Purification and Biochemical Characterization of Lectin from Viscum album (겨우살이 Lectin의 정제 및 생화학적 특성)

  • The lectin was purified through 0.15 M NaCl extraction, ammonium sulfate precipitation, sepharose 4B affinity chromatography and gel filtration using sephadex G-150 from the leaves of Visum album collected in Mt. Duk Yu. The final gel filtration step resulted in 11.64 folds purification with 0.14% of recovery yield. We also performed biochemical characterization of the purified Visum album lectin. HPLC analysis of lectin purified by gel filtration revealed a singel peak. The analysis of the purified lectin by SDS-PAGE showed a tetramer composed of two identical subunits with molecular weights of 32 and 30 kDa. The lectin was a glycoprotein containing 14.4% carbohydrate, which consist of glucose, fructose, arabinose and xylose, and the amino acids such as phenylalanine, lysine and tyrosine. The purified lectin agglutinated human red blood cell types with similar potency, but when tested against red blood cells from mouse, bovine, rabbit, chicken and porcine, significant difference in potency were observed. Hemaggluting activity was inhibited by D-galactose, D-mannose, D-lactose and D-raffinose, but not by D-glucose, D-glucosamine, D-mannosamine, L-fructose, D-xylose, D-arabinose, D-galacturonic acid, D-fructose, L-rhamnose and N-acetyl-D-galactosamine. The optimal pH and thermal stability of the purified lectin were pH 4.0-7.0 and 20-5$0^{\circ}C$, respectively.

Purification and Characterization of a New Galactoside Specific Lectin from Trichosanthes kirilowii Root

  • A new lectin, named TRA, was purified from Trichosanthes kirilowii root by acid-treated Sepharose 6B, Mono-Q, and TSK-gel 3000SW column sequential chromatography. The lectin appeared homogeneous by native gel electrophoresis at pH 4.3 and gave two protein bands of Mr=31 and 28 kDa by SDS-PAGE. The N-terminal amino acid sequences of the polypeptides of TRA have not been reported in amino acid sequences of the lectins. TRA lectin formed a precipitate with asialofetuin, neuraminidase-treated fetuin. A sugar inhibition assay indicated that N-acetyl-D-galactosamine, among the monosaccharides tested, was the most potent inhibitor of TRA-induced hemagglutination. Asialofetuin showed a 260-times stronger inhibitory activity than N-acetyl-D-galactosamine. TRA lectin also showed agglutination with normal leukocytes and lymphoma cells, but not with premature hemopoietic cells. These results suggest that TRA is a novel plant lectin.

Effects of Physical Parameters on Water Quality in Agricultural Reservoirs (농업용 저수지의 물리적 인자가 수질에 미치는 영향)

  • The effect of physical parameters on water quality was analyzed using monitoring data of 193 agricultural reservoirs. The retention time of reservoirs ($t_d$) ranged between 10 and 140 days, and the ratio of drainage area (DA) to reservoir surface area (SA) was between 10 and 120. Both ratios of DA/SA and total area (TA)/ reservoir storage (ST) in Korean agricultural reservoirs were relatively greater than those in natural lakes in other countries. As retention time was plotted against DA/SA ratio, it was shorter in Korean reservoirs than natural lakes. The semi-logarithmic relationship between TA/SA and t>$t_d$ was $t_d\;=\;42.21(TA/ST)^{-1}$ (n = 50, $R^2\;=\;0.89$). While areal loading of total phosphorus (TP) was below $4\;gTP{\cdot}m^{-2}{\cdot}yr^{-1}$ in general, it exceeded $10\;gTP{\cdot}m^{-2}{\cdot}yr^{-1}$ in reservoirs where DA/SA ratio was greater than 100, which implies that areal loading of TP increases as DA/SA ratio increases. Chl-a concentration was positively related with the mean depth of reservoir, implying the higher Chl-a concentration with deeper the mean depth. Therefore, the deeper reservoir might be advantageous in water quality management perspective if other morphological conditions are similar. The empirical regression equation using physical parameters was also suggested in the estimation of TP concentration in the reservoirs. Combined information presented in this paper might be applicable to the water quality management in agricultural reservoirs.

