  • Objective: In the present study, an liquid chromatography/mass spectrometry (LC/MS) metabolomics approach was performed to investigate potential biomarkers of milk production in high- and low-milk-yield dairy cows and to establish correlations among rumen fluid metabolites. Methods: Sixteen lactating dairy cows with similar parity and days in milk were divided into high-yield (HY) and low-yield (LY) groups based on milk yield. On day 21, rumen fluid metabolites were quantified applying LC/MS. Results: The principal component analysis and orthogonal correction partial least squares discriminant analysis showed significantly separated clusters of the ruminal metabolite profiles of HY and LY groups. Compared with HY group, a total of 24 ruminal metabolites were significantly greater in LY group, such as 3-hydroxyanthranilic acid, carboxylic acids, carboxylic acid derivatives (L-isoleucine, L-valine, L-tyrosine, etc.), diazines (uracil, thymine, cytosine), and palmitic acid, while the concentrations of 30 metabolites were dramatically decreased in LY group compared to HY group, included gentisic acid, caprylic acid, and myristic acid. The metabolite enrichment analysis indicated that protein digestion and absorption, ABC transporters and unsaturated fatty acid biosynthesis were significantly different between the two groups. Correlation analysis between the ruminal microbiome and metabolites revealed that certain typical metabolites were exceedingly associated with definite ruminal bacteria; Firmicutes, Actinobacteria, and Synergistetes phyla were highly correlated with most metabolites. Conclusion: These findings revealed that the ruminal metabolite profiles were significantly different between HY and LY groups, and these results may provide novel insights to evaluate biomarkers for a better feed digestion and may reveal the potential mechanism underlying the difference in milk yield in dairy cows.

  • This study was conducted to investigate reasons for culling of Chungcheong dairy herds and to investigate the relationships between cow parity, interval from calving to culling, or season and culling reasons. Lactation data from 826 cows on 9 dairy farms from 2013 to 2015, including reasons for culling, dates of birth, last calving, and culling, were analyzed. The overall culling rate was 29.8% (246/826), with mammary problems (23.2%) being the most prevalent reason for culling, followed by infertility (14.2%) and downer cow syndrome (11.0%). The culling rate varied among farms (18.8-43.1%, p < 0.05), but was greater in parity $4{\leq}(40.7%)$ than parities 1, 2, or 3 (17.9-23.6%), whereas the rate was less frequent during mid lactation (21.1%) than in early (36.2%) or late (42.7%) lactation. Moreover, the culling rate was greater during summer (35.8%) than other seasons (20.3-23.2%). Mammary problems, downer cow syndrome, and metabolic disorders were the more common reasons for culling during early lactation, whereas infertility was predominant during late lactation. Our data show that the culling rate in Chungcheong dairy herds was associated with cow parity, interval from calving to culling, and season, with the main reasons for the removal of cows from dairy herds being mammary problems, infertility, and downer cow syndrome.

  • The study was conducted to evaluate the effect of vitamin E and zinc supplementation on energy metabolites, lipid peroxidation, and milk production in peripartum Sahiwal cows. For this, thirty-two pregnant dry Sahiwal cows were selected at sixty days prepartum and divided into four groups viz control, $T_1$, $T_2$, and $T_3$ of eight each. Group $T_1$ were supplemented with zinc at 60 ppm/d/cow, group $T_2$ were supplemented with vitamin E at 1,000 IU/d/cow and group $T_3$ were supplemented with combination of vitamin E at 1,000 IU/d/cow and zinc at 60 ppm/d/cow during d 60 prepartum to d 90 postpartum. Blood samples were collected on d -60, -45, -30, -15, -7, -3, 0, 3, 7, 15, 30, 45, 60, 90, and 120 with respect to day of parturition and analysed for glucose, non esterified fatty acid, and thiobarbituric acid reactive substance. Body condition score was maintained significantly better (p<0.05) in $T_3$ than in the control, $T_1$ and $T_2$ groups. Overall glucose level was higher (p<0.05) in $T_3$ than control, $T_1$, and $T_2$ groups. Levels of nonesterified fatty acid, and thiobarbituric acid reactive substance were lower (p<0.05) in $T_3$ than control, $T_1$, and $T_2$ groups. Milk yield was higher (p<0.05) in $T_3$ than control, $T_1$, and $T_2$ groups. In conclusion, the present study indicated that the supplementation of vitamin E and zinc in peripartum Sahiwal cows enhanced milk production by reducing negative energy balance.

  • This study was conducted to determine the microorganisms inhabitating in cow genitalia and their antimicrobial drug susceptibility. During the period between July, 1985 and February, 1986, a total of 111 cow genitalia, 58 from Korean native and 53 from dairy cow, were sampled at three abattoirs. Gross pathological examination and bacterial isolation and identification were performed from the genital samples. In addition antimicrobial drug susceptibility test for the microorganisms isolated, some synergistic activity among drugs were examined on the major organism isolated from the cases of endometritis and pyometra. The results are summerized as follows: 1. Among the bacteria isolated from the genitalia, Staphylococcus spp., C. pyogenes, E. coli, Proteus spp., Streptococcus spp., Bacillus spp. were most frequently isolated whereas the genera of Pasteurella, Pseudomonas, Klebsiella and Yersina were detected far less frequently. 2. In Korean native cow the genera of Straphylococcus and Steptococcus were more isolated than dairy cow while in dairy cow the genera of Corynebacterium, Proteus, Escherichia were more of ten isolated than Korean native cow. 3. From cow genital organs showing lesions of endometritis and prometra, C. pyogenes was most frequently isolated, the isolation rate being 60 percent, and follow by Staphylococcus spp., Proteus spp., E. coli and Pasteurella spp. in the order. 4. Antimicrobial drug susceptibility test conducted on the major organisms isolated showed that all the isolates were susceptible to gentamicin, cephalosporin and sulfisoxazole, but resistant to tetracycline and penicillin. 5. Twenty-nine isolates of C. pyogenes were submitted to the synergistic activity test of cephalosporin, kanamycin and streptomycin with penicillin. Synergists were demonstrated in 90 percent, 31 percent and 27 percent of isolates examined by the combined use of penicillin and cephalosporin, penicillin and kanamycin, penicillin and streptomycin, respectively. About 10 percent of the isolates were found to be indifferent by the synergism test.

  • The present study was conducted to investigate effect of dietary protected amino acid on milk yield and composition in dairy cow using meta-analysis. Total 21 research papers were employed in analysis, and mixed model was used for the analysis of effects. Effect of protected methionine (PM) and combination of protected methionine and lysine (PML) were investigated under two different levels of dietary crude protein (CP, <18% and >18%). For performance of dairy cow, milk yield, milk composition including milk fat and protein content and yield and 4% FCM (fat corrected milk) production were used for analysis. In case of milk yield, a trend of increment was found at PM supplementation at low CP (P=0.055). However, the effect of PM at high CP was detected as not significant (P>0.05). In case of milk protein, inclusion of PM at low CP showed significant decrement (P<0.05). However, there was no significant effect of MP on milk protein at high CP (P>0.05). Supplementation of MP at high CP level showed significant increment of milk fat (P<0.05). MP supplementation represented significant increment of 4% FCM production (P<0.05) regardless of dietary CP levels. Effects of PML on milk yield and composition at both of low and high dietary CP were not significant in this study. However, it seem to be that there was a possible positive effect of MPL application at high dietary CP on performance of dairy cow.
