음이온성 및 비이온성 계면활성제 혼합물의 Thin Layer Chromatography에 의한 분리 (Separation of Mixtures for Anionic and Nonionic Surfactants by Thin Layer Chromatography)

  • Thin layer chromatography를 사용하여 음이온성 및 비이온성 계면활성제의 신속한 분리 및 정성분석에 필요한 용매조건들을 조사하였다. 알킬사슬의 종류와 alkylene oxide의 분포에 따른 분리특성을 이용하여 polyoxyalkylene 계 비이온성 계면활성제들을 간단히 정성 분석 할 수 있었으며, densitometer를 사용하여 polyoxyethylenated nonyl phenol의 ethylene oxide(EO)에 따른 분포를 쉽게 비교할 수 있었다. 일련의 음이온성 계면활성제들을 $R_f$ 값과 발색되는 색으로 정성분석하였으며, 음이온성 및 비이온성 계면활성제 혼합물을 각각 분리하였다. 지방산계 계면활성제를 diester, monoester 및 미반응된 polyethylene glycol의 세 부분으로 농축 분리하여 그 혼합비율을 구하였다. 이상의 모든 실험은 전개용매를 80mm정도 전개시킨 상태에서 이루어졌고 분리에 걸린 시간은 13-20분 정도였다.

회수 Boric Acid를 이용한 Dicarboxylic Acid의 환경친화적 에스터화 반응 (Eco-friendly Esterification of Dicarboxylic Acid Using Recovered Boric Acid)

  • 본 연구에서는 norbornene 디에스터 유도체를 합성하는 공정에서 반응 부산물로 생성되는 boric acid를 회수하여 norbornene diacid의 에스터화 반응에 재사용 가능성을 조사하였다. Boric acid를 4가지의 alcohol과 에스터화 시켜 4종류의 trialkyl borate (tributyl borate, tripentyl borate, triisopentyl borate, trihexyl borate)를 합성한 다음 이를 norbornene dicarboxylic acid와 반응시키는 2단계 공정으로 디에스터 유도체를 합성하였다. 이 때 norbornene dicarboxylic acid의 전환율은 89.50~99.31%였다. 디에스터화 반응의 부산물인 boric acid를 NaCl염과 함께 회수하여 trialkyl borate를 합성한 후 정제한 결과 회수율은 92.43~99.35%이었다. 이를 다음 반응에 사용한 결과 디에스터의 수율은 43.70~64.39%였고 순도 97.70~99.24%로 수율이 일부 감소되었으나 손실이 크지 않음을 확인하였다. 주요 반응 부산물인 boric acid를 공정내에서 재사용이 가능하므로 화합물 배출에 의한 환경오염을 사전에 예방할 수 있는 친환경공정으로 적용 가능함을 확인하였다.

분광학적 방법에 의한 계면활성제의 확인 (Qualitative Identification of Surfactants by Spectroscopic Method)

  • Our study is aimed at proposal of systematic verification method of molecular structure using measuring method of selective ionic determination and spectrometry on 34 kinds of surfactants such as sodium dodecyl sulfate(SDS) which are most widely used today. In the IR spectrum, unsaturated fatty acids reveal themselves by HC= at $3000{\sim}3020cm^{-1}$, and intensity of $720cm^{-1}$ depends on carbon length of alkyl group. Also ethylene oxide(EO) adducts exhibit weak characteristic bands by $-CH_{2}-CH_{2}-O$ at 1350, 1100 and $950cm^{-1}$. Isethionate can be distinguished from diester succinate by intensity ratio of 1740 and $1200cm^{-1}$ spectrums, the ratio of latter is close to 1 due to 2 carboxylate radical in diester succinate. Quaternary ammonium salts exhibit characteristic band of $C_{4}N^{+}$ at $1000-900㎝^{-1}$. In the case of dialkyl dimethyl ammonium salts in quaternary ammonium surfactants, the spectrum of $3000cm^{-1}$ by $N-CH_{3}$ collapses to a very weak band at $3020cm^{-1}$. In ammonium heterocyclic derivatives, pyridinium salts show characteristic bands at 1640 and $1460cm^{-1}$, while imidazolinium salts exhibit characteristic band at $1620-1610cm^{-1}$. In the characteristic spectrum at $1080-1050cm^{-1}$ on OH radicals of the alkyl esters, primary alcohol appears as weak band and the 2 bands show in almost same intensity when primary and secondary alcohols exist together in one molecule. Also, alkyl ester of polyhydric alcohols appears as various broad band.

Synthesis of New Boron Derived Compounds; Anticancer, Antioxidant and Antimicrobial Effect in Vitro Glioblastoma Tumor Model

  • Objective : The aim of our study is to investigate the cytotoxic, antioxidant, and antimicrobial effects of newly synthesized boron compounds in U87MG glioblastoma cell treatment. Methods : We synthesized boron glycine monoester (BGM) and boron glycine diester (BGD) structures containing boron atoms and determined their cytotoxic activities on glioblastoma by the MTT method. The inhibitory concentration 50 (IC50) value was calculated with GraphPad Prism 5.0 program. The IC50 values were administered 48 hours on U87MG glioblastoma cell. Catalase (CAT), acid phosphatase (ACP) and alkaline phosphatase (ALP) enzyme activity, malondialdehyde (MDA), total glutathione (GSH), and total protein levels were detected using spectrophotometric methods. We determined the antimicrobial activities of BGM and BGD with the disc diffusion method. Results : After 48 hours of BGM and BGD application to U87MG glioblastoma cells, we found the IC50 value as 6.6 mM and 26 mM, respectively. CAT and ACP enzyme activities were decreased in BGM and BGD groups. MDA which is a metabolite of lipid peroxidation was increased in both boron compounds groups. GSH level was reduced especially in BGD group. BGM and BGD have been found to be antimicrobial effects. Conclusion : Boron compounds, especially the BGM, can provide a new therapeutic approach for the treatment of glioblastoma with their anticancer, antioxidant, and antimicrobial effects.

