Ginsenoside profiles and related gene expression during foliation in Panax ginseng Meyer

  • Panax ginseng is one of the most important medicinal plants in Asia. Triterpene saponins, known as ginsenosides, are the major pharmacological compounds in P. ginseng. The present study was conducted to evaluate the changes in ginsenoside composition according to the foliation stage of P. ginseng cultured in a hydroponic system. Among the three tested growth stages (closed, intermediate, and opened), the highest amount of total ginsenoside in the main and fine roots was in the intermediate stage. In the leaves, the highest amount of total ginsenoside was in the opened stage. The total ginsenoside content of the ginseng leaf was markedly increased in the transition from the closed to intermediate stage, and increased more slowly from the intermediate to opened leaf stage, suggesting active biosynthesis of ginsenosides in the leaf. Conversely, the total ginsenoside content of the main and fine roots decreased from the intermediate to opened leaf stage. This suggests movement of ginsenosides during foliation from the root to the leaf, or vice versa. The difference in the composition of ginsenosides between the leaf and root in each stage of foliation suggests that the ginsenoside profile is affected by foliation stage, and this profile differs in each organ of the plant. These results suggest that protopanaxadiol- and protopanaxatriol(PPT)-type ginsenosides are produced according to growth stage to meet different needs in the growth and defense of ginseng. The higher content of PPT-type ginsenosides in leaves could be related to the positive correlation between light and PPT-type ginsenosides.

The Calmodulin-Binding Transcription Factor OsCBT Suppresses Defense Responses to Pathogens in Rice

  • We previously isolated the OsCBT gene, which encodes a calmodulin (CaM)-binding protein, from a rice expression library constructed from fungal elicitor-treated rice suspension cells. In order to understand the function of OsCBT in rice, we isolated and characterized a T-DNA insertion mutant allele named oscbt-1. The oscbt-1 mutant exhibits reduced levels of OsCBT transcripts and no significant morphological changes compared to wild-type plant although the growth of the mutant is stunted. However, oscbt-1 mutants showed significant resistance to two major rice pathogens. The growth of the rice blast fungus Magnaporthe grisea, as well as the bacterial pathogen Xanthomonas oryzae pv. oryzae was significantly suppressed in oscbt-1 plants. Histochemical analysis indicated that the hypersensitive-response was induced in the oscbt-1 mutant in response to compatible strains of fungal pathogens. OsCBT expression was induced upon challenge with fungal elicitor. We also observed significant increase in the level of pathogenesis-related genes in the oscbt-1 mutant even under pathogen-free condition. Taken together, the results support an idea that OsCBT might act as a negative regulator on plant defense.

담배에서 병원균에 반응하는 MAPK 신호전달체계에 의해 매개되는 방어 유전자들의 분리 및 특성화 (Isolation and Characterization of Defense Genes Mediated by a Pathogen-Responsive MAPK Cascade in Tobacco)

  • SIPK와 WIPK의 상위 단계 인산화 효소로 알려진 NtMEK2가 DEX 유도성 시스템에 의해 밝혀졌다. 이 NtMEK2 유전자가 지속적으로 활성화된 돌연변이체인 $NtMEK2^{DD}$의 발현은 SIPK와 WIPK를 활성화 시켜 주므로 과민감 반응과 같은 세포 괴사를 야기하는 것으로 나타나 NtMEK2-SIPK/WIPK 체계가 담배에서 방어 반응을 조절하고 있음을 알 수 있었다. 그러나 NtMEK2-SIPK/WIPK 체계에 의해서 조절 되는 하위 기질이나 방어관련 유전자들에 대한 연구는 아직 미비한 상태이다. 그래서 본 연구는 NtMEK2-SIPK/WIPK 체계에 매개되는 하위 유전자들을 분리하기 위하여 $NtMEK2^{DD}$ 형질전환 식물체를 이용해 ACP에 기초한 DDRT-PCR을 수행하였다. 그 결과 본 연구를 통해 처음으로 pI2-4, MTS2, SINA, CDM1, HRGP 및 DEG45를 포함해 여섯 개의 DEG들을 선발하였다. 이 유전자들의 발현은 $NtMEK2^{DD}$ 형질전환에서 다시 확인하였으며 특히 pI2-4, CDM1, HRGP의 유전자 발현은 다른 유전자들과 비교해 볼 때 살리실산과 담배모자이크바이러스에 강하게 반응하여 증폭됨을 알 수 있었다. 이러한 결과를 볼 때 NtMEK2-SIPK/WIPK 체계에 의해 조절되는 세 개의 유전자는 병저항성에 관여하고 있음을 제시한다 하겠다.

