Cortex mori (Morus alba L.), the root bark of mulberry tree, has been used as an antiphlogistic, diuretic, and expectorant in herbal medicine. The purpose of this study was to determine whether Cortex mori could inhibit the ovalbumin (OA) -induced late asthmatic reaction in guinea pigs. Guinea pigs were sensitized by two exposures to an aerosol of OA(1.0%) and then challenged with aerosolized antigen(2.0%), The animals were pretreated by three inhalations of the aerosoled Cortex mori either before antigen sensitization or cahllenge. Bronchoalveolar lavage fluid(BALF) and peripheral blood were collected at 17 hours after OA challenge. The cell populations in BALF and peripheral blood were examined to determine the changes of the relative proportions of eosinophils, neutrophils and mononuclear cells etc. Beta-glucuronidase activity in BALF was measured to evaluate the alveolar macrophage activation. OA-induced histamine release from guinea pig peritoneal fluid cells was measured by radioisotope enzymatic asssay. Results were as follows. The number of eosinophils, neutriphils and lymphocytes recovered in BALF were significantly increased in the 17h following aerosol challenge with OA. Among them, eosinophil and neutriphils were decreased remarkably in group that had been preinhalated with Cortex mori. The number of lymphocytes in BALF were not decreased in group pretreated with CM before sensitization but decreased in Group pretreated with CM before challenge. After OA challenge, the number of eosinophils in peripheral blood were markedly increased, but Cortex mori inhibited significantly the OA-induced eosinophilia. Beta-glucuronidase activity in the supernatants of BALF were significantly increased in the 17h following aerosol challenge with OA, however, pretreatment of Cortex mori had no influence on Beta-glucuronidase activity, suggesting that Cortex mori had no inhibitory effect on OA-induced alveolar macrophage activation. Cortex mori inhibited the OA-induced histamine release from guinea pig peritoneal fluid cells. From the above results, it is suggested that Cortex mori contains some substances with an activity to inhibit the the OA-induced late phase reaction of the bronchial asthma in guinea pigs.
Zea mays Linne, which belongs to Graminae family, has been ordinarily used as edibles and fodder. Its stigma, Maydis Stigma, has been discarded without any special application with the exception of being used as diuretic in the sphere of Chinese medicine. Moreover, the pharmacological research toward Maydis Stigma as a medicinal herb has not been regretfully undertaken at all. The authors hereby paid attention to this point of view and made experiment to find out whether the water extract of Maydis Stigma might possess some pharmacological effect on the blood pressure of the rats or not. $LD_{50}$ to the mice was determined and the experiment was performed making the use of several agents related to the blood pressure such as atropine, Avil, dichloroisoproterenol and hexamethonium. The blood pressure of the rats and rabbits was recorded with the equipment of polygraph (Grass model 7 DAB) and its accessory, Statham pressure transducer. The results of the experiment are as follows. 1. $LD_{50}$ to the mice weighing 20g is determined 1,125 mg/kg. 2. The blood pressure manifests the gradual decrease by the rate of $-15.50{\pm}3.32,\;-22.14{\pm}8.73$ and $-46.25{\pm}5.17\;mmHg$ in proportion to ZW 25, 50 and 150mg/kg administered to the rats. 3. The blood pressure also manifests the same gradual decrease in proportion to ZW administered to the rabbits. 4. It is of significant difference to manifest the increase of $+5.14{\pm}4.65\;mmHg$ in the rats pretreated with atropine as compared with $-22.14{\pm}8.73\;mmHg$ of control group when administered each ZW 50mg/kg. 5. The administration of ZW 50 mg/kg to the rats pretreated with Avil, dichloroisoproterenol and hexamethonium does not manifest the significant difference in comparison with the control group.
The dried roots of Rehmannia glutinosa (RG) have been used in the traditional medicine for treatment of diabetes, fever and dysuria, etc. In order to investigate the quality evaluation of the dried roots of RG, we conducted the physico-chemical and biological evaluation method. The amount of catalpol from commercial samples is very diverse about $0.00{\sim}3.89%$ by using HPLC method, because it is easily decomposed by processing of RG. So, we should try to identify the correlation with the contents of catalpol and biological activities of RG. We chose 3 samples which were a wide difference of catalpol contents between each sample (Sample-I; 3.4%, Sample-II; 2.8%, Sample-III; 0.05%). Sample-I and Sample-II were found to be more effective than Sample-III on the DPPH free radical scavenging effect and inhibitory effect on $H_2O_2-catalyzed$ lipid peroxidation in rat liver in vitro. And, Sample-I and Sample-II exhibited more significant effects than Sample-III on accelerating actions of the small and large intestinal transport, and diuretic action in mice. So, it is suggested that the quantitative determination of catalpol should be required for the tandardization of the dried roots of RG.
