Decomposition of Triclosan onto E-beam Process using a Design of Experiment(DOE) (전자빔을 이용한 triclosan 제거에 있어서 실험계획법의 이용)

  • This study investigated on the photolytic degradation of Triclosan by E-beam process. The optimization of process was investigated during a series of batch experiments by design of experiments(DOEs). The DOE was one of the statistical application that was used for designed the response surface to determine the effects of each parameters. The responses were applied as removal rate of Triclosan(%, $Y_1$) and TOC removal rate(%, $Y_2$). Two independent variables were concentration of Triclosan and irradiation intensity that were designed as "$x_1$" and irradiation intensity was designed as "$x_2$". The regression equation in coded parameter between the Triclosan removal efficiencies(%) and TOC removal efficiencies(%) was $Y_1=63-12.4335x_1+15.1835x_2+5.8125x{_1}^2-5.6875x{_2}^2-0.75x_1x_2(R^2=95.1%,\;R^2(Adj)=91.7%)$ and $Y_2=46-8.8462x_1+11.7175x_2-0.75x{_1}^2-6.25x{_2}^2(R^2=98.7%,\;R^2(Adj)=97.7%)$, respectively. The model predictions agreed well with the experimentally observed results $R^2$ and $R^2(Adj)$ over 90% within both of $Y_1$ and $Y_2$. This result shows that the regression model express well about the effects of parameters on E-beam process and the statistical method was successfully applied.

Studies on the Stabilities of Red Pepper Oleoresin (고추 oleoresin의 품질안정성(品質安定性))

  • This experiment was carried out to study the preparations of red pepper oleoresin, the effects of pH and heat treatment on the stabilities of capsanthin and capsaicin in oleoresin state, and the interacting effects of ascorbic acid, metal salts and EDTA on the stabilities of capsanthin in the oleoresin-linoleate aqueous model system. The results were as follows: 1. Acetone was the most effective solvent to extract capsanthin and capsaicin from red pepper powder. The yield of oleoresin extracted with acetone was 14.27%. 2. Capsaicin was more stable at high temperature than capsanthin in oleoresin state. Capsanthin and capsaicin in oleoresin state were comparatively stable in the range (ron) pH 3 to pH 8. 3. Ascorbic acid acted as a prooxidant on the capsanthin oxidation reaction at concentrations up to $10^{-3}M$, but acted as an antioxidant at $10^{-1}M$. 4. The addition of $Cu^{+2}M$ and $Fe^{+3}M$ ions at all concentration increased the prooxidant activity on the degradation of capsanthin in oleoresin state. 5. EDTA showed a strong antioxidation the stability of capsanthin in oleoresin state.

Remediation of Water Quality Using Up-Flow Circulation Apparatus(UFCA) in a Reservoir (용승순환장치를 이용한 호소의 수질개선에 관한 연구)

  • This study was conducted to analyse the effects of the UFCA for treating polluted water in a reservoir. The UFCA mixes water by circulation of surface and bottom water layers. The circulation supplies oxygen to bottom of the reservoir, resulting in water quality improvement. With a UFCA in use, we surveyed the changes of temperature, pH, transparency, depth, conductivity, DO, COD, BOD, T-N, T-P and Chlorophyll-a for 7 months from Feb. to Aug. in 2004 in our experimental reservoir. There was little difference in the surface and bottom temperatures of the reservoir because of water mixing by the UFCA. However, pH was changed from 7 to 9. The transparency of water was about 80 cm through the all periods. Conductivity was $150\;{\mu}S/cm$ in early Feb., but increased to $270\;{\mu}S/cm$ in early March. Little change was seen in DO with depth, but it was maintained above 6 mg/l in June and July. BOD increased from 2.1 to 12.2 mg/l. The study reservoir did not undergo any eutrophication during the period of our experiment, but the comparison reservoir had an algae-bloom. The COD in the experimental reservoir increased from 5.4 to 14.5 mg/l. The COD concentration of the experimental reservoir was higher than comparison reservoir at the beginning of the study but in August this situation was reversed. SS concentration increased from 13.5 to 23.5 mg/l in Feb., but it fell from between 8.5 to 11.2 mg/l in July. T-N was increased from 1.3 to 4.9 mg/l. It increased up to 3 times in the rainy season as compared to other components. However the comparison reservoir increased up to 40 times higher than the experimental reservoir in the same period. T-P increased from 0.04 to 0.17 mg/l. The ratio of T-N to T-P increased from 20:1 to 40:1 which means that T-P was a growth limiting factor for algae and aquatic plants. Chlorophyll-a increased from 20 to 120 mg/l, and its concentration was correlated with T-P, such that Chlorophyl-a concentration increased with increased of T-P concentration. The concentrations of COD, T-N, T-P and other parameters were higher in the experimental reservoir than in the comparison reservoir but this situation was reversed in July, when the most severe eutophication occurred. The results show that overall the experimental reservoir was greatly remedied by UFCA. The UFCA accelerated the degradation of aquatic organic materials through effective supply of air with up-flow and circulation of water. We conclude that the UFCA can be very effective in aspect of the remediation of water quality incontaminated reservoirs and lakes.

