A Possible Physiological Role of Caspase-11 During Germinal Center Reaction

  • Caspase-11 has been known as a dual regulator of cytokine maturation and apoptosis. Although the role of caspase-11 under pathological conditions has been well documented, its physiological role has not been studied much. In the present study, we investigated a possible physiological function of caspase-11 during immune response. In the absence of caspase-11, immunized spleen displayed increased cellularity and abnormal germinal center structure with disrupted microarchitecture. The rate of cell proliferation and apoptosis in the immunized spleen was not changed in the caspase-11-deficient mice. Furthermore, the caspase-11-deficient peritoneal macrophages showed normal phagocytotic activity. However, caspase-11-/-splenocytes and macrophages showed defective migrating capacity. The dysregulation of cell migration did not seem to be mediated by caspase-3, interleukin-$1{\alpha}$ or interleukin-$1{\beta}$ which acts downstream of caspase-11. These results suggest that a direct regulation of immune cell migration by caspase-11 is critical for the formation of germinal center microarchitecture during immune response. However, humoral immunity in the caspase-11-deficient mice was normal, suggesting the formation of germinal center structure is not essential for the affinity maturation of the antibodies.

  • Approximately 200 individual skeletal elements, which differ in shape and size, are the building blocks of the vertebrate skeleton. Various features of the individual skeletal elements, such as their location, shape, growth and differentiation rate, are being determined during embryonic development. A few skeletal elements, such as the lateral halves of the clavicle and parts of the skull are formed by a process called intramembranous ossification, whereby mesenchymal cells differentiate directly into osteoblasts, while the majority of skeletal elements are formed via endochondral ossification. The latter process starts with the formation of a cartilaginous template, which eventually is being replaced by bone. This requires co-regulation of differentiation of the cell-types specific for cartilage and bone, chondrocytes and osteoblasts, respectively. In recent years it has been demonstrated that Wnt family members and their respective intracellular pathways, such as non-canonical and the canonical $Wnt/{\beta}$-catenin pathway, play important and diverse roles during different steps of vertebrate skeletal development. Based on the recent discoveries modulation of the canonical Wnt-signaling pathway could be an interesting approach to direct stem cells into certain skeletal lineages.

Utilization of Ferroproteins by Candida albicans during Candidastasis by Apotransferrin

  • Many reports have stated that some of the pathogenic bacteria can obtain iron from ferroproteins, such as cytochrome C, ferritin, hemin, hemoglobin, and myoglobin. These reports prompted us to determine if an opportunistic pathogenic fungus, Candida albicans, can utilize ferroproteins to circumvent the iron-regulatory effect of transferrin. The following assays were carried out to measure in vitro growth stimulation by the ferroproteins: as an initial step, C. albicans was cultured in iron-free (pretreated with apotransferrin for 24h) culture medium. Once Candida albicans yeast cell growth reached stasis from iron starvation, individual ferroproteins were added to the culture media. Results showed that hemin, hemoglobin, and myoglobin supported a partial growth recovery. Additional studies with haptoglobin, a serum protein that interacts with the globin moiety of certain ferroproteins, established that C. albicans could obtain iron from the haptoglobin-ferroprotein complexes. These data indicate that the heme part of the ferroproteins is the source of iron. This implies that heme oxygenase, CaHMX1 might be involved in bringing about dissociation of heme-containing protein for iron-acquisition. In addition, anticandidal activity of transferrin takes place not only by the process of iron regulation, but also by direct interaction with the yeast cells.

Autophagy as an Innate Immune Modulator

  • Autophagy is a fundamental cellular process in eukaryotic cells for maintaining homeostasis by degrading cellular proteins and organelles. Recently, the roles of autophagy have been expanded to immune systems, which in turn modulate innate immune responses. More specifically, autophagy acts as a direct effector for protection against pathogens, as well as a modulator of pathogen recognition and downstream signaling in innate immune responses. In addition, autophagy controls autoimmunity and inflammatory disorders by negative regulation of immune signaling. In this review, we focus on recent advances in the role of autophagy in innate immune systems.

  • GDI(Gasoline Direct Injection( engine technology is well known as a new technology since it can improve fuel consumption and meet future emission regulations. But the GDI has many difficulties to be solved, such as complexity of injection control mode, unburned hydrocarbon, and restricted power. A 2-D shape combustion chamber was adopted to investigate mixture formation and combustion characteristics of GDI engine. Spray and combustion experiments were performed by changing the injection timing. injection pressure an din-cylinder flow in Rapid Compression and Expansion Machine(RCEM).Through the experiments, the detailed characteristics of fuel spray and combustion was analyzed by visualizing the in-cylinder phenomena according to the change of injection condition, and the optimal fuel injection timing and fuel injection pressure were obtained.

