Autophagy down-regulates NLRP3-dependent inflammatory response of intestinal epithelial cells under nutrient deprivation

  • Dysregulation of inflammation induced by noninfectious stress conditions, such as nutrient deprivation, causes tissue damage and intestinal permeability, resulting in the development of inflammatory bowel diseases. We studied the effect of autophagy on cytokine secretion related to intestinal permeability under nutrient deprivation. Autophagy removes NLRP3 inflammasomes via ubiquitin-mediated degradation under starvation. When autophagy was inhibited, starvation-induced NLRP3 inflammasomes and their product, IL-1β, were significantly enhanced. A prolonged nutrient deprivation resulted in an increased epithelial mesenchymal transition (EMT), leading to intestinal permeability. Under nutrient deprivation, IL-17E/25, which is secreted by IL-1β, demolished the intestinal epithelial barrier. Our results suggest that an upregulation of autophagy maintains the intestinal barrier by suppressing the activation of NLRP3 inflammasomes and the release of their products, including pro-inflammatory cytokines IL-1β and IL-17E/25, under nutrient deprivation.

Neuroprotective Effect of Lilii bulbus, Nelumbins semen on the Sleep Deprivation-induced Oxidative Stress in the Hippocampus Dentate Gyrus (수면박탈로 유도한 Hippocampus Dentate gyrus의 산화 스트레스에 대한 백합, 연자육의 신경세포보호효과)

  • Sleep deprivation is an extremely common event in today's society. It has caused learning cognitive skill deterioration and poor concentration, increased disease such as heart disease, diabetes and obesity, sexual function decrease, infertility increase, depression and autonomic nervous system disorder. Sleep deprivation-induced stress caused NADPH oxidase and oxidative stress. And this oxidative stress induces apoptosis. Lilii bulbus and Nelumbins semen are known to mental and physical relaxation effects. In this study, we induced sleep deprivation(SD) in Sprague-Dawley rats in water for 5 days and thereafter administered orally L. bulbus and N. semen for 5 days. Brain tissues were observed by histochemical, immunohistochemical and tunel staining. The immunoreactives of Tumor necrosis factor ${\alpha}$, Neuronal nitric oxide synthases, Phospho-SAPK/JNK and gp91-phox of the L. bulbus administered group and N. semen administered group were weaker than those of sleep deprivation group. In the L. bulbus administered group and N. semen administered group, apoptosis was decreased than that of sleep deprivation group. Proapoptotic p53, Bax, Cleaved caspase 3 immunoreactives of the administered group were weaker than those of sleep deprivation group, whereas anti-apoptotic Bcl-2 immunoreactity was stronger in the L. bulbus administered group and N. semen administered group. Antioxidant mechanism such as DJ-1, superoxide dismutase 1, Nuclear factor-like 2 immunoreactives of the L. bulbus and N. semen administered group were stronger than those of sleep deprivation group. These results demonstrate that L. bulbus, N. semen had the neuroprotective effects on the sleep deprivation-induced oxidative stress in the hippocampus.

Factors Influencing Consumption Patterns of Household Recognizing Relatively Deprivation (상대적 박탈 인지가구의 소비유형과 영향요인)

  • This study identifies the relation between recognition relatively deprivation and consumption patterns. The data used were the seventh Korea Welfare Panel Study (KOWEPS). The final sample of 1,597 households was selected based on several sampling steps standard weights suggested by KOWEPS were applied. The final sample was subdivided into household recognizing relatively deprivation (51.7%) and non-household recognizing relatively deprivation (48.3%) per level of subjective income and objective income. We derived six consumption patterns from the cluster analysis as per consumption ratio: transportation dominant consumption pattern, essential consumption pattern, other dominant consumption pattern, care attention consumption pattern, private transfer consumption pattern, and education dominant consumption pattern. The results of this study are as follows. The analysis of the determinants of recognizing relatively deprivation in regards to question one found that the household was likely to be relatively deprived when: the head of a household was older, a household had a dual income, children and more total assets then those assessed 1 year ago compared to household living conditions, result of comparing the consumption patterns difference between household recognizing relatively deprivation and non-household recognizing relatively deprivation indicated that household recognizing relatively deprivation was more likely to belong to the transportation dominant consumption pattern, other dominant consumption pattern, and private transfer consumption pattern. The multinomial logistic analysis conducted to understand the determinants affecting the consumption patterns of household recognizing relatively deprivation indicated differences in demographic characteristics, household-related variables, financial variables and perception of economic conditions.

