Protective Effect of Korean Medicinal Plants on Ethanol-Induced Cytotoxicity in HepG2 Cells

  • The purpose of this study is to evaluate cytoprotective effect of Korean medicinal plants on alcohol-induced cytotoxicity in liver cells. Out of the 120 plant extracts tested in this study, 53 plant extracts enhanced alcohol-induced cytotoxicity in liver cells by 50~80%, while other 11 plant extracts including Crataegus pinnatifida reduced cytotoxicity by 1~68%. The results of DPPH free radical test and LDL lipid peroxidation test on the plant extracts that sharply reduced cytotoxicity in liver cells shows that Crataegus pinnatifida and Cinnamomum cassia had antioxidative effect. This study reports that the plant extracts that enhance or reduce ethanol-induced cytotoxicity in liver cells can be research objects as cytotoxic plants or cytotoxicity-protective plants.

Protective Effect of Kaempferol on Cultured Neuroglial Cells Damaged by Induction of Ischemia-like Condition

  • This study was performed to evaluate the cytotoxicity induced by ischemia-like condition (ILC) in cultured neuroglial cells (C6 glioma cells). The protective effect of kaempferol (KAE), flavonoid against the cytotoxicity induced by ILC induction was assessed. In addition, antioxidative effects of KAE were done by colorimetric assays. Cell viability and the antioxidative effects such as DPPH-radical scavenging activity, superoxide dismutase (SOD)-like activity and inhibitory activity of lipid peroxidation (LP) were analyzed. ILC induction decreased cell viability in a dose-dependent manner, and the $XTT_{90}$ value (low cytotoxicity value) and $XTT_{50}$ value (high cytotoxicity value) were determined during ILC induction for 15 and 40 minutes, respectively. The butylated hydroxytoluene (BHT) antioxidant significantly increased cell viability damaged by the ILC-induced cytotoxicity. In the protective effect of KAE on ILC-induced cytotoxicity, KAE protected the ILC-induced cytotoxicity by the significant increase of cell viability, and also it showed DPPH-radical scavenging ability, SOD-like ability and inhibitory ability of LP. From these results, it is suggested that ILC induction showed cytotoxicity in these cultures and the oxidative stress is involved in the ILC-induced cytotoxicity. While, KAE prevented ILC-induced cytotoxicity by antioxidative effects. In conclusion, natural products like KAE may be a putative therapeutic agent for the treatment of disease associated with oxidative stress such as ischemia.

Antioxidant Effect of Poncirin and Cytotoxicity on Cultured Human Skin Fibroblast Damaged by Methyl Mercury

  • In order to evaluate on the cytotoxicity of methyl mercury (MM) and antioxidant effect of phenolic compound, poncirin against MM-induced cytotoxicity, XTT assay was performed to determine the cell viability after human skin fibroblasts (Detroit 51) were grown in the media containing various concentrations of methylmercuric chloride (MMC). And also, the antioxidant effect of poncirin on the cytotoxicity induced by MMC was examined by cell viability and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity in these cultures. MMC decreased cell viability in dose-dependent manner in these cultures and the midcytotoxicity value was determined at concentration of 30 ${\mu}M$ MMC after human skin fibroblasts were treated with $10\sim50{\mu}M$ MMC for 72 hours, respectively. MMC was highly toxic on cultured human skin fibroblasts by toxic criteria. MMC-mediated cytotoxicity was related with oxidative stress by the diminution of toxic effect according to the treatment of vitamin E. In the antioxidant effect of poncirin, it showed vitamin E-like DPPH radical scavenging activity at 90 ${\mu}g/ml$ poncirin and also, remarkably increased cell viability compared with MMC-treated group. From these results, it is suggested that MMC-mediated cytoxicity was highly toxic and was related with oxidative stress in cultured human skin fibroblasts, and also phenolic compound such as poncirin showed the protection on MMC-induced cytotoxicity by antioxidant effect in these cultures.

