Shinisan has been used for treatment of allergic disease in Oriental Medicine. However, its effect in experimental models remains unknown. The mast cell plays a pivotal role in initiating allergic response by secreting intracytoplasmic granular mediators such as histamine. The present report describes an inhibitory effect of Shinisan on mast cell-mediated immediate type allergic reactions. Topical application of compound 48/80 can induce an ear swelling response in normal ($WBB6F_1-+/+$) mice but not in the congenic mast cell-deficient $WBB6F_1-W/W^v$ mice. Shinisan inhibited concentration-dependently mast cell-dependent ear swelling response induced by compound 48/80. Shinisan inhibited concentration-dependently passive cutaneous anaphylaxis induced by anti-dinitrophenyl (DNP) IgE in rats by topical application. Shinisan also inhibited concentration-dependently the histamine release from the rat peritoneal mast cells by compound 48/80 or anti-DNP IgE. Moreover, Shinisan had a significant inhibitory effect on compound 48/80-induced systemic anaphylactic reaction. These results indicate that Shinisan inhibits immediate-type allergic reactions by inhibition of mast cell degranulation in vivo and in vitro.

The present report describes an inhibitory effect of Magnoliae flos on mast cell-mediated immediate-type allergic reactions. Topical application of compound 48/80 can induce an ear swelling response in normal($WBB6F_1-+/+$) mice but not in the congenic mast cell-deficient $WBB6F_1-W/W^v$ mice. The purpose of this research was to investigate the effect of Kamidangkwieumja(KDEJ) water extract on the allergy reaction in mice. The results were obtained as follows: 1. The passive cutaneous anaphylaxis induced by egg albumin in fat was not affected. 2. The lethal anaphylaxis induced by platelet activating factor in mice. was inhibited. 3. The degranulation of peritoneal mast cells induced by compound 48/80 was not affected. 4. The acute hind paw edema was inhibited after 2hours later when it was induced by histamine. 5. The permeability of evans blue into peritoneal cavity induced by acetic acid was not affected. 6. Arthus reaction in mice was not affected. 7. The delayed type hypersensitivity induced by SRBC was inhibited. 8. The contact dermatitis induced by DNFB was not affected. 9. The hemagglutination titer induced by SRBC was inhibited. 10. The writhing syndrome induced by acetic acid was inhibited. 11. The population of heper T cells in mice thymus was enhanced. 12. The phagocytic activity of peritoneal macrophages was enhanced. 13. The production of nitric oxide from peritoneal macrophages was not affected. These results suggest that the anti-allergy effect of KDEJ is caused by steroidlike and enhanced immune action. The steroidlike action of KDEJ correspond with steroid-applying-method that frequently used in clinic, so it is used io treatment of psoriasis. The research on anti-allergy of KDEJ might has to be continued.