The Protective Effects of Chilgi-tang on Oxidative Stress by Glucose Deprivation in Neuro 2A Cells (Glucose deprivation으로 유발된 Neuro 2A 세포의 산화적 손상에 대한 칠기탕(七氣湯)의 보호효과)

  • Objective: The water extract of Chilgi-tang (CGT) has been traditionally used in treatment of heart diseases caused by stress in Oriental Medicine. However, little is known about the mechanism by which CGT rescues neuronal cells from injury damage. Therefore, this study was designed to evaluate the protective effects of CGT on Neuro-2A cells by glucose deprivation-induced cell death. Methods: We investigated how cell death induced by glucose deprivation was associated with increased reactive oxygen species (ROS) generation. Result: The CGT treatment prior to glucose deprivation insult significantly reduced the number of cell deaths and the glucose deprivation-induced increase in ROS. Nitric oxide (NO) was also attenuated by CGT treatment. In addition, we demonstrated that the anti-cell death effect of CGT was blocked by heme oxygenase-1 (HO-1) activation. Finally, pretreatment of cells with a hemin, HO-1 inducer, reduced glucose deprivation-induced cell death. In contrast, pretreatment of cells with a ZnPP, HO-1 activity inhibitor, attenuated CGT-induced inhibition of cell death. Conclusions: These findings indicate that ROS plays an important role in glucose deprivation-induced cell death and that CGT may prevent glucose deprivation-induced cell death by inhibiting the ROS generation through HO-1 activation in Neuro-2A cells.

Difference of Area-based deprivation and Education on Cerebrovascular Mortality in Korea (교육수준과 지역결핍지수에 따른 뇌혈관질환 사망률 차이)

  • This study was performed to identify the difference of the area-based deprivation and the educational level on the cerebrovascular mortality in Korea. Data used in this study was obtained from the Death Certificate Data 2000 and the 2000 Census produced by Korean National Statistics(NSO). We classified the whole country into 246 areas based on the administrative districts. Then, the Standardized Mortality Ratio (SMR) in cerebrovascular disease was calculated according to the sex, education level and 246 areas. Its Predicted SMR was calculated by the Empirical Bayes Methods to reduce the variation of the SMR values. The area-based deprivation of 246 areas were measured using the modified Carstairs index in which the 5 indicators consisted of overcrowding, the unemployment ratio of men, the percentage of households classified low social class, the percentage of non home owners, and finally those houses lacking basic amenities. The correlation between the area-based deprivation and the SMR of the whole country and the correlation between the area-based deprivation and the SMR of each metropolitan cities or provinces was analyzed by the Pearson correlation analysis method. After classifying the deprivation of 246 areas into 5 levels, we performed the random intercept Poisson regression analysis after adjusting education level and age using Empirical Bayes Method to investigate the relationship between the 5 deprivation levels and the cerebrovascular mortality. The SMR was increased in lower education level. Each 246 areas had different values in SMR, Predicted SMR and area-based deprivation. The area-based deprivation and the SMR of the whole country was not correlated in both sexes. The education level of an individual was associated the risk of cerebrovascular mortality in men. The risk of cerebrovascular mortality increased with age compared to the reference(<30). The area-based deprivation was not associated with the risk of cerebrovascular in both sexes. The findings of this study suggest that the SMR had positive and negative correlations with area-based deprivation depending on the metropolitan cities or province. It also suggests that the individual education level and age were related with mortality and finally that the area-based deprivation was not associated to the cerebrovascular mortality in Korea.

The Relative Deprivation of the Households in Poverty through Cost of Living (생계비를 통해 본 빈곤가구의 상대적 박탈)

  • The purpose of this study was to examine the relative deprivation of the households in poverty, comparing with the households in non-poverty, and to find out the characteristics of household groups by the level of relative deprivation. The data for this study was from the 6th Korea Welfare Panel Survey(KOWEPS), consisting of total 5,552 households. The results are as follows: There were statistically significant differences in the objective cost of living, the subjective minimum cost of living, and the subjective adequate cost of living between households in poverty and households in non-poverty. There was statistically significant difference in the relative deprivation between households in poverty and households in non-poverty. Poverty was the factor for the relative deprivation of households.

Effect of Bile Deprivation on Serum Lipids and Gastrin in the Rat (흰쥐의 담즙 상실이 혈청 지질과 Gastrin 대사에 미치는 영향)

  • The effect of bile deprivation on serum lipid and gastrin contents was investigat\ulcornered after choledocho-urinary cystostomy in Sprague-Dawley rats. Bile deprivated rats were compared with sham operated control group. Gastrin levels in serum and antral tissue were measured and serum lipid concentrations were also measured. Gastrin levels of serum and tissue after bile deprivation were increased significantly compared with those of the controL At the end of 1st and 2nd week after bile deprivation, serum cholesterol and triglyceride contents were significantly lower han those of the control. By 4th week, there was no significant difference between two groups. Increases in serum and antral gastrin levels temporarily coincided well with decreases in serum lipid contents after bile deprivation. These results suggest that there is increase in biosynthesis and release of gastrin and decrease in fat absorption at early stage of bile deprivation.

