Insect Cell Cultures for Recombinant Protein Production (재조합 단백질 생산을 위한 곤충세포의 배양)

  • Insect cell cultures were performed in laboratory-scale vessels. The batch growth of insect cells was affected by such parameters as serum content, other nutrients, seeding density, and mechanical agitation. Lactate and ammonium were not likely to be environmental factors that inhibited cell growth at the concentrations observed at the end of batch cultures. In addition, redox potential was found to be a useful index in monitoring low-level dissolved oxygen during the cultivation of insect cells. Recombinant protein production by cells infected with a genetically-modified baculovirus was also demons treated. The maximum beta-galactosidase synthesis of 2800 units per reactor volume was achieved at the dilution rate of $0.006hr^{-1}$.

Estimation on the Physical Habitat Suitability of Benthic Macroinvertebrates in the Gapyeong Stream (가평천 저서성 대형무척추동물의 물리적 서식처 적합성 평가)

  • Habitat suitability index (HSI) of 17 benthic macroinvertebrate taxa, which were lotic insects of generic category except Potamanthidae in mayfly, was developed for three physical habitat factors (current velocity, water depth and substrate) based on an ecological monitoring in a Korean stream (Gapyeong). Weibull model was used as a probability density function to analyze the distribution of individual abundance related with physical factors, which showed it was so available. Number of species and total individual abundance increased along with the increase of current velocity and the mean diameter of substrate, and decreased along with the increase of water depth. Most taxa showed a clear preference for a fast current velocity, shallow water depth and coarse substrate except Ephemera, Potamanthidae (mayfly), and Plectrocnemia (caddisfly) which were rheophobic, potamophilic and lithophobious. Based on the canonical correspondence analysis, the relative importance of each factor was determined as follows: current velocity > substrate > water depth.

  • Using modified Lincoln solution, hydroponic system was developed to compare simultaneously a range of pasture species in terms of relative growth rate(RGR), rate of roots to shoots, and relative root volume and root area. Modified Lincoln solution achieved optimum growth of nineteen forage species tested, and was proved to be a good method for studying plant root characteristics. RGR was measured sucesshlly showing reasonably consistant values over time. There was a much differences in rootkhoot ratio between species within groups as between groups. Root tissue density was lower in grasses than legume or herb group. Root area index was very high for grasses and herbs, and this index should be useful for evaluating the comparative root system of different species.

Physicochemical Properties of Rice Extrudate with Added Ginger Powder by the Response Surface Regression Analysis (반응 표면 분석에 의한 생강 분말을 첨가한 쌀 압출 성형물의 이화학적 성질)

  • This research was attempted to investigate changes in physicochemical properties of rice extrudate with added ginger powder extruded by single screw extruder. Graphic three dimensional analysis on response surface regression was used to evaluate effects of extrusion variables on quality factors of the extrudate according to two independent variables, ginger consent 0∼12%, moisture content 14∼26%. The summarized results are as follows : 1) Regarding proximate composition of rice extrudate with added ginger powder, as ginger powder content of raw material Increased, crude tat, crude protein, crude ash and crude fiber increased, while soluble nitrogen free extract decreased. 2) Graphic three dimensional analysis on response surface regression was conducted for each dependent variable which revealed statistically significant relationship with independent variables, 0∼120A ginger and 14∼26% moisture content. Expansion ratio had a critical point as moisture content changed. As ginger and moisture content Increased, bulk density, break strength and water absorption Index Increased, while water solubility Index decreased. The predicted maximum degree of gelatinization in 6.15% ginger and 15.56% moisture content is 88.27%, and lightness decreased as ginger content Increased. According to the microstructure for the cross section of extrudate obsorbed with image analyzer, air cell number and perimeter revealed saddle point, meanwhile total area and fractarea of air cell had critical points as moisture content changed. In view of the results, quality of rice extrudate with added ginger powder was optimum when rice flour was fed to the extruder with 2∼7% singer powder and 15∼20% moisture content.

  • The fluid ensemble in the liquid lens, which is composed of electrolyte and silicone oil, is the key material system to vary the focal length processing of the electrochemical desorption. In order to characterize the capability of the liquid lens according to response time and optical range, we prepared a fluid ensemble comprising the electrolyte and oil. To elucidate the physical mechanism of the effective response time, we examined the viscosity dependency while satisfying the requirements for the density and refractive index of the electrolyte and oil, respectively. The characterization results indicated that the response time (up and down) is influenced by the viscosity of the electrolyte and oil. On this basis, we prepared a fluid ensemble capable of reversibly adjusting for the focal length of the liquid lens, as well as the response time. The ensemble is applicable to various systems such as micro-lens and optical sensors.

A study on the anti-oxidative effect of electroacupuncture at Wijung (BL40) in rats (위중전침의 항산화 효과에 대한 실험적 연구)

  • Objectives : The purpose of this study is to observe the anti-oxidative effects of electroacupuncture at Wijung (BL40) in rats. Methods : The author performed several experimental items including measurements of body weight, liver index, levels of albumin, total bilirubin, LDL-cholesterol, LDH, GOT and GPT in blood serum, and levels of SOD, glutathione, catalase, No and MDA (malondialdehyde) in liver, histological analysis of liver. Results : 1. In the BL40-EA group, liver index was decreased significantly compared to those of the control group, the holder group and the sham-EA group. 2. In the BL40-EA group, the albumin level was increased significantly compared to those of the control and holder group. LDL-cholesterol and GOT level were increased significantly compared to those of the control group and the holder group. 3. In the BL40-EA group, the SOD activity & the Catalase activity were decreased significantly compared to those of the control group and the holder group. 4. In the BL40-EA group, the density of liver tissue was more similarly maintained to the normal group compared to those of the control group and the holder group. Conclusion : These results suggest that electroacupuncture at BL40 has an antioxidant effect in human, and hereafter continuous study & clinical application is needed.