염증성 질환에 대한 Crocin의 치료 효과 (Therapeutic Effect of Crocin in Inflammatory Diseases)

  • Crocin은 여러 가지 요리에 향미와 색깔을 주는 치자 열매나 사프란에 함유되어 있는 적노란색의 수용성 색소이다. 사프란과 치자는 전통의학 분야에서 부종, 해열, 해독 작용이 있어 바이러스성 간염, 식도염, 관상동맥심장병, 신경쇠약, 불면증, 퇴행성 신경질환, 호흡기 질환, 비뇨기 질환 등을 치료하는데 사용되어 왔다. Crocin (C44H64O24)은 카로테노이드의 복합체로, dicarboxylic acid crocetin과 disaccharide gentiobiose로 이루어진 diester이다. Crocin은 혈액학적인, 병리학적인 독성이나 유전독성이 없다. 현재까지 수많은 생체 내 및 생체 외 연구들을 통해 Crocin의 생물학적인 약리작용이 밝혀지고 있다. 본 총설에서는 염증성 장질환, 위염, 천식, 동맥경화, 류머티스 관절염, 다발성 경화증, 당뇨, 알츠하이머병, 파킨슨병, 우울증 등의 염증 관련 질환에서 Crocin의 보호 효과를 요약한다. Crocin은 다양한 작용 기전을 통해 항염, 항산화, 세포 자살 방지 기능을 함으로써 이들 질환을 개선하는 것으로 추론된다.

Fatty Acid Analysis from Leech Skin

  • The lipids of a Korean native blood-sucking leech (Hirudo nipponia) were isolated and analyzed. Cholesterol, fatty acids, triglycerides, glycerylether diester and cholesteryl esters were found from epidennal extracts. The major fatty acids in the leech skin were $C_{16:0}$, $C_{18:0}$, $C_{20:1}$. $C_{20:3}$ components. These components were derived from cholesteryl esters, glycerides, fatty acids and other polar lipids. After 60 days fasting, cholesterol, fatty acids and triglycerides in the skin were reduced in number and the total fatty acid composition was changed slightly. The data of total fatty acids in fasted and fed leech showed that medium-chain fatty acids were more affected than long-chain fatty acids. Other trace fatty acids appeared to be decreased by fasting.

  • Polyalkylene phosphates - polymers that are built on the repeating unit of the diester of phosphoric acid: -[OP(O)(OH)Oalkylene]-, are known to form backbones of nucleic and teichoic acids. Various synthetic ways will be reported for the synthesis of the bare chains, where "alkylene" in the formula above means mostly two or three methylene groups. Some other structures have also been prepared. Several applications of these polymers are to be discussed, namely as liquid membranes, as components of two-block copolymers (ionic-nonionic diblock dihydrophilic) used as modifiers of CaCO3 crystallization, and as components of the inorganic-polymer hybrid materials. Some other applications in the biomedical field will also be discussed.

Monoterpenoid계의 새로운 항암제 합성 및 In vitro 세포독성 평가 (Synthesis and in vitro Cytotoxicity Monoterpenoid as New Antitumor Agents)

  • Many attention has been focused on developing new chemotherapeutic agents for a treatment of cancer from natural products. From Carpesium divaricatum S. et Z. (Compositae), various monoterpenoid compounds were isolated and exhibited mild antitumor activity against human tumor cell lines. These facts prompted us to explore the structure-activity relationship of these compounds. The synthesis of monoterpenoid compound was accomplished by Fries rearrangement, Grignard reaction, elimination, allylic oxidation, esterification and epoxidation as key steps. The results of in vitro cytotoxicity (A549, SK-OV-3, SK-MEL-2, XF498, HCT15) of the synthesised compounds are as follows: First of all, epoxide moiety is prerequisite for cytotoxic activity in diester compound. Any kind of compounds with olefin or diol moiety instead of epoxide ring exhibited poor or mild cytotoxic activity respectively. Of o-acetoxy and isobutoxy epoxy esters, p-sub-stituted phenylacetate compounds exhibited high cytotoxic activities against SK-MEL-2 and HCT15.

Diester계 치환기를 이용한 분산염료의 알칼리 영향성에 관한 연구 (Characterization of Diester functional-based Alkali-clearable Azo Disperse dyes)

  • 일반적인 분산염료, Phthalimide계 중간체로 사용한 분삼염료, Diester계 커플러만 이용한 분산염료와 비교한 Diester계 치환기를 이용한 분산 염료들은 R/C조건과 EtOAc와 증류수로 층분리하여 증류수층의 UV-Vis absorbance은 떨어지고 용해성은 증가한다. 그리고 Thiopene ring만 가진 염료와 비교한 염료들은 같은 파장에서의 UV-Vis absorbance은 떨어져 용해도는 증가하고 Thiopene ring과 Azo group의 분해에 의한 화합물 때문에 단파장에서의 UV-Vis absorbance가 크게 증가하여 염색 폐수의 색도를 감소시킬 수 있다.

Synthesis of novel disperse dyes derived from phthalimide containning diester groups and their dyeing properties

  • In this study, 12 azo disperse dyes based on phthalimide were synthesized including their precursors. The chemical structures of the dyes and the corresponding precursors were analyzed by the GC-MASS, $^1H$ NMR spectra and elemental analysis. The dyeability on PET, such as step dyeing and build-up properties and color fastness were examined using conventional exhaust dyeing procedures.
