소의 친자감정을 위한 Microsatellite markers의 유전적 다양성 분석 (Analysis of genetic diversity for cattle parentage testing using microsatellite markers)

  • The objective of present study was to ascertain genetic diversity for cattle parentage testing. A total of 59 random cattle samples(29 Korean native cattle and 30 dairy cows) were genotyped by using 11 microsatellite loci(BM1824, BM2113, ETH10, ETH225, EH3, INRA23, SPS115, TGLA122, TGLA227, TGLA53, and TGLA126). This method consisted of multiplexing PCR procedure and showed reasonable amplification of all PCR products. Genotyping was performed with an ABI 310 genetic analyzer. The number of alleles per locus varied from 5 to 11 with a mean value of 6.73 in the Korean native cattle(KNC), 4 to 9 with a mean of 5.91 in dairy cows(DC). Expected heterozygosity was ranged 0.534~0.855(mean 0.732), 0.370~0.866(mean 0.692) in the KNC and DC, respectively. PIC value was ranged 0.485~0.821(mean 0.684), 0.336~0.834(mean 0.640) in the KNC and DC, respectively. Of the 11 markers, 7 markers(ETH10, EH3, INRA23, SPS115, TGLA122, TGLA227, TGLA53) and 3 markers(INRA23, TGLA227, TGLA53) have relatively high PIC value (>0.7) in the KNC and DC, respectively. The total exclusion probability of 11 microsatellite loci was 0.9997 and 0.9991 in the KNC and DC, respectively. These results present basic information for developing a system for parentage verification and individual identification in the KNC and DC.

한우 미경산우의 인공수정 적기 예측을 위한 체측활용 (Use of morphometric measurement for estimation of AI timing of Hanwoo heifer (Korean native cattle))

  • The aim of the present study was to evaluate and estimate timing of artificial insemination (AI) in Hanwoo heifer (Korean native cattle) that is the most popular breed of beef cattle in Korea. To determine changes in body weight of heifers around AI, body weight were measured at different stages either before or after AI. We found that daily body weight gain was higher in the pregnant cows after AI. We also investigate correlation between body mass measured by shoulder height and body length, and conception rates, used (body length+ height)2 instead of height2 for body mass index (BMI), and found that relatively more BMI heifers (>55) showed higher conception rates. Finally, we estimated body weight by measuring should height (SH), heart girth (HG), and body length (BL); $BW=3.93372^*HG-2.90985^*SH-0.021^*BL$. In addition, we observed that HG is most closely correlated with BW; $y(BW)=1.77355^*x(HG)$, $R^2=0.98881$. In summary, we can determine the best timing of AI using body measurement and its application including BMI.

부산지역 한우의 요네병 감염 실태 조사 (Sero-prevalence of Paratuberculosis (Johne's disease) of Korean Native Cattle in Busan Area)

  • Johne's disease is a chronic inflammatory bowel disease of cattle, sheep, goats and other ruminants. Mycobacterium paratuberculosis is the etiologic agent of this disease. Many studies have been carried out on paratuberculosis from Korean native cattle and dairy cattle in multiple areas around nation, but there is no report in Busan area. The purpose of this study is to investigate the seroprevalence of bovine paratuberculosis in Busan area from March in 2011 to October in 2012. A total of 863 Korean native cattle of 213 farms were tested by ELISA method. The 287 (33.3%) Korean native cattle of 119 (55.9%) farms were positive in ELISA. In regional analysis, 234 (33.6%) out of 696 cows in Kijang-gun, 35 (39.3%) out of 89 cows in Gangseo-gu and 15 (20.8%) out of 72 cows in Geumjeong-gu were positive. In sexual analysis, 277 (33.6%) out of 824 cows in Female and 10 (25.6%) out of 39 cows in Male were positive. In aga-related analysis, 13 (22.4%) out of 58 cows in 1 year, 33 (32.0%) out of 103 cows in 2 years, 87 (34.1%) out of 255 cows in 3 years, 118 (36.6%) out of 322 cows in 4 years, 21 (36.8%) out of 57 cows in 5 years, 8 (29.6%) out of 27 cows in 6 years, 6 (31.6%) out of 19 cows in 7 years and 1 (4.5%) out of 22 cows in 8-11 years were positive.