Cloning and Expression of the Aminopeptidase Gene from the Bacillus lichenformis In Bacillus subtilis

  • A gene (hap) encoding aminopeptidase from the chromosomal DNA of Bacillus licheniformis was cloned. The gene is 1,347 bp long and encodes a 449 amino acid preproprotein with a major mature region of 401 amino acids (calculated molecular mass 43,241 Da). N-Terminal sequence of the purified protein revealed a potential presence of N-terminal propeptide. The deduced primary amino acid sequence and the mass analysis of the purified protein suggested that a C-terminal peptide YSSVAQ was also cleaved off by a possible endogeneous protease. Tho amino acid sequence displayed 58% identity with that of the aminopeptidase from alkaliphilic Bacillus halodurans. This bacterial enzyme was overexpressed in recombinant Escherichia coli and Bacillus subtilis cells. Clones containing the intact hap gene, including its own promoter and signal sequence, gave rise to the synthesis of extracellular and thrmostable enzyme by B. subtilis transformants. The secreted protein exhibited the same biochemical properties and the similar apparent molecular mass as the B. lichenzyormis original enzyme.

Corrosion Fatigue Crack Propagation Behaviour of TMCP Steel Plate at Ballast Tank of Ship Structure under the Condition of Cathodic Overprotection (선체구조 Ballast Tank 고장력 TMCP강판의 과방식중 부식피로균열 전파거동)

  • For the steel structures those are used in harsh sea environments, corrosion fatigue is a challenging issue in connection with design life. In this research, in order to investigate the influence of cathodic overprotection on the corrosion fatigue crack propagation behavior, corrosion fatigue crack propagation test under the condition of -950mV vs SCE was conducted by using of high tensile TMCP steel plate and the relationships between da/dN-${\Delta}K$ were obtained. At this test, when ${\Delta}K$ is low, the crack propagation rates were accelerated compared to those of seawater condition, however, when ${\Delta}K$ is high, the crack propagation rates were lower than those of seawater condition. As the cause for the acceleration and deceleration of corrosion fatigue crack propagation rates under the condition of cathodic overprotection, the role of hydrogen and calcareous deposits are discussed.

A Fatigue Life Prediction by Growth Characteristics of a Small Surface Crack (작은 표면균열의 성장특성에 의한 수명예측)

  • This paper deals with a fatigue life prediction of a surface crack based on the experimentally obtained relationship between surface crack length ratio $a/a_{f}$ and cycle ratio $N/N_{f}$ using micro computer. Firstly $a/a_{f}$-$N/N_{f}$ curves obtained from experimental tests, were assumed as three curves UC(the upper limit curve), LC(the lower limit curve) and MC(the middle curve), and these were utilized to predict the fatigue life. Comparing the calculated values which represent the characteristics of crack growth behaviors from the three assumed curves with the experimental ones, it has been found that in the stable crack growth region, they coincide reasonably well each other. And the differences between the fatigue lives obtained from the assumed curves and the experimental fatigue life did not exceed 20%. Using the characteristics of $a/a_{f}$-$N/N_{f}$ curves, it is possible to predict the da/dN-Kmax curves and the S-$N_{f}$ curves.

  • Fatigue crack growth and life have been estimated based on established empirical equations. In this paper, an alternative method using artificial neural network (ANN) -based model developed to predict fatigue damages simultaneously. To learn and generalize the ANN, fatigue crack growth rate and life data were built up using in-plane bending fatigue test results. Single fracture mechanical parameter or nondestructive parameter can't predict fatigue damage accurately but multiple fracture mechanical parameters or nondestructive parameters can. Existing fatigue damage modeling used this merit but limited real-time damage monitoring. Therefore, this study shows fatigue damage model using backpropagation neural networks on the basis of X -ray half breadth ratio B / $B_o$, fractal dimension $D_f$ and fracture mechanical parameters can estimate fatigue crack growth rate da/ dN and cycle ratio N / $N_f$ at the same time within engineering limit error ($5\%$).