  • This study was to evaluate the cytotoxic effect of three root canal disinfectants (formocresol, camphorated phenol and eugenol) and ten root canal sealers(Cavitec, Hypo-cal, Vitapex, AH26, Canals, Mynol, $N_2$, $N_2$-Medical, Z. O. E. and Calvital) in vitro. The experiments were performed in four differrent modes. In the first and second experiment, the "long-distance" cytotoxicity of three root canal disinfectants were tested on L cells. In the third exeriment, ten root canal sealers were tested for cytotoxicity by means of the tissue culture-agar overlay method immediately, 4 and 24 hours after the experiment. In the fourth experiment, the study with radioactively labeled L cells were employed to determine the relative cytotoxicity of ten root canal sealers. The results were as follows; 1. Every vapors from disinfectants showed more or less cytotoxicity. Of the three disinfectats, formocresol appeared to be the highest cytotoxic effect and camphorated phenol was the lowest. 2. Root canal sealers tested in tbis study showed cytotoxicity at every stage of time intervals. 3. The highest cytotoxic effect was freshly mixed $N_2$ meaical and $N_2$ also revealed the highest cytotoxic effect after 4 or 24 hours among these materials. Vitapex was found the lowest cytotoxic effect at all experimental stage. 4. Root canal sealers except N2 and Mynol showed cytotoxic effect were decreased cytotoxicity according to the time elapsed.

Effect of Water Extract from Saururus Chinensis (Lour.) Bail Water Extracts on the Cancer Cells and Antioxidative Activity in Cytotoxicity (삼백초(Saururus Chinensis (Lour.) Bail) 열추출물의 항암 및 세포독성 저해 효과)

  • Chemoprevetive effect of Saururus Chinensis (Lour.) Bail water extract on several tumor cells and Chinese hamster V79 cells were investigated. The water extracts of Saururus Chinensis (Lour.) Bail showed a higher cytotoxicity effect on the human histiocytic leukemia cells(U937) and protective effects against the cytotoxicity of H$_2$O$_2$. These results suggest that Saururus Chinensis (Lour.) Bail may useful as potential soures of chemopreventive and antioxidative agents.

Effect of Allopurinol on Methylmercuric Chloride-Induced Cytotoxicity in $C_6$ Cultured Glioma Cells

  • It is demonstrated that inorganic mercury has cytotoxic effect on glial cells. Recently, oxygen radicals is involved in methylmercuric chloride (MMC)-induced cytotoxicity. But, the toxic mechanism of MMC is left unknown. The purpose of this study was to examine the cytotoxicity of MMC on $C_6$ glioma cells. The cytotoxicy was measured by cell viability using XTT assay in $C_6$ glioma cells. Colorimetric assay is regarded as a very sensitive screening method for the determination of the cell viability on various agents. In this study, MMC decreased cell viability according to the dose- and time dependent manners after $C_6$ glioma cells were grown with various concentrations of MMC for 48 hours. In the protective effect of allopurinol on MMC-induced cytotoxicity, allopurinol was effective in the prevention of MMC-induced cytotoxicity in these cultures. These results suggest that MMC has highly cytotoxic effect on $C_6$ glioma cells by the decrease of cell viavility, and free radical scavenger such as allopurinol was effective on organic mercury-induced cytotoxicity in these cultures.

Cytotoxicity of Ligularia fischeri Extracts (곰취 추출물의 세포독성 효과)

  • This study was investigated to observe the cytotoxicity effect of Ligularia fischeri extracts against cancer cell lines including human lung carcinoma(A549), human cervix epitheloid carcinoma(HeLa) and human hepatocellular carcinoma(HepG2) using SRB(sulforhodamine B) method. The ethanol and methanol extracts of 1$\mu\textrm{g}$/${mu}ell$ showed approximately 79.2% and 86.4% cytotoxicity effects on HepG2 cell line and the ethyl acetate fracton fractionated from ethanol extracts showed the strongest cytotoxicity effect with 94% inhibition. The inhibitory effect of ethanol extract on HeLa cell line was somewhat low with 50~56% inhibition, but ethyl acetate fraction showed higher cytotoxicity effect with 91% and 91.9% inhibition on the HeLa and A549 cell line. On the contrary, the ethanol and methanol extracts showed the lower inhibition effects on the normal liver cell, WRL68, compared to human cancer cell lines.

