  • WR 2721 (S-2-(3-aminopropylaminoethylphosphorothioate) is a radioprotective drug that is now undergoing clinical trials in the United States and Japan. a liquid chromatographic electrochemical method for the determination of WR 2721 an WR 3689 [S-2-(3-methylaminopropylamino)ethylphorothioate] in human plasma was developed in this study. This method includes the use of a Hg/Au electrochemical detector and a cyano column for the direct measurement of WR 2721 and WR 3689 in plasma. An analog of WR 2721, WR 149846 was used as an internal standard. WR 2721 and WR 3689 could be well separated from the solvent front, with a mobile phase of acetonitrile-water (20:80), 0.1M acetic acid and 1.2 mM sodium octane sulfonate. This method was shown to be precise. Both intra-day and inter-day results were within 10% CV. Also, sample preparation was fairly simple. Since WR 2721 and WR 3689 were unstable at room temperature, it was essential to use an automatic sample processor with a refrigerator, especially for carrying out routine analyses.

  • Streptomyces venezuelae ATCC 15439 is notable in its ability to produce two distinct groups of macrolactones. It has been reported that the generation of two macrolactone structures results from alternative expression of pikromycin (Pik) polyketide synthase (PKS). It was previously reported that the hybrid pikromycin-tylosin PKS can also produce two different macrolactones but its mechanistic basis remains unclear. In order to address this question, a series of site-directed mutagenesis of tentative alternative ribosome binding site and translation start codons in tylGV were performed. The results suggest that macrolactone ring size is not determined by the alternative expression of TylGV but through other mechanism(s) involving direct interaction between the PikAIII and TE domain or skipping of the final chain elongation step. This provides new insight into the mechanism of macrolactone ring size determination in hybrid PKS as well as an opportunity to develop novel termination activities for combinatorial biosynthesis.

  • The apartment will be much more convenient than the other one in the application of green technologies, providing the merits by mass production. For example, pre-manufactured building materials can be effectively adapted to the formation of buildings. Recently, the form of double-enveloped window system has been developed for the purposed of minimizing energy loss occurred around windows. On the other hand, the expansion of balcony area was legalized and thus, a visual buffer area does not exist any more. All-glass window wall on apartment houses without a balcony produces pretty harmful area with direct sunbeam. A various types of glass can be applied to the window system and it plays a critical role in the determination of inner visual environment. For the purpose of verifying the potential of its daylighting, a mock-up model has been constructed and tested. The refurbished version of conventional window shows its illuminated characteristics. Clear and Low-e glass have been adapted for window area and daylighting ratio has been compared in both test cells.

  • The present work proposes a simple electrochemical method applicable to any immobilization processes of oxidase using a Clark type oxygen electrode as a base transducer. The present work suggests an optimal immobilization technique among three different methods of glucose oxidase(GOD) onto one side of $37[\mu}$mthick blend membranes, composed o 80% of cellulose triacetate and 20% of polycaprolactone, on the basis of the maximum Michaelis-Menten parameter(Vm) determined by either steady state or transient analyses. The electrode system was made of disk type gold cathode(4mm diameter) and Ag/AgCl anode. One side of the blend membrane was in contact with the cathode surface while the other side was immobilized with GOD either in covalent-bond or cross-linked forms, the latter being covered by $25{\mu}$m thick dialysis membrane of cellulose acetate. The resultant current density was on-line monitored by a potentiostat while glucose level was varied from 1 to 20 mM. The present study shows that direct cross-linking of GOD with glutaraldehyde was mostly preferred for fabrication of glucose sensor, on the basis of resultant kinetic parameters from either steady state or transient analyses.

  • Offshore floater such as FPSO, drillship is composed of topside and hull side, and the interface structure is called topside module support. In this study, practical considerations were investigated for the design of topside module supports, from the concept design stage to the final stage of structural determination, in view of design efficiency and construction productivity. The effects of welding design factors of topside module support, such as welding throat thickness, sectional welding area, and welding man-hours, were compared and analyzed closely with respect to productivity. The current status and problems regarding the application of deep or full penetration welding are discussed, and a direct-calculation method is suggested as a possible solution to these problems.

Compositional Study of Surface, Film, and Interface of Photoresist-Free Patternable SnO2 Thin Film on Si Substrate Prepared by Photochemical Metal-Organic Deposition

  • The direct-patternable $SnO_2$ thin film was successfully fabricated by photochemical metal-organic deposition. The composition and chemical bonding state of $SnO_2$ thin film were analyzed by using X-ray photoelectron spectroscopy (XPS) from the surface to the interface with Si substrate. XPS depth profiling analysis allowed the determination of the atomic composition in $SnO_2$ film as a function of depth through the evolution of four elements of C 1s, Si 2p, Sn 3d, and O 1s core level peaks. At the top surface, nearly stoichiometric $SnO_2$ composition (O/Sn ratio is 1.92.) was observed due to surface oxidation but deficiency of oxygen was increased to the interface of patterned $SnO_2/Si$ substrate where the O/Sn ratio was about 1.73~1.75 at the films. This O deficient state of the film may act as an n-type semiconductor and allow $SnO_2$ to be applied as a transparent electrode in optoelectronic applications.

