Inhibitory Effect of Methanol Extracts and Solvent Fractions from Meju on Mutagenicity and Growth of Human Cancer Cells (메주 메탄올 추출물 및 분획물의 항돌연변이 및 인체 암세포 성장 억제 효과)

  • Inhibitory effects of methanol extracts and several solvent fractions from meju on mutagenicity in vitro genotoxicity (SOS chromotest) and growth of human cancer cells (AGS gastric adenocarcinoma and Hep 3B hepatocellular cancinoma cells) were studied. The treatment of meju methanol extracts $(100{\mu}g/assay)$ to SOS chromotest system inhibited N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) induced mutagenicity by 36%. However, the ethylacetate and dichloromethane fractions from meju methanol extracts showed the stronger antimutagenic effects (91% and 91%, respectively) in SOS chromotest. In sulforhodamine B (SRB) assay, the treatments of ethylacetate and dichloromethane fractions (2 mg/assay) significantly inhibited the growth of AGS and Hep 3B cancer cells by 64% and 71%, respectively. These results indicated that meju had inhibitor)r effects on MNNG in SOS mutagenic system and growth of human cancer cells, suggesting that its antimutagenic effect may be relative to activity of doenjang.

Antioxidative Effects of Soybean Extracts by using Various Solvents (다양한 용매를 이용한 대두 추출물의 항산화효과)

  • The effective extraction of antioxidative substances from soybean was investigated by using various solvents, such as water, ethanol, methanol, acetone, chloroform, benzene, ethyl acetate, ether, dichloromethane, and hexane. Extraction was performed by cold method at $30^{\circ}C$ and by reflux method at $85^{\circ}C$. The antioxidative effect of the extracts was determined by peroxide value during the oxidation of soybean oil containing the extracts at $105^{\circ}C$ for 10 hours, and also by TBARS(thiobarbituric acid reactive substances) formed during the peroxidation of egg lecithin liposomes. The antioxidant activity of the extracts from raw soybean was higher than that from defatted soybean. The antioxidant activity of the extracts by reflux method was higher than that by cold method. The methanol extract from defatted and roasted soybean(DRS) showed the highest antioxidative effect against oxidation of soybean oil, while the water extract from DRS in egg lecithin liposomes. In the peroxidation of egg lecithin liposomes, the antioxidative effect of polar solvents extracts were higher than those by nonpolar solvents extracts.

Inhibitory Effect of Solvent Fraction of Various Kinds of Kimchi on Ultraviolet B Induced Oxidation and Erythema Formation of Hairless Mice Skin (김치종류별 용매 획분의 자외선 B 조사에 의해 유도된 피부산화 및 홍반 생성 억제 효과)

  • Kimchi is well known as a traditional Korean food containing various antioxidative compounds and it has been demonstrated that dietary kimchi inhibit the tissue oxidation and aging in many related studies. Ultraviolet B (UVB) radiation is the most potent among the many related factor to skin photo-aging. This experiment is designed to elucidate the inhibitory effect of solvent fractionation of various kimchi on UVB induced oxidation and erythema formation. Among solvent fractions, phenol content mostly existed in dichloromethane / ethylacetate fraction of buchu kimchi, dichloromethane fraction of Korean cabbage kimchi and ethylacetate / dichloromethane fraction of mustard leaf kimchi. Free radical scavenging activities were higher in dichloromethane and ethylacetate fraction from buchu kimchi, dichloromethane fraction from Korean cabbage kimchi, and ethylacetate fraction from mustard leaf kimchi. When the damage of photo-oxidation by UVB irradiation was tested in the presence of solvent fractions of kimchi, dichloromethane / ethylacetate fraction of buchu kimchi, ethylacetate fraction from Korean cabbage and from mustard leaf kimchi showed the higher protective activities than others. The erythema formations which were induced by UVB irradiation were decreased in solvent fractions of kimchi that hate higher antioxidant activities. In conclusion, kimchi solvent fractions having antioxidant compounds mostly inhibited photo-oxidation and erythema formation by UV-irradiation, therefore kimchi consumption can retard skin aging due to the presence of antioxidative compounds.