Regulation of Cyclic AMP-Response Element Binding Protein Zhangfei (CREBZF) Expression by Estrogen in Mouse Uterus

  • CREBZF (cAMP-response element binding protein zhangfei) is a member of ATF/CREB family, and which regulates various cellular functions by suppressing major factors with direct interaction. In this study, we have examined the expression of CREBZF on mouse endometrium during uterus estrous cycles and estrogen (E2) treatment. In uterus, CREBZF mRNA expression was higher than other organs and mRNA and protein of CREBZF was increased in proestrus phase and decreased in estrus phase. The expression of CREBZF in 3-weeks old mouse uterus was reduced by E2 injection in endometrium. In addition, the expression of progesterone receptor, a marker of E2 in ovariectomized mice was found to be strongly expressed in stroma, while CREBZF was only expressed in epithelium. Also, we conformed that E2-suppressed CREBZF was restored by co-injection of ICI 182,780, an estrogen receptor antagonist. Overall, these results suggest that CREBZF is regulated by estrogen and involved in ER signaling pathway in mouse uterus.

Numerical Analysis for Development of Vertical Falling Prevention System

  • Various falling prevention systems with vertical guided rail have been supplied in order to prevent falling accidents and acquire the long life and cost down for the maintenance. Almost the imported system is composed of a guided rail and the comfort components. It has the mechanism that cannot be dismounted from the guided rail at the accident site. Thus, the aim of this study is to develop a falling prevent system that has a direct dismounting function, which can release easily from the falling accident point. This system has a high durability and robust. And the cost efficiency of the system and safety for workers also are improved remarkably. From the results, we showed the proposed new falling system is sufficiently safety and satisfies the KOSHA regulation through the numerical stress analysis. Also it is expected as the useful and effective safety device for workers.

열차운행에 따른 철도역사의 진동영향 평가 (The Vibrational Evaluation of Railway Station by the Train Service)

  • The high interest for the ground vibrations which is caused by the train service is risen. When the trains are passed, a serious force is applied to ground and the caused vibration is propagated to the area of the building by the ground and rocks. This vibration comes to feel in the residents with the direct vibration, it is radiated in sound. The caused vibration and radiated sound affect the human's life, and this vibration brings about the operation interruption of the equipment which is sensitive to the vibration, or will bring about the structural damage of the building. In this study, the effect of the vibrations caused by the passing trains on the railway station and buildings is investigated by vibration measurement. Indoor and outdoor measurement is carried out by each trains.

Fast Screening of Harmful Disinfectants in Household Products via Low-Temperature Plasma Ionization-Mass Spectrometry

  • Isothiazolinone derivatives are widely used in consumer products as disinfectants or preservatives, but there are growing concerns about their impact on human health. Therefore, rapid screening of these biocides is very important for proper control and regulation of potentially hazardous substances. To this end, low-temperature plasma (LTP) ionization mass spectrometry (MS) was investigated to demonstrate its potential for direct and selective analysis of isothiazolinones from sprayed aerosol samples. Benzisothiazolinone (BIT) was clearly identified from a commercial fabric deodorant using LTP ionization MS and MS/MS. LTP allowed selective ionization of BIT directly from the simply sprayed aerosol sample and illustrated its potential for fast screening without sample pre-treatments. Selective nature of LTP ionization, on the other hands, implicates use of LTP ionization MS as a general screening method for specific groups of hazardous chemicals in commercial products.

Understanding insomnia as systemic disease

  • Sleep plays a critical role in homeostasis of the body and mind. Insomnia is a disease that causes disturbances in the initiation and maintenance of sleep. Insomnia is known to affect not only the sleep process itself but also an individual's cognitive function and emotional regulation during the daytime. It increases the risk of various neuropsychiatric diseases such as depression, anxiety disorder, and dementia. Although it might appear that insomnia only affects the nervous system, it is also a systemic disease that affects several aspects of the body, such as the cardiovascular, endocrine, and immune systems; therefore, it increases the risk of various diseases such as hypertension, diabetes mellitus, and infection. Insomnia has a wide range of effects on our bodies because sleep is a complex and active process. However, a high proportion of patients with insomnia do not seek treatment, which results in high direct and indirect costs. This is attributed to the disregard of many of the negative effects of insomnia. Therefore, we expect that understanding insomnia as a systemic disease will provide an opportunity to understand the condition better and help prevent secondary impairment due to insomnia.
