Hepatoprotective Activity of Bacopa monniera on D-galactosamine Induced Hepatotoxicity in Rats

  • Hepatoprotective action of alcoholic extract of Bacopa monniera (BME) was evaluated on Dgalactosamine (D-GalN) induced rat liver toxicity. Bacopa monniera extract reduced the elevated serum enzyme activities of ALT, AST, ALP, LDH, ${\gamma}-GT$ and the formation of hepatic malondialdehyde induced by D-GalN. The alcholic extract of Bacopa monniera also significantly restored the decreased levels of glutathione and the decreased activities of glutathione peroxidase, glutathione reductase, superoxide dismutase, catalase and glucose-6-phosphatase. Therefore these results suggest that Bacopa monniera has hepatoprotective effect against D-GalN induced hepatotoxicity.

지유생간탕(地楡生肝湯)이 흰쥐의 간(肝)에 미치는 영향(影響) (Experimental Study on the protective effects of Jiyusaenggan-Tang on liver injury.)

  • In order to investigate the protective effects of Jiyusaenggan-Tang by F-I(Jiyusaenggan-Tang aspirated by distilled water), F-II(Non-polysaccharides in Jiyusaenggan-Tang) and F-III(Polysaccharides in Jiyusaenggan-Tang), experimental studies were performed in mice with liver injury induced by $CCI_4$, and d-galactosamine. The results were summarized as follows : 1. The increases of the serum GOT activities in mice induced by $CCI_4$ were inhibited significantly by the administration of highly concentrated FIII. The increases of the serum GPT, LDH, and ALP activities in mice induced by $CCI_4$ were inhibited significantly by the administration of highly concentrated FI and FIII. 2. The increases of the serum total-cholesterol and triglyceride activities in mice induced by $CCI_4$ were inhibited significantly by the administration of highly concentrated FI, FII and FIII. 3. On liver injury induced by the $CCI_4$, the activities of GOT, GPT, ALP, LDH were inhibited by highly concentrated FIII, F-I, F-II, and F-I, F-II, F-III on total cholesterol level and F-III, F-I, F-II on triglyceride level in order of their efficacies. And F-III is considered to be the most effective component. 4. The increases of the serum GOT, GPT activities in mice induced by d-galactosamine were inhibited significantly by the administration of FI and FIII. The increases of the serum LDH, ALP activities in mice induced by d-galactosamine were inhibited significantly by the administration of FI and FIII. 5. The increases of the serum Total-cholesterol, Triglyceride activities in mice induced by d-galactosamine were inhibited significantly by the administration of FI, FII and FIII. 6. The increases of the serum GOT, GPT activities in mice induced by d-galactosamine were inhibited significantly in F-III administrated group and then F-I in order of efficacies. FII does not showed significant effect, on inhibiting the activities of serum GOT and GPT levels. The activity of ALP was inhibited by F-III, F-I, F-II, LDH by F-III, F-I in order of efficacies, and F-II did not showed significant effect on LDH activity. The triglyceride level was decreased significantly by F-III, F-II, F-I and the total cholesterol level by F-II, F-I, F-III in order of their efficacies. Judging from the above results, it is considered that Jiyusaenggan-Tang has protective effect against liver injury, and that Polysaccharides in Jiyusaenggan-Tang is more effective.

인진호탕합시호소간탕가감방(茵陳蒿湯合柴胡疏肝湯加減方)이 간손상(肝損傷)에 미치는 영향(影響) (The effects of Injinhotang-sihosogantang-gagambang on liver injury)