Prunellae Spica, the herbaceous plant in the genus Prunella, is a traditional herbal medicine and has been reported to have diuretic, anti-bacterial and anti-oxidant effects. However, the mechanism of its action was not clearly identified. In the present study, we investigated the hepatoprotective effect of Prunellae Spica extract (PSE) against the damage of mitochondria and death in hepatocyte induced by oxidative stress. Treatment of arachidonic acid (AA)+iron significantly induced oxidative stress and apoptosis in the hepatocytes. However, PSE protected cells and inhibited apoptosis by altering the protein levels such as poly(ADP-ribose) polymerase and pro-caspase 3. Moreover, AA+iron induced reactive oxygen species production and mitochondrial dysfunction, and Both of them were inhibited by PSE treatment. PSE markedly activated AMP-activated protein kinase (AMPK), an important regulator in cell survival. Furthermore, this activation by PSE was mediated with liver kinase B1, a major upstream kinase that phosphorylates Thr 172 of AMPKα, and this activation was associated with its cell protection, as assessed by an experiment of a chemical inhibitor. In conclusion, this study demonstrate that PSE protects hepatocytes against oxidative stress as mediated with activation of LKB1-dependent AMPK pathway.
lntrarenal resistive index (RI) was determined in normal, sedated and diuretic kidneys. The mean RI values of normal dogs were $0.64{\pm}0.03$ without statistically significant difference between left and right kidney. Interrenal RI difference (${\Delta}RI$) was $0.03{\pm}0.01$. No significant relationship was found between mean intrarenal RI versus body weight. The mean intrarenal RI values after administration of acepromazine, xylazine, thiopental, and ketamine were $0.66{\pm}0.03$, $0.64{\pm}0.03$, $0.56{\pm}0.05$, and $0.50{\pm}0.05$, respectively. After administration of acepromazine or xylazine, a significant change of the RI was not found. But, a significant decrease of the RI following thiopental or ketamine administration could be observed as compared with the normal RI. The mean intrarenal RI values were $0.63{\pm}0.06$ and $0.62{\pm}0.04$ at 10 minutes and 30 minutes after injection of furosemide, respectively. No significant change of intrarenal RI was found after administration of furosemide. But, mannitol significantly decreased mean intrarenal RI to $0.57{\pm}0.02$ and $0.58{\pm}0.03$ at 30 minutes and 60 minutes, respectively. Based on the obtained results, values of 0.72 and 0.05 may be proposed as the reasonable upper limits of RI and ${\Delta}RI$ of normal average. Acepromazine or xylazine are recommended as anesthetics for renal diseases because they have less effect on the RI. Further study using mannitol injection to increase ${\Delta}RI$ may be suggested in unilateral urinary tract obstruction since mannitol more effectively changed mean RI than furosemide.
Background: Taraxacum platycarpum has been used in traditional medicine in Korea to treat intoxication and edema and as a diuretic. According to previous reports, it has anti-cancer, anti-gastritis, and anti-inflammation effects. However, the improvement effect of T. platycarpum on rheumatoid arthritis has not been investigated. The anti-oxidative and anti-inflammation effects of the aerial parts of T. platycarpum are different from those of its subterranean parts. Thus, we evaluated the effect of the water extracts of Taraxaci radix (WTR) on type II collagen-induced rheumatoid arthritis (CIA) in animal models. Methods and Results: Rheumatoid arthritis was induced by type II collagen. WTR (100 mg/kg and 500 mg/kg) was administered to the animal models. Methotrexate was used as the positive control. The levels of interleukin-6, TNF-alpha, and type II collagen IgG in the animals were measured by using enzyme-linked immunosorbent assay. Treatment with 500 mg/kg WTR decreased the serum levels of interleukin-6, TNF-alpha, and collagen IgG in the CIA models. Moreover, treatment with WTR diminished the arthritisinduced swelling of the hind legs and monocyte infiltration in the bloodvessels of the animal models. Conclusions: These results indicate that WTR has the potential to improve rheumatoid arthritis by reducing the levels of inflammatory cytokines such as interleukin-6 and TNF-alpha. However, further experiments are required to elucidate the influence of WTR on signal transduction in vitro and in vivo.