Antimicrobial Effects of Natural Plant and Mushroom, Dicyophora indusiata Extracts on Fish Pathogenic Bacteria (어류질병세균에 대한 천연식물 및 망태버섯 (Dicyophora indusiata) 추출물의 항균활성)

  • Ethanol extract obtained from bark, roots, stem, leaves and seeds of 30 species of plants and mushroom, Dicyophora indusiata were examined antimicrobial effect on fish pathogenic bacteria, Listoneria anguillamm, Lactococcus garvieae, Streptococcus iniae by disk method. Ethanol extract of D. indusiata was found to be the most active against all fish pathogenic bacteria in test system. Ethyl acetate fraction from ethanol extract of D. indusiata showed the strongest compared with those from other solvent fractions such as dichloromethane, n-buthanol and water. The antimicrobial effect of ethyl acetate fraction was particularly evident against S. iniae.

Cytotoxicity and Antimicrobial Effects of Extracts from Salvia miltiorrhiza (단삼 추출물의 세포독성과 항균효과)

  • This study was carried out to evaluate cytotoxic effects of Salvia miltiorrhiza extracts on NIH 3T3 fibroblasts and KB cell lines. Disruptions in cell organelles were determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. The comparison of $IC_{50}$ of values of Salvia miltiorrhiza extracts in KB cell lines showed that their susceptibility to these extracts decreased in the following order: hexane extract > chloroform extract > methanol extract> dichloromethane extract > ethyl acetate extract>ethanol extract by the MTT method. The dried roots of Salvia miltiorrhiza was extracted several solvents, and then antimicrobial activity was investigated. The minimal inhibitory concentrations (MIC's) of the extract against microorganisms were also examined. Amtimicrobial activity of ketoconazol as reference was compared to those of extracts of hexane, chloroform, dichloromethane, ethyl acetate, ethanol and methanol. The antimicrobial activity of all extracts from the sample had growth inhibition activity against gram-negative bacteria, gram-positive bacteria and fungi. These results suggest that the hexane and chloroform soluble extracts of Salvia miltiorrhiza may be a valuable choice for the studies on the tumor cell lines and growth inhibition activity.

Genotoxicological Safety Evaluation of the Solvent Extracts for Medicinal Herbs that are of Highly Domestic Spendings (국내 소비 주요 생약재의 유기용매 추출물에 대한 유전독성평가)

  • This study is carried out to evaluate the genotoxicity of herbs (Angelica decursiva, Polypori umbellate, Astragalus membranaceua, Paeonia iactiflora, Glycyrrhiza uralensis, Cnidium officinale, Rehmannia glutinosa, Cyperus rotundus, Dioscorea batatas and Platycodi Radix) by using the Ames test. The Salmonella typhimurium reversion assay is being performed by using the Sal. typhimurium TA98, TA100, TA102, TA1535 and TA1537 as tester strains. Among 70% of ethanol extracts from 10 herbs, the number of revertant colonies is being increased in Astragalus membranaceua, Cnidium officinale and Dioscorea batatas in a dose-dependent manner, as compared with negative controls of the metabolic activation. In case of dichloromethane and ethyl acetate fraction from 70% of ethanol extracts, the number of revertant colonies is increased in Angelica decursiva, Astragalus membranaceua, Cnidium officinale, Rehmannia glutinosa and Dioscorea batatas in a dose-dependent manner, as compared with negative controls of metabolic activation. Such results indicate that Angelica decursiva, Astragalus membranaceua, Cnidium officinale, Rehmannia glutinosa and Dioscorea batatas all show genotoxic effects when being extracted with the solvent extractions such as 70% of ethanol, dichloromethane and ethyl acetate, and thus, they might be genotoxically- non-safe.