  • In order to investigate the protective effect of Injinhotanghapsihosogantang-gagambang on the liver injury of rats induced by $CCl_4$ and d-galactosamine, the serum transaminase(GOT&GPT) alkaline phosphatase(ALP), lactic dehydrogenase(LDH) for enzyme activities, triglyceride for serum component, liver weight and glutathione S-transferase(GST), Superoxide dimutase(SOD) were measured. All animals were divided into 5 groups, those were normal group(untreated), control group(treated with vehicle 0.9% Saline solution), sample I group(10mg/kg administrated), sample II group(30mg/kg administrated), Silymarin 200 administrated group. The results were as follows: 1. The inhibitory effects of the serum GOT activities in rats induced by $CCl_4$ were noted in both sample I (p<0.001) and sample II group(p<0.001). In serum GPT activities, sample I (p<0.01) and sample II group(p<0.01). 2. The inhibitory effects of the serum LDH activities in rats induced by $CCl_4$ were noted in both sample I (p<0.001) and sample II group(p<0.001). 3. The increased effects of the serum ALP activities in rats induced by $CCl_4$ were not recognized. 4. The inhibitory effects of the serum triglyceride content level in rats induced by $CCl_4$ were inhibited in only sample II group(p<0.05). 5. The increased effects of the liver weight level in rats induced by $CCl_4$ were inhibited in both sample I (p<0.05) and sample II group(p<0.05). 6. The inhibitory effects of the GST activities in rats induced by $CCl_4$ were not recognized. In SOD activities, both sample I (p<0.05) and sample II group(p<0.001) showed the inthbitory effects. 7. The inhibitory effects of in the serum GOT, GPT activities in rats induced by d-galactosamine were not recognized. 8. The increases of the serum LDH level in rats induced by d-galactosamine were noted in both sample I (p<0.01) and sample II group(p<0.001). 9. The inhibitory of the serum triglyceride content level in rats induced d-galactosamine were noted in only sample II group(p<0.05). According to the above results, it is considered that Injinhotanghapsihosogantang-gagambang has protective effect against liver injury in rats induced by $CCl_4$ and d-galactosamine.

흰쥐에서 DWP-04가 D-galactosamine에 의해 유도된 간독성의 보호효과 (Protective Effect of DWP-04 Against Hepatotoxicity Induced by D-galactosamine)

  • 간기능 보호 작용이 있는 것으로 알려진 DDB, selenium과 glutathione의 혼합제제인 DWP-04의 간보호 작용을 검토할 목적으로 DWP-04를 실험동물에 경구로 투여하고서 D-galactosamine으로 간독성을 유발하여 혈액의 변동 및 간 조직에서의 활성산소 생성계 및 해독계의 활성에 미치는 영향을 관찰한 결과 GaIN의 단독투여는 대조군에 비하여 혈중 간 기능 지표효소 및 간 조직에서의 지질과산화의 함량이 현저히 증가하였으나, DWP-04의 전처리로 현저히 감소되었다. CaIN의 단독 투여로 활성산소의 생성계인 phase 1계의 효소가 현저히 증가하던 것이 DWP-04의 처리로 억제되었으며 해독계인 phase II계의 효소는 GaIN의 투여로 대조군에 비하여 억제되던 것이 DWP-04의 전처리로 정상군에는 미치지 않으나 유의성 있게 증가되었다. 간 조직중 glutathine의 함량은 CaIN의 투여로 현저히 억제되었으며 DWP-04의 투여로 증가하였는데 이러한 결과는 DWP-04의 투여로 glutathione peroxidase의 활성보다는 $\gamma-glutamylcysteine$ synthetase의 활성을 조절한 결과로 생각된다. 이상의 결과를 종합하여 볼 때 DWP-04의 투여는 활성산소의 생성 및 해독계를 조절하므로서 GaIN으로 인한 간손상을 보호하는 효과가 있는 것으로 사료된다.