국가 종축우에 대한 요네병 항체 양성률 조사 (Seroprevalence of Paratuberculosis in Pure-bred Breeding Cattle in Korea)

  • 요네병은 Mycobacterium avium subspecies paratuberculosis에 의해 발생하며 반추동물에서 만성 쇠약을 일으켜 전세계적으로 양축산업에 커다란 경제적 손실을 일으키는 질병이다. 본 연구에서는 2008년부터 2009년까지 2년동안 의뢰된 총 3,927건의 국가 종축우의 혈청에 대한 요네병의 항체 양성률에 대해 조사한 결과 21두(0.5%)에서 항체 양성을 나타내었으며 이는 국내 일반 농가의 양성률(1.2-16.4%) 보다 현저히 낮은 수준이었다. 또한 연령별 양성률은 2세 미만에서 0.2% (3두), 2-3세에서 0.4% (2두), 3-4세에서 0.7% (3두), 4세 초과에서 0.9% (13두)로 전 연령에서 양성이 나타났으나 연령이 높아짐에 따라 양성률이 증가하는 경향을 보였다. 품종 별로는 젖소는 모두 음성이었으며 한우에서만 양성률 0.6%를 나타내었다.

  • Sixteen Korean native bulls weighing about 211 kg were randomly allotted to investigate the effect of different dietary calcium levels on performance. Feeding period was divided into growing period and fattening period for 180 days. Four calcium levels (0.44, 0.58, 0.72 and 0.86%) were used to each period. Bulls fed the diet containing 0.72% calcium had greater body weight gain and consumed more feed than bulls fed diets containing 0.44, 0.58 and 0.86% calcium and improved in feed efficiency. Bulls fed the diet containing 0.44% calcium were worst in performance. It can be concluded that Korean native bulls in growing and fattening period required no more than 0.72% calcium in diet that contained 0.55% phosphorus, and appeared that when ratio of calcium to phosphorus intake was 1.45, performance of Korean native bulls was most beneficial.

분만간격과 산차를 중심으로 한국 재래종인 한우의 번식능력 분석 (Reproductive performance of Korean native cattle (Hanwoo) focusing on calving interval and parity)

  • 한우는 한국 고유재래종으로 가장 많이 사육되고 있으나 번식성적에 대한 자료는 제한적이다. 본 연구는 총 163,613두의 번식성적에 대하여 통계 분석하였다. 산차 및 계절별로 분만 간격과 임신기간의 차이는 없었으나 매년 분만간격이 증가하고 있었다. 또한 생시 체중이 23 kg 이었을 때 생애 번식성적이 가장 우수 하였다.

한우의 번식실태평가 및 번식우 생산성 분석 (An Analysis of Evaluation for Korean Native Cattle (Hanwoo) Reproductive Performance and Cow-Calf Profitability)

  • Calculating break-even price of calf production is closely associated with reproductive efficiency. To determine the price, we need data from reproduction records including number of claves weaned, number of cows exposed for breeding, and annual cash coast per cow, and average weaning or market weight of claves sold and retained. Unfortunately, the data were not available in Korea native cow (Hanwoo). To evaluate the performance and the price, we collected calving interval from about 60,000 cows for last 10 years and estimated reproductive performance. Calving interval was increased 4.3% and pregnancy rate was decreased about 1.4~2.8% year-on-year. Increases in growth rates of number of cow and semen per calf supported the low reproductive performance. Finally, break-even price was calculated using estimated percent calf crop and demonstrated that growth rate of break-even price is larger than that of annual cash per cow, suggesting cow-calf profitability and financial efficiency in Korea native cow (Hanwoo) is getting worse.

