Effect of dietary Platycodon grandiflorum on plasma glucose and lipid metabolism in $KK-A^y$ mice and streptozotocin-induced diabetic rats

  • This study was designed to investigate the effect of dietary Platycodon grandiflorum on plasma glucose and lipid metabolism in $KK-A^y$ mice and STZ-induced diabetic rats. Both plasma triglyceride and plasma cholesterol levels in STZ-induced diabetic rats were significantly decreased by dietary Platycodon grandiflorum feeding for 4 weeks compared to those of control rats, but there were no marked differences in $KK-A^y$ mice. However, for plasma glucose values, Platycodon grandiflorum feeding resulted in a significant decrease in both STZ-induced diabetic rats and $KK-A^y$ mice. Also, dietary Platycodon grandiflorum slightly decreased the postprandial glucose level at 30 and 60 mins during oral glucose tolerance test in $KK-A^y$ mice. Although there was no statistical significance, the fasting plasma insulin levels of Platycodon grandiflorum dieted $KK-A^y$ mice tended to decrease when compared to that of control mice. Therefore, the present results suggested that dietary Platycodon grandiflorum may have a beneficial effect on preventing hypercholesterolemia and hyperlipidemia.

Effectso fvitamin E Supplementation on the Lipid Perosides and Activities of Antioxidative Enzymes in the Pancreas of diabetic KK Mice (비타민 E 보강식이가 당뇨 KK 마우스의 췌장에서 지잘과 산화물의 항산화 효소 활성에 미치는 영향)

  • The purpose of this study was to investigate the effect of vitamin E supplementation on the lipid peroxidation and activities of antioxidative enzymes in the pancreas of diabetic KK mice. KK mice were fed high ft diet containing 20% corn oil(wt/wt), and sacrificed at 2 months of diabetes. A hish vitamin E diet consisted of the high fat diet supplemented with an excessive amount of 이-$\alpha$-tocopheryl acetate (2080IU/kg diet). The incidence of diabetes mellitus was 61% when mice were fed the high fat diet, but was 44% when mice were fed the high vitamin E diet, Vitamin E supplementation fhus seems to have the effect of decreasing of decreasing the onset of diaetes. In the diabetic group, we found increases of MDA (malondialdehyde) and antioxidative enzyme activities. Treatment with vitamin E did not modify the level of fasting blood glucose. However, MDA and antiosicative enzyme activities in diabetic mice were decreased by the high vitamin E diet. Increased levels of lipid peroxidation products suggests the occurrence of oxidative damage in the pancreas of diabetic mice. The increased level of antiosicative enzyme activities could be due to an adaptive response to conditions of increased peroxidative stress. Significant normalization on catalase activity was noted in vitamin E supplemented animals.

  • This study was carried out to investigate the supplementary effects of ${\gamma}$-oryzanol extracted from rice bran on lipid metabolism in diabetic mice. We supplied 2 kinds of experimental diets (CO without and GO with 0.2% ${\gamma}$-oryzanol) to diabetic mice for 8 weeks. Diet intake, body weight, organ weight, contents of serum and hepatic lipid profiles, and fecal lipid levels were measured. Though there was no significant difference in diet intake, body weight change and organ weight between experimental groups, the concentration of serum total cholesterol and hepatic total lipid, total cholesterol and HMG-CoA reductase activity was significantly lower in GO group treated with 0.2% ${\gamma}$-oryzanol of diet than CO group after supplementary period of experimental diets. And total lipid, triglyceride, total cholesterol, and bile acid levels excreted to feces were significantly higher in GO group than CO group. These results suggest that ${\gamma}$-oryzanol decrease the serum and hepatic lipid levels by lowing HMG-CoA reductase activity or increasing the contents of fecal lipid in diabetic KK mice.

Effects of Vitamin E Supplementation on Glycosylation Products in Diabetic KK Mice (비타민 E 보강식이가 당뇨 KK마우스에서 당화단백질 생성에 미치는 영향)

  • We investigated the effects of vitamin E supplementation on the protein glycosylation in vivo. Weaned KK-mice were fed high fat diet containing 20% corn oil(wt/wt), and sacrificed at 4, 6, and 0 months of age. High vitamin E diet was the high fat diet supplemented with an excess amount of 이-$\alpha$-tocopheryl acetate(2080IU/kg diet). We measured $HbA_{1C}$ as a glycosylation early product, and collagen-linked fluorescence (CLF) of skin as a glycosylation and product. We found that diabetic group had increased levels of $HbA_{1C}$ within 2 months after onset of diabetes and during the experiments. The skin CLF increased dramatically 5 months after onset of diabetics. Treatment with vitamin E did not modify the level of blood glucose. However, we observed a significant lowering in CLF and $HbA_{1C}$ in diabetic mice.