The Antioxidative Effect of Eclipta prostrata L. Extract on Cultured NIH3T3 Fibroblasts Injured by Manganese-Induced Cytotoxicity

  • Manganese (Mn) is used as main materials in various chemical processes of industry, but it suggested that Mn brings about its toxicant by fume or dust through respiratory system and skin barrier. Mn toxicant induces the loss of mental health and life quality by cerebrovascular and skin diseases. Nevertheless, it lefts much unknown on the mechanism and the effectively therapeutic methods about Mn toxicant. Therefore, this study was evaluated the cytotoxicity induced by manganese dioxide ($MnO_2$) in cultured NIH3T3 fibroblasts, and also, the correlation between $MnO_2$-induced cytotoxicity and oxidative stress was examined. While, the effect of Eclipta prostrata L. (EP) extract belong to Compositae was assessed against $MnO_2$-induced cytotoxicity in the view of antioxidative effect for searching the natural resources mitigating or preventing the $MnO_2$-induced cytotoxicity. In this study, $MnO_2$-induced cytotoxicity was revealed as mid-toxic by Borenfreud and Puerner's toxic criteria, and catalase (CAT), an antioxidant prevented $MnO_2$-induced cytotoxicity by the remarkable increase of cell viability in these cultures. While, in the protective effect of EP extract on $MnO_2$-induced cytotoxicity, EP extract effectively prevented the cytotoxicity induced by $MnO_2$ via antioxidative effects such as xanthine oxidase (XO) inhibitory ability and DPPH-radical scavenging ability. From the above results, EP extract showed the effective prevention against $MnO_2$-induced cytotoxicity correlated with oxidative stress by antioxidative effects. Conclusively, this study may be useful to research or development the alternatively therapeutic agent from natural resources like EP extract for the treatment of diseases resulted in oxidative stress.

Antimicrobial Effect and Cytotoxicity of Chinensis Galla (오배자의 항균활성 및 세포독성에 미치는 영향)

  • Chinensis galla has been used as an multi-functional herb, such as anti-inflammatory, anti-virus, and antitumor agent. This study was performed to antimicrobial and cytotoxicity effect in vitro. The results were summarized as follows : Chinensis galla was antimicrobial effect on Staphylococcus aureus subsp. aureus, Bacillus cereus, Bacillus subtilis, Staphylococcus epidermidis. Chinensis galla was antimicrobial effect on Kocuria rhizophila, Corynebacterium ammoniagenes. The extracts of Chinensis galla exhibited cytotoxicity on human dermal fibroblast at $10\;{\mu}{\ell}$ but not at $5\;{\mu}{\ell}$, and the same results was known under a microscope. Accordingly the results show Chinensis galla could antimicrobial effect but exhibited cytotoxicity on human dermal fibroblast at high concentration and it needs more research.

Cytoprotective Effect of Phenolic Compounds Against Hexavalent Chromium-Induced Cytotoxicity (페놀 화합물의 Hexavalent Chromium독성에 대한 세포 보호효과)

  • Objectives : In order to evaluate the cytotoxicity of hexavalent chromium, the cytoprotective effect of phenolic compounds against hexavalent chromium-induced cytotoxicity, cell viability, cell adhesion ability, lactate dehydrogenase(LDH) activity, and morphological changes of cells were examined. Methods : We measured the cytotoxicity of hexavalent chromium with 3-[4,5-dimethyl-thiazol-2-yl]-2,5-diphenyltetrazolium bromide(MTT), 2,3-bis-[2-methoxy-4-nitro-5-sulfophenyl]-2H-tetrazolium-5-caboxanilide (XTT), LDH and DPPH methods. Results : The cytotoxicity of hexavalent chromium($IC_{50}$, $44.0-51.0{\mu}M$) was high according to the toxic criteria. Cytoprotective effect of phenolic compounds against $IC_{50}$ value of hexavalent chromium in cell morphology increased in a concentration-dependent manner. Conclusions : These results suggest that 3,4,5-trihydroxybenzoic acid may be used as a cytoprotective agent against chromium(IV)-mediated cytotoxicity.
