Antidiuretic action of 5-HT infused into the vein was not influenced by ketanserin, 5-H $T_2$receptor blockade, given into a renal artery, vein or carotid artery, by methysergide, 5-H $T_1$receptor blockade, given into a renal artery, whereas above antidiuretic action was inhibited by methysergide given into vein or carotid artery. Diuretic action of 5-HT infused into a renal artery in only experimental kidney was blocked by ketanserin injected into a renal artery, was not influenced by methysergide administered into a renal artery. Above results suggest that 5-hydroxytryptamine(5-HT) produced the antidiuretic action through central 5-H $T_1$receptor and the diuretic action through 5-H $T_2$receptor located in renal tubules of kidney.ney.

Influence of Oxytocin on the Renal Function of the Chicken (닭의 신장기능(腎臟機能)에 미치는 Oxytocin의 영향(影響))

  • The existence of oxytocin in the pituitary gland of chicken has been ascertained, but its physiological roles are still obscure. In the study the action of oxytocin on renal function of the chicken was investigated during water diuresis, utilizing clearance and the Sperber technique. The results obtained are summarized as follows: Oxytocin, like in many species of mammals, elicited a profound diuretic response in the chicken. Urine flow, excretion of electrolytes, as well as glomerular filteration rate increased, with intravenous infusion of $3{\sim}10\;m{\mu}/kg/min$. Oxtocin, infused into the renal portal circulation via hindleg vein in a dose of $3{\sim}13\;m{\mu}/kg/min$. elicited marked increase in urine flow, glomerular filteration rate and sodium excreted in the urine. The diuretic effect was more pronounced in the infused side. It is suggested that diuretic response to oxytocin in the chicken results from dual action of oxytocin: increase of GFR and inhibition of sodium reabsorption on the renal tubule. The possibility that oxytocin might act through some endogenous substances could be ruled out.

  • In anesthetized dogs, antidiuretic action of intravenously administered BRL 34915 (10.0~30.0 $\mu$/kg) was blocked by renal denervation, whereas it was not affected by glibenclamide, a selective $K_{ATP}$ blocker, given into renal artery. Diuretic action in ipsilateral kidney produced by intrarenal administration of BRL 34915 was not influenced by renal denervation, but blocked completely by glibenclamide given into the vein. Above results suggest that the antidiuretic action of BRL 34915 is mediated by renal sympathetic nerves and the diuretic action is caused by opening of $K^+$ channel within kidney.

  • In dogs, renal denervation did not affect the diuretic action accompanied with renal hemodynamic chanties and inhibition of electrolytes reabsorption rates in renal tubules by methoxyverapamil infused into the vein or into a renal artery, while renal denervation blocked the antidiuretic action due to the decreased free water and osmolar clearances along with the reduced sodium amounts excreted in urine by methoxyverpamil infused into the carotid artery. These experimental results suggest that methoxyverapamil may cause diuresis by direct action in kidney but the antidiuretic action through central function mediated by renal nerves.

  • The influence of penicillin G infusion(7.5 units/min/kg) on the diuretic action of furosemide$(250{\mu}g/kg)$ in rabbits was studied to investigate the drug interaction between penicillin G and furosemide. The results were as follows: 1) There was no significant change in renal function in the penicillin G infused rabbits compared with the normal ratbit. 2) In comparison with the control group, a significant increase in ${\triangle}U_{flow},\;{\triangle}U_{Na}V,\;{\triangle}U-kV\;and{\triangle}U_{cl}V$ was noted by 30 minutes following the administration of furosemide in the penicillin G infused group. There was no significant difference in ${\triangle}C_{In},\;{\triangle}C_{PAH}$ between the two groups and ${\triangle}Na^+$ reabsorption rate was significantly decreased in the latter. The potentiating effect of penicillin G on the diuretic action of furosemide should be due to the increased luminal concentration of free form of furosemide, and it may be related to the competitive inhibition of plasma protein binding between the two drugs.

  • This study was attempted to investigate on renal effect of ($\pm$)6-chloro-7,8-dihydroxy-1-phenol 2,3,4,5-tetrahydro-lH-3 benzazepine (SKF 81297), dopamine $D_1$ receptor agonist, in dog. SKF 81297, when gluten intravenously, produced diuretic action along with the increases of renal plasma flow (RPF), glomerular filtration rate (GFR), amounts of N $a^{+}$ and $K^{+}$ excreted into urine ( $E_{Na}$ , $E_{K}$) and osmolar clearance ( $C_{osm}$). It also decreased the reabsorption rates of N $a^{+}$ and $K^{+}$ in renal tubule ( $R_{Na}$ , $R_{K}$) and free water clearance ( $C_{H2O}$), whereas ratios of $K^{+}$ agonist N $a^{+}$ in urine and filtration fraction (FF) was not changed. SKF 81297, when administered into a renal artery, elicited diuresis both in experimental kidney given the SKF 81297 and control kidney not given, while the effect was more remarkable in experimental kidney than those exhibited in control kidney. SKF 81297 given into carotid artery also exhibited diuresis, the potency at this time, compared to those induced by intravenous SKF 81297, was magnusgreat. Above results suggest that SKF 81297 produces diuresis by both indirect action through changes of central function and direct action being induced in kidney. Central diuretic action is mediated by improvement of renal hemodynamics, but direct action by inhibition of electrolytes reabsorption in renal tubule.enal tubule. tubule.