Factors Influencing Suicidal Ideation According to the Deprivation Indices of Different Regions in Korea

  • Objectives: This study examined how deprivation differed by region and the effect those differences had on suicidal ideation among the local population. Methods: Data collected over 10 years (2012-2021) in the Korea Welfare Panel Study were organized into 3 categories: metropolitan, city, and rural. A panel analysis was conducted on the impact of deprivation indices, socio-demographic characteristics, and life satisfaction on suicidal ideation in each category. Results: Income, divorce status, family relationship satisfaction, and medical deprivation had a significant impact on suicidal ideation in metropolitan areas, whereas these variables did not have significant effects in rural areas. In other words, income, family, and medical support were more impactful in city areas. Conclusions: Although the deprivation index was higher in rural areas than in city areas due to an aging population and reduced income levels, the mental health of rural residents was found to be generally better than that of city residents. The possibility that this is related to the strength of relationships within the respective communities should be considered in light of recent discussions on relational welfare.

The Effects of Balance Training with Visual Cue Deprivation on Gait Function in Patients with Stroke (시각차단 균형훈련이 뇌졸중 환자의 보행기능에 미치는 영향)

  • PURPOSE: The purpose of this study was to investigate the effect of balance rehabilitation training with the visual cue deprivation on gait function in stroke patients in comparison with balance training without the visual cue deprivation. METHODS: Twenty two stroke patients participated in this study. Patients were randomly assigned to one of the two balance training program with and without the visual cue deprivation. Balance training session for each group lasted 50 minutes, 3 times a week for a total of 6 weeks. Gait function was measured with the Functional Gait Assessment (FGA), the self-selective comfortable gait speed (CGS), the maximal gait speed (MGS), and the Gait Analysis System. Temporal and spatial gait parameters of each evaluation were measured before and after the balance training program respectively. RESULTS: After the program, the visual cue deprivation group improved significantly in the FGA, the CGS, the gait velocity, the step time, the step length, the stride length, and the Functional Ambulation Performance (FAP) in comparison with the balance training group with the visual cue (p<.05). CONCLUSION: The gait function of the participants with the visual cue deprivation showed more improvement after the balance training program compared to the patients group without the visual cue deprivation, Therefore, the balance training program with the visual cue deprivation may be useful for rehabilitation of patients with chronic stroke.

The Effect of Media Richness, Social Presence, and Life Satisfaction on Continuance Usage Intention or Withdrawal Intention of SNS Users via Relative Deprivation (매체 풍요도, 사회적 존재감 및 생활 만족도가 상대적 박탈감을 통해 SNS 이용자의 이용 지속 의도 또는 이탈 의도에 미치는 영향)

  • Purpose - This study aims to empirically verify the impact of media richness, social presence, and prior life satisfaction on various continual usage or withdrawal behaviors of SNS users via both a positive path of satisfaction and a negative path of relative deprivation. By identifying these causal paths, we observe dynamic interactions of SNS user psychology in a balanced view, and provide some implications about design principles for SNS providers. Research design, data, and methodology - We developed 16 hypothesis based on media richness theory, social presence theory, social comparison theory, the literature about relative deprivation, and the literature about the various reactions of IS users. The rich SNS media, social presence recognition among peer SNS users, and prior life satisfaction could generate positive experience, attitude, and virtuous behavioral intentions among SNS users. At the same time, rich media, low social presence, and low prior life satisfaction could generate relative deprivation and could increase withdrawal behavioral intentions such as refusal to provide information, misrepresentation of information, and removal of uploaded information in SNS. Scenario surveys were conducted to collect data from potential SNS users. Data from 357 surveys were collected and analyzed through a PLS algorithm to test the hypotheses. Results - Media richness, social presence, and prior life satisfaction could significantly increase perceived enjoyment, satisfaction, and behavioral intention of continual usage and knowledge sharing. They also could significantly decrease refusal and misrepresentation intention. Relative deprivation is significantly decreased only by prior life satisfaction. Relative deprivation could not significantly decrease satisfaction, but it could significantly increase misrepresentation and removal intention, which could be regarded as information distortion intention. Conclusions - SNS providers should focus on developing rich media and social presence support because these two variables could impact the positive experiences of SNS users. Moreover, the positive experiences could heavily influence SNS user behavior. Some management is needed to prevent relative deprivation and its consequences of misrepresentation and removal intention. SNS providers should prevent SNS users from excessive image misrepresentation and removal as this information distortion could be the source of relative deprivation.