Antioxidative and Antimicrobial Activities of Castanopsis cuspidata var. sieboldii Extracts (구실잣밤나무 추출물의 항산화 및 항균 활성)

  • This study was designed to investigate the possible utilization of Castanopsis cuspidata as a source of antiseptic agents. The leaves of C. cuspidata, extracted by 80% ethanol, were sequentially fractionated with n-hexane, dichloromethane, ethylacetate, and n-butanol. In order to effectively screen for a natural preservative agent, we first investigated the antioxidant activities such as DPPH radical scavenging capacity, superoxide radical scavenging capacity, and xanthine oxidase inhibitory activity of the C. cuspidata ethanol extracts and fraction. Using a screening system, we found that the ethylacetate fraction had the strongest antioxidant activity, which followed a dose-dependent manner. The antimicrobial activities were shown in the ethylacetate fraction of C. cuspidata. Among the five fractions, the ethylacetate fraction showed the highest antimicrobial activities against microorganisms tested, which were Bacillus sublitis, Listeria monocytogenes, Staphylococcus aureus, Escherichia coli, Salmonella enteritidis and Salmonella typhimurium. In addition, leaf extracts of C. cuspidata could be suitable for the development of food preservatives.

STUDIES ON ANTICANCER EFFECTS OF EXTRACTS CAESALPINIA SAPPAN ON ORAL CARCINOMA AND OSTEOSARCOMA CELLS (소목 추출물의 구강암 및 골육종 세포주에 대한 항암작용에 관한 연구)

  • Anticancer effect of methanol extract of Caesalpinia sappan L. on oral carcinoma (KB) and osteosarcoma (HOS) cells were investigated in this study. In order to elucidate the anticancer mechanism of Caesalpinia sappan L, we analyzed telomerase inhibitory effect of the methanol extract of Caesalpinia sappan L. In addition we prepared 5 fraction samples according to its polarity differences and analyzed anticancer effects on oral carcinoma and osteosarcoma cells. Following results are obtained in this study. 1. 50% cell proliferation inhibitory value ($IC_{50}$) of the methanol extract of Caesalpinia sappan L. against oral carcinoma (KB) cells and osteosarcoma (HOS) cells were $9.0{\mu}g/ml$ and $10.9{\mu}g/ml$, respectively. 2. The methanol extract of Caesalpinia sappan L. showed inhibitory effect of telomerase which is required for cancer cell immortality. Therefore, it seems that the anticancer effect of methanol extract of Caesalpinia sappan is at least partially due to telomerase inhibitory effect. 3. Five fraction samples were prepared according to its polarity and 88.7% of ingredient of total methanol extract was transferred to ethylacetate fraction. Thin layer chromatography analysis showed that dichloromethane fraction contained ingredient with relatively high polarity and ethylacetate fraction contained similar ingredient found in total methanol extract. 4. Anticancer effect was observed in n-hexane, dichloromethane, and ethylacetate fractions. The highest anticancer effect was found in dichloromethane fraction which had $IC_{50}$ value of 4.4 and $>4.0{\mu}g/ml$ against oral carcinoma (KB) cells and osteosarcoma (HOS) cells, respectively.

Feasibility of Fibrous Solid Phase Extraction to Alkylphenols Analysis (섬유상 고상 추출물질을 이용한 알킬페놀 화합물 분석 가능성)

  • p-phenylene-2,5-benzobisoxazole (PBO; Zylon$(R)$) fibers as an adsorbent were employed for solid phase extraction of aqueous alkylphenols. The removal ratios for 10 kinds of alkylphenols at initial concentration of $100{\mu}gL^{-1}$ were in the range of 16.8-96.3% and the removals increased with the increase of the phase ratio (fiber weight/solution volume). The plots of the logarithm of partition coefficient (log K) were correlated with the logarithm of the n-octanol/water partition coefficient (log P). The adsorbed alkylphenols were completely desorbed with the mixture of acetonitrile and dichloromethane.