다슬기 추출물이 D-galactosamine에 의해 손상된 간에 미치는 효과 (Effect of Semisulcospira libertina Extract on Hepatic Injury Induced by D-galactosamine)

  • 본 연구는 D-galactosamine에 의해 간 손상이 유도된 흰쥐에 다슬기(Semisulcospira libertina) 추출물이 손상된 간세포의 개선 및 회복에 미치는 효과에 관해 조사하였다. 다슬기 추출물은 간 손상에 따른 간 조직 내 국소적 지방 변성과 염증세포 침윤을 크게 감소시켜 대조군과 유사한 수준으로 회복시키는 경향을 나타내었다. 또한 다슬기 추출물은 간 손상 지표 효소인 AST와 ALT, LDH 및 ALP의 증가된 효소 활성을 대조군 수준으로 완화시키며, 간 조직 내 지질과 과산화지질의 함량을 감소시켜 간 손상으로 인한 혈중 효소 활성과 조직 내 지질 함량을 개선하는 것으로 조사되었다. 이와 더불어 다슬기 추출물은 염증반응을 촉진시켜 조직상해 및 괴사를 유도하는 $TNF-{\alpha}$의 발현을 억제시키며, 염증 시 세포를 보호하는 HO-1의 발현을 촉진시켜 염증 반응에서 손상된 세포를 대조군 상태의 세포로 회복시키는데 관여하는 것으로 조사되었다. 따라서 다슬기 추출물은 간조직의 괴사 및 섬유화를 회복시키고 혈액 내 효소의 활성과 조직 내 지질 함량을 개선할 뿐만 아니라 염증 반응 인자의 발현을 조절하고 있어 간 손상으로 인한 간세포의 개선 및 회복에 효과가 높은 기능성 소재로서의 가능성을 제시해 주고있다.

과체 성분의 간질환 모델에서의 효과 (Effects of the Constituents of Melonis Pedicellus in the Animal Models of Hepatic Diseases)

  • In order to investigate the hepatoprotective constituents, Melonis Pedicellus was systematically extracted and fractionated with n-hexane, ethyl acetate, butanol and water Treatment of rats with ethyl acetate fraction reduced hepatic injuries induced by $\alpha$-naphthylisothiocyanate or D-galactosamine, whereas the components in water fraction showed protective effect only against D-galactosamine-induced hepatitis in rats. Two cucurbitacins and three sterols were isolated from ethylacetate fraction and their chemical structures were identified as cucurbitacin B, isocucurbitacin B, $\alpha$-spinasterol, stigmast-7-en-3-ol and stigmast-7-en-3-ol-0-$\beta$-D-glucopyranoside. Cucurbitacin B at the dose of 1 mg/kg (p.o.) signifcantly increased in bile flow in rats with ANIT-induced cholestasis. Isocucurbitacin B at 5 mg/hg (p.o.) showed signilicant protective effects against ANIT-induced cholestasis. These results showed that cucurbitacin B and isocucurbitacin B from Melonis Pedicellus may have hepatoprotective effect in rats with experimental cholestasis.

  • Objectives : Propolis and Hovenia dulcis Thunb. has been used as treatment of diseases in the Korean medicine. In this study, we investigated that the hepatoprotective effects of Propolis oral administration according to mixture with Hovenia dulcis Thunb. on ${\gamma}-GTP$, GOT, GPT, Total bilirubin, LDH, ALP, Total cholesterol, Triglyceride, SOD, activity of catalase and Glutathione Peroxidase in galactosamine (GalN)-induced liver in rats. Methods : The animals were divided into 5 groups. Control, the liver injury-induced and not treated group. Pro1, liver injury and administrated propolis. Pro2, liver injury and administrated propolis capsule. Pro3, liver injury and administrated mixture of propolis capsule with Hovenia dulcis Thunb. Pro4, liver injury and administrated mixture of Propolis capsule with Hovenia dulcis Thunb. and Artemisia capillaris Thunb.. Animals were treated by Oral administration of Propolis, Hovenia dulcis Thunb., and Artemisia capillaris Thunb. mixture ltime 2 days for 14 days. Results : The Pro1 group was significantly increased on ${\gamma}-GTP$ and activity of Glutathione peroxidase but decreased on GOT in serum as compared with the control group. The Pro2 group was significantly increased on WBC, RBC, Hct, HGB in serum and activity of CuZnSOD as compared with the control group. The Pro 3 group was decreased on Total bilirubin, increased on LDH, WBC, RBC, Hct and HGB in serum as compared with the control group. The Pro 4 group was decreased on GOT in serum as compared with the control group. Conclusion : By evaluating the liver function and lipid metabolism, Pro3 had a hepatoprotective effect on the prevention of hepatotoxity.