A Study on Distance Estimation in Virtual Space According to Change of Resolution of Static and Dynamic Image (가상현실공간에서 정적 및 동적 이미지의 해상도 변화에 따른 거리추정에 관한 연구)

  • The virtual reality (VR) technology has been used as the application of architectural presentation or simulation tool in the field of industry. The high immersion and intuitive visual information are the great merits of design evaluation or environmental simulation when we are using the virtual environments. But the distortion of distance perception in VR is still a big problem when the accuracy of distance presentation is strictly required. For example, distance estimation is especially important when the virtual environments are applied to the presentational tool for evaluation the space design or planning in the field of architecture. If there are some perception error between the built space in real and represented space in virtual, the accurate design evaluation or modification of design is hard to be carried out during the design development stage. In this paper, we have carried out some experiments about distance estimation in the immersive virtual environments to verify the factors and their influence. We made a hypothesis that the lack of the information for the user in VR causes the different distance estimation from the real world because users are usually comfortable with moving fast and long distance in VR environments compared with moving slow and short distance in real space. So, we carried out basic experiment to prove our hypothesis that the lack of information makes subjects estimate the distance of walking in VR shorter compared with the same distance in real. Also, among the factors that probably affect the distance estimation in VR, we have verified the influence of the image resolution. The influence of resolution degradation of image on the distance estimation was verified with the condition of static and dynamic images. The results showed that the resolution has deep relation with the distance estimation. For example, the subject underestimated the distance at the lower resolution condition. We also found the methods of the making the lower resolution image could affect on the visual perception of subjects.

Studies on Cheese Ripening Part V. Degradation of Nitrogenous Compounds During the Ripening of Gouda-type Cheese (치이즈 숙성(熟成)에 관(關)한 연구(硏究) 제5보(第5報) Gouda-type 치이즈의 질소화합물(窒素化合物)의 변화(變化))

  • This experiment has been carried out to study the changes of caseins is Gouda-type cheese during ripening by the use of DEAE-cellulose column and polyacrylamide gel electrophoresis. The results obtained were as follow: 1. The amount of water soluble nitrogenous compounds in the cheese ripened for 2 and 4 months was 19.03% and 30.61% for total nitrogen in the cheese, respectively. On the other hand, the amount of 5% NaCl soluble nitrogenous compounds in the cheese increased to 41.13% for 2 months ripening, but it decreased to 22.61% at 4th month. 2. By DEAE-cellulose column, various nitrogenous compounds of Gouda-type cheese were fractionated into 5 major and several minor peaks. The proportion of some fraction areas to the whole increased with the ripening period and the others decreased. 3. Electrophoretic patterns of various nitrogenous compounds in a 4-month-old cheese showed 5 and 8 bands, repectively. 4. Sixteen amino acids were identified in soluble and precipitable compounds at 12% TCA in a 4-month-old cheese.