  • Many researchers have studied woven fabric carbon-fiber-reinforced composite (CFRP) materials but the study of fatigue crack propagation in composites has been insufficient. It has known that the crack propagation behavior differs depending on the load and the fiber direction. In this study, the fatigue crack propagation along two different fiber array directions ($0^{\circ}$, $45^{\circ}$) in plain woven CFRP composite was investigated. Fatigue crack propagation tests were conducted on the woven CFRP composite under a sinusoidal waveform load with stress ratios of 0.1 at a frequency of 10 Hz. Once the results of the tests were obtained, fatigue crack propagation rates (da/dN) were plotted against the energy release rate amplitude (${\Delta}G$), and it was observed that either mode I crack propagation or mixed mode crack propagation occurs depending on the fiber array direction.

  • Recently, with rapid increase of gas demand, there occurs much interest their security of safety in the gas storage tanks and pressure vessels etc. In order to solve the problems, the occurrence of corrosion fatigue crack and the propagation behavior must be investigated. Especially the occurrence of corrosion fatigue crack and the propagation behavior in the part which has concentrated stress or defects, must be studied more carefully. In this paper, the high-tensile steel of SPV 50 which is much used for building the LPG storage tanks was tested by the use of a plane bending corrosion fatigue tester under the various marine environment and in the air. These experiments were carried out to investigate the surface crack propagation behavior, the value of experimental constant for Paris' rule(da/dN=$C(K)^m$), the crack depth propagation rate and the accelerative factor of the surface crack propagation rate. The main results obtained are as follows ; 1) As the specific resistances of marine environment decreases, the exponential value of slope m of Paris' rule(da/dN=$C(K)^m$) decreases and the value of intercept C increases. 2) The surface crack propagation rate and the crack depth propagation rate are delayed, as the specific resistances of marine environment is increased. 3) The accelerative factor of the surface crack propagation rate by corrosion fatigue is higher, according as the stress intensity factor range ${\Delta}K_A$ is small.

Antioxidant Principles of Nelumbo nucifera Stamens

  • In our ongoing study to identity antioxidants from natural sources, the antioxidant activity of Nelumbo nucifera stamens was evaluated for their potential to scavenge stable 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radicals, inhibit total reactive oxygen species (ROS) generation, in kidney homogenates using 2 ,7 -dichlorodihydrofluorescein diacetate (DCHF-DA), and scavenge authentic peroxynitrites ($ONOO^-$). A methanol (MeOH) extract of the stamens of N. nucifera showed strong antioxidant activity in the $ONOO^-$system, and marginal activity in the DPPH and total ROS systems, so were therefore fractionated with several organic solvents, such as dichloromethane ($CH_2 Cl_2$), ethyl acetate (EtOAc) and n-butanol (n-BuOH). The EtOAc soluble fraction, which exhibited strong antioxidant activity in all the model systems tested, was further purified by repeated silica gel and Sephadex LH-20 column chromatographies. Seven known flavonoids [kaempferol (1), kaempferol 3-Ο-$\beta$-D-glucuronopyranosyl methylester (2), kaempferol 3-Ο-$\beta$-D-glucopyranoside (3), kaempferol 3-Ο-$\beta$-D-galactopyranoside (4), myricetin 3 ,5 -dimethylether 3-Ο-$\beta$-D-glucopyranoside (5), kaempferol 3-Ο-$\alpha$-L-rhamnopyranosyl-(1$\rightarrow$6)-$\beta$-D-glucopyranoside (6) and kaempferol 3-Ο-$\beta$-D-glucuronopyranoside (7)], along with $\beta$-sitosterol glucopyranoside (8), were isolated. Compound 1 possessed good activities in all the model systems tested. Compounds 2 and 7 showed scavenging activities in the DPPH and $ONOO^-$ tests, while compounds 3 and 4 were only active in the $ONOO^-$ test. Conversely, compound 8 showed no activities in any of the model systems tested.