In order to examine that the effect of Sam Hwa San, circulating the vital energy of Sam Cho and controlling body fluid metabolism, gives any influence on renal function, changes in the urine flow, eletrolytes excretion, plasma aldosterone concentration and renin activity were observed after intravenous infusion of the Sam Hwa San extract in rabbit. Also in vitro effect of the herb extract on oxygen consumption in renal cortical slices and ATPase activity in kidney microsomes was measured. The following results were obtained : 1. The urine flow was markedly increased at 10 min after intravenous infusion of the Sam Hwa San extract($0.134{\pm}0.015$ vs. $0.433{\pm}0.046ml/min.kg$), but return ed to normal value after 40 min of infusion. 2. The glomerular filtration rate was significantly increased at 10 min after in travenous infusion of the Sam Hwa San extract, and the renal plasma flow at 10 and 20 min after infusion of the Sam Hwa San extract, following return to normal value. 3. $Na^+$ excretion was significantly increased during 10-40 min after intravenous infusion of the Sam Hwa San extract, although showed the maximal rate at 10-20 min. The fractional $Na^+$ excretion was also increased during 10-40 min. $K^+$ excretion was rapidly increased at 10 min after the intravenous Infusion of the Sam Hwa San extract and then gradually decreased to normal level at 40 min. The fractional $K^+$ excretion was significantly increased during 10-40 min after the intravenous infusion of the Sam Hwa San extract. 4. The plasma aldosterone concentration and renin activity were not altered by the infusion of the Sam Hwa San extract. 5. The ouabain-sensitive oxygen consumption of renal cortical slices was significantly reduced by the Sam Hwa San extract(0.5 and 1.0 vol.%). 6. The Na-K-ATPase activity of renal microsomes was strongly inhibited by the Sam Hwa San extract(0.5 and 1.0 vol.%). These results suggest that the Sam Hwa San causes a strong diuretic effect which results from reduction of Na reabsorption in renal tubule by a direct inhibition of Na-pump and, in part, from all increase in renal blood flow. In clinic, it is considered to obtain the therapeutic effect in body fluid metabolism disharmony to cause the circular disorder of vital energy.
Astragalus membranaceus has a long history of medicinal use in Chinese herbal medicine. It has been shown to have immunostimulant, tonic, antioxidant, antiperspirant, diuretic, anti-diabetic, expectorant properties, and a supplementary medicine during cancer therapy. In this study, we investigated the effect of anti-oxidation of Astragalus membranaceus root extract. The anti-oxidative activities of water, 80% methanol, and 100% methanol extracts from Astragalus membranaceus were analyzed by DPPH free radical scavenging activity, Superoxide dismutase-like activity, reducing power, and crude ash. The water extract demonstrated to be more effective than methanol extract for a DPPH radicals scavenging activities and reducing power. Superoxide dismutase-like activity showed higher efficiency in 80% methanol extract. Our results indicate that Astragalus membranaceus extracts could be used as a source of antioxidant ingredients in the food industry.
Aconiti Tuber is the root of Aconitum sp (Ranunclaceae) which has been considered as one of the most important medicinal plant having cordiotonic, diuretic and analgesic effect. On the other hand, it has been known that Aconiti Tuber contained toxic agent, aconitine alkaloids so that only processed Aconiti Tubers have been used as herbal drug traditionally. For the safety evaluation of processed Aconiti Tuber, quantitative determination of aconitine and acute, subacute toxicity test were performed on 5 commercial processed Aconiti Tubers. Arapid and precise method using HPLC has been developed for the separation and determination of aconitine. Samples were extracted with hydrochloric acid (pH3) and hot water decoction. In case of d-HCL extracts, the contents of aconitine were from 0.08 mg/g to trace. But in case of hot water decoction extracts, the contents of aconitine were not detected. For the investigation of Aconiti Tuber toxicity in rats, hot water decoction samples and methanol extracts were tested. 1) Acute toxicity test Hot water decoction sample and methanol extracts from Aconiti Tuber did not show any toxic effects in rats by an oral administration. $LD_50values of 2 extracts were above 10.0 g/kg. 2) Subacute toxicity study In the repeated administration study, hot water decoction samples were given orally to Sprague-Dawlay rats for 2 week at daily doses of 5.0 g/kg. The results are as follows; No toxic manifestation, body weight changes and lethality were observed during wxperimental period. There were no significant changes in serum enzyme activities such as GOT, GPT, LDH, ALP between treated and control groups. However CPK values were decreased in the Subuja-treated group. (P<0.01). In addition, no gross and microscopic changes were noted in Aconiti Tuber-treated groups.