Direct digital radiography versus conventional radiography for estimation of canal length in curved canals

  • Purpose : The purpose of this study was to compare the conventional and digital radiography in the estimation of working length in mandibular molars. Materials and Methods : Sixty molar teeth were selected and divided into three groups in the basis of canal curves (0-$15^{\circ}$, 15-$30^{\circ}$, > $30^{\circ}$). After the placement of a 15 K-file, radiographs were taken with a conventional film (Fspeed) and a digital sensor. Canal lengths were measured in these images by two observers. Statistical analysis was performed with repeated measures of ANOVA and paired sample t-test with 95% confidence. Results : There was a high inter-observer agreement on the measurements of working length in conventional and digital radiographs. There was no significant difference between the mean values of measurements in conventional and digital radiography. Moreover, there was no significant difference between conventional and digital radiography with the actual values in the basis of canal curves. Conclusion : The accuracy of conventional and digital radiography in the determination of the working length was in an acceptable range.

  • Since carbohydrates are major mediators in cell-to-cell adhesion and communication, the development of specific and strong binders against them could generate promising therapeutics. As the first step towards that goal, sugar molecules have to be immobilized to be used as an affinity matrix. The amino functionality in sugar is the most active nucleophile for the immobilization, if the amino group is available. An alternative and general method is to use the hydroxyl group as a direct nucleophile, but the quantitation of immobilized hydroxyl groups is not easily done. To overcome this limitation, we have developed a method to immobilize various isomers of monosaccharides with p-nitrophenyl groups to the beads by using their hydroxyl groups. It was found that the amount of immobilized sugar was independent of the structure of the sugar, but was dependent on the number of hydroxyl groups. We also developed a sensitive method to quantify the amount of immobilized sugar at the picomolar scale by utilizing commercially available glycosidases to release a sensitive reporter molecule, p-nitrophenol, and detect it by HPLC. This new technique would allow a facile quantitation method for immobilized sugar molecules, which could be used as the affinity matrix to develop strong binders against biologically important sugars.

Cavity as a New Passive Device for Reduction of Skin Friction and Heat Transfer (새로운 수동제어소자인 공동을 이용한 마찰력과 열전달 감소에 관한 연구)

  • In order to examine the possibility of using a cavity as a passive device for reduction of skin friction and heat transfer, an intensive parametric study over a broad range of the cavity depth and length at different Reynolds numbers is performed for both laminar and turbulent boundary layers in the present study. Direct and large eddy simulation techniques are used for turbulent boundary layers at low and moderate Reynolds numbers, respectively. for both laminar and turbulent boundary layers over a cavity, a flow oscillation occurs due to the shear layer instability when the cavity depth and length are sufficiently large and it plays an important role in the determination of drag and heat-transfer increase or decrease. For a cavity sufficiently small to suppress the flow oscillation, both the total drag and heat transfer are reduced. Therefore, the applicability of a cavity as a passive device for reduction of drag and heat transfer is fully confirmed in the present study. Scaling based on the wall shear rate of the incoming boundary layer is also proposed and it is found to be valid in steady flow over a cavity.

A Study on the Distributions of Paraoxonase Activity and the Factors affecting Paraoxonase Activity of a Rural Population (일부 농촌주민 혈중 Paraoxonase 활성도의 분포 및 관련인자에 관한 조사연구)

  • The paraoxonase (E. C. 3.1.1.2) is a major enzyme to detoxicate the organophosphorus and carbamate which are the most widely used as the agricultural spraying insecticides. To investgate the distributions of plasma paraoxonase activity and the factors affecting the enzyme activity, the plasmas of 945 Korean rural population were analysed with the modified Krisch's direct sphectrphotometry method. Three indices of the enzyme activity - basal activity, stimulated activity (by NaCl), % stimulation - were obtained from the analysis. Three indicies suggested unimodal distributions, so we couldn't identify the low activity group risk group to organophosphorus & carbamate insecticides poisoning. There is no significant relation between 3 actvity indicies and sex, age, or history of insecticide use (p>0.05). The basal activity and the stimulated activity have significant relationship and high coefficient of determination with the activities of their parents ($r^2$=0.30, 0.24 ; p<0.05), but the % stimulation does not ($r^2$=0.02 ; p<0.05). These results suggest that the activity of paraoxonase is determined mainly by the genetic factor.