The relationship between age and the mandibular cortical bone thickness by using panoramic radiograph (파노라마방사선사진을 이용한 연령별 하악 피질골 두께에 관한 연구)

  • Purpose : This study was to determine the relationship between age and the mandibular cortical bone thickness on panoramic radiograph. Materials and Methods : Panoramic radiographs of 360 patients (180 men and 180 women) over 20 years old, who visited the Chonbuk National University Hospital from January to December in 2007, were assessed. The subjects were divided into 5 age groups. Five indices such as cortical bone thickness at the gonion (GI), antegonion (AI), and below the mental foramen (MI), the panoramic mandibular index (PMI), the mandibular cortical index (MCI) were measured on panoramic radiographs. Results : All five indices including GI, AI, MI, PMI, and MCI showed significant differences between third decade and over 8 decade groups (p<0.05). PMI, MI and GI showed significant differences with gender statistically (p<0.05). Conclusion : The mandibular cortical bone thickness showed negative correlation with age, and the value of the thickness (PMI, MI, and GI) was greater in men than in women.

Short Reads Phasing to Construct Haplotypes in Genomic Regions That Are Associated with Body Mass Index in Korean Individuals

  • Genome-wide association (GWA) studies have found many important genetic variants that affect various traits. Since these studies are useful to investigate untyped but causal variants using linkage disequilibrium (LD), it would be useful to explore the haplotypes of single-nucleotide polymorphisms (SNPs) within the same LD block of significant associations based on high-density variants from population references. Here, we tried to make a haplotype catalog affecting body mass index (BMI) through an integrative analysis of previously published whole-genome next-generation sequencing (NGS) data of 7 representative Korean individuals and previously known Korean GWA signals. We selected 435 SNPs that were significantly associated with BMI from the GWA analysis and searched 53 LD ranges nearby those SNPs. With the NGS data, the haplotypes were phased within the LDs. A total of 44 possible haplotype blocks for Korean BMI were cataloged. Although the current result constitutes little data, this study provides new insights that may help to identify important haplotypes for traits and low variants nearby significant SNPs. Furthermore, we can build a more comprehensive catalog as a larger dataset becomes available.

Evaluating the effective spectral seismic amplification factor on a probabilistic basis

  • All contemporary seismic Codes have adopted smooth design acceleration response spectra, which have derived by statistical analysis of many elastic response spectra of natural accelerograms. The above smooth design spectra are characterized by two main branches, an horizontal branch that is 2.5 times higher than the peak ground acceleration, and a declining parabolic branch. According to Eurocode EN/1998, the period range of the horizontal, flat branch is extended from 0.1 s, for rock soils, up to 0.8 s for softer ones. However, from many natural recorded accelerograms of important earthquakes, the real spectral amplification factor appears to be much higher than 2.5 and this means that the spectrum leads to an unsafe seismic design of the structures. This point is an issue open to question and it is the object of the present study. In the present paper, the spectral amplification factor of the smooth design acceleration spectra is re-calculated on the grounds of a known "reliability index" for a desired probability of exceedance. As a pilot scheme, the seismic area of Greece is chosen, as it is the most seismically hazardous area in Europe. The accelerograms of the 82 most important earthquakes, which have occurred in Greece during the last 38 years, are used. The soil categories are taken into account according to EN/1998. The results that have been concluded from these data are compared with the results obtained from other strong earthquakes reported in the World literature.

Prediction of osteoporosis using fractal analysis et cetera on panoramic radiographs (파노라마 방사성사진에서 프랙탈 분석 등을 이용한 골다공증 예측)

  • Purpose: The purpose of this study was to investigate whether panoramic radiographs were useful in predicting osteoporosis. Materials and Methods: 50 postmenoposal women between the age of 41.8 and 78.5 were classified as normal and osteoporosis groups according to the bone mineral density of lumbar vertebrae. Panoramic radiographs were taken. Age, body mass index, remaining mandibular teeth, mandibular cortical thickness and morphology, and fractal dimensions at periapical areas of mandibular first molars were evaluated to differentiate the two groups. Results: The age of osteoporotic group was statistically significantly higher than that of normal group (p<0.05), but not the body mass index or number of remaining mandibular teeth. The mean fractal dimension of osteoporotic group was $1.391{\pm}0.085$, and was significantly lower than that of the normal group, which was $1.523{\pm}0.725$ (p<0.01). Thick mandibular cortical thickness was common in normal group, whereas thin or very thin mandibular cortical thickness was common in osteoporotic group and the difference was significant (p < 0.05). C2 pattern was common in normal group followed by C1, whereas C2 was common in osteoporotic group followed by C3. The difference was statistically significant (p< 0.0 1). Conclusion: Age, mandibular cortical thickness and shape, fractal dimension on panoramic radiographs were useful in predicting osteoporosis.