가미청간탕의 간보호 및 면역조절효과 (A Study on the Immune Modulation and Hepatoprotection of Gamichunggan-tang (GCT))

  • Objectives : This study was to examine the efficacy of GCT on the hepatoprotective effect in the liver function and immune octivity. Methods : The experiment to investigate the hepatoprotective effect of GCT on the liver damage was conducted with D-galactosamine. The experiments to verify the effects of GCT on the immune activity were conducted by carbon clearance assay, plaque-forming cell SRBC assay of IgM, lymphoproliferation assay of T and B cells, and adherence and phagocytosis of mocrophages. Results: In the damage of liver induced by D-galactosamine, GCT carried hepatoprotective effect on AST. In carbon clearance assay GCT showed significant effect on phagocytosis of Kuffer cells. In the plaque-forming cell assay, GCT improved the formation of IgM. In the lymphoproliferation assay, GCT activated the formation of T and B lymphocytes. In macrophages, GCT activated adherence and phagocytosis. Conclusion : Though further study is needed, our findings suggest that GCT could be recommended as hepatoprotector and immune modulator for liver disease.

  • The roots and rhizome of Acanthopanax koreanum are used as folk medicine to ameliorate hepatitis in Korea. The ability of A. koreanum to protect mice from fulminant hepatitis induced in mice by D-galactosamine and lipopolysaccharide was evaluated. Preparations of A. koreanum used were an ethanol extract, and the ethanol-soluble and ethanol-insoluble components of the water extract. (omitted)

  • 기문혈(期門穴)에 대한(對) 오공자약침(蜈蚣雌藥鍼)이 D-Galactosamine에 의하여 유발된 간손상(肝損傷)에 미치는 영향을 실험적으로 규명하고자 흰쥐에게 약침을 처치한 후 혈청내의 G0T GPT, ${\gamma}$-GTP, Total bilirubin, LDH, ALP, Total cholesterol, Triglyceride 및 혈 액액내의 WBC, RBC, HGB, Hct 등을 관찰한 바 다음과 같은 결과를 얻었다. 1. GPT변화에서 대조군에 비하여 SHA-2군과 SHA-3군이 유의한 감소를 보였다. 2. Total bilirubin변화에서 대조군에 비하여 SHA-1군, SHA-2군과 SHA-3군이 유의한 감소를 보였다. 3. ${\gamma}$-GTP변화에서 대조군에 비하여 SHA-1군 과 SHA-2군이 유의한 감소를 보였다. 4. Total cholesterol변화에서 대조군에 비하여 SHA-2 군이 유의한 증가를 보였다. 5. Triglyceride변화에서 대조군에 비하여 SHA-1군, SHA-2군과 SHA-3군이 유의한 감소를 보였다. 6. HDL-cholesterol변화에서 대조군에 비하여 SHA-2군이 유의한 증가를 보였다. 7. 혈액내 변화에서 대조군에 비하여 WBC는 SHA-2군에서 유의한 감소를 보였으며, RBC는 SHA-2군과 SHA-3군에서 유의한 증가를 보였고, HGB는 SHA-2군과 SHA-3군에서 유의한 증가를 보였으며, Hct는 SHA-3군에서 유의한 증가를 보였다. 이상의 실험결과를 종합해보면 오공약침이 D-galactosamine으로 유발된 백서의 간손상에 효과적이라고 생각되며 간손상에 대한 예방, 치료적인 부분에서 향후 지속적인 연구가 필요하리라고 사료된다.
