Jab1 as a Mediator of Nuclear Export and Cytoplasmic Degradation of p53

  • Jun activation domain-binding protein 1 (Jab1) is involved in various cellular mechanisms including development in Drosophila and mouse, cell cycle control and signal transduction pathways. Recent studies also determined that Jab1 functions as a nuclear exporter and inducer of cytoplasmic degradation for several proteins including p53, p27, capsid of West Nile virus, and Smad4/7 proteins. In particular, p53 is shown to bind to and to be exported into the cytoplasm by Jab1, which helps to maintain low levels of p53 under normal conditions. This review was undertaken in an effort to understand the biological significance of the homeostasis of p53 as maintained in the presence of Jab1. Based on our observations, we have provided potential mechanistic hypotheses for the nuclear export of p53 in coordination with Jab1 and the role of other factors in these processes.

Origin of Somatic Embryo Induced from Cotyledons of Zygotic Embryos at Various Developmental Stages of Ginseng

  • Excised cotyledon segments of ginseng zygotic embryos at various developmental stages were cultured on MS basal medium from which somatic embryos were directly induced. The frequency of somatic embryo formation on the segments declined with the advancing zygotic embryo maturity. All of the cells in the cotyledons of immature zygotic embryos were smaller and more densely cytoplasmic than those in mature embryos. Histological examinations revealed that the poly-somatic embryos formed on immature embryos were of multi-cell originand derived from the epidermal and subepidermal cell layers. However, in the cotyledon of germinating zygotic embryos, only theepidermal cells were densely cytoplasmic and singularly competent to develop into somatic embryos resulting into single embryos at a frequency of 100%.

  • Fine structural changes of the germ cells during spermatogenesis in the goldeye rockfish, Sebastes thompson; were examined by means of the transmission and scanning electron microscopy. A spermatogonium has a large nucleus with a single nucleolus in the interphase. Primary spermatocytes are characterized by the formation of chromatin clumps and presence of the synaptonemal complex in the nucleus. The nucleoplasm of secondary spermatocytes is more condensed than that of primary spermatocytes, and the cytoplasm contains numerous mitochondria, endoplasmic reticulum and Golgi complex. The nuclei of spermatids in metamorphosis show sickle-like shape as the nucleoplasm becomes more condensed. In the cytoplasm of spermatids, the proacrosomal granules are not found at all. A spermatozoon consists of head, neck and tail. The acrosome is absent in the head. Four to five cytoplasmic collars are observed in the posterior portion of the head of spermatozoon. The well developed axonemal lateral fins are observed in the flagellum of spermatozoon.

Glucose Transporters and Insulin Action : Some Insights into Diabetes Management

  • Insulin stimulates glucose uptake in muscle and adipose cells primarily by recruiting GLUT4 from an intracellular storage pool to the plasma membrane. Dysfunction of this process known as insulin resistance causes hyperglycemia, a hallmark of diabetes and obesity. Thus the understanding of the mechanisms underlying this process at the molecular level may give an insight into the prevention and treatment of these health problems. GLUT4 in rat adipocytes, for example, constantly recycles between the cells surface and an intracellular pool by endocytosis and exocytosis, each of which is regulated by an insulin-sensitive and GLUT4-selective sorting mechanism. Our working hypothesis has been that this sorting mechanism includes a specific interaction of a cytosolic protein with the GLUT4 cytoplasmic domain. Indeed, a synthetic peptide of the C-terminal cytoplasmic domain of GLUT4 induces an insulin-like GLUT4 recruitment when introduced in rat adipocytes. Relevance of these observations to a novel euglycemic drug design is discussed.

화상분석기를 이용한 정도별 이형성증에 대한 연구 (The Study of Dysplasic Grades to Digital Image Analyzer)

  • The purpose of this study was to develop discriminant analysis models for predicting cervical normal/dysplasia case diagnoses using cytometric features derived from the digital image analysis of cell monolayers. The database consisted of 19 cases diagnosed either as normal (n=5), moderate dysplasia (n=7), severe dysplasia (n=7) on monolayer preparations. We studied the nuclear and cytoplasmic characteristics of cells in the normal, moderate dysplasia and severe dysplasia on cervical samples. The morphometric parameters selected for the analysis were nuclear/cytoplasmic ratio and the nuclear variations measured by image analysis on normal and precancerous lesions of cervical smears; several shape factors; area; perimeter; maximal, minimal and equivalent circle diameters. The results showed that the dysplasia samples exhibited changes in both cellular and nuclear form and size but lacked substantial differences in the tumor grades. The coefficient of nuclear variation is as follows to normal cell $21.8{\pm}3.2%$, moderate dysplasia $33.5{\pm}6.1%$, severe dysplasia $27.7{\pm}5.8$ of cervical smears.