배 검은별무늬병(Venturia nashicola) 고도 저항성 '93-3-98' 유래 PR-10 유전자의 특성 (Characterization of PR-10 gene derived from highly resistant '93-3-98' pear inoculated with scab (Venturia nashicola))

  • 배 검은별무늬병 고도저항성 '93-3-98'과 고도감수성 '스위트스킨'간의 suppression subtractive hybridization 분석을 통해 '93-3-98'에서 특이적으로 발현되는 pathogenesis-related 10 (PR-10) 유전자를 분리하여 PyrcpPR-10으로 명명하고 기관 및 품종별 발현양상을 분석하였다. 단편염기서열의 rapid amplification of cDNA ends PCR을 통해 PyrcpPR-10 유전자는 전체길이가 743bp이고, 480bp의 ORF와 159개의 아미노산을 가지는 것으로 확인되었다. PyrcpPR-10 유전자의 염기서열은 '황실리'(저항성), '감천배'(중도저항성), '원황'(중도감수성), '신고', '스위트스킨'(고도감수성)은 동일하였으나 'Bartlett'(고도저항성)은 일부 염기서열의 차이를 보였다. BLAST X를 통한 다른 식물 종의 PR-10 아미노산과 비교에서 64 ~ 98%의 상동성을 보였고 공통적으로 GXGGXG motif를 가지고 있었다. 기관 및 조직별 PyrcpPR-10 유전자의 발현량은 꽃잎이 가장 높았으며 다음으로 잎, 꽃대, 눈, 수피 순이었다. 저항성과 감수성 품종에 따른 PyrcpPR-10 유전자의 발현양상은 모든 품종에서 접종 24시간 후 급격히 증가였으며, 특히 'Bartlett', '93-3-98', '황실리'에서 높게 발현되었고 '감천배', '원황'의 경우 저항성 품종에 비해 상대적으로 낮았으며, 고도 감수성 '신고', '스위트스킨'은 발현이 가장 낮았다. 배에서 분리한 PyrcpPR-10 유전자는 검은별무늬병 저항성에 직접 연관되는 것으로 추정된다.

  • 고염 스트레스는 식물의 성장과 수확량에 치명적인 영향을 야기한다. 그와 같은, 환경 스트레스에 의하여 식물은 다양한 유전자의 발현으로 저항성을 가지게 하는 기작이 발달되어 있다. 본 연구에서는 애기장대에서 다양한 환경 스트레스에 관여하는 유전자를 분리할 목적으로 GGM(Graphical Gaussian Model) program을 사용한 후, BLH8(BEL1-Like Homeodomain Gene 8) 유전자의 돌연변이 식물체를 구축하였다. atblh8-1 돌연변이체는 고농도의 $Na^+$$K^+$ 이온에 특이적으로 백화현상을 보이지만, 뿌리 성장에는 변화를 보이지 않았다. 그러므로, BLH8 단백질은 $Na^+$$K^+$과 같은 환경스트레스 저항성에 관여하는 중요한 인자임을 시사한다. 이와 같이, GGM program은 환경 스트레스에 관여하는 유전자를 분리하기 위한 유용한 도구일 것으로 사려된다.

닭의 성숙/미성숙란에서 RNA Sequencing을 이용한 유전자 발현 양상 고찰 (Gene Expression Profiling by RNA Sequencing in Mature/Immature Oocytes of Chicken)

  • 조류의 난포 성장은 호르몬의 작용에 따라 크기가 달라져 각각의 단계를 이루며 성장하게 된다. 난의 성숙에 관련된 유전자는 난 단백질 생산과 산란률에 밀접한 관련이 있으며, 이를 유전자 발현 측면에서 심도 있는 고찰이 필요가 있다. 본 연구는 NGS를 이용한 RNA-seq 데이터를 이용하여 유전자의 발현량과 유전자 상호 구조에 대한 분석을 실시하여 난의 발달 과정에 필요한 유전자군을 조사하였다. 본 실험에 사용된 개체는 한국 재래계 흑색계통이 사용되었고, 비교조직은 미성숙란과 성숙란의 RNA를 추출하여 유전자의 발현 양상을 살펴봄으로 난의 성숙에 필요한 유전자의 발현 양상을 보고자 하였다. 실험을 위해 Total RNA를 추출하였고, HiSeq 2000 platform을 사용하여 염기서열을 분석하고, Tuxedo Protocol과 DAVID 프로그램을 통해 유전자의 기능과 상호간의 연관관계를 예측하였다. 탐색된 유전자군은 미성숙란과 성숙란 간에 많은 차이를 보이고 있는 유전자군을 탐색한 결과, 315개의 발현이 다르게 나타나는 것으로 보이고 있으며, GO 분석을 통하여 기능면에서 미성숙란과 성숙란에서 확연히 구분되는 유전자 발현 양상을 확인할 수 있었다. 이들 결과를 통하여 향후 난성숙 과정을 이해하고, 계란 품질 향상을 위한 마커 개발을 기여할 수 있을 것으로 사료된다.

