In Vitro Free Radical and ONOO- Scavengers from Sophora flavescens

  • Activity-guided fractionation of the CH$_2Cl_2$-soluble fraction of the roots of Sophora flavescens furnished five 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scaveng ers: trans-hexadecyl ferulic acid (1) cis-octadecyl ferulic acid (2), trans-hexadecyl sinapic acid (3), (-)-4-hydroxy-3-methoxy-(6aR,11aR)-8, 9-methylenedioxypterocarpan (4) and desmethylanhydroicaritin (8), along with nine known inactive compounds: (-)-maackiain (5), xanthohumol (6), formononetin (7), (2S)-2'-methoxykurarinone (9), (2S)-3${\beta}$,7,4'-trihydroxy-5-methoxy-8-(${\gamma},{\gamma}$- imethylallyl )-flavanone (10), (2S)-7,4'-dihydroxy-5-methoxy-8- (${\gamma},{\gamma}$-dimethylallyl ) -flavanone (11), umbelliferone (12), kuraridin (13), and trifolirhizin (14). Compounds 1-4 and 8 exhibited DPPH free radical scavenging effects at IC$_{50}$ values of 33.01 ${\pm}$ 0.20, 57.06 ${\pm}$ 0.16, 39.84 ${\pm}$ 0.36, 35.83 ${\pm}$ 0.47, and 18.11 ${\pm}$ 0.04${\mu}$M, respectively. L-Ascorbic acid, when used as a positive control, exhibited an IC$_{50}$ value of 7.39 ${\pm}$ 0.01 ${\mu}$M. Compounds 1-4 and 8 also appeared to exert significant scavenging effects on authentic ONOO-, with IC$_{50}$ values of 5.76 ${\pm}$ 1.19, 15.06 ${\pm}$ 1.64, 8.17 ${\pm}$ 4.97, 1.95 ${\pm}$ 0.29 and 4.06 ${\pm}$ 2.41 ${\mu}$M, respectively. Penicillamine (IC$_{50}$= 2.36 ${\pm}$ 0.79${\mu}$M) was used as a positive control. In addition, compounds 2,4,6,8, and 10 were isolated from this plant for the first time.

Chungkookjang Fermentation of Mixture of Barley, Wormwood, Sea Tangle, and Soybean (보리, 쑥, 다시마, 대두 혼합물의 청국장 발효)

  • Barley, wormwood, sea tangle, and soybean were fermented by Bacillus licheniformis B 1 to make Chungkookjang with better flavor and aroma. Maximal protease activity in mixed grains was observed one day after inoculation. pH increased to 8.4 two days after inoculation. Browning material derived from interaction between sugar and amino acids increased 20-fold. Thus, it is proved that Chungkookjang can be made in the mixed grains. Antioxidant activities of mixed fermented grains dissolved in ethanol or methanol (0.2 and 1 %) increased depending on their concentrations. Antioxidant activities were determined using 1,1-diphenyl-2-picrylhydrazyl (DPPH). One % of powder Chungkookjang dissolved in methanol showed highest antioxidant activity. Systolic blood pressure of hypertensive volunteers who took 20 g of mixed fermented powder decreased on average by 10 mmHg in 2 h. Preference of mixed fermented soybean containing barley, wormwood, sea tangle to fermented soybean was demonstrated by t-test analysis. Mixed fermented grains can be developed as a functional food to lower hypertension.

Antioxidant Activity of Fermented Barley, Wormwood, Sea Tangle, and Soybean (발효 보리, 쑥, 다시마, 대두의 항산화효과)

  • Superoxide is involved in causing inflammation, cancer, and arteriosclerosis in many cases. Taking antioxidant material can be helpful in preventing the diseases. Natural food such as barley, wormwood, sea tangle, and soybean contain antioxidant ingredients. Antioxidant activity increase was determined by fermenting them with microorganism. To determine the activity, 1,1-diphenyl-2-picrylhydrazyl (DPPH) solution was used. When barley, wormwood, sea tangle, and soybean were fermented with Bacillus lichenifomis Bl, antioxidant activities of each fermented product increased 2.6, 1.6, 2.7, and 1.7 folds, respectively. Also, absorbance of fermented soybean was higher than that of soybean at the range of 250~290nm, which might be involved in differences of antioxidant activity of the two. Paraquat suppressed Esherichia coli DH5$\alpha$ growth by making superoxide inside the strain. However, when ethanol extract from fermented soybean was added into the GM (glucose-mineral) media containing the strain, its growth was recovered, suggesting that ethanol extract can move across E. coli, and can function as anti-oxidant material in vivo. Thus, it will be possible to develope antioxidant material from fermented soybean which can be taken orally.