Weaned KK mice were fed high fat a diet containing 20% corn oil(wt/wt), and were sacrificed at 4, 6, and 9 months of age. The high vitamin E diet consisted of the high fat diet supplemented with an excessive amount of dl-$\alpha$-tocopheryl acetate (2080 IU/kg diet) . In the diabetic groups, we found an increase in lipofuscin and decrease in antioxidative enzyme activities with aging. Treatment with vitamin E did not modify the level of fasting blood glucose. However, a significant decrease in lipofuscin and increase in antioxidative enzyme acitvities were observed in diabetic mice.

  • We investigated the effects of vitamin E supplementation on protein glycosylation in early and end stage product, and light microscopic studies were done on the renal glomeruli of KK-mice of various ages and various duration of diabetes. Weaned KK-mice were fed high fat diets containing 20% corn oil(wt/wt), and sacrificed at 4,6, and 9 months of age. The high vitamin E diet was a high fit diet supplemented with an excess amount of d1-$\alpha$-tocopheryl acetate (2080IU/kg diet). We measured Hemoglobin $A_{IC}$ (Hb $A_{IC}$) as a glycosylation early product, and renal collagen-linked fluorescence as a glycosylation end product. In the diabetic group, levels of Hb $A_{IC}$ were increased within 2 months after onset of diabetes and remained at a constant level for the duration of experiment. 5 months after onset of diabetes, renal collagen linked fluorescence(CLF) was markedly increased. A quantative, morphologically demonstratable, progressive thickening of the basement membrane and calcification occured in the diabetic KK-mice. There is a statiscally positive correlation between CLF and histologic grade of diabetic nephropathy. Hepatic vitamin E levels correlated with those of Hb $A_{IC}$, renal CLF, and renal calcification. Treatment with vitamin I did not modify the level of blood glucose. However, we observered a significant lowering of CLF and Hb $A_{IC}$ in diabetic mice. Supplementation of vitamin E was found to delay the progression of diabetic nephropathy.   • Effects of water extracts of antler on blood sugar levels were studied in normal rats and KK-mice. a spontaneously diabetic strain of murine. Blood sugar levels were determined at 30, 60, and 120 minutes after intraperitoneal administration of 1 ml/kg or 2ml/kg AE into normal rats. A moderate but significant decrease in blood sugar level was noticed only in 30 minutes following 2 ml/kg AE administration. On the other hand no significant changes in blood sugar level was observed in KK-mice to which 1ml/kg/day AE had been given for 10 weeks. The results of present study strongly indicate that antler extract exerts no significant effect on blood sugar levels in experimental animals.

  • This study was carried out to investigate the supplementary effects of the rice germ oil compared with soy bean oil on blood glucose level of non-insulin dependent diabetic mice. Forty diabetic KK mice were fed two kinds of experimental diets with 20% lipid from soy bean oil as a control(CO) and rice germ oil(RG) for 8 weeks, respectively. Diet intake, body weight, organs weights and lipids levels of serum, liver and feces were measured. There was no significant difference in food and water intake, body weight gain and organs weights between experimental groups. The concentrations of fasting and random blood glucose were similar between CO and RG groups. There was no significant difference in blood glucose levels after glucose treatment during the glucose tolerance test between two groups. The levels of $HbA_{1c}$ as the index of blood glucose status, and insulin were similar in two groups. These results suggested that rice germ oil can't reduce blood glucose concentration of non-insulin dependent diabetic mice compared with soybean oil. But we need to investigate the hypoglycemic effect of rice germ oil by changing supplementary level and period.

These results suggested that rice germ oil can reduce serum total cholesterol and LDL-cholesterol and hepatic total lipid concentration of non-insulin dependent diabetic mice compared with soybean oil due to increasing fecal lipid excretion. But we need to investigate the hypolipidemic effect of this oil by supplementary level and period.

  • This study was carried out to investigate the effect of Lentinus edodes which were harvested at different times of maturity on blood glucose and lipid levels in diabetic mice. The diabetic KK mice were fed diets supplemented with Lentinus edodes harvested early (LE) or late (LL) for eight weeks, and control (CO) mice were fed regular diet without Lentinus edodes. Diet intake, body weight, organ weight, blood glucose and serum lipid levels were measured. The mice which were fed diets supplemented with LE and LL showed significantly lower body weights compared to the CO group. There were no significant differences in the diet intake and the weights of different organs including liver, heart, kidneys, and epididymal fat pad among three different groups. Furthermore, the concentrations of serum triglycerides and total cholesterol were significantly lower in LE and LL groups compared to the CO group. These results clearly demonstrate that Lentinus edodes, regardless of their harvested period reduced body weight and serum lipid without affecting blood glucose level in diabetic mice.
