Neuroprotective Effect of Scopoletin from Angelica dahurica on Oxygen and Glucose Deprivation-exposed Rat Organotypic Hippocampal Slice Culture

  • This study examined the neuroprotective effect of scopoletin from Angelica dahurica against oxygen and glucose deprivation-induced neurotoxicity in a rat organotypic hippocampal slice culture. Scopoletin reduced the propidium iodide (PI) uptake, which is an indication of impaired cell membrane integrity. In addition, it inhibited the loss of NeuN, which represents the viability of neuronal cells. The results suggests that scopoletin from A. dahurica protects neuronal cells from the damage caused by oxygen and glucose deprivation.

Behavioral and physiological changes during heat stress in Corriedale ewes exposed to water deprivation

  • This study was conducted to investigate the behavioral and physiological changes of heat stressed Corriedale ewes exposed to water deprivation. Nine Corriedale ewes (average $BW=45{\pm}3.7kg$) were individually fed diets based on maintenance requirements in metabolic crates. Ewes were assigned into three groups (9 sheep per treatment) according to a $3{\times}3$ Latin square design for 3 periods with 21-d duration for each period. The control (CON) group was given free access to water, 2 h water deprivation (2hWD), and 3 h water deprivation (3hWD) following feeding. No differences were found in fecal excretion frequency, standing frequency (number/d), and sitting frequency among the groups (p > 0.05). Measurements of standing duration (min/d) and urine excretion frequency (number/d) showed a significant decrease whereas sitting duration (min/d) showed a significant increase in the 2hWD and 3hWD groups when compared with the CON group (p < 0.05). Fecal score and heart rate (number/min) were not different among the groups (p > 0.05). However, respiratory rate (number/min) and panting score were found to be significantly higher in the 2hWD and 3hWD groups than in the CON group (p < 0.05). It is concluded that water deprivation following feeding intensifies physiological heat stress related indicators such as respiratory rate and panting score and changes behavioral parameters such as water intake and urine excretion frequency in heat stressed ewes. Daily adaptation to the extreme environmental conditions may occur actively in ewes.

Effect of Post Hatch Feed Deprivation on Yolk-sac Utilization and Performance of Young Broiler Chickens

  • An experiment was conducted to study the effect of post-hatch feed deprivation on yolk sac utilization andsubsequent performance of young broiler chickens (280) up to 35 days of age. The experimental treatments included access to feed at 8 h intervals after hatch, up to 48 h (0, 8, 16, 24, 32, 40 or 48 h). Water was offered ad libitum to all the groups immediately after placement. Results indicated that chicks with access to feed immediately after hatch used up the residual yolk more quickly. Access to feed between 8-24 h post-hatch, supported faster utilization of residual yolk compared to those chicks that remained unfed for 40-48 h (p<0.05). Further, deprivation of feed up to 24 h did not alter the lipid and protein contents in residual yolk, but fasting of chicks beyond 24 h (32, 40 and 48 h) led to retention of higher lipid and lower protein content in the yolk sac (p<0.05). At 7 days of age, the weights of proventiculus and gizzard were not affected by feed deprivation up to 48 h. However, the liver, pancreas and jejunum recorded significantly (p<0.05) heavier weights in chicks that were fed during the initial 24 h period compared to delayed feeding (32-48 h). Chicks fed within 24 h after hatch gained significantly (p<0.05) higher weight at 5 weeks of age than those that received feed between 32 and 48 h. Feed deprivation for 48 h was more detrimental to growth than 24-40 h. This study revealed the significance of early posthatch feeding (<24 h) on faster utilization of yolk sac nutrients and optimum development of intestines and organs, culminating in improved weight gain (>10.5%) of broilers at 5 weeks of age.

Nitric Oxide Prevents the Bovine Cerebral Endothelial Cell Death Induced by Serum-Deprivation

  • Endothelial cells play a central role in the inflammatory processes, and activation of nuclear factor kappa B ($NF-_{\kappa}B$) is a key component in that inflammatory processes. Previously, we reported that tumor necrosis factor alpha($TNF{\alpha}$) had protective effect of cell death induced by serum deprivation and this protection was related to $NF-_{\kappa}B$ activation. Inducible nitric oxide synthase (iNOS) is a member of the molecules which transcription is regulated mainly by $NF-_{\kappa}B$. And the role of nitric oxide (NO) generated by iNOS on cell viability is still controversial. To elucidate the mechanism of $TNF{\alpha}$ and $NF-_{\kappa}B$ activation on cell death protection, we investigate the effect of NO on the cell death induced by serum- deprivation in bovine cerebral endothelial cells in this study. Addition of $TNF{\alpha}$, which are inducer of iNOS, prevented serum-deprivation induced cell death. Increased expression of iNOS was confirmed indirectly by nitrite measurement. When selective iNOS inhibitors were treated, the protective effect of $TNF{\alpha}$ on cell death was partially blocked, suggesting that iNOS expression was involved in controlling cell death. Exogenously added NO substrate (L-arginine) and NO donors (sodium nitroprusside and S-nitroso-N-acetylpenicillamine) also inhibited the cell death induced by serum deprivation. These results suggest that NO has protective effect on bovine cerebral endothelial cell death induced by serum-deprivation and that iNOS is one of the possible target molecules by which $NF-_{\kappa}B$ exerts its cytoprotective effect.