Determination of Radical Scavenging Activity of Aster yomena (Kitam.) Honda (쑥부쟁이 추출물의 라디칼 소거활성 평가)

  • In this study, we investigated the antioxidative effects of AY by measuring 1.1-Diphenyl-2-picrylhydrazyl (DPPH), hydroxyl radical ($^{\cdot}OH$), and superoxide radical ($O_2{^-}$) scavenging activities in vitro. AY was extracted with ethanol and then partitioned with n-hexane, methylene chloride ($CH_2Cl_2$), ethylacetate (EtOAc) and n-butanol (n-BuOH). In the DPPH radical scavenging assay, AY at concentrations of 10 to $100{\mu}g/mL$ dose-dependently increased inhibition of DPPH oxidation, with the EtOAc fraction of AY showing the highest DPPH radical scavenging activity fractions. The $^{\cdot}OH$ radical scavenging activities of the extract and four fractions of AY increased by over 80% at a concentration of $50{\mu}g/mL$. In particular, the IC50 value of the EtOAc fraction was $0.03{\mu}g/mL$, which was the lowest value among all fractions. We also found that the EtOAc fraction of AY was better at $O_2{^-}$ radical scavenging than other fractions. Taken together, these results suggest that AY, especially the EtOAc fraction, can be used as a natural antioxidant against free radicals.

Isolation of Flavonoids and a Cerebroside from the Stem Bark of Albizzia julibrissin

  • From the EtOAc fraction of the MeOH extract of Albizzia julibrissin (Leguminosae), a rare 5-deoxyflavone (geraldone, 1), isookanin (2), luteolin (3), an isoflavone (daidzein, 4), five prenylated flavonoids [soph6f1avescenol (5), kurarinone (6), kurarinol (7), kuraridin (8) and kuraridinol (9)], a cerebroside (soya-cerebroside I, 10), and $(-)-syringaresinol-4-O-{\beta}-D-glucopyranoside$ (11) were isolated and characterized on the basis of spectral data. Compounds 2, 3, and 11, showed 1, 1-diphenyl-2-picrylhydrazyl radical scavenging activity.

Antioxidation activity of residue after omija (Schisandra chinensis) juice extract (오미자(Schisandra chinensis) 착즙박 추출물의 항산화 활성)

  • The physicochemical properties and schizandrin contents of various solvent ($H_2O$, 50% EtOH, 75% EtOH, 95% EtOH) extracts from residue after Omija juice was investigated using total polyphenol contents (TOC), 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity (RAS), anthocyanin contents (ANC), and schizandrin contents level (SCL). Total polyphenol contents, radical scavenging activity, and anthocyanin contents of 50% EtOH extract were the highest among all residue after Omija juice extracts, and was 16.70 mg/mL in the TOC and 86.16% in the DPPH-RAS. This meant that 50% EtOH extract from residue after Omija juice had more available antioxidant matters. As extraction time increases all extract treatments significantly reduced in the ANC contents (p <0.05). Amount of the SCL were observed higher value in 95% EtOH extract of residue after Omija juice.