Our preliminary aim is to elucidate the pharmacokinetic features of the PNS(Panax notoginseng Buck F.H. Chen. (Arialiaceae) root). First, we assessed the prevention of neurtrophil functions. A Panax notoginseng inhibited neutrophil functions, including degranulation, superoxide generation, and leukotriene B4 production, without any effect on 5-lipoxygenase activity. This Panax notoginseng reduced nitric oxide (NO) and prostaglandin E2 production in mouse peritoneal macrophages stimulated with lipopolysaccharide, whereas no influence on the activity of inducible NO synthase, cyclo-oxygenase-2 or cyclo-oxygenase-1 was observed. Panax notoginseng significantly reduced mouse paw oedema induced by carrageenan. The results indicate that Panax notoginseng exerts anti-inflammatory effects related to the inhibition of neutrophil functions and of NO and prostaglandin E2 production, which could be due to a decreased expression of inducible NO synthase and cyclo-oxygenase-2.
Allergic diseases have increased rapidly over the past decades, affecting an estimated 20~30% of the population in developed countries. In this study, we investigated whether or not a typical costal sand dune plant Carex pumila (CPE) suppresses the activation of mast cells and IgE-mediated allergic response in vitro and in vivo. As the results, the extract of Carex pumila inhibited antigen-stimulated degranulation in RBL-2H3 cells and Bone marrow-derived mast cells (BMMCs), and IgE-mediated passive cutaneous anaphylaxis (PCA) in mice. CPE also suppressed the production of pro-inflammatory cytokines, TNF-${\alpha}$ and IL-4, in antigen-stimulated mast cells. As its mechanism of action, CPE inhibited the activation of Syk in $Fc{\varepsilon}RI$-mediated signalling pathway, and that of LAT, a downstream adaptor molecule of Syk, in a dose-dependent manner. CPE also suppressed the activation of mitogen-activated protein (MAP) kinases, p38, ERK1/2, JNK, and Akt. Altogether, CPE inhibited mast cell activation and IgE-mediated allergic response by antigen through suppressing the activation of Syk. These results suggest that CPE may be useful for the treatment of allergic diseases.
Malus sieboldii is a dicotyledonous plant that grows widely in Jeju Island and Ganghwa Island in Korea. Malus sieboldii has been known as a detoxifying and antioxidant plant, but study on allergic diseases is not known. In this study, we investigated the effect of Malus sieboldii extract (MSE) on the activation of mast cells, which is well known to be a critical causative cell to induce allergic diseases. As a result of our experiments, MSE inhibited the degranulation and inflammatory cytokine secretion from mast cells by antigen stimulation. As the mechanism of MSE in mast cells, it inhibited the activation of Syk kinase, a essential signaling protein activated by antigen, and further inhibited activation of $PLC{\gamma}$ and MAP kinase(P38, ERK1/2, and JNK). Furthermore, in vivo animal studies showed that MSE significantly inhibited IgE-mediated passive cutaneous anaphylaxis and passive systemic anaphylaxis in a dose-dependent manner. Taken together, the results of this study showed for the first time that MSE inhibited IgE-mediated allergic responses by suppressing Syk kinase in mast cells. Therefore, it could be considered that MSE is worth developing as an anti-allergic material.
Mast cells are crucial as effector cells in the immediate-type allergic reaction. Lentinus edodes has been the popular edible mushroom in oriental countries and reported to have immunomodulatory, anti-tumor, anti-atherogenic, anti-viral, and anti-allergic activities. However, the roles of L. edodes in mast cell-mediated anaphylactic reaction have not been fully elucidated. In this research, we have demonstrated the effects of the methanol extract of L. edodes (MELE) on mast cell-mediated anaphylaxis-like and anaphylactic reactions. MELE suppressed systemic anaphylaxis-like reaction, plasma histamine levels, and ear swelling response in mice treated with compound 48/80. MELE also suppressed passive systemic and cutaneous anaphylaxis mediated by anti-dinitrophenyl IgE. In accordance with these findings, MELE dose-dependently decreased histamine release from RPMC evoked by compound 48/80 or the antigen-antibody reaction. To clarify the mechanism of degranulation system, intracellular cAMP levels as well as calcium influx in RPMC was evaluated. In compound 48/80-treated RPMC, MELE blocked calcium uptake into the cells. In addition, MELE elevated the intracellular cAMP content and significantly attenuated compound 48/80-induced cAMP reduction in RPMC. Taken together, we propose the clinical use of MELE in mast cell-mediated immediate-type allergic diseases.
Mast cells play a key role in IgE-mediated allergic response. We investigated whether Swertia pseudochinensis Hara extract (SPE) inhibits IgE-mediated allergic response in mast cells and an allergic animal model. Additionally, we explored SPE's mechanism of action in mast cells. Our results showed that SPE inhibited both antigen-stimulated degranulation and the production of TNF-α and IL-4 in bone marrow-derived mast cells (BMMCs) and rat basophilic leukemia (RBL)-2H3 cells. SPE also suppressed allergic response in IgE-mediated passive cutaneous anaphylaxis (PCA) in mice. As for the mechanism of action of SPE in mast cells, it inhibited the activation of Syk kinase, a critical signaling protein in the FcεRI-mediated signaling pathway, and also the activation of LAT, a downstream adaptor protein of Syk. We further observed the reduced activation of mitogen-activated protein (MAP) kinases (P38, ERK1/2, and JNK) and Akt in mast cells. Our results described for the first time that SPE has an anti-allergic effect by suppressing mast cells through the inhibition of Syk kinase. Therefore, SPE may be useful for the treatment of type I allergic diseases.

Lobelia chinensis Lour. has commonly been used in traditional Chinese medicine for the treatment of antidote, diuretic, diarrhea, and inflammation. This study aimed to identify the active compounds in an aqueous extract of L. chinensis responsible for its anti-atopic effect in vitro using RBL-2H3 cells. A chemical investigation of secondary metabolites in an aqueous extract of L. chinensis led to the isolation of nine chemical constituents, which included the four marker compounds, and these were evaluated for their inhibitory effects on IL-4 mRNA expression and the release of β-hexosaminidase in propidium iodide-induced RBL-2H3 cells. We found diosmetin and fraxidin inhibited cellular IL-4 mRNA expression, and that diosmetin and 6,8-dimethoxycoumarin inhibited DNP-specific IgE-induced degranulation in these cells. Our study suggests that diosmetin, fraxidin, and 6,8-dimethoxycoumarin are potential candidates for the treatment of atopic diseases.
Neutrophils, the most abundant innate immune cells, play essential roles in the innate immune system. As key innate immune cells, neutrophils detect intrusion of pathogens and initiate immune cascades with their functions; swarming (arresting), cytokine production, degranulation, phagocytosis, and projection of neutrophil extracellular trap. Because of their short lifespan and consumption during immune response, neutrophils need to be generated consistently, and generation of newborn neutrophils (granulopoiesis) should fulfill the environmental/systemic demands for training in cases of infection. Accumulating evidence suggests that neutrophils also play important roles in the regulation of adaptive immunity. Neutrophil-mediated immune responses end with apoptosis of the cells, and proper phagocytosis of the apoptotic body (efferocytosis) is crucial for initial and post resolution by producing tolerogenic innate/adaptive immune cells. However, inflammatory cues can impair these cascades, resulting in systemic immune activation; necrotic/pyroptotic neutrophil bodies can aggravate the excessive inflammation, increasing inflammatory macrophage and dendritic cell activation and subsequent TH1/TH17 responses contributing to the regulation of the pathogenesis of autoimmune disease. In this review, we briefly introduce recent studies of neutrophil function as players of immune response.