Effects of pH Control Methods on Removal Efficiency in Electrokinetic Bioremediation of Phenanthrene-contaminated Soil (Phenanthrene-오염토양의 동전기 생물학적 복원에서 pH 조절방법이 제거효율에 미치는 영향)

  • In this study, problems related with pH control in electrokinetic(EK) bioremediation of phenanthrene contaminated soil were observed, and the effects of pH control methods on the removal efficiency were investigated to search a further application strategy. In a preliminary experiment, it was found out by flask cultivation that a certain sulfate concentration was needed to degrade phenanthrene well using Sphingomonas sp. 3Y. However, when $MgSO_4$ was used as sulfate source in EK bioremediation, the bacterial activity reduced seriously due to the abrupt decrease of pHs in soil and bioreactor by the combination of magnesium and hydroxyl ions. When another strong buffering compound was used to control the pH problem, the good maintenance of the bacterial activity and pHs could be observed, but the removal efficiency decreased largely. When a low concentration of $MgSO_4$ was added, the removal efficiency decreased somewhat in spite of the good maintenance of neutral pHs. With the addition of NaOH as a neutralizing agent, the removal efficiency also decreased because of the increase of soil pH. Consequently the selection of electrolyte composition was a very important factor in EK bioremediation and some sulfate sources suitable for both bacterial activity and contaminant degradation should be investigated.

Application and perspectives of proteomics in crop science fields (작물학 분야 프로테오믹스의 응용과 전망)

  • Thanks to spectacular advances in the techniques for identifying proteins separated by two-dimensional electrophoresis and in methods for large-scale analysis of proteome variations, proteomics is becoming an essential methodology in various fields of plant sciences. Plant proteomics would be most useful when combined with other functional genomics tools and approaches. A combination of microarray and proteomics analysis will indicate whether gene regulation is controlled at the level of transcription or translation and protein accumulation. In this review, we described the catalogues of the rice proteome which were constructed in our program, and functional characterization of some of these proteins was discussed. Mass-spectrometry is a most prevalent technique to identify rapidly a large of proteins in proteome analysis. However, the conventional Western blotting/sequencing technique us still used in many laboratories. As a first step to efficiently construct protein data-file in proteome analysis of major cereals, we have analyzed the N-terminal sequences of 100 rice embryo proteins and 70 wheat spike proteins separated by two-dimensional electrophoresis. Edman degradation revealed the N-terminal peptide sequences of only 31 rice proteins and 47 wheat proteins, suggesting that the rest of separated protein spots are N-terminally blocked. To efficiently determine the internal sequence of blocked proteins, we have developed a modified Cleveland peptide mapping method. Using this above method, the internal sequences of all blocked rice proteins (i. e., 69 proteins) were determined. Among these 100 rice proteins, thirty were proteins for which homologous sequence in the rice genome database could be identified. However, the rest of the proteins lacked homologous proteins. This appears to be consistent with the fact that about 30% of total rice cDNA have been deposited in the database. Also, the major proteins involved in the growth and development of rice can be identified using the proteome approach. Some of these proteins, including a calcium-binding protein that fumed out to be calreticulin, gibberellin-binding protein, which is ribulose-1,5-bisphosphate carboxylase/oxygenase activate in rice, and leginsulin-binding protein in soybean have functions in the signal transduction pathway. Proteomics is well suited not only to determine interaction between pairs of proteins, but also to identify multisubunit complexes. Currently, a protein-protein interaction database for plant proteins (http://genome .c .kanazawa-u.ac.jp/Y2H)could be a very useful tool for the plant research community. Recently, we are separated proteins from grain filling and seed maturation in rice to perform ESI-Q-TOF/MS and MALDI-TOF/MS. This experiment shows a possibility to easily and rapidly identify a number of 2-DE separated proteins of rice by ESI-Q-TOF/MS and MALDI-TOF/MS. Therefore, the Information thus obtained from the plant proteome would be helpful in predicting the function of the unknown proteins and would be useful in the plant molecular breeding. Also, information from our study could provide a venue to plant breeder and molecular biologist to design their research strategies precisely.