Extracellular Signal-Regulated Kinase (ERK1/2) Regulate Glucose Deprivation-Induced Cell Death in Immunostimulated Astrocytes

  • In our previous study, glucose deprivation was reported to induce the potentiated death and ATP loss in immunostimulated astroglia. And this vulnerability to glucose deprivation was due to overproduction of nitric oxide (NO) and hydrogen peroxide (H$_2$O$_2$). In the present study, the role of extracellular signal-regulated kinase 1/2 (ERK1/2) in the glucose deprivation-induced death of immunostimulated astroglia was examined. We showed that immunostimulation with LPS+IFN-ν activated the ERKl/2 signal pathway and produced a large amount of NO and H$_2$O$_2$. Generation of NO and H$_2$O$_2$ in immunostimulated astroglia was mediated via ERK1/2 signal pathways, since addition of the ERK kinase (MEKl) inhibitor PD98059 reduced NO and H$_2$O$_2$production. ERK1/2 activation-mediated NO and H$_2$O$_2$ production is due to an activation of iNOS and NADPH oxidase, respectively. Finally, we found that glucose deprivation caused ATP depletion and the augmented death in immunostimulated astroglia, which was also prevented by PD98059 treatment. These results demonstrate that the ERK1/2 signal pathways play an important role in glucose deprivation induced the death in immunostimulated astroglia by regulating the generation of NO and H$_2$O$_2$.

Income Differences in Smoking Prevalences in 245 Districts of South Korea: Patterns by Area Deprivation and Urbanity, 2008-2014

  • Objectives: The aim of this study was to measure income differences in smoking prevalence at the district level and to investigate correlations among area deprivation, smoking prevalence, and income differences in smoking prevalence, stratified by urbanity. Methods: Data were pooled from the Community Health Survey data of South Korea between 2008 and 2014. The age-standardized prevalence of smoking and its interquintile income differences were calculated. We conducted correlation analyses to investigate the association of the deprivation index with smoking prevalence and interquintile differences in smoking prevalence. Results: Across 245 districts, the median prevalence of smoking in men was 45.9% (95% confidence interval [CI], 43.4 to 48.5%), with an interquartile range (IQR) of 4.6% points. In women, the median prevalence was 3.0% (95% CI, 2.4 to 3.6%) and IQR was 1.6% points. The median interquintile difference in smoking prevalence was 7.4% points (95% CI, 1.6 to 13.2% points) in men and 2.7% points (95% CI, 0.5 to 4.9% points) in women. The correlation coefficients for the association between the deprivation index and smoking prevalence was 0.58, 0.15, -0.22 in metropolitan, urban, and rural areas, respectively, among men, and 0.54, -0.33, -0.43 among women. No meaningful correlation was found between area deprivation and interquintile difference in smoking prevalence. The correlation between smoking prevalence and interquintile difference in smoking prevalence was more evident in women than in men. Conclusions: This study provides evidence of geographical variations in smoking prevalence and interquintile difference in smoking prevalence. Neither smoking prevalence nor the deprivation index was closely correlated with interquintile income difference in smoking prevalence. Measuring inequalities in smoking prevalence is crucial to developing policies aimed at reducing inequalities in smoking.

Glucose Deprivation and Immunostimulation Induced Death in Rat Primary Astrocytes is Mediated by Their Synergistic Effect on the Decrease in Cellular ATP Level

