Implementation of a self-mixing type LDF probe and blood flow simulator (자기혼합형 LDF 프로브와 혈류 시뮬레이터의 구현)

  • In this pager, the authors have implemented a blood flow simulator and a LDF(laser Doppler flowmeter) probe using self-mixing effect of the laser diode. The purpose of the blood flow simulator is to simulate microvascular blood flow in tissue. It consists of melinex film (thickness = $123\;{\mu}m$) which has similar optical characteristics to epidermis and porous polyethylene filter (Vyon, porosity 35%, mean pore size $50\;{\mu}m$, thickness=1 mm) which has similar optical characteristics to dermis. The blood flowmeter probe consists of laser diode(5 mW, 780 nm wavelength), CD lens(focal length 12 mm). current-to-voltage converter, highpass filter, and premplifier. It doesn't need optical fiber, therefore, implementation of the probe is simpler than conventional probe using optical fiber.

  • The effects of scald bum Injury ($40-45\%$ of total body surface area), there were not inhalation and secondary infection, on the histological changes and the levels of oxygen free radical generating and scavenging enzymes have been determined in liver tissue of rat models. It was found that dermal epithelium was left out with edema of dermis layer and hydorpic swelling of hepatocytes, Burn injury increased liver weight (L.W./B.W.) and serum aspartate aminotransferase content (pThe data of this study suggest that liver damage Induced by scald burn injury leads to dysbalance of oxygen free radical generating and scavenging enzymes.

  • Antisense phosphorothioate oligodeoxynucleotide(PS-ODN) against $TGF-{\beta}$ was developed as scar formation inhibitor. The scar was caused collagen deposition due to overexpression of $TGF-{\beta}$ in wounded skin. The percutaneous absorption of partially modified PS-ODN(25 mer) was investigated for the purpose of its effective delivery. Though PS-ODN has high molecular weight (MW=8,000) and polyanionic charge, it was permeated through skin. The skin permeation of PS-ODN was markedly increased by the removal of stratum corneum and dermis. Moreover, the skin permeation of PS-ODN was decreased in the following order; hairless mouse skin>rat skin>human cadaver skin. Thus, PS-ODN represents a logical candidate for further evalution due to the potential for delivery into the wounded skin.

  • Skin biopsy with investigation of small nerve fiber in human epidermis and dermis has been proven to be a useful method for demonstration of small fiber neuropathy. Quantification of intraepidermal nerve fiber density using anti-Protein Gene Product 9.5 (PGP 9.5) antibody is standardized method to diagnose the small fiber neuropathy. Skin biopsy method also makes it possible to differentiate the type of nerve fibers by using different antibodies. Quantification of dermal structures with different type of nerve fibers could be used to invest pathophysiologic mechanism of diseased state.

  • Dendritic cells(DC) are a system of highly efficient antigen-presenting cells that initiate the primary immune response. There are two kinds of dendritic cells in the skin, Langerhans cell in the epidermis and dermal dendritic cell in the dermis. The knowledge of DC, which are very important in the immune reponse of cancer, autoimmune disease, transplantation and infection, has been known through the study about Langerhans cells. In this paper, the role of Langerhans cell in the contact hypersensitivity and atopic dermatitis is discussed and culture methods of mouse Langerhans cells and human U from pheripheral blood monocytes are described.

Transdermal Permeation of Riboflavin in Ointment Bases Using Gums & Enhancers (Gum류의 연고제제와 흡수촉진제가 Riboflavin의 경피흡수에 미치는 영향)

  • We investigated characters of transdermal therapeutic system(TTS) and the skin permeability of that with applying drug delivery system(DDS). Natural gums were selected as material of TTS. The permeation of natural gums ointment containing drug in rat skin using diffusion cell model. Permeation properties of materials were investigated for water soluble drug such as riboflavin in vitro. We used glycerin, PEG 600 and oleic acid as enhancers. Since dermis has more hydration than the stratum corneum, skin permeation rate at steady state was highly influenced when glycerin was used in riboflavin. The permeation rate of content enhancer and drug was found to be faster than that of content riboflavin only. These results showed that skin permeation rate of drug across the composite was mainly dependent on the property of ointment base and drug. All the gum ointment tested showed good safety. Proper selection of the materials which resemble and enhance properties of the delivering drug was found to be important in controlling the skin permeation rate.

  • Glomus tumors are small vascular tumors that are usually benign and rarely occur. They originate from glomus bodies and present in the reticular dermis. They are clinically distinguished by their small size and their ability to cause extreme pain. Most of these tumors are subungually located. However, atypical locations of the tumors sometimes cause misdiagnosis, particularly when the lesion is rarely reported. Therefore, we report a case of glomus tumor which presented with chronic abdominal pain, found in the abdominal wall that has never been reported before.

  • Plantar corn is a circumscribed and sharply demarcated hard mass of traumatic hyperkeratosis, which has a central core impacting the dermis deeply and causes pain while walking. The cause of the corn is the mechanical stress to the skin induced by several causative factors; extrinsic(tight shoes) or intrinsic(bony prominence), or combined(the claw toe). We found 7 cases of patients with a painful recurrent plantar corn which had an epidermal cyst under its lesion. These corns and cysts were excised totally and there was no recurrence in all cases during two to four months follow-up period. We think that an epidermal cyst may be another intrinsic factor for the development of a plantar corn. So, an epidermal cyst should be considered once in a recurrent plantar corn to be recalcitrant against popular treatments.

  • Structure of the skin in amphibious fish, Periophthalmus modestus, was described in relation to cutaneous respiration. The epidermis has no gland cell. The epidermis consists of three regions: outermost layer of one to five layers of flattened epithelial cells, middle layer of swollen epithelial cells instead of glandular cells and stratum germinativum of cuboidal cells. There are numerous blood capillaries in the outermost layer of the epidermis and diffusion distance between the blood of capillaries and the epidermis is about 1.4 Um. The middle layer of the epidermis appears to be a web-like structure due to the swollen epithelial cells. The stratum germinativum has a well-developed lymphatic space containing lymphocytes. There are numerous blood capillaries and elliptical area with acid mucopolysaccharides in stratum laxum of the dermis. The skin of Periophthalmus modestus may be an accessory respiratory organ for oxygen uptake during terrestrial or aquatic life.

  • Aplasia cutis congenita (ACC) is a rare congenital disorder characterized by skin defects involving the epidermis, dermis, subcutaneous tissue, bone, and sometimes dura. It commonly affects the scalp in approximately 70% of cases, but the neck, trunk, and the extremities can also be affected. ACC can occur either as an isolated condition or associated with other anomalies and congenital syndromes, and it can be acquired either genetically or sporadically. Morbidity and mortality are associated with the defects of skull bone, dura, and other multiple anomalies. We herein report the case of a female infant, with a large scalp defect accompanied by a skull defect noted at birth, who developed mental retardation in the preschool years.