Antioxidant and Blood-Pressure Reduction Effects of Fermented Soybean, Chungkookjang (청국장의 항산화 및 혈압강하 효과)

  • Fermented soybean, Chungkookjang has diverse bioactive compounds including antioxidants and peptides. Ethanol extract from Chungkookjang exhibited absorbance of 0.55 at 285 nm, where amino acids and peptides containing phenol are known to exist. Antioxidant activity of Chungkookjang was determined using the 1,1-diphenyl-2-picrylhydrazyl (DPPH) method. With increasing concentrations of ethanol extracts, their antioxidant activities increased. Blood pressure was determined every two hours after taking raw Chungkookjang which does not contain salts. In 6 h, systolic blood pressure dropped by 14 mmHg, and diastolic one dropped by 8 mmHg, which was statistically significant. Daidzein, antioxidants, angiotensin I-converting enzyme (ACE) inhibitor such as Lys-Pro which are rich in Chungkookjang might contribute to the reduction of blood pressure.

  • The extract of EA lacks studies showing its efficacy other than that it contains caffeic acid, an active compound that has antioxidant and neuroprotective effects on nerve cells. Therefore, in this study, we attempted to determine the effectiveness of EA extraction. In this study, we performed a DPPH assay to determine the antioxidant potential of EA. And then, the cytotoxic concentration of EA in HaCaT keratinocytes was determined, and the antioxidant effect was determined by measuring the malondialdehyde (MDA). The results of DPPH, a chemical antioxidant assay, clearly demonstrated the antioxidant capacity of EA extracted with distilled water. In addition, cell-based assays provide useful information on the protective effect of EA on oxidative stress-induced apoptosis.

DPPH Free Radical Scavenging Effect of the Aerial Parts of Desmodium oldhami (큰도둑놈의갈고리 지상부의 DPPH Free Radical 소거효과)

  • The antioxidant activity of Desmodium oldhami (Leguminosae) was determined by measuring the radical scavenging effect on DPPH (1,1-diphenyl-2-picrylhydrazyl). The methanolic extract of D. oldhami showed the strong antioxidant activity. Five compounds, kaempferol-3-O-rhamnopyranoside (afzeline) (1), quercetin-3-O-[$\alpha$-L-rhamnopyranosyl($1{\rightarrow}6$)-$\beta$-Dglucopyranoside] (rutin) (2), kaempferol-3-O-glucopyranoside (astragalin) (3), genistein-7-O-$\beta$-D-glucopyranoside (genistin) (4), kaempferol-3-O-rutinoside (5) were isolated from the active ethyl acetate soluble fraction of D. oldhami through repeated silica gel and Sephadex LH-20 column chromatography. Among them, compound 2 showed the most significant antioxidative effect on DPPH free radical scavenging test. Compounds 1-5 are reported for the first time from this plant.

Antioxidant Potential of Enzymatic Extracts from Blueberry (Vaccinium corymbosum L.) (블루베리 (Vaccinium corymbosum L.) 유래 효소 추출물의 항산화성)

  • Enzymatic extracts were prepared from the blueberry (Vaccinium corymbosum L.) collected in Jeju, Korea. Five carbohydrases namely AMG, Celluclast, Termamyl, Ultraflo and Viscozyme, and five proteases namely Alcalase, Flavourzyme, Kojizyme, Neutrase and Protamex were used to prepare the enzymatic extracts. Antioxidant properties of each extracts were studied using stable 1,1-diphenyl 2-picrylhydrazyl (DPPH), reactive oxygen species (ROS), nitric oxide (NO) scavenging, metal chelating assays and lipid peroxidation inhibitory activity in hemoglobin-induced linoleic acid system. The phenolic content of all enzymatic extracts was in the range of 517.85-597.96 mg/100 g dried sample. DPPH and NO${\cdot}$scavenging, and metal chelating assays exhibited prominent activities. Viscozyme showed the highest DPPH activity $(0.046{\pm}0.002\;mg/mL)$ while AMG Showed the highest activity in NO${\cdot}$scavenging $(0.339{\pm}0.011\;mg/mL)$. All the extracts exhibited strong metal chelating activities. Blueberry enzymatic extracts also showed relatively good activity in hydrogen peroxide scavenging. AMG showed the highest lipid peroxidation inhibitory activity $(0.28{\pm}0.01\;mg/mL)$ in hemoglobin-induced linoleic acid system. In this results, the blueberry, which has potential antioxidant components, may be a good candidate as a natural antioxidant source.