Generation of Chemically Active Species in Hybrid Gas-Liquid Discharges (기체-액체 혼합 방전에 의한 화학적 활성종 생성 특성)

  • We carried out a laboratory scale experiment about the characteristics of chemically active species produced in hybrid gas-liquid discharges. The electrode configuration which had high voltage electrode in the gas phase and ground electrode in the liquid was utilized while high voltage electrode has been typically positioned in the liquid in other studies. Our electrode was configured in such a way as to increase the energy efficiency of chemical reactions by creating a higher electrical field strength and a narrower pulse width than the typical electrode configuration. The highest ozone concentration was obtained at 45 kV which was the medium value in operating voltages. The decrease of solution conductivity increased the resistance of liquid phase and the electric field strength through the gas phase, so ozone gene-ration rate was enhanced. The increase of voltage promoted the production rate of hydrogen peroxide by increasing the electric field strength. In a lower voltage, the increase of solution conductivity increased the degradation rate of $H_2O_2$, so the $H_2O_2$ generation rate decreased. On the other hand, the effects of UV radiation, shock waves etc. increased the $H_2O_2$ generation rate as the solution conductivity increased. A higher rate of $H_2O_2$ generation can be achieved by mixing argon to oxygen which generates a stronger and more stable discharges.

Mineralization and Adsorption of $^{14}C$-Lablled Imazapyr in Soil (($^{14}C$ 표지 Imazapyr의 토양중 무기화와 흡착)

  • In order to elucidate the degradation characteristics of the herbicide imazapyr in soil, mineralization to $^{14}CO_2$and adsorption were investigated using eight types of soils with the different physico-chemical properties. The results obtained were as follows: 1. During the incubation period of 12 weeks after the treatment of imazapyr, the amounts of $^{14}CO_2$ evolved from 8 types of soils with different properties ranged from 1.5 to 4.9% of the originally applied $^{14}C$ activities. Soil C, G, and H with low pH and high organic matter showed low $^{14}CO_2$evolution, whereas soil B and D with high pH and low organic matter did high $^{14}CO_2$ evolution. 2. Time for reaching the equilibrium concentrations in the adsorption experiment of imazapyr in soils was about 3 hours at $25^{\circ}c$ in soil C, D, G, and H. Imazapyr was adsorbed in the range of 0.25${\sim}$28.32% in soils with different physico-chemical properties. Among the soil parameters, organic matter content was the most influential in imazapyr adsorption on soil. The Freundlich adsorption coefficient $(K_f)$ increased 5.5 to 25.6 times as organic matter content increased 2.0 to 21.3 times. Hence it seems that the extent to which soil organic matter contributes to imazapyr adsorption is greater than that of clay mineral. $K_f$ values for the soils tested were 0.44, 0.08, 0.65, and 2.05 in soil C, D, G, and H, respectively. In all the soils tested, $K_f$ values had a strong resemblance to K_$K_d$.

Application of DNA Probe Method for Detection of 2,4-Dichlorophenoxyacetic Acid Degrading Bacteria in Soil (DNA Probes에 의한 토양의 이사디 (2,4-D) 분해세균의 검출)

  • Total bacterial community DNA, which was extracted from microcosm soil and field soil after 2,4-D amendments, was analyzed on Southern blots, using the tfdA gene probe derived from plasmid pJP4 and the Spa probe from Sphingomonas paucimobilis. Southern blot analyses with total bacterial DNA extracted from soils Inoculated with Pseudomonas cepacia/pJP4 revealed that DNA probe method could detect the 2,4-D degrading bacteria down to $10^5\;cells/g$ dry soil. In the microcosm experiment, there was a good correlation between 2,4-D degradation and banding patterns in hybridization analyses performed after each 2,4-D treatment using the two probes. When bacterial DNA extracted from microcosm soil was hybridized with the Spa probe, a change in the position of hybrid bands was observed over time in a Southern blot, suggesting that population change or possibly genetic rearrangement in 2,4-D degrading microbial populations occurred in this soil. With the Spa probe, one hybrid DNA band was persistently observed throughout the five 2,4-D additions. When bacterial DNA isolated from the field soil was probed with the tfdA and Spa, strong hybridization signal was observed in the 100 ppm-treated subplot, weak signal In the 10 ppm-treated subplot, and no significant signal in the 1 ppm-treated and control subplots. The data show that DNA probe analyses were capable of detecting and discriminating the indigenous 2,4-D degrading microbial populations in soil amended with 2,4-D under laboratory and field conditions.