  • Phytophthora blight is a devastating disease of pepper and occurs almost anywhere peppers are grown. Phytophthora blight is caused by Phytophthora capsici and this pathogen can infect every part of the plant by moving inoculum in the soil, by infecting water on surface, by aerial dispersal to sporulating lesions. Management of Phytophthora blight currently relies on cultural practices, crop rotation, and use of selective fungicides. Since these treatments are a short-term management, a classical breeding for development of resistant pepper against the Phytophthora is an alternative. So far some of the resistant cultivars have been on the market, but those are limited regionally and commercially. Therefore, ultimately an elite line resistant against this disease should be developed, if possible, by biotechnology. We have set out a series of work recently in order to develop Phytophthora resistant pepper cultivar. For the first time, the cDNA microarray analysis was peformed using an EST chip that holds around 5000 pepper EST clones to identify genes responsive to Phytophthora infection. Total RNA samples were obtained from Capsicum annuum PI201234 after inoculating P. capsici to roots and soil and exposed to the chip. .Around 900 EST clones were up-regulated and down-regulated depending on the two RNA sample tissues, leaf and root. From those, we have found 55 transcription factors that may be involved in gene regulation of the disease defense mechanism. Further and in detail information will be provided in the poster.

Biocontrol Efficacies of Bacillus Species Against Cylindrocarpon destructans Causing Ginseng Root Rot

  • Two antifungal bacteria were selected from forest soils during the screening of microorganisms antagonistic to Cylindrocarpon destructans, a cause of ginseng root rot. The antifungal bacteria were identified as Bacillus subtilis (I4) and B. amyloliquefaciens (yD16) based on physiological and cultural characteristics, the Biolog program, and 16S rRNA gene sequencing analyses. Antagonistic activity of both bacterial isolates to C. destructans increased with increasing temperature. More rapid starch hydrolytic activity of the bacteria was seen on starch agar at higher temperatures than at lower temperatures, and in the higher density inoculum treatment than in the lower density inoculum treatment. The bacterial isolates failed to colonize ginseng root the root tissues inoculated with the bacteria alone at an inoculum density of $1{\times}10^6$ cfu/ml, but succeeded in colonizing the root tissues co-inoculated with the bacteria and C. destructans. Scanning electron microscopy showed that the pathogen was damaged by the low-density inoculum treatment with the bacterial isolates as much as by the high-density inoculum treatment. Both bacterial isolates were more effective in reducing root rot when they were treated at a concentration of $1{\times}10^6$ cfu/ml than at $1{\times}10^8$ cfu/ml. Also, only the former treatment induced prominent wound periderm formation, related to structural defense against pathogen infection. The results suggest that the bacterial antagonists may have high potential as biocontrol agents against ginseng root rot at relatively low-inoculum concentrations.

Identification of another calmodulin-binding domain at the C-terminal region of AtCBP63

  • Calcium signals can be transduced by binding calmodulin (CaM), a $Ca^{2+}$ sensor in eukaryotes, is known to be involved in the regulation of diverse cellular functions. We isolated a CaM-binding protein 63 kD (AtCBP63) from the pathogen-treated Arabidopsis cDNA expression library. Recently, AtCBP63 was identified as a CaM bining protein. The CaM binding domain of AtCBP63 was reported to be located in its N-terminal region, In this study, however, we showed that ACaM2 could specifically bind to second CaM-binding domain (CaMBD) of AtCBP63 at the C-terminal region. The specific binding of CaM to CaM binding domain was confirmed by a gel mobility shift assay, a split ubiquitin assay, site-directed mutagenesis, and a competition assay using a $Ca^{2+}$/CaM-dependent enzyme. The gene expression of AtCBP63 was induced by pathogens and pathogens related second messengers. This result suggests that a CaM binding protein, AtCBP63, may play role in pathogen defense signaling pathway.

Effect of salt stress on the anthocyanin content and associated genes in Sorghum bicolor L.

  • Abiotic stress is one of the most serious problems in plant productivity because it dramatically delays plant growth and development. One of the abiotic stresses, soil salinity, has an adverse effect on plant growth, particularly in areas where irrigation is necessary like semiarid Asia and Africa. Among several physiological parameters, anthocyanin accumulation is a valuable indicator of the condition of the plant, and it tends to increase under salt stress conditions because of its protective role in such an environment. Consequently, it may be important to search for well adapted genotypes for upcoming climate changes. Anthocyanins are known to have important roles in defense against biotic and abiotic stresses, providing important functions for protecting plant cells from reactive oxygen species. In this study, we investigated the anthocyanin accumulation between two Korean sorghum genotypes, Sodamchal and Nampungchal. The two genotypes were subjected to a regulated salinity condition, and the anthocyanin contents were evaluated in both. In Nampungchal, the anthocyanin content increased with 150 mM NaCl treatment during the time course of the experiment. However, the anthocyanin content of Sodamchal decreased in the same condition. The measured values of the anthocyanin content should be useful to identify the intensity of the salt tolerance in Sorghum bicolor L. Furthermore, we studied gene expression profiling of salt stress related genes with qRT-PCR. These results suggest that Nampungchal is a more tolerant genotype to salt stress compared to Sodamchal. This information should be useful for breeding salt-resistant cultivars in sorghum.