  • Assisted reproductive technologies (ART) merely depend on improving the oocyte maturation and their developmental competence to produce good quality embryos. Oocyte maturation passes through long and complex molecular steps starts from the early embryonic life and ends with sperm fertilization. Oocyte developmental competence can be attained by improving the nuclear and cytoplasmic mechanisms together with some epigenetic maturation. In this review, we highlight the cornerstones of oocyte maturation on both nuclear and cytoplasmic levels. Interfering or supporting these molecular mechanisms would help in the development of novel regulating agents for reproductive performance of humans and livestock species.

1H, 15N, and 13C backbone assignments and secondary structure of the cytoplasmic domain A of mannitol trasporter IIMannitol from Thermoanaerobacter Tencongensis phosphotransferase system

  • The mannitol transporter Enzyme $II^{Mtl}$ of the bacterial phosphotransferase system has two cytoplasmic phosphoryl transfer domains $IIA^{Mtl}$ and $IIB^{Mtl}$. The two domains are linked by a flexible peptide linker in mesophilic bacterial strains, whereas they are expressed as separated domains in thermophilic strains. Here, we carried out backbone assignment of $IIA^{Mtl}$ from thermophilic Thermoanaerobacter Tencongensis using a suite of heteronuclear triple resonance NMR spectroscopy. We have completed 94% of the backbone assignment, and obtained secondary structural information based on torsion angles derived from the chemical shifts. $IIA^{Mtl}$ of Thermoanaerobacter Tencongensis is predicted to have six ${\beta}$ strands and six ${\alpha}$ helices, which is analogous to $IIA^{Mtl}$ of Escherichia coli.

Methimazole-Induced Anti-Neutrophil Cytoplasmic Antibody-Associated Vasculitis in the Peripheral Nerves

  • Anti-neutrophil cytoplasmic antibody (ANCA)-associated vasculitis has been reported in Graves' disease patients treated with antithyroid drugs (ATDs), especially propylthiouracil. ATD-induced ANCA-associated vasculitis usually involved the kidneys followed by the respiratory organs and skin. The treatment of ANCA-associated vasculitis induced by ATDs is to stop ATD therapy immediately, which often leads to an overall good prognosis. We report a case of ANCA-associated vasculitis in the peripheral nerves of the lower extremities in a 66-year-old woman who was treated with methimazole (MMI) for Graves' disease. To our knowledge, this is the third case of peripheral nervous system (PNS) involvement of ATD-induced vasculitis and the first case of PNS vasculitis associated with MMI.

Oxidative stress causes Alu RNA accumulation via PIWIL4 sequestration into stress granules

  • The Alu element, the most abundant transposable element, is transcribed to Alu RNA. We hypothesized that the PIWI protein regulates the expression of Alu RNA in retinal pigment epithelial (RPE) cells, where accumulated Alu RNA leads to macular degeneration. Alu transcription was induced in RPE cells treated with $H_2O_2$. At an early stage of oxidative stress, PIWIL4 was translocated into the nucleus; however, subsequently it was sequestered into cytoplasmic stress granules, resulting in the accumulation of Alu RNA. An elevated amount of Alu RNA was positively correlated with the disruption of the epithelial features of RPE via induction of mesenchymal transition. Therefore, we suggest that oxidative stress causes Alu RNA accumulation via PIWIL4 sequestration into the cytoplasmic stress granules.

Direct and functional interaction between dopamine D2 receptor and ALY

  • The signaling pathway of dopamine D$_2$ receptor was studied using yeast two-hybrid system. The 3rd cytoplasmic loop of rat D$_2$ receptor was fond to interact with ALY. The interaction in the yeast was observed only with the 3rd cytoplasmic loop of D$_2$ receptor but not with that of D$_3$ or D$_4$ dopamine receptor. The interaction between two proteins was also confirmed by GST pull-down assay. Co-expression of D$_2$ receptor and ALY enhanced the expression of Lef-1 promoter in C6 cells and the promoter of D$_2$ dopamine receptor itself.