국내 서식 한우에서 큐열 항체 양성율 조사 (Seroprevalence of Q-fever in Korean native cattle)

  • Q-fever is a vector-borne (Coxiella [C.] burnetii) zoonotic disease that is an increasing public health concern. To date, some research about Q-fever prevalence in dairy herds and human patients has been reported in Korea, but information about Korean native cattle is scarce. To measure the prevalence rates of C. burnetii in Korean native cattle, a total of 1,095 bovine serum samples collected during 2010~2013 were analyzed with an enzyme-linked immunosorbent assay. Sixty-eight heads of cattle were diagnosed as positive and while 19 heads were suspected (positive rate = 6.2%). Interestingly, Jeju province had a seropositivity rate six times greater than that of other provinces (18.9% vs. 3.2%). High seroprevalence might be caused by wide distribution of ticks in Jeju province compared to other regions. Based on these data, extensive monitoring of C. burnetii infection in cattle, tick distribution, and climate changes is required.

Analysis of seasonal effect on Korean native cattle (Hanwoo) birth weight

  • Recently, summer temperatures have frequently been abnormal in Korea owing to global warming. In summer, a decrease in feed intake rate and biological activity were observed in Hanwoo (Korean Native Cattle), leading to lower production rates in the industry. However, the precise scale of damage was not reported as with other animals of economic value. This study was conducted to investigate the effects of birth season on birth weight in Hanwoo. Data were collected from 100 local breeding farms from 2016 to 2019. A total of 41,081 Hanwoo calves were classified and analyzed by sex, year, month, and season (March-May, spring; June-August, summer; September-November, fall; and December-February, winter) of birth. The birth weight of Hanwoo calves differed according to birth month. The average birth weight of male calves was 30.47 kg and that of female calves was 28.16 kg. Hanwoo birth weight was the highest in March-born calves and the lowest in July-born calves. The birth weights of calves born in February, March, April, November, and December were significantly larger than those of calves born in July. In addition, the birth weight of Hanwoo calves from the summer was significantly lower than that of calves born in other seasons. Furthermore, Hanwoo steer slaughter age showed a negative correlation, whereas carcass weight had a positive correlation with birth weight. In the beef cattle industry, birth weight is a very important economic characteristic that is related to growth rate. These data will contribute toward planning the reproduction of Hanwoo and analysis of changes in characteristics of economic value owing to high temperatures.

Effect of Timed Artificial Insemination Protocols on the Pregnancy Rate Per Insemination and Pregnancy Loss in Dairy Cows and Korean Native Cattle under Heat Stress

  • We aimed to determine the effect of timed artificial insemination (TAI) protocols on the pregnancy rate per insemination and pregnancy loss compared with AI performed at detected estrus in dairy cows and Korean Hanwoo cattle under heat stress. In dairy cattle, 1,250 sets of data that underwent AI during heat stress (temperature-humidity index ≥ 72) were categorized according to their TAI protocols or as controls: 1) PGF-36 h-estradiol benzoate (EB)-36 h-TAI (PG-EB group, n = 113); 2) GnRH-7 days-PGF-56 h-GnRH-16 h-TAI (Ovsynch group, n = 455); or 3) GnRH-6 days-Ovsynch (G6G group, n = 136). The remaining cows underwent AI at detected estrus (AIDE group, n = 546). The probability of pregnancy per AI 45 days after AI was higher (P < 0.01) in the PG-EB (odds ratio [OR]: 1.68), Ovsynch (OR: 1.48), and G6G (OR: 1.79) groups than in the AIDE group. However, the prevalence of pregnancy loss between 30 and 45 days after AI did not differ among the groups. In Hanwoo cattle, 617 sets of data inseminated artificially under heat stress were categorized into AIDE (n = 281), PG-EB (n = 194), and combined Ovsynch or G6G (n = 142) groups. The probability of pregnancy per AI 45 days after AI and the prevalence of pregnancy loss between 30 and 45 days after AI did not differ among the groups. Thus, implementation of a TAI protocol (PG-EB, Ovsynch, or G6G) in dairy cows under heat stress improves the pregnancy rate per AI versus AIDE, whereas there is no beneficial effect of TAI on the pregnancy rate of Hanwoo cattle under heat stress.