  • In this study we investigated whether ATP loss was involved in the potentiated death of immunostimulated rat primary astrocytes in glucose-deprived condition. Rat primary astrocytes immunostimulated with LPS plus IFN-${\gamma}$ for 48 h underwent death upon glucose deprivation, which dependent on the production of peroxynitrite. Intracellular ATP level synergistically decreased by glucose deprivation in immunostimulated astrocytes but not in control cells, and the loss of ATP occurred well ahead of the LDH release. The synergistic cell death and ATP loss by immunostimulation and glucose deprivation were inhibited by iNOS inhibitor (L-NAME and L-NNA) or peroxynitrite decomposition catalyst (also a superoxide anion scavenger), Mn(III)tetrakis(N-methyl-4'-pyridyl)porphyrin (MnTMPyP). Exogenous addition of peroxynitrite generator, SIN-l timedependently induced ATP loss and cell death in the glucose-deprived astrocytes. Depletion of intracellular glutathione (GSH) and dis겨ption of mitochondrial transmembrane potential (MTP) were also observed under same conditions. Supply cellular ATP by the addition of exogenous adenosine or ATP during glucose deprivation inhibited ATP depletion, GSH depletion, MTP disruption and cell death in SIN-l treated or immunostimulated astrocytes. This study showed that perturbation in the regulation of intracellular ATP level in immunostimulated astrocytes might make them more vulnerable to energy challenging stimuli.

과중채무자의 사회경제적 박탈에 관한 연구 (Indebtedness and Socioeconomic Deprivation : A Study of Debt Relief Program Users)

  • 본 연구의 목적은 빠르게 증가하는 가계부채가 중요한 사회적, 경제적 이슈가 된 현 시점에 과중채무자들의 생활상을 물질적 결핍과 사회적 결핍을 포괄하는 '사회경제적 박탈' 개념을 활용하여 조명하는 것이다. 본 연구는 채무조정제도를 이용하는 과중채무자들을 면접조사한 데이터(N=209)를 분석하였고, 한국복지패널의 일반집단 및 저소득집단과의 비교를 실시하였다. 연구결과는 과중채무집단이 일반인구 및 저소득집단에 비하여 생계, 고용, 주거, 건강, 사회적 관계 차원에서 모두 결핍수준이 월등히 높으며, 과중채무집단 내부에서는 개인파산집단의 결핍수준이 개인회생, 개인워크아웃, 기타집단보다 높은 것으로 나타났다. 결과를 토대로 과중채무자와 채무조정제도를 위한 제도적, 실천적 함의를 논의하였다.

일부 치위생과 학생들의 스마트폰 과의존과 수면부족 및 자기통제력 융합연구 (The Convergence Study of Smartphone Overdependence, Sleep Deprivation and Self-control in Some Dental Hygiene Students)

  • 본 연구는 치위생과 학생들의 스마트폰 과의존과 수면부족을 파악하여 자기통제력에 미치는 영향을 알아보고자 실시하였다. 충청지역에 소재하고 있는 치위생과 대학생 267명을 대상으로 조사하여 분석하였다. 스마트폰 사용 시간이 길수록 스마트폰 과의존은 높게, 자기통제력은 낮게 나타났고, 대인관계와 대학생활에 만족할수록 스마트폰 과의존은 낮게 나타났다. 스마트폰 과의존과 수면부족은 양의 상관관계를, 스마트폰 과의존과 자기통제력은 음의 상관관계를 보였다. 수면부족과 자기통제력은 음의 상관관계를 보였다. 스마트폰 과의존의 하위영역인 문제적 결과와 수면부족이 자기 통제력에 영향을 미치는 것으로 나타났다. 그러므로 치위생과 학생들의 자기통제력을 높일 수 있는 교육과 프로그램을 개발하고 수면문제와 스마트폰 과의존을 예방할 수 있는 방안을 마련해야 할 것이다.

청년의 물질적 박탈감이 주관적 정신건강에 미치는 영향: 사회적 지지의 조절효과 (The Effects of Material Deprivation on Subjective Mental Health of Youth: Moderating Effects of Social Support)

  • 본 연구는 청년의 물질적 박탈감이 주관적 정신건강에 미치는 영향과 이들 관계에서 사회적 지지의 조절효과를 살펴 볼 목적으로 수행되었다. 이를 위해 '2021 청년 사회·경제 실태조사'에 참여한 청년 2,041명의 응답데이터를 활용하여 빈도분석, 기술통계, 상관분석, 조절효과 모형에 대한 분석을 실시하였다. 분석결과, 첫째, 청년의 물질적 박탈감이 주관적 정신건강에 부적으로 유의한 영향을 미치는 것으로 나타났다. 둘째, 청년의 물질적 박탈감이 주관적 정신건강에 미치는 영향을 사회적 지지가 조절하는 것으로 나타났다. 본 연구결과를 통해 청년의 주관적 정신건강 수준 제고를 위해서는 청년의 물질적 박탈상황을 미연에 방지하기 위한 공공정책 및 제도를 보다 강화할 뿐 아니라 사회적 지지수준을 높여 사회적 지지체계를 강화하는 방안이 필요함을 알 수 있었다.
